Interferons (IFN) are antiviral cytokines that mitigate the effects of invading viruses early on during the infection process. SARS-CoV and MERS induce weak IFN responses; hence, the clinical trials which included recombinant IFN accompanied with other antiviral drugs exhibited improved results in terms of shortening the duration of illness. The aim of the present study was to evaluate the type I IFN response in COVID-19 patients to determine whether it is sufficient to eliminate or reduce the severity of the infection, and whether it can be recommended as a potential therapy. Total RNA sample

The ten-eleven translocation (TET) family genes, which encode 5-methylcytosine (5mC) dioxygenases, play a "double-edged sword" role in tumor initiation and progression. However, the functional role and molecular mechanism of tumor cell-intrinsic TET3 in anti-tumor immunity remain incompletely understood. Here, we uncover that TET3 mRNA expression was aberrantly elevated in multiple cancer types and correlated with poor overall survival. Transcriptomic analysis reveals that TET3 depletion upregulated the expression of innate immune response genes, including numerous interferon-stimulated genes

While the long-term efficacy of checkpoint inhibition is established, the lack of response in IFN-rich tumors presents a critical challenge. These findings suggest that immune sensitivity depends on variables beyond cytokine levels and should be assessed in a temporal context. We propose the "IFN clock," in which efficacy is determined by the kinetics of interferon signaling rather than its level. The IFN clock is structured into three phases: a fast-on activation phase, a fast-off recovery phase, and a persistent-on resistant phase. In this model, short-lived interferon signaling supports ant

OBJECTIVE: Autoimmune haemolytic anaemia (AIHA) occurs in approximately 5-10% of patients with SLE and it is traditionally associated with higher disease activity, increased damage accrual and reduced survival. SLE management recommendations partially address disease-related haematological manifestations without specific indication for AIHA treatment. Indeed, therapeutic approaches are prevalently extrapolated from the haematology field. In the present report, we aimed at describing a case series of patients with SLE and AIHA successfully treated with anifrolumab (ANF).
METHODS: We described

Blau syndrome is a rare autoinflammatory disorder caused by NOD2 mutations, characterized by granulomatous arthritis, uveitis, and dermatitis. While type I interferon signatures are biomarkers in several autoinflammatory diseases, their role in Blau syndrome remains unclear. We assessed the interferon score in 11 patients with Blau syndrome and correlated it with disease activity. Our results demonstrate that type I interferon signatures were detected in only a minority of patients and showed no significant correlation with clinical disease activity, inflammatory markers, or treatment response

Macrophages coordinate inflammatory and immune responses to threats, yet how they interpret diverse danger signals to tailor inflammation remains unclear. Disturbances in extracellular and intracellular homeostasis alter cell volume, but the consequences for macrophage inflammatory responses are poorly understood. We demonstrate that macrophages use cell volume control as a danger-sensing mechanism to promote and augment inflammation. Using volume-regulated anion channel (VRAC)-deficient macrophages, which lack cell volume control under hypo-osmotic conditions, we show that cell volume disrupt

Sweet syndrome (SS) is an autoinflammatory neutrophilic dermatosis characterized by abrupt-onset inflammatory skin lesions and systemic symptoms, yet its molecular pathogenesis remains incompletely defined. To delineate disease-specific inflammatory programmes, we performed NanoString-based transcriptomic analysis of SS skin lesions and compared them with healthy control skin and pyoderma gangrenosum, a related neutrophilic dermatosis. SS exhibited a distinct inflammatory transcriptional signature marked by robust upregulation of type I and II interferon-stimulated genes, including CXCL9, CXCL

BACKGROUND: Various functions of activated microglia play crucial roles in the progression of retinitis pigmentosa (RP). This study aims to investigate the mechanisms underlying microglial migratory responses and phagocytic activity and their effects on photoreceptor degeneration.
METHODS: Trem2-deficient rd10 mice (Trem2-/-:rd10) were used in this study. N-methyl-N-nitrosourea (MNU)-induced retinal degeneration was established in microglia-specific Trem2 overexpression mice (Tmem119CreERT2:Rosa26CAG - LSL-Trem2). IFN-α/β receptor I (IFNAR1) neutralizing antibody was used to achieve type I in

STING signaling is emerging as a critical component of neurodegenerative diseases. While microglial STING-type I interferon (IFN-I) signaling is well-established, the role of STING signaling in neurons remains unclear. Here, we show that the STING protein is expressed in Purkinje cells of the cerebellum. Selective activation of STING signaling only in Purkinje cells using a conditional constitutively active N153S allele results in progressive neuronal loss and cerebellar atrophy, astrogliosis, but no microgliosis, leading to severe motor impairments in mice. Surprisingly, Purkinje cell STING a

Exposure to influenza A virus, respiratory syncytial virus, and human metapneumovirus is well known to increase the risk of Streptococcus pneumoniae (SPn) pneumonia in humans. Type I IFN (IFN-I) is a hallmark response to acute viral infections, and alveolar macrophages (AMs) constitute the first line of airway defense against opportunistic bacteria. Our study reveals that virus-induced IFNAR1 (IFN-I receptor) signaling directly impairs AM-dependent antibacterial protection. Using Ifnar1 conditional knockout mouse models, in vivo antibodies, bone marrow chimeric mice, and AM reconstitution, we

