Electrocatalytic Reductive Amination of Aldehydes and Ketones with Aqueous Nitrite.

The electrocatalytic utilization of oxidized nitrogen waste for C-N coupling chemistry is an exciting research area with great potential to be adopted as a sustainable method for generation of organonitrogen molecules. The most widely used C-N coupling reaction is reductive amination. In this work, we develop an alternative electrochemical reductive amination reaction that can proceed in neutral aqueous electrolyte with nitrite as the nitrogenous reactant and via an oxime intermediate. We develop a selection criterion for nitrite reduction electrocatalysts suited for oxime electrosynthesis and

Harnessing Electrochemistry for Direct Deoxygenative Silylation of Alcohols and Ketones.

Alcohols and ketones are abundant and structurally diverse feedstocks, yet their direct transformation into organosilicon compounds remains challenging due to the difficulty in selective cleaving C─OH and C═O bonds. Here, we report an electrochemically driven deoxygenative C─Si bond formation strategy that converts alcohols and ketones directly to organosilicon compounds. The reactions operate under mild conditions without external redox reagents and sacrificial electrodes. A wide range of alcohols, including primary, secondary, and tertiary alcohols, as well as ketones, are efficiently conver

Molecular Editing of Ketones Derived by Pro-aromatic of Dihydroquinazolinones.

Ketones represent a vital class of feedstock chemicals in organic synthesis. Researchers have explored a versatile deacylation strategy utilizing ketones as radical precursors, involving activation through condensation with 2-aminobenzamide to form dihydroquinazolinones (DHQZ). This aromatization-driven deconstructive reaction of ketones offers an efficient approach for the selective construction of diverse bonds, including C-H/D, C-C (sp, sp2, sp3), C-F, C-S/Se, C-N, and C-Si under mild conditions. As a result, ketones have found widespread application in the rapid and efficient assembly of s

Friedel-Crafts acylation via interrupted Beckmann fragmentation of activated ketones.

Friedel-Crafts (FC) acylation has long been a fundamental electrophilic arene substitution reaction. In this work, we report a new procedure for FC acylation utilizing stable and user-friendly acylium precursors, α-oximinoketones. The key to this methodology is the selective Csp2 -Csp2 bond cleavage in α-oximinoketones, facilitated by triflic anhydride, leading to the formation of acylium ions under mild conditions. This approach demonstrates compatibility with a variety of substituted alkyl, (hetero)aryl ketones, and cyclic ketones as acylating reagents, where cyclic ketones allow for unique

Access to Alkylative/Hydrodefluorination of Trifluoromethyl Ketones Using Photoexcited Dihydropyridines.

A convenient strategy for selective and challenging single C-F bond functionalization of trifluoromethyl ketones has been introduced under visible light irradiation. Photoexcited 1,4-DHP serves a dual purpose as a reductant and a source of alkyl radicals (or H atoms) in the alkylative defluorination and hydrodefluorination processes. The solvent hexafluoroisopropyl alcohol (HFIP) plays a crucial role in the selective single C-F bond cleavage. Overall, a transition-metal- and photocatalyst-free, mild protocol for accessing a broad range of functionalized fluorinated products of high pharmaceuti

1,4-Reduction of α,β-Unsaturated Ketones Catalyzed by Tetraarylhydrophosphoranes.

We demonstrate that a pentavalent phosphorus compound, tetraarylhydrophosphorane, is capable of catalyzing 1,4-reduction of α,β-unsaturated ketones in the presence of pinacolborane. The structure and properties of tetraarylhydrophosphorane and its participation in the chemoselective 1,4-reduction were elucidated through both experimental and theoretical approaches.

Catalytic Asymmetric Barbier Reaction of Ketones with Unactivated Alkyl Electrophiles.

The Barbier reaction is a reductive-type addition of an aldehyde or ketone with an organic electrophile in the presence of a terminal metal reductant, providing a straightforward and efficient method for carbon-carbon bond formation. This reaction possesses the advantage of circumventing the preparation of moisture- and air-sensitive organometallic reagents. However, the catalytic Barbier reaction of ketones to construct tetrasubstituted stereogenic centers is largely underdeveloped, despite its great potential for accessing synthetically challenging chiral tertiary alcohol. Particularly, the

Iron-Catalyzed Reductive Ring Opening/gem-Difluoroallylation of Cyclopropyl Ketones.

By merging C-C and C-F bond cleavage, we developed a regioselective ring opening/gem-difluoroallylation of cyclopropyl ketones with α-trifluoromethylstyrenes, which proceeds under the catalysis of iron with the combination of manganese and TMSCl as the reducing agents, providing a new entry to the synthesis of carbonyl-containing gem-difluoroalkenes. Remarkably, the ketyl radical-induced selective C-C bond cleavage and the following generation of more-stable carbon-centered radicals enable complete regiocontrol of this ring opening reaction for various substitution patterns of the cyclopropane

Photochemical Selective Oxidation of Benzyl Alcohols to Aldehydes or Ketones.

Using Eosin Y as a metal-free photocatalyst and O2 as an oxidant, the present study reports a new photochemical protocol that enables efficient aerobic oxidation of various benzyl alcohols to the corresponding aldehydes or ketones in excellent yields under mild reaction conditions. The catalyst system presents good functional-group tolerance and exquisite chemoselectivity, which also can easily be scaled-up to gram scale. Moreover, the methodological applications in practical synthesis of several organic molecules and the primary reaction mechanism were also discussed.

