The effect of storage delay and storage temperature on orthopaedic surgical samples contaminated by Staphylococcus Epidermidis.

BACKGROUND: Prosthetic Joint Infection (PJI) is a rare but devastating complications with high morbitity and mortality. The identification of the causal microorganism remains crucial and determines therapeutic strategies and success. Microbiology cultures remain the common method to diagnose PJI. Unfortunately, 14% of intra-articular punctures remain negative after culture. The microorganisms are best detected by inoculation of microbiology samples in blood culture bottles (Bactec), or after sonication of the implant and polymerase chain reaction (PCR). The identification of the causal microor

Different Behavior and Response of Staphylococcus Epidermidis and Streptococcus Pneumoniae to a Ventriculoperitoneal Shunt: An in vitro Study.

BACKGROUND: Pneumococcal shunt infection is a rare event. There is no consensus on the therapeutic management of this kind of shunt infection according to literature reviews, and it seems to be different from infection with Staphylococcus epidermidis. We studied 2 shunted patients with pneumococcal meningitis, both of whom were treated with only antibiotics. The management of these cases seems to be different from that of shunt catheter infection due to these bacteria. We conducted a laboratory study to show the different behavior of pneumococcus compared to S. epidermidis regarding shunt cath

Different sensitivity levels to norspermidine on biofilm formation in clinical and commensal Staphylococcus epidermidis strains.

Biofilm formation on medical and surgical devices is the main virulence factor of Staphylococcus epidermidis. A recent study has shown that norspermidine inhibits and disassembles the biofilm in the wild-type Bacillus subtilis NCBI3610 strain. In this study, the effect of norspermidine on S. epidermidis biofilm formation of clinical or commensal strains was tested. Biofilm producing strains of S. epidermidis were isolated from healthy skin (HS; n = 3), healthy conjunctiva (HC; n = 9) and ocular infection (OI; n = 19). All strains were treated with different concentrations of norspermidine, spe

Clinical significance of the isolation of Staphylococcus epidermidis from bone biopsy in diabetic foot osteomyelitis.

INTRODUCTION: Coagulase-negative staphylococci are considered as microorganisms with little virulence and usually as contaminants. In order to establish the role of Staphylococcus epidermidis as a pathogen in diabetic foot osteomyelitis, in addition to the isolation of the sole bacterium from the bone it will be necessary to demonstrate the histopathological changes caused by the infection. METHODS: A consecutive series of 222 diabetic patients with foot osteomyelitis treated surgically in the Diabetic Foot Unit at La Paloma Hospital (Las Palmas de Gran Canaria, Canary Islands, Spain) between

Anaerobic co-reduction of chromate and nitrate by bacterial cultures of Staphylococcus epidermidis L-02.

Industrial wastewater is often polluted by Cr(VI) compounds, presenting a serious environmental problem. This study addresses the removal of toxic, mutagenic Cr(VI) by means of microbial reduction to Cr(III), which can then be precipitated as oxides or hydroxides and extracted from the aquatic system. A strain of Staphylococcus epidermidis L-02 was isolated from a bacterial consortium used for the remediation of a chromate-contaminated constructed wetland system. This strain reduced Cr(VI) by using pyruvate as an electron donor under anaerobic conditions. The aims of the present study were to

Use of ribotyping to investigate tracheal colonisation by Staphylococcus epidermidis as a source of bacteremia in ventilated newborns.

Ribotyping was used to determine whether a relationship exists between endotracheal tube colonisation with Staphylococcus epidermidis and bacteremia with this organism. Over a three-week period, four mechanically ventilated preterm babies presented with five episodes of infection and bacteremia. For each blood specimen obtained for culture a tracheal aspirate sample was collected at the same time by suctioning. After DNA extraction and cleavage by EcoRI, hybridisation was performed with a digoxigenin-labelled 16S-rDNA probe from Escherichia coli. Five different band patterns were recognised on

Formation of HCN by human phagocytosing neutrophils--1. Chlorination of Staphylococcus epidermidis as a source of HCN.

