Simultaneous determination of aesculin and aesculetin and their interactions with DNA using carbon fiber microelectrode modified by Pt-Au bimetallic nanoparticles.

Electrode sensitivity and selectivity in complex biological matrices are major challenges in the development of electrochemical sensors. Bimetallic nanoparticles provide a new perspective for enhancing electrocatalytic property because of some specific synergetic effects. In this work, platinum nanoparticles (PtNPs) and gold nanoparticles (AuNPs) modified carbon fiber microelectrode (PtNPs/AuNPs/CFME) was fabricated to determine aesculin and aesculetin simultaneously. Differential pulse voltammetry (DPV) method was conducted for the electrochemical sensing of aesculin and aesculetin, the modif

Conformable Microelectrode Arrays Integrated with a Scoop-Shaped Slide-Well for Dynamic Electrophysiological Profiling of Patient-Derived Cardiac Organoids.

We present an integrated microelectrode array (MEA) platform for high-precision electrophysiological characterization of patient-derived 3D cardiac organoids (COs), enabling dynamic recordings from both healthy and Duchenne muscular dystrophy (DMD) "mini-hearts." A deformable MEA, fabricated by photolithographic patterning of support layers and microelectrodes, is conformally assembled onto a customized scoop-slide well. The concave geometry enables passive self-alignment of millimeter-scale COs within a multichannel electrode array, allowing simultaneous extracellular field potential (EFP) re

A high-density microelectrode array integrated microfluidic platform for quantitative investigation of functional connectivity in interconnected human dopaminergic neurospheroids.

The quantitative investigation of functional connectivity across spatially organized neuronal networks remains a major challenge in human three-dimensional (3D)in vitromodels. Although neurospheroids, brain organoids, and microfluidic systems enable controlled structural organization and guided neurite growth, their integration with high-resolution electrophysiological interfaces remains limited, often requiring post hoc adaptations that compromise recording stability and long-term reproducibility. Here, we present a polydimethylsiloxane (PDMS) microfluidic platform natively engineered for sea

Utilizing GO/PEDOT:PSS/PtNPs-enhanced high-stability microelectrode arrays for investigating epilepsy-induced striatal electrophysiology alterations.

The striatum plays a crucial role in studying epilepsy, as it is involved in seizure generation and modulation of brain activity. To explore the complex interplay between the striatum and epilepsy, we engineered advanced microelectrode arrays (MEAs) specifically designed for precise monitoring of striatal electrophysiological activities in rats. These observations were made during and following seizure induction, particularly three and 7 days post-initial modeling. The modification of graphene oxide (GO)/poly (3,4-ethylenedioxythiophene):polystyrene sulfonate (PEDOT:PSS)/platinu-m nanoparticle

Microelectrode clusters enable therapeutic deep brain stimulation without noticeable side-effects in a rodent model of Parkinson's disease.

BACKGROUND: Deep Brain Stimulation (DBS) is an established treatment for motor symptoms in Parkinson's disease (PD). However, side effects often limit the usefulness of the treatment. NEW METHOD: To mitigate this problem, we developed a novel cluster of ultrathin platinum-iridium microelectrodes (n = 16) embedded in a needle shaped gelatin vehicle. In an established rodent PD-model (6-OHDA unilateral lesion), the clusters were implanted in the subthalamic area for up to 8 weeks. In an open field setting, combinations of microelectrodes yielding therapeutic effects were identified using statis

Hierarchical ZnO@Hybrid Carbon Core-Shell Nanowire Array on a Graphene Fiber Microelectrode for Ultrasensitive Detection of 2,4,6-Trinitrotoluene.

A hierarchical architecture composed of nitrogen (N)-rich carbon@graphitic carbon-coated ZnO nanowire arrays on a graphene fiber (ZnO@C/GF) was fabricated by direct growth of a ZnO@zeolitic imidazolate framework-8 (ZIF-8) core-shell nanowire array on a GF followed by annealing and used as a microelectrode for detection of 2,4,6-trinitrotoluene (TNT). In such a design, ZnO accumulated TNT through a strong nitroxide-zinc interaction and ZIF-8 served as the precursor of the N-rich carbon@graphitic carbon layer that seamlessly connected ZnO with the GF to improve the poor conductivity of ZnO, thus

A Chitosan-Carbon Nanotube-Modified Microelectrode for In Situ Detection of Blood Levels of the Antipsychotic Clozapine in a Finger-Pricked Sample Volume.

