A screening of a range of common laboratory antibiotics for inhibition of the hammerhead ribozyme has shown that in addition to certain aminoglycosides (most notably neomycin B) the tetracyclines are also effective inhibitors, with chlorotetracycline being more effective than tetracycline. Inhibition by chlorotetracycline is not as strong as that by neomycin B but is more complicated, with at least two binding sites apparent. As with hammerhead inhibition by neomycin B, chlorotetracycline inhibition can be overcome by raising the concentration of the Mg2+ ion cofactor. We find that around six

SPR techniques can provide a wealth of insight into the nature of protein-carbohydrate interactions. Information not obtained readily by other methodologies can be gathered relatively quickly in a label-free manner with low sample consumption. This chapter focused on two applications in which SPR has been used to map conformational epitopes on bacterial polysaccharides recognized by protective antibodies. The glycoside composition and sequence of an extracellular polysaccharide flocculant of Klebsiella pneumoniae H12 was analyzed. GC and HPLC analysis of the acid-hydrolysate identified its constituent monosaccharides as D-Glc, D-Man, D-Gal, and D-GlcA in an approximate molar ratio of 3.9:1.0:2.3:3.6. Oligosaccharide alditols were released from bovine submaxillary mucin by alkaline borohydride treatment. Aminoglycoside antibiotics such as gentamicin and amikacin are well recognized as a clinically important antibiotic class because of their reliable efficacy and low cost. We identified a novel metabolic system of morphine in the opium poppy (Papaver somniferum L.). In response to stress, morphine is quickly metabolized to bismorphine consisting of two morphine units, followed by accumulation in the cell wall. A new triterpenoid saponin named bredemeyeroside B has been isolated from the roots of Bredemeyera floribunda and its structure elucidated as 3-O-beta-D-glucopyranosyl-2 beta, 3 beta, 27-trihydroxyolean-12-en-23,28-dioic acid 28-O-beta-D-xylopyranosyl-(1-->4)-[beta-D-apiofuranosyl-(1-->3)]-a lpha -L-rhamnopyranosyl-(1-->2)-[alpha-L-rhamnopyranosyl-(1-->3] -beta-D-fucopyranoside by means of MS, NMR and chemical evidence. The best yield of lipopolysaccharide (LPS) of Pseudomonas pseudomallei GIFU 12046 was obtained by extraction of defatted cells by phenol/chloroform/petroleum ether.