Plasmacytoid dendritic cells (pDCs) mount powerful antiviral type I interferon (IFN-I) responses, yet only a fraction of pDCs produces high levels of IFN-I. Here we report that peripheral pDCs in naive mice comprise three subsets (termed A, B and C) that represent progressive differentiation stages. This heterogeneity was generated by tonic IFN-I signaling elicited in part by the cGAS/STING and TLR9 DNA-sensing pathways. A small 'IFN-I-naive' subset (pDC-A) could give rise to other subsets; it was expanded in STING deficiency or after the IFN-I receptor blockade, but was abolished by exogenous

Dysregulation of type I interferon (IFN-I) signaling has been implicated in the pathogenesis of multiple systemic autoimmune diseases, including systemic lupus erythematosus, Sjögren's syndrome, and rheumatoid arthritis. Tripartite motif-containing protein 21 (TRIM21) serves as both a major autoantigen and a pivotal E3 ubiquitin ligase. In the IFN-I pathway, TRIM21 plays a dual regulatory role by targeting transcription factors such as interferon regulatory factor 3 (IRF3), interferon regulatory factor 5 (IRF5), and interferon regulatory factor 7 (IRF7) through multiple ubiquitination mechanis

PD-1 and PD-L1 are crucial regulators of immunity expressed on the surface of T cells and tumour cells, respectively. Cancer cells frequently use PD-1/PD-L1 to evade immune detection; hence, blocking them exposes tumours to be attacked by activated T cells. The synergy of PD-1/PD-L1 blockade with type I interferon (IFN) can improve cancer treatment efficacy. Type I IFN activates immune cells boosts antigen presentation and controls proliferation. In addition, type I IFN increases tumour cell sensitivity to the blockade. Combining the two therapies increases tumoral T cell infiltration and acti

OBJECTIVE: The type 1 interferon (IFN) pathway is up-regulated in dermatomyositis (DM). We sought to define how organ-specific disease activity as well as autoantibodies and other clinical factors are independently associated with systemic type I IFN activity in adult patients with DM.
METHODS: RNA sequencing was performed on 355 whole blood samples collected from 202 well-phenotyped DM patients followed up during the course of their clinical care. A previously defined 13-gene type I IFN score was modeled as a function of demographic, serologic, and clinical variables using both cross-section

Cryptosporidium infects gastrointestinal epithelium and is a leading cause of infectious diarrhea and diarrheal-related death in children worldwide. There are no vaccines and no fully effective therapy available for the infection. Type II and III interferon (IFN) responses are important determinants of susceptibility to infection but the role for type I IFN response remains obscure. Cryptosporidium parvum virus 1 (CSpV1) is a double-stranded RNA (dsRNA) virus harbored by Cryptosporidium spp. Here we show that intestinal epithelial conditional Ifnar1-/- mice (deficient in type I IFN receptor) a

Interferon regulatory factor (IRF) 3 and IRF7 are master regulators of type I interferon (IFN-I)-dependent antiviral innate immunity. Upon viral infection, a positive feedback loop is formed, wherein IRF7 promotes further induction of IFN-I in the later stage. Thus, it is critical to maintain a suitably low level of IRF7 to avoid the hyperproduction of IFN-I. In this study, we find that early expression of IFN-I-dependent STAT1 promotes the expression of XAF1 and that XAF1 is associated specifically with IRF7 and inhibits the activity of XIAP. XAF1-knockout and XIAP-transgenic mice display res

DYT-PRKRA (dystonia 16 or DYT-PRKRA) is caused by mutations in the PRKRA gene that encodes PACT, the protein activator of interferon (IFN)-induced double-stranded (ds) RNA-activated protein kinase (PKR). PACT participates in several cellular pathways, of which its role as a PKR activator protein during integrated stress response (ISR) is the best characterized. Previously, we have established that the DYT-PRKRA mutations cause enhanced activation of PKR during ISR to sensitize DYT-PRKRA cells to apoptosis. In this study, we evaluate if the most prevalent substitution mutation reported in DYT-P

In addition to SARS-CoV-2 and its variants, emerging viruses that cause respiratory viral infections will continue to arise. Increasing evidence suggests a delayed, possibly suppressed, type 1 interferon (IFN-I) response occurs early during COVID-19 and other viral respiratory infections such as SARS and MERS. These observations prompt considering IFN-β as a prophylactic or early intervention for respiratory viral infections. A rationale for developing and testing intranasal interferon beta (IFN-β) as an immediately available intervention for new respiratory viral infections that will arise un

Type I interferons (IFNs-I) fulfil multiple protective functions during pathogenic infections, but they can also cause detrimental effects and enhance immunopathology. Here, we report that IFNs-I promote the dysregulation of iron homeostasis in macrophages during systemic infections with the intracellular pathogen Candida glabrata, leading to fungal survival and persistence. By engaging JAK1, IFNs-I disturb the balance of the transcriptional activator NRF2 and repressor BACH1 to induce downregulation of the key iron exporter Fpn1 in macrophages. This leads to enhanced iron accumulation in the
