Tabernaemontana arborea (Apocynaceae) is a Mexican tree species known to contain ibogan type alkaloids. This study aimed at determining central nervous system-related activities of an alkaloid extract obtained from the root bark of T. arborea. A gas chromatography-mass spectrometry (GC-MS) analysis was performed to describe the alkaloid profile of the extract. A wide dosing range (0.1 to 56.2 mg/kg) of this extract was evaluated in different murine models. Electrical brain activity was examined by electroencephalography (EEG). The extract's effects on motor coordination, ambulatory activity, a

Pityriasis versicolor is the most common skin mycosis in humans worldwide. Yeasts of the genus Malassezia, particularly M. furfur, a saprophyte occurring widely on human skin, are generally regarded as the causative agents. M. furfur is able to convert tryptophan into a variety of indole alkaloids, some of them showing biological properties that correlate well with certain clinical features of pityriasis versicolor. This suggests a possible role for these compounds in the pathophysiology of the disease. We here report that the novel pityriarubins A, B and C, isolated from cultures of the yeast

The 6/5/6 to 6/6/5 ring system rearrangement from indole alkaloids strictosidine (1) and strictosamide (2) to quinoline alkaloid pumiloside (3) is an essential but unelucidated process in the biosynthesis of camptothecin, especially, the intermediates involved in the process have not been identified. Herein, the nonenzymatic conversions including spontaneous reaction from 1 to 2, and light- or flavin-catalyzed skeletal rearrangement from 2 to 3 have been discovered. Particularly, the crucial biosynthetic intermediate strictosamide ketolactam (2a) with dicarbonyl nine-membered ring involved in

The spirocyclopropanyl cyclohexadienone alkaloids are among the most potent antitumor natural products yet discovered. The intriguing biological activity and unique structures of these alkaloids have made them attractive targets for total synthesis. Herein, we report an efficient and convergent total synthesis of (+)-CC-1065, the most structurally complex congener within this family of natural products. The synthetic strategy employs a novel application of our vinyl- and arylketene-based benzannulation, employing a visible light-promoted reaction of 2-iodoynamides with α-diazo ketones to acces

Extracellular enzymes secreted by Candida albicans are claimed to be virulence factors responsible for penetration of the yeast into host cells. Substances able to inhibit lipolytic and proteinase activities of the fungus might be of therapeutic use in some pathologic conditions caused by C. albicans. In the present work, we have tested the influence of the flavonoid compounds apigenin and kaempferol, the indole alkaloid ibogaine, and the protoberberine alkaloid berberine on the in vitro enzyme activity of C. albicans. The substances showed complex suppressive effects concerning the processes

C-H insertion offers a powerful route to C-C bond formation, yet it has often proven challenging to integrate into cascade synthetic sequences for further molecular elaboration. This limitation stems primarily from the aggressive conditions required for carbene generation and the high sensitivity of carbene species, which lead to premature termination and incapability with downstream transformations. Although intramolecular C-H insertion provides direct access to indolines, conventional approaches tend to produce ungoverned mixtures of indolines and indoles, owing to harsh conditions. To addre

Extracts of the bark of the central African tree Pausinystalia yohimbe contain yohimbine, an indole alkaloid, which is used to treat erectile dysfunction. The reported side effects of over-the-counter preparations of yohimbine include gastrointestinal upset, anxiety, increased blood pressure, headache, agitation, rash, tachycardia, and frequent urination. In this report, we describe a severe case of intractable priapism associated with the ingestion of yohimbe extract. Management required insertion of a proximal cavernosal spongiosum shunt (Quackles shunt) in the operating room.

Inflammatory bowel disease (IBD) remains one of the most prevalent gastrointestinal diseases worldwide. Purinergic signaling has emerged as a promising therapeutic target of inflammation-associated diseases. However, little is known about the specific roles of purinergic receptors in IBD. In the present study, expression profile of purinergic receptors was screened in the public Gene Expression Omnibus (GEO) datasets, and we found that expression of P2RX1 was significantly upregulated in inflamed colon tissues. Then, purinergic receptor P2RX1 was genetically ablated in the background of C57BL/

The growing need for effective HBV treatments and lead compounds with novel mechanisms prompted us to explore synthetic strategies for generating skeletally diverse alkaloidal Michael acceptors. Our approach uniquely embeds Michael acceptors directly within multicyclic alkaloid-inspired frameworks, exploiting the azepinoindole scaffold-a privileged structure in indole alkaloids. A single-step assembly between the versatile intermediate 13 with methyl propiolate 14 or its derivatives enabled the rapid and divergent synthesis of six alkaloidal Michael acceptors (15-20). This strategy facilitated

Starting from 1H-indole-3-carbaldehyde, 19 indole oxazole alkaloid analogues were synthesized in excellent yields. The results of bioassays showed that compound Z-2 possessed potent activity against Rhizoctonia solani and Botrytis cinereaPers with EC50 values of 7.03 and 13.5 mg/L, respectively. Compound Z-3 (EC50 = 6.81 mg/L) exhibited significant activity against Sclerotinia sclerotiorum. In vivo antifungal bioassays suggested that compounds Z-2 and Z-3 could serve as novel agricultural antifungals. Cell membrane permeability and PI staining experiments demonstrated that compounds Z-2 and Z-

The first total synthesis of scarce alkaloid indigotinoline A from Isatis indigotica was achieved in six steps from a known carboxylic acid (13% overall yield). The distinctive 6/5/6/5/6 N,O-heterocycle was efficiently assembled via a one-pot triple cascade, Povarov-type cyclization, intramolecular substitution, and visible-light-mediated oxidative rearrangement, followed by bioinspired N-oxidation/1,2-migration. This synthesis also prompted re-examination of proposed biosynthetic precursor isatindigotindoline B, leading to revision of its stereochemistry based on spectroscopic analysis.

Cyanobacteria are well-known for producing a diverse array of specialised metabolites, with increasing occurrences due to our changing climate. Though some cyanobacterial proliferations are harmless, others produce toxic compounds with detrimental effects to their surrounding environment including human health. A large cyanobacterial accumulation proliferating at Lough Urlár, Co. Mayo, Ireland, was selected for further chemical analysis owing to the presence of several polychlorinated metabolites detected by LC-MS in the methanolic fraction, which also displayed cytotoxic activity. After deep