The antipsychotic clozapine is the most effective medication available for schizophrenia and it is the only antipsychotic with a known efficacious clinical range. However, it is dramatically underutilized due to the inability to test clozapine blood levels in finger-pricked patients' samples. This prevents obtaining immediate blood levels information, resulting in suboptimal treatment. The development of an electrochemical microsensor is presented, which enables, for the first time, clozapine detection in microliters volume whole blood. The sensor is based on a microelectrode modified with mic

A novel flexible cuff-like microelectrode for dual purpose, acute and chronic electrical interfacing with the mouse cervical vagus nerve.

OBJECTIVE: Neural reflexes regulate immune responses and homeostasis. Advances in bioelectronic medicine indicate that electrical stimulation of the vagus nerve can be used to treat inflammatory disease, yet the understanding of neural signals that regulate inflammation is incomplete. Current interfaces with the vagus nerve do not permit effective chronic stimulation or recording in mouse models, which is vital to studying the molecular and neurophysiological mechanisms that control inflammation homeostasis in health and disease. We developed an implantable, dual purpose, multi-channel, flexib

Use of intraoperative CT to predict the accuracy of microelectrode recording during deep brain stimulation surgery. A proof of concept study.

OBJECTIVES: Intraoperative computed tomography (iCT) is currently used to confirm the target location of the microelectrode (ME) during microelectrode recording (MER) and ultimate location of deep brain stimulation (DBS) leads at our institution. We evaluated whether iCT can be used to predict the trajectory and accuracy of the ME track. PATIENTS AND METHODS: Intraoperative imaging profiles of ten consecutive patients who had undergone DBS surgery were retrospectively reviewed. We found that cranial iCT, in addition to visualizing the target, also visualizes the extra-cranial segment of the g

The use of microelectrode array (MEA) to study the protective effects of potassium channel openers on metabolically compromised HL-1 cardiomyocytes.

The microelectrode array (MEA) was used to evaluate the cardioprotective effects of adenosine triphosphate sensitive potassium (K(ATP)) channel activation using potassium channel openers (KCOs) on HL-1 cardiomyocytes subjected to acute chemically induced metabolic inhibition. Beat frequency and extracellular action potential (exAP) amplitude were measured in the presence of metabolic inhibitors (sodium azide (NaN(3)) or 2-deoxyglucose (2-DG)) or KCOs (pinacidil (PIN, a cyanoguanidine derivative, activates sarcolemmal K(ATP) channels) or SDZ PCO400 (SDZ, a benzopyran derivative, activates mitoc

K(+) transport in the mesonephric collecting duct system of the toad Bufo bufo: microelectrode recordings from isolated and perfused tubules.

We studied the mechanisms of K(+) transport in cells from isolated and perfused collecting tubules and ducts from the mesonephric kidney of the toad Bufo bufo. Cells were impaled with microelectrodes across the basal cell membrane. The basolateral membrane potential (V(bl)) depolarized upon change of bath [K(+)] from 3 to 20 mmol l(-1) demonstrating a large K(+) conductance in this membrane. In collecting tubules and collecting ducts a V(bl) of -66+/-2 mV and -74+/-4 mV depolarized by 30+/-2 mV and 36+/-3 mV, respectively (N=23; 15). The K(+) channel inhibitor Ba(2+) (1 mmol l(-1)) inhibited t

Extensive coronary venous system mapping for outflow tract ventricular arrhythmias: Utility of a dual over-the-wire microelectrode catheter technique.

BACKGROUND: During catheter ablation of epicardial or intramural outflow tract ventricular arrhythmia (OTVA), the presumed site of origin must often be inferred from coronary venous system (CVS) mapping, which is anatomically constrained. OBJECTIVE: This study sought to evaluate the feasibility and clinical utility of a dual over-the-wire (OTW) microelectrode catheter technique for extensive CVS mapping in OTVAs. METHODS: We prospectively enrolled 30 patients with OTVA exhibiting either a right bundle branch block or a left bundle branch block pattern with V2 or V3 transition. 2 2.7Fr OTW mi

Feasibility of Basal Ganglia Microelectrode Recordings under General Anesthesia with Combined Nitrous Oxide and Sevoflurane: A Retrospective Single-Center Experience.

