Catatonia in Ugandan children with nodding syndrome and effects of treatment with lorazepam: a pilot study.

BACKGROUND: Nodding syndrome (NS) is a severe neuropsychiatric syndrome of an unknown etiology affecting children and adolescents mostly in Eastern Africa. Symptoms of NS and catatonia seem to overlap. We investigated the presence and types of catatonic symptoms in NS and their response to one or two doses of lorazepam, the first-line treatment for catatonia. METHODS: A cross-sectional descriptive study with systematic assessment of catatonia in 33 patients with NS using a modified version of the Bush Francis Catatonia Rating Scale. Sixteen patients met criteria for catatonia and were observe

Effects of Lorazepam and citalopram on human defensive reactions: ethopharmacological differentiation of fear and anxiety.

Drugs that are clinically effective against generalized anxiety disorder preferentially alter rodent risk assessment behavior, whereas drugs that are clinically effective against panic disorder preferentially alter rodent flight behavior. The theoretical principle of "defensive direction" explains the pattern of associations between emotion and defensive behavior in terms of the differing functional demands arising from cautious approach to threat (anxiety) versus departure from threat (fear), offering the prospect that clinically important emotions may be explained using a single rubric of de

Effects of Lorazepam on the automatic online evaluation of sleep EEG data in healthy volunteers.

In earlier publications we described an automatic algorithm to detect rapid eye movement (REM) sleep from a single-channel EEG recording without using EMG or EOG information. This system consisted of an artificial neural network operating on the basis of preprocessed EEG data and was composed to provide a maximum of robustness for online applications. In the present study the influence of acute administration of lorazepam on the performance of the REM detection procedure was evaluated. Following an adaptation to laboratory conditions, sleep EEG data were obtained from healthy subjects in three

Influence of certain calcium-channel blockers on some aspects of lorazepam-dependence in mice.

The effect of acute and chronic treatments of the calcium-channel blockers, isradipine, diltiazem and flunarizine in protecting against lorazepam dependence has been demonstrated in mice. Dependence was induced by twice-daily administration of lorazepam (1 mg kg-1) for 10 days, doubling the dose during the next 10 days. Withdrawal symptoms and changes in the noradrenaline, dopamine and 5-hydroxytryptamine content of different regions of the brain were observed after either 24-h withdrawal or flumazenil administration. Isradipine inhibited lorazepam withdrawal symptoms, the effect being accompa

[A controlled double-blind study of tetrabamate versus lorazepam and placebo in generalized anxiety].

The anxiolytic efficacy of tetrabamate was evaluated in a multicentric double-blind study versus lorazepam and placebo, in 269 patients with a generalized anxiety disorder according to DSM III-R criteria. The anxiolytic activity of tetrabamate (at 900 mg/day) was significantly superior than that of placebo from day 7 of treatment and equivalent to lorazepam efficacy (at 4.5 mg/day). In the tetrabamate group, 55.3% were considered as "good responders" (as defined by a HARS score reduction equal or superior to 50%), versus 51.3 and 32.9% respectively in the lorazepam and the placebo groups (chi-

Differential generalization to pentobarbital in rats trained to discriminate lorazepam, chlordiazepoxide, diazepam, or triazolam.

In drug discrimination studies benzodiazepine-trained animals have typically responded on the drug lever when tested with barbiturates. In a recent study, greater specificity appeared to be shown when lorazepam was used as a training drug. The generality and limits of this finding were explored in the present set of experiments. The asymmetrical cross-generalization found in lorazepam- and pentobarbital-trained baboons was replicated in rats and was shown not to be a function of either lorazepam (0.1., 0.32, or 1.0 mg/kg) or pentobarbital (10 or 25 mg/kg) training dose (i.e., pentobarbital-tra

Effects of barbital and lorazepam in the AT and ANT rats: motor performance and brain monoamine functions.

The motor impairing effects and central monoaminergic neuronal effects of barbital (120 mg/kg, IP) and lorazepam (3 mg/kg, IP) were studied in the alcohol tolerant (AT) and alcohol non-tolerant (ANT) rat lines developed for low and high sensitivity to motor impairment from ethanol (2 g/kg, IP), respectively. Like ethanol, both barbital and lorazepam impaired the performance of the ANT rats more than that of the AT rats. Ethanol increased the synthesis of catecholamines in the striatum and hypothalamus, but barbital and also lorazepam decreased it in both rat lines. Ethanol increased or did not

Lorazepam, a new benzodiazepine derivative, in the treatment of anxiety: a double-blind clinical evaluation.

Ninety-five adult outpatients suffering from moderate to severe anxiety were randomly assigned to treatment with lorazepam or placebo in a double-blind four-week study. Patients were evaluated at pretreatment and after two and four weeks of treatment using three rating scales. Lorazepam, at an average daily dose of 3.2 mg twice daily, was highly effective in relieving anxiety as documented by clinically and statistically significant differences over placebo in most of the items on all rating scales at all rating periods. No serious adverse effects were reported, and there were no adverse inter

Effective Dose 50 of Intranasal Lorazepam for Clinical Sedation in Zebra Finches (Taeniopygia guttata).

Sedation is a common procedure in avian practice and benzodiazepines are often used as sole agents or in combination with other drugs to sedate avian patients. Midazolam and diazepam have been evaluated in zebra finches (Taeniopygia guttata); however, lorazepam has not. Our aim was to determine the sedation effective dose 50 (ED50) of intranasal (NAS) lorazepam in zebra finches. Using the Dixon up-and-down method, the first randomly selected finch received 0.25 mg/kg NAS lorazepam. Sedation was scored after 20 minutes: 1) recumbent = 0 versus not recumbent = 1; 2) captured at first attempt = 0

Management of Relapsing Catatonia After Lorazepam Discontinuation: A Systematic Review of Published Case Reports.