Theoretical insights into the spectroscopic properties of ferrocenyl hetaryl ketones.

Quantum chemical calculations have been carried out to elucidate the electronic structure as well as to draw structure-property relationships for a series of ferrocenyl hetaryl ketones by means of simulated NMR, IR and UV-vis spectra. In this series, the list of hetaryl groups included furan-2-yl, thiophen-2-yl, selenophen-2-yl, 1H-pyrrol-2-yl and N-methylpyrrol-2-yl. Density functional theory was employed to determine the ground-state properties of the five ketones while their excited-state properties were modeled using a broad range of theoretical methods, namely from time-dependent density

C-H/N-H cross-coupling has become a key technology for the selective conjugation of azole drug molecules. However, the development of new synthetic models and green chemical methods is imperative to enhance the construction of multi-functional compounds and compounds with unique functional groups. We herein reported an electrochemical synthesis of α-tetrazolyl ketones with excellent yields and broad substrate scope, encompassing electron-donating and electron-withdrawing groups of aryl ketones, heterocycles, and alkyl and various ketone drugs. It was further proved that α-iodoketone was involv

Catalytic Asymmetric α-Alkylation of Ketones with Unactivated Alkyl Halides.

A catalytic, enantioselective method for direct α-alkylation of ketones with unactivated alkyl halides is realized by employing an α-enolizable ketone in a nickel-catalyzed C(sp3)-C(sp3) cross-coupling reaction. The key to the success is attributed to a unique bimetallic ligand. A variety of acyclic ketones and unactivated alkyl iodides can serve as suitable substrates under mild conditions to generate chiral ketones with α-quaternary carbon stereocenters in high yields with good enantioselectivities. A range of transformations based on the ketone moiety are also demonstrated to show the poten

Photoredox/nickel dual catalyzed stereospecific synthesis of distal alkenyl ketones.

The selective C-C bond deconstruction/refunctionalization via a photoredox/nickel dual-catalyzed hydroalkylation of alkynes is developed under mild reaction conditions. In this protocol, a broad range of alkyl- and aryl-alkynes could react smoothly with cycloalkanols, affording the corresponding distal and site-specific vinyl-substituted ketones with high yields and excellent regioselectivities. Moreover, DFT calculations verified that the electron-rich behavior of aromatics and weak Brønsted bases have a common effect on the photocatalytic oxidant ring-opening of cyclobutanols.

Diverse reactivity of an Al(I)-centred anion towards ketones.

The reactivity of a seven-membered cyclic potassium diamidoalumanyl toward a variety of ketone small molecules has been assessed. Whilst acetophenone generates an aluminium pinacolate derivative, reductive C-C coupling is induced between the ketyl and ortho-carbon centres of two equivalents of benzophenone. In contrast, whereas oxidative addition of an enolisable proton is observed with 2,4-dimethyl-3-pentanone, 2,2,4,4-tetramethyl-3-pentanone undergoes an unprecedented hydroalumination process, where the reducing hydride may be traced to intramolecular oxidative addition of a (sp3)C-H bond.

Palladium-catalyzed stereoselective ring-opening reaction of aryl cyclopropyl ketones.

Herein, we report that α,β-unsaturated ketones could be obtained by palladium-catalyzed ring-opening of mono-substituted cyclopropyl ketones efficiently and systematically. (E)-1-Arylbut-2-en-1-ones were generated from aryl cyclopropyl ketones stereoselectively in yields of 23-89% by the Pd(OAc)2/PCy3 catalytic system. The reaction exhibited stereoselectivity (only E products were found) and was suitable for both phenyl and heteroaryl cyclopropyl ketones.

NickelII-catalyzed asymmetric photoenolization/Mannich reaction of (2-alkylphenyl) ketones.

A diastereo- and enantioselective photoenolization/Mannich (PEM) reaction of ortho-alkyl aromatic ketones with benzosulfonimides was established by utilizing a chiral N,N'-dioxide/Ni(OTf)2 complex as the Lewis acid catalyst. It afforded a series of benzosulfonamides and the corresponding ring-closure products, and a reversal of diastereoselectivity was observed through epimerization of the benzosulfonamide products under continuous irradiation. On the basis of the control experiments, the role of the additive LiNTf2 in achieving high stereoselectivity was elucidated. This PEM reaction was prop

Boron-Catalyzed, Diastereo- and Enantioselective Allylation of Ketones with Allenes.

The diastereo- and enantioselective allylation of ketones remains a synthetic challenge, with transition metal catalysis offering the most applied methods. Here, a boron-catalyzed allylation of ketones with allenes is presented. Excellent yield, regioselectivity, and diastereoselectivity were found across functionalized substrates. The reaction was further developed to accommodate an enantioenriched boron catalyst and thus gave asymmetric ketone allylation in good yield, diastereoselectivity, and enantioselectivity. Mechanistic studies supported a hydroboration-allylation-transborylation pathw

Iridium-Catalyzed and pH-Dependent Reductions of Nitroalkenes to Ketones.

A highly chemoselective conversion of α,β-disubstituted nitroalkenes to ketones is developed. An acid-compatible iridium catalyst serves as the key to the conversion. At a 2500 S/C ratio, nitroalkenes were readily converted to ketones in up to 72% isolated yields. A new mechanistic mode involving the reduction of nitroalkene to nitrosoalkene and N-alkenyl hydroxylamine is proposed. This conversion is ready to amplify to a gram-scale synthesis. The pH value plays an indispensable role in controlling the chemoselectivity.