Phagocytosis of Staphylococcus epidermidis by human neutrophils is accompanied by HCN liberation. The amount of HCN evolved is significantly higher when the bacteria are damaged by penicillin. One of the substrates yielding HCN during phagocytosis are N-dichloroglycyl residues of bacterial peptidoglycan formed by the chlorinating myeloperoxidase-H2O2-Cl- system of neutrophils. HCN deriving from the bacterial structures constitutes 6-12% of total HCN liberated during phagocytosis, which indicates that there are other substrates for HCN production. Chlorination process within phagocytosing neutr

Relationship between resistance to metallic ions and production of beta-lactamase in strains of Staphylococcus epidermidis.

Thirty five strains of Staphylococcus epidermidis were tested for resistance to penicillin G, erythomycin, metallic ions Zn, As3, As5, Cd, Hg, Pb and activity of beta-lactamase. These studies have shown that the majority of tested staphylococci were resistant to penicillin G, erythromycin, and produced beta-lactamase. No correlation between the activity of beta-lactamase and the resistance to metallic ions has been shown.

A Molecularly Imprinted Polymer Nanobodies (nanoMIPs)-Based Electrochemical Sensor for the Detection of Staphylococcus epidermidis.

Methicillin-resistant Staphylococcus epidermidis (MRSE) contamination is commonly found on human skin and medical devices. Herein, we present a sensor utilizing molecularly imprinted polymer nanobodies (nanoMIP) for recognition and electrochemical impedance spectroscopy (EIS) to detect S. epidermidis. Sensor manufacturing involves synthesizing nanoMIP via solid-phase synthesis using whole bacteria as templates. Screen-printed gold electrode (AuSPE)-modified 16-mercaptohexadecanoic acid (MHDA) served to immobilize the nanoMIPs on the sensor surface through an amide bond, with the remaining func

Genomic characterization of Staphylococcus epidermidis Se252 isolated from the rhizosphere of a Brazilian endemic plant.

BACKGROUND: Staphylococcus epidermidis (Se) is commonly regarded as a commensal organism; however, under specific conditions, it may act as an opportunistic pathogen. Here, we report the whole-genome sequencing and comparative genomic analysis of Se strain 252 (Se252), isolated from the rhizosphere of an endemic Brazilian plant. RESULTS: Se252 exhibits a unique repertoire of genes associated with environmental adaptation and virulence. These include two putative Type VII secretion system (T7SS) effectors and thirteen proteins involved in adhesion, toxin production, and immune evasion—among the

Flying under the radar: vancomycin heteroresistance in Staphylococcus epidermidis peritonitis is associated with treatment failure.

BACKGROUND: Antimicrobial resistance (AMR) is a global One Health problem and a growing threat to human and animal health. Antimicrobial susceptibility tests (AST) such as the broth-microdilution method (BMD) guide appropriate antimicrobial therapy but may fail to detect various forms of AMR, such as heteroresistance. We report vancomycin treatment failure in a Staphylococcus epidermidis peritoneal dialysis (PD) associated peritonitis case with vancomycin heteroresistance undetected by conventional AST and whole-genome sequencing. CASE SUMMARY: A patient with cloudy PD effluent presented to h

Biofilm and the effect of sonication in a chronic Staphylococcus epidermidis orthopedic in vivo implant infection model.

BACKGROUND: In diagnosing chronic orthopedic implant infections culture of sonicate represents a supplement to tissue cultures. However, the extent to which biofilm forms on implant surfaces and the degree of dislodgement of bacteria by sonication remains unclear. In this in vivo study using a low bacterial inoculum, we aimed to determine whether a variable effect of sonication could be observed in a standardized in vivo model. MATERIALS AND METHODS: Seven Wistar rats underwent surgery with intramuscular implantation of two bone xenograft implants, each containing two steel plates. The grafts

Staphylococcus epidermidis bloodstream infections are a cause of septic shock in intensive care unit patients.

OBJECTIVES: Staphylococcus epidermidis (SE) is a supposedly low-virulence agent, which may cause proven bloodstream infections (BSIs), with little-known consequences on intensive care unit (ICU) patients. We aimed at studying ICU patients diagnosed with BSIs caused by SE (SE-BSIs). METHODS: We constituted a retrospective cohort in two medical ICUs. SE-BSIs were defined by two or more independent SE-positive blood cultures of the same strain, within 48 hours, without concurrent infection. RESULTS: We included 59 patients; 58% were men (n = 34), with median age of 67 (interquartile range 60-74

Staphylococcus epidermidis protease EcpA can be a deleterious component of the skin microbiome in atopic dermatitis.

