Developmental changes in the liver weight- and body weight-normalized clearance of theophylline, phenytoin and cyclosporine in children.

AIMS: Body weight- (BW) normalized pediatric dosages of metabolically eliminated drugs often exceed the corresponding adult values. We aimed to clarify whether such findings would be attributable either to an augmented hepatic drug-metabolizing activity or to a systematic bias introduced by adopting BW as a size standard of clearance. MATERIALS AND METHODS: We chose 3 model drugs that are metabolized by distinct cytochrome P450 (CYP) isoforms (theophylline, phenytoin and cyclosporine for CYPIA2, CYP2C9/2C19 and CYP3A4, respectively). The MEDLINE database covering 1966 to May 2001, was searche

Organ weights in human fetuses after formalin fixation: standards by gestational age and body weight.

This study provides new standards of fetal organ weights (brain, heart, liver, pancreas, spleen, lungs, kidneys, adrenals, thymus, and thyroid) and body dimensions (crown-heel and crown-rump lengths, head circumference, and foot length). Subjects came from a large dataset including more than 4,000 fetuses autopsied in fetopathology units of pediatric hospitals in Paris between 1986 and 2001. From this dataset, 673 subjects were carefully selected by exclusion of multiple pregnancies, macerated and malformed fetuses, subjects with abnormal karyotypes, and those with severe infections. Fetal age

Segmental Variation in Lumbar Nerve Root Weight: A Donor-Body Study.

Introduction The cauda equina consists of lumbosacral nerve roots extending from the conus medullaris within the lumbar cistern. Although radiographic imaging is essential for evaluating lumbar spinal pathology, normal anatomical variation in the cauda equina may affect its appearance. This donor-body study quantified normalized segmental linear mass per root across the L1-L2 through L4-L5 vertebral intervals and evaluated level-specific and demographic patterns. These measurements provide exploratory anatomical data for future imaging and clinical correlation studies. Methods Thirty-two donor

Body-Weight Changes and Physical Activity After Anterior Cruciate Ligament Reconstruction.

CONTEXT: Recovery after anterior cruciate ligament reconstruction (ACLR) can lead to weight gain and reduced physical activity (PA), potentially increasing long-term health risks. Understanding the relationship between PA patterns and weight changes after ACLR is essential for optimizing rehabilitation strategies. OBJECTIVE: To examine the relationship between PA patterns and weight changes in the first 6 months after ACLR. We hypothesized that lower PA levels and longer sedentary time would predict greater weight gain. DESIGN: Case series. SETTING: Academic hospital and private sports medi

Comparison of Ventilatory Ratios Calculated Using Predicted Versus Actual Body Weight.

BACKGROUND: Ventilatory ratio (VR) is a surrogate marker of dead-space fraction that is increasingly used to assess ventilation impairment and predict clinical outcomes. However, the current VR equation uses on predicted body weight (PBW) to estimate reference minute ventilation, which may lead to an overestimation of dead-space fraction in patients with extreme body weights. We hypothesized that VR calculated using PBW and actual body weight (ABW) might differ in their ability to predict outcomes, particularly in obese patients. METHODS: This retrospective study compared the discrimination c

Astrocyte Loss Augments Body Weight Through Reduction in Adipose Sympathetic Outflows.

Emerging evidence indicates that astrocytes modulate energy metabolism and homeostasis. However, one important but poorly understood element is the necessity of astrocytes in the control of body weight. Here, we apply viral vector-assisted brain-region selective loss of astrocytes to define physiological roles played by astrocytes in the arcuate nucleus of the hypothalamus (ARH) and to elucidate the involved mechanism. We find that astrocyte loss potently augments body weight in adult mice fed chow or high-fat diet. Mechanistically, we find that the loss of astrocytes reduces adipose tissue no

Impact of body weight on IVF: pathophysiology, outcomes, and clinical considerations.

