Ocular surface disease in patients with ocular hypertension and glaucoma.

PURPOSE: To review the prevalence, diagnosis, causes, and treatment of ocular surface disease (OSD) in patients with ocular hypertension or primary open-angle glaucoma. METHODS: A review of the literature pertaining to OSD and glaucoma. RESULTS: Recent studies indicate that OSD demonstrates an overall prevalence in glaucoma of 42% (range 20-59%) and is severe in 36% (range 14-66%). Further, the prevalence appears to increase with the greater the number of glaucoma drugs prescribed. Symptoms and signs are non-specific to the anterior surface of the eye and are thought to result from allergic,

Non-preserved 1% lidocaine solution has less antibacterial properties than currently available anaesthetic eye-drops.

PURPOSE: Some anaesthetics inhibit bacterial growth, and thus may lead to low rates of positive cultures from bacterial keratitis. Antibacterial properties of lidocaine were compared with those of oxybuprocaine or tetracaine, either in current commercial eye lotions or in extemporaneous solutions. METHODS: Forty-eight bacterial strains were used to determine the minimum inhibitory and bactericidal concentrations of four commercial eye lotions containing oxybuprocaine or tetracaine, of a non-ophthalmic 1% lidocaine commercial solution and of extemporaneously prepared solutions of oxybuprocaine

Epidermal damage induced by irritants in man: a light and electron microscopic study.

Irritant contact dermatitis may be induced by many chemicals and has a far greater incidence than allergic contact dermatitis. Despite this, it receives relatively little attention and its pathogenesis remains poorly understood. To gain a greater understanding of the interaction of irritants with the skin, we investigated the histopathological changes resulting from the topical application of a series of structurally unrelated irritants. Human volunteers were patch-tested with appropriate concentrations of nonanoic acid, sodium lauryl sulphate, dithranol, benzalkonium chloride, croton oil, and

Ultrastructural changes to corneal stromal cells due to ophthalmic preservatives.

Normal and keratectomised rabbit corneas received drops containing preservatives over a two day period. All preservatives investigated caused some change in the morphology of the stromal keratocytes, particularly in the absence of the anterior epithelial barrier. Thimerosal (0.004%) or disodium ethylenediaminetetraacetate (0.1% Na2EDTA) produced only minimum change. Chlorbutanol (0.5%) applied to intact corneas resulted in swelling of the cytoplasm and occasionally breaks in the external cell membrane, whilst in the keratectomised corneas some mitochondria showed swelling and distortion of cri

An animal model for altering the irritability threshold of normal skin.

Theory behind conditioned hyperirritability (autoeczematization) predicts the lowering of the irritation threshold in the presence of a pre-existing dermatitis. We have attempted to develop an animal model that parallels the syndrome seen in man. Groups of 10 guinea pigs were shaved and depilated; irritation thresholds to benzalkonium chloride and trichloroacetic acid were determined using 1 cm diameter open patches. Reactions were scored 24 h later on the basis of erythema and induration. Animals having as little as 1.56 cm2 of skin acutely inflamed with a known irritant had lowered irritatio

Conjunctivitis due to soft lens solutions.

In cases of conjunctivitis associated with the use of soft lenses, delayed hypersensitivity to disinfecting components of the used soft lens liquids has to be considered. In 7/15 patients, positive patch test reactions to merthiolate, chlorhexidine or the soaking solutions were observed. The significance of these findings is discussed.

Multilocus sequence typing and antimicrobial resistance in Enterococcus faecium isolates from fresh produce.

The purpose of the present study was to determine the relatedness of Enterococcus faecium isolates from fresh produce to E. faecium strains from other sources by using multi-locus sequence typing (MLST) and to determine the antimicrobial resistance of the isolates. MLST analysis of 22 E. faecium isolates from fresh produce revealed 7 different sequence types (ST 22, ST 26, ST 43, ST 46, ST 55, ST 94 and ST 296). Most isolates belonged to ST 296 (40.9 %), followed by ST 94 (27.3 %). All isolates were sensitive to vancomycin and to imipenem, and only one was resistant to ampicillin (MIC 32 mg/l)

Oxygen-reducing microbial cathodes monitoring toxic shocks in tap water.

Electroactive biofilms (EABs) have recently attracted considerable research interest for their possible use as amperometric biosensors in environmental or bioprocess monitoring, for example for in situ detection of toxic compounds. Almost exclusively, corresponding research has focused on heterotrophic, anodic EABs. These biofilms require sufficiently high organic loads and anoxic conditions to deliver a stable baseline current. Conversely, electroautotrophic O2-reducing EABs have recently been proposed to monitor toxic shocks in oxic solutions that are poor or devoid of organic substrate. Thi

The Listeria monocytogenes transposon Tn6188 provides increased tolerance to various quaternary ammonium compounds and ethidium bromide.

Tolerance of the foodborne pathogen Listeria monocytogenes to sublethal concentrations of disinfectants has been frequently reported. Particularly, quaternary ammonium compounds (QACs) such as benzalkonium chloride (BC) are often used in disinfectants and also as antiseptics in food industry and hospitals. Recently, we described Tn6188, a novel transposon in L. monocytogenes harbouring the transporter QacH, a molecular mechanism leading to increased tolerance to BC. In this study, we investigated the presence of Tn6188 within the genus Listeria spp. Our screening indicates that the distributio

Methemoglobinemia induced by a topical anesthetic spray (cetacaine).

In two seriously ill patients, cyanosis developed shortly after a topical anesthetic spray (Cetacaine) was used. In both cases the presence of methemoglobinemia was suggested by a discrepancy between the arterial oxygen tension and the oxygen saturation of hemoglobin, as measured spectrophotometrically. The characteristic responses of the spectrophotometric oximeter to blood containing different concentrations of methemoglobin are described. Physicians administering this topical anesthetic spray (Cetacaine) should be aware of the possible development of methemoglobinemia.

Benzalkonium Chloride Tolerance Among Listeria innocua from Food and Food Processing Environments in Poland.

Benzalkonium chloride (BC) is widely used as a disinfectant in the food industry; however, increasing reports of Listeria innocua isolates exhibiting tolerance to this compound highlight the need to better understand their adaptive mechanisms. This study aimed to evaluate BC tolerance in 51 L. innocua isolates originating from raw and processed meat products (n = 32) and meat-processing environments in Poland (n = 19). Phenotypic tolerance was assessed using the agar diffusion method on two media: Brain Heart Infusion (BHI) agar and Mueller-Hinton (M-H) agar supplemented with 1.2% sheep blood,

Reference genes validation in tear fluid for RNA analysis in ocular surface disease.

Proper normalization of gene expression data is essential for detecting early molecular alterations in eye disorders. In this study, we assessed seven potential reference genes (18S, RER1, ACTB, GAPDH, PGK1, UBC, and AP3D1) in tear fluid collected from individuals with dry eye disease and glaucoma patients using benzalkonium-preserved topical medications, as well as from healthy controls. Utilizing various stability analysis methods (geNorm, NormFinder, comparative ∆CT method, BestKeeper, and RefFinder), we determined that 18S, RER1, and ACTB were the most stable reference genes, while UBC and