BACKGROUND: Staphylococcus aureus and Staphylococcus epidermidis are the most abundant bacteria found on the skin of patients with atopic dermatitis (AD). S aureus is known to exacerbate AD, whereas S epidermidis has been considered a beneficial commensal organism. OBJECTIVE: In this study, we hypothesized that S epidermidis could promote skin damage in AD by the production of a protease that damages the epidermal barrier. METHODS: The protease activity of S epidermidis isolates was compared with that of other staphylococcal species. The capacity of S epidermidis to degrade the barrier and i

Staphylococcus epidermidis Contributes to Healthy Maturation of the Nasal Microbiome by Stimulating Antimicrobial Peptide Production.

The composition of the human microbiome profoundly impacts human well-being. However, the mechanisms underlying microbiome maturation are poorly understood. The nasal microbiome is of particular importance as a source of many respiratory infections. Here, we performed a large sequencing and culture-based analysis of the human nasal microbiota from different age groups. We observed a significant decline of pathogenic bacteria before adulthood, with an increase of the commensal Staphylococcus epidermidis. In seniors, this effect was partially reversed. In vitro, many S. epidermidis isolates stim

In-host evolution of Staphylococcus epidermidis in a pacemaker-associated endocarditis resulting in increased antibiotic tolerance.

Treatment failure in biofilm-associated bacterial infections is an important healthcare issue. In vitro studies and mouse models suggest that bacteria enter a slow-growing/non-growing state that results in transient tolerance to antibiotics in the absence of a specific resistance mechanism. However, little clinical confirmation of antibiotic tolerant bacteria in patients exists. In this study we investigate a Staphylococcus epidermidis pacemaker-associated endocarditis, in a patient who developed a break-through bacteremia despite taking antibiotics to which the S. epidermidis isolate is fully

A case of blastomycosis-like pyoderma caused by mixed infection of Staphylococcus epidermidis and Trichophyton rubrum.

Blastomycosis-like pyoderma (BLP) is a type of chronic pyoderma characterized histologically by specific epidermal changes namely: pseudoepitheliomatous hyperplasia and intraepithelial abscesses. These epidermal changes are also seen in blastomycosis (referred to as deep dermatophytosis in North America). Here, we describe the case of a 53-year-old male with prurigo nodularis, diabetes, and chronic lymphocytic leukemia who presented with multiple yellowish-red colored papules that coalesced to form a vegetating plaque. In addition to the typical features of BLP, spores with budding were seen h

Drug susceptibility and clonality of methicillin-resistant Staphylococcus epidermidis in hospitalized patients with hematological malignancies.

PURPOSE: To evaluate the clonal relatedness and drug susceptibility of Streptococcus epidermidis isolated from hematological patients. METHODS: All S. epidermidis isolated from hematological patients who developed bloodstream infections between June 2005 and December 2007 were included. The clonal relationship was tested by means of pulsed-field gel electrophoresis (PFGE) analysis. RESULTS: Fifteen methicillin-resistant S. epidermidis (MRSE) isolates were examined from patients' blood culture samples. Two subgroups that differed approximately by 40% in their PFGE banding were identified. In

[Genetic transfer of methycilline resistance in Staphylococus epidermidis and Staphylococcus aureus strains in mixed cultures].

In mixed cultures of staphylococci a transfer of the resistance to methicillin and penicillinase plasmids as well as tetracycline and chloramphenicol plasmids was investigated. It was shown that the resistance to methicillin was transferred in mixed cultures from one strain of S. aureus to another and from S. epidermidis to S. aureus. In both cases transfer of methicillin resistance required, the presence of penicillinase plasmid in recipient or donor strain. In the case of other markers transmission was independent. Moreover it was shown that the transfer of resistance genes in mixed cultures

Changing resistance to antimicrobial drugs, and resistance typing in clinically significant strains of Staphylococcus epidermidis.

Resistance to 11 antimicrobial drugs was assessed in 532 clinically significant strains of Staphylococcus epidermidis received in the years 1978, 1979 and 1980 and compared with that of strains collected in 1976 and 1977 for an international study. Only 14% of strains were sensitive to all the drugs tested. Resistance to gentamicin increased significantly from 7% to 33% during the study period. The degree of association between pairs of drugs was assessed. There was strong association between resistance to methicillin, aminoglycosides and macrolide antibiotics and only weak association between