PURPOSE OF REVIEW: This study aimed to examine if excess body weight impacts the outcomes of IVF treatment cycles. RECENT FINDINGS: While data on the effect and/or association of excess weight on IVF outcomes continue to produce mixed results, recent studies stratifying analyses by age are yielding clarity on a detrimental potential of obesity on IVF outcomes. Specifically, IVF outcomes in the third decade of life are susceptible to adverse implications of excess weight, as reflected in lower clinical pregnancy and live birth rates. Plausible pathogenic mechanisms underlying the detrimental i

Anti-Factor Xa Activity of Apixaban in Extremely Low Body Weight.

Background: Direct oral anticoagulants (DOACs) are generally preferred over warfarin for preventing arterial and venous thromboembolism. However, the efficacy and safety of DOACs in patients with extremely low body weight (BW) are uncertain. This study investigates anti-factor Xa (anti-FXa) activity of apixaban and compares it between patients with normal BW (>50 kg) and underweight (≤50 kg). Methods: We enrolled 150 patients on branded generic apixaban (ApixanTM) for atrial fibrillation (AF), venous thromboembolism, and intracardiac thrombus. Anti-FXa activity of apixaban was measured at peak

Refeeding Edema in Restrictive Eating Disorders: Beyond Acute Body Weight Gain.

BACKGROUND: One study recently suggested that phosphate supplementation might contribute to the occurrence of refeeding edema in patients with restrictive eating disorders (EDs) with severe malnutrition complications. OBJECTIVE: This commentary aims to provide insight into the study while suggesting a more detailed approach to defining malnutrition, refeeding syndrome, and edema. MAIN DISCUSSION: There are several diagnostic criteria for diagnosing malnutrition, some of which might overlap with the criteria of refeeding syndrome risks. A precise nutritional and hydration status assessment is

Effects of Body Weight Support in Running on Achilles Tendon Loading.

Achilles tendon (AT) tendinopathy is common in runners. Repetitive AT loading may play a role in etiology. Interventions such as body weight support (BWS) may reduce loading on the AT in running. Examine how ground reaction force, AT loading, foot strike, and cadence variables change in running with BWS. Twenty-four healthy female runners free from injury were examined. Participants ran on an instrumented treadmill with and without BWS using a harness-based system at a standardized speed. The system has 4 elastic cords affixed to a harness that is attached to a frame-like structure. Kinematic

Human muscle and spinal activation in response to body weight loading.

Human standing is the anatomical and functional framework for independent movement. The study of weight bearing during standing and movement is crucial to support the development of technology aimed at restoring independent gait, where unsupported weight bearing is still elusive. This study aims to determine muscle and spinal activation at different gravitational loads in young, healthy individuals to provide potential patterns of spinal stimulation for standing. Muscle activity was recorded with surface electromyography (EMG) from 18 healthy participants at different body angles while on a mo

Height, weight, and body mass index in patients with familial dysautonomia.

BACKGROUND: Children with familial dysautonomia (FD) are smaller and grow more slowly than the general population. It is unknown whether this abnormal growth is due to comorbidities that patients with FD live with, or if it is a direct effect of the disease-causing homozygous ELP-1 mutations. Here, we created growth curves for weight, height, and body mass index (BMI) in male and female children with FD to monitor the nutritional status of patients with FD. METHODS: We used the New York University (NYU) FD Registry which includes data from 680 individuals with FD who were followed longitudina

Downregulation of Bdnf Expression in Adult Mice Causes Body Weight Gain.

Gain or loss of appetite and resulting body weight changes are commonly observed in major depressive disorders (MDDs). Brain-derived neurotrophic factor (BDNF) is broadly expressed in the brain and is thought to play a role in the pathophysiology of MDDs and obesity. Congenital loss of function of BDNF causes weight gain in both humans and rodents; however, it is not clear whether acquired loss of function of BDNF also affects body weight. Thus, we exploited mutant mice in which the Bdnf expression level is regulated by the tetracycline-dependent transcriptional silencer (tTS)-tetracycline ope

The Potentially Modifiable Risk Factor in Idiopathic Intracranial Hypertension: Body Weight.