INTRODUCTION: Deep brain stimulation (DBS) is an established treatment for Parkinson's disease (PD). The traditional method for accurate implantation is awake microelectrode recordings (MERs) to map out the borders of the target nucleus. However, a significant portion of patients are unable to tolerate awake surgical procedures. Asleep MER techniques under different general anesthesia regimens have been described with variable effects on recording quality and required a lower inhaled sevoflurane level to obtain single unit recordings. Hence, a reliable method for asleep MER mapping is needed w

Electrochemical HPLC Determination of Piperazine Antihistamine Drugs Employing a Spark-Generated Nickel Oxide Nanoparticle-Modified Carbon Fiber Microelectrode.

In this work, we demonstrate a sensitive high-performance liquid chromatography (HPLC) method for the determination of piperazine antihistamine drugs employing innovative electrochemical detection based on a spark-generated nickel oxide nanoparticle-modified carbon fiber microelectrode built into a miniaturized electrochemical detector. The direct carbon fiber-to-nickel plate electrode spark discharge, carried at 0.8 kV DC, with the nickel electrode connected to the negative pole of the high-voltage power supply, provides extremely fast (1 s) in situ tailoring of the carbon fiber microelectrod

Microelectrode Arrays Measure Blocking of Voltage-Gated Calcium Ion Channels on Supported Lipid Bilayers Derived from Primary Neurons.

Drug studies targeting neuronal ion channels are crucial to understand neuronal function and develop therapies for neurological diseases. The traditional method to study neuronal ion-channel activities heavily relies on the whole-cell patch clamp as the industry standard. However, this technique is both technically challenging and labour-intensive, while involving the complexity of keeping cells alive with low throughput. Therefore, the shortcomings are limiting the efficiency of ion-channel-related neuroscience research and drug testing. Here, this work reports a new system of integrating neu

Does the Use of Intraoperative Microelectrode Recording Influence the Final Location of Lead Implants in the Ventral Intermediate Nucleus for Deep Brain Stimulation?

To determine if the use of intraoperative microelectrode recording (MER) influences the final location of lead implant in deep brain stimulation (DBS) of the ventral intermediate nucleus (VIM), and to evaluate the incidence of associated complications. The usefulness of intraoperative MER in DBS is debated, some centers suggesting it increases complications without additional benefit. We conducted a retrospective chart review of all patients who underwent VIM DBS with MER at the University of Texas Health Science Center in Houston from June 1, 2009 to October 1, 2013. Initial (MRI determined)

Treatment of Macro-Reentry Atrial Tachycardia with Very High-Power, Short-Duration, Temperature-Controlled Ablation of Anterior Line Using an Open-Irrigated Ablation Catheter with Microelectrodes.

BACKGROUND Treatment of atrial fibrillation and atrial tachycardia (AT) with catheter ablation results in high rates of success with the procedure and on long-term follow-up. A novel ablation catheter with a very high-power, short-duration (vHPSD) ablation mode using 90 W for 4 s has been introduced, which could improve safety and efficacy of catheter ablation, especially for pulmonary vein isolation (PVI). To date, vHPSD mode has only been evaluated for treatment of PVI, but it could be an efficient technique for linear lesions. Here, we present the first use of the novel vHPSD mode alone for

Peptide substrate optimization toward multiplex protease activity profiling under neutral pH conditions using Au microelectrode arrays.

Cathepsin B (CB), a lysosomal cysteine protease, is associated with cancer and other diseases. Sensitive multiplex detection of CB and other extracellular proteases at neutral pH compatible with human serum is essential to advance biosensing platforms for disease diagnosis. Here, we present a strategy to enhance protease activity profiling based on a 3×3 gold (Au) microelectrode array (MEA). Ferrocene-tagged peptide substrates are functionalized on Au MEA. The proteolytic cleavage induces an exponential decay of the electrochemical signal, with the inverse of the decay time constant (1/τ) refl