BACKGROUND: Lorazepam is the mainstay of pharmacological treatment of catatonia. It is recommended that lorazepam, when effective, be tapered and gradually stopped depending upon maintenance of clinical improvement. This recommendation is not supported by any controlled studies. There are case reports on relapses of catatonia while tapering lorazepam; these patients required long-term maintenance treatment for sustained symptomatic management. OBJECTIVE: This is a review of published literature focusing on relapse of catatonia following lorazepam discontinuation after maintenance treatment.

Effects of lorazepam on saccadic eye movements - evidence from prosaccade and free viewing tasks.

RATIONALE: Peak velocities of saccadic eye movements are reduced after benzodiazepine administration. Even though this is an established effect, past research has only examined it in horizontal prosaccade tasks. OBJECTIVES: The spectrum of saccadic eye movements, however, is much larger. Therefore, we aimed to make a first attempt at filling this research gap by testing benzodiazepine effects on saccades under different experimental task conditions. METHODS: 1 mg lorazepam or placebo was administered (within-subjects, double-blind, in randomised order) to n = 30 healthy adults. Participants

Implication of UGT2B15 Genotype Polymorphism on Postoperative Anxiety Levels in Patients Receiving Lorazepam Premedication.

BACKGROUND: Lorazepam is used as premedication for its anxiolytic properties. The UGT2B15 genotype is of importance for the metabolism of lorazepam. The clinical effect of genetic polymorphisms in UGT2B15 genotype on the treatment of anxiety levels in same-day surgery patients receiving lorazepam, however, is unknown. METHODS: Three hundred ninety-eight same-day surgery patients of mixed sex (from a previous double-blinded randomized controlled trial who were assigned to either lorazepam [n = 198] or placebo [n = 200]) were assessed for the UGT2B15*2 variant allele. Anxiety was measured preop

[Prolonged toxic coma and anisocoria secondary to doxepin, lorazepam and phenobarbital poisoning--case study].

Doxepin is a tricyclic antidepressant from the group of dibenzoxepines. Apart from the antidepressant effect, it has also the sedative and anxiolytic effect, so it is used in the treatment of anxiety disorder in the course of psychosis, organic diseases and alcoholism. Doxepin increases concentration of norepinephrine and serotonin in the brain by preventing inactivation and blocking their reuptake. In addition, the drug has an antagonistic effect on receptors in CNS (muscarinic M1, histamine H1, alpha-1adrenergic, serotonergic 5-HT2) and also blocks sodium and potassium channels in cardiomyoc

Apolipoprotein E epsilon4 allele and lorazepam effects on memory in high-functioning older adults.

CONTEXT: The apolipoprotein E (APOE) epsilon4 allele has been implicated as a significant risk factor in the development of late-onset Alzheimer disease, but the evidence of cognitive sequelae in healthy individuals has been mixed. OBJECTIVE: To determine if the APOE epsilon4 allele increases susceptibility to lorazepam-induced verbal learning impairment in nondemented older adults. DESIGN: A placebo-controlled crossover design. SETTING: A community-based sample of subjects. PARTICIPANTS: Sixty-four cognitively intact and highly educated (>12 years) adults. The Use of Lorazepam for Acute Episodes in a Patient with Kleine-Levin Syndrome: A Longitudinal Case Report.

Kleine-Levin Syndrome (KLS) is a rare, neurological disorder characterized by episodes of hypersomnia, cognitive impairment, and behavioral abnormalities. The etiology of KLS remains unclear, and treatment responses are highly variable. This report describes a 22-year longitudinal experience with a 40-year-old male diagnosed with KLS at 15 years of age. Treatment trials of all standard KLS medications (lithium, valproic acid, selective serotonin reuptake inhibitors, modafinil) yielded no benefit in attenuating or preventing episodes. Following the onset of overlapping an acute KLS exacerbation

A case of comorbidity of schizophrenic catatonia and chronic intestinal pseudo-obstruction Successfully managed with lorazepam.

It is challenging to manage schizophrenic catatonia and comorbid chronic intestinal pseudo-obstruction (CIPO). The pathology of catatonia is unclear. There are few reports or research on this issue. In this case, we present a middle-aged woman diagnosed with schizophrenia with catatonic features and comorbid CIPO. In the treatment process, modified electroconvulsive therapy (mECT) improved her stupor and CIPO partially. Lorazepam alleviated her stupor and CIPO completely. It is the first report describing complete remission with lorazepam in patient suffering from comorbid schizophrenic catato

Concomitant lorazepam use and antidepressive efficacy of repetitive transcranial magnetic stimulation in a naturalistic setting.

BACKGROUND/OBJECTIVES: Repetitive transcranial magnetic stimulation (rTMS) has been established as an effective therapeutic intervention for the treatment of depression. Preliminary data suggest that the efficacy of rTMS is reduced in patients taking benzodiazepines (BZD). Here, we use real-world data from a large sample to investigate the influence of lorazepam on the effectiveness of rTMS. METHODS: From a retrospective cohort of clinically depressed patients that were treated with rTMS, we compared 176 patients not taking any BZD with 73 patients taking lorazepam with respect to changes in