PURPOSE OF REVIEW: Idiopathic intracranial hypertension (IIH) prevalence increased in conjunction with rising obesity rates. Here, we highlight the importance of weight management in IIH and introduce glucagon-like peptide 1 (GLP-1) receptor agonists (RAs) as potential treatment strategy for IIH. RECENT FINDINGS: Weight gain is a risk factor for IIH, and weight loss (via any treatment strategy) plays a key role in IIH management. GLP-1 is an incretin secreted by the distal small intestine in response to a meal. GLP-1 RAs have been shown to improve glycaemic control (no hypoglycaemia) and lowe

MTHFR Polymorphisms in Girls with Anorexia Nervosa: Implications on Body Weight.

The development of atypical vs typical anorexia nervosa (AN) might be explained by the genetic background. We assessed the link between the subtypes of AN and the genetic polymorphisms of the thrombotic panel and the methyltetrahydrofolate reductase (MTHFR) gene. This cross-sectional pilot study recruited 48 girls with AN and 10 age-matched control girls with normal menstruation. We recorded anthropometric parameters and obtained blood samples for genotyping and hormonal assessment. Classification of AN was performed according to the DSM-V criteria. Girls with AN had 2.66 times higher odds of

Sympathetic overactivation predicts body weight loss in patients with heart failure.

Neurohumoral activation is frequently observed in chronic heart failure (HF) patients who develop body weight (BW) loss. We therefore hypothesized that sympathetic overactivation can predict progression of BW loss in HF patients with reduced ejection fraction. We prospectively evaluated BW loss in 108 non-edematous HF in whom muscle sympathetic nerve activity (MSNA) was measured. Follow-up began on the day of first MSNA measurement. Patients with BW loss of ≥5% of baseline BW during the first year of follow-up were considered to be experiencing BW loss. Maximal BW loss (%) and time to first BW

High protein diet promotes body weight loss among Brazilian postpartum women.

There is evidence in the general population that adhering to a high protein and low carbohydrate diet may help in losing weight. However, there is little evidence among postpartum women. The aim of this study is to evaluate the effect of a high protein diet on weight loss among postpartum women. A parallel-randomized controlled trial with 94 postpartum women was conducted in a maternity ward in Mesquita county (recruitment from February 2009 to December 2010) and in a polyclinic in Rio de Janeiro city (recruitment from December 2010 to December 2011). Women were randomized to the intervention

Distinct Glucose-Lowering Mechanisms of Ipragliflozin Depending on Body Weight Changes.

BACKGROUND: Sodium-glucose co-transporter 2 inhibitors have been shown to reduce body weight. However, little is known about whether a reduction in body weight affects glycemic and non-glycemic parameters. OBJECTIVES: The aim of this study was to investigate the link between the changes in body weight and those in metabolic parameters in drug-naïve subjects with type 2 diabetes mellitus (T2DM) receiving ipragliflozin monotherapy. METHODS: Subjects received ipragliflozin monotherapy 25-50 mg/day for 3 months (n = 33). They were then divided into two groups: group L ('lost'; n = 17) comprised

Probability of Vitamin D Deficiency by Body Weight and Race/Ethnicity.

INTRODUCTION: While most physicians recognize that vitamin D status varies by skin color because darker skin requires more light to synthesize vitamin D than lighter skin, the importance of body weight to vitamin D status is a newer, less recognized, finding. The purpose of this study was to use nationally representative US data to determine the probability of vitamin D deficiency by body weight and skin color. METHODS: Using data for individuals age ≥6 years from the 2001 to 2010 cycles of the US National Health and Nutrition Examination Survey, we calculated the effect of skin color, body w
