NAD+ and its derivatives, which act as redox coenzymes, are crucial for cellular metabolism and energy production. Nevertheless, the processes by which Streptococcus mutans, a bacterium known for causing dental caries, synthesizes NAD+ are not well elucidated. Through a genome-wide screen, we identified the nicotinic acid salvage pathway and the evolutionarily incomplete PnuC-NadR pathway involved in NAD+ biosynthesis in S. mutans UA159. The nicotinic acid pathway is regulated by SmNiaR, a nicotinic acid-responsive transcription regulator featuring an N-terminal DNA-binding winged helix-turn-h

Streptococcus mutans, a Gram-positive, facultative anaerobic bacterium, is a key contributor to dental caries. It forms biofilms to colonize the tooth surface, has stress resistance mechanisms to survive the fluctuating oral environment, and produces virulence factors, all of which cause enamel demineralization, acid production, and caries development. Traditional antimicrobial strategies target central metabolic pathways in S. mutans, but most enzymes are conserved between humans and bacteria. However, one prospective approach is to target methionine biosynthesis. This pathway is essential fo

Fructose catabolism by Streptococcus mutans is initiated by three PTS transporters yielding either fructose-1-phoshate (F-1-P) or fructose-6-phosphate (F-6-P). Deletion of one such F-1-P-generating PTS, fruI, has been shown to reduce the cariogenicity of S. mutans in rats fed a high-sucrose diet. Moreover, a recent study linked fructose metabolism in S. mutans to a reactive electrophile species (RES) methylglyoxal. Here, we conducted a comparative transcriptomic analysis of exponentially grown S. mutans shocked with 50 mM fructose, 50 mM glucose, 5 mM methylglyoxal, or 0.5 mM hydrogen peroxide

Dental caries is a chronic infectious disease that occurs in the hard tissue of teeth under the influence of multiple factors, among which bacteria being a key factor. Streptococcus mutans ( S. mutans) is considered a major pathogen that causes caries. Secondary metabolites, including bacteriocins and polyketides/non-ribosomal peptides, are a class of small-molecule compounds synthesized by S. mutans. To date, polyketides/non-ribosomal peptides identified in S. mutans include mutanobactin, mutanocyclin, and mutanofactin, which are synthesized by the mub, muc, and muf biosynthetic gene clusters

The commensal pathogen Streptococcus mutans uses AgI/II adhesins to adhere to gp340 adsorbed on teeth. Here we analyzed isolates of S. mutans (n = 70 isolates) from caries and caries-free human extremes (n = 19 subjects) by multilocus sequence typing (MLST), AgI/II full-length gene sequencing, and adhesion to parotid saliva matched from the strain donors (nested from a case-control sample of defined gp340 and acidic proline-rich protein [PRP] profiles). The concatenated MLST as well as AgI/II gene sequences showed unique sequence types between, and identical types within, the subjects. The mat

Adhesion of bacteria to pellicle-coated tooth surfaces is the first step in biofilm formation on teeth. The aim of this study was to explore whether human milk prevents or promotes adhesion of cariogenic Streptococcus mutans to saliva-coated hydroxyapatite (HA) using an in vitro model system. S. mutans binding to HA coated with human parotid saliva (s-HA) or human milk was studied, in addition to binding inhibition to s-HA by human milk. S. mutans did not bind to HA coated with milk. S. mutans binding to s-HA was inhibited by milk from 15 (71 %) of 21 women, whereas milk from the remaining 6 m

The oral microbe Streptococcus mutans uses adaptive mechanisms to withstand the fluctuating pH levels in its natural environment. The regulation of protein synthesis is part of the mechanism of acid adaptation and tolerance in S. mutans. Here, we demonstrate that the organism's acid-inducible protein repertoire includes an AP endonuclease activity. This abasic site-specific endonuclease activity is present at greater levels in cells grown at low pH than in cells grown at pH 7, and is apparently independent of the RecA protein. Experiments using tetrahydrofuran or alpha-deoxyadenosine-containin

Forty-five subjects contributing 150 root surfaces with gingival recession were sampled seven times over a 32-month period. We calculated isolation frequencies of lactobacilli on selective Rogosa SL agar and S. mutans in a sensitive enrichment broth and on mitis salivarius agar. Both S. mutans and lactobacilli were isolated more frequently from surfaces which became carious than from those remaining caries-free. Isolation frequencies were also higher for caries-free surfaces in caries-active subjects than for caries-free surfaces in caries-inactive subjects. The presence or absence of S. mutan

The success of oral vaccines based on Strep. mutans might be influenced by its presence in the gastrointestinal (GI) tract. Enumeration of Strep. mutans at various sites of the GI tract of sucrose-fed monkeys revealed a localized distribution. High numbers of Strep. mutans were recovered from only the dental plaque, caecum and the large intestine. Where Strep. mutans was not detectable in the dental plaque it could not be isolated from regions of the GI tract sampled. The absence of Strep. mutans from the small intestine suggests that the disparate results obtained from experiments in which th

Sodium fluoride caused inhibition of growth rate and growth levels of Streptococcus mutans with glucose as the primary energy and carbon source. Stannous fluoride increased growth lag nad caused a much greater inhibition of growth rate than did sodium fluoride. Neither compound was found to be bactericidal when culture viability was measured after 6 days of incubation. Leuconostoc mesenteroides, which lacks a phosphotransferase system for sugar transport, showed less inhibition of growth rate with both inhibitors than did S. mutans, which possesses a phosphotransferase system. Shikonin is extracted from the roots of Lithospermum erythrorhizon, and shikonin extracts have been shown to have inhibitory effects on several bacteria. However, shikonin extracts are difficult to formulate because of their poor water solubility. In the present study, we prepared a shikonin dispersion, which was solubilized by the inclusion of β-1,3-1,6 glucan, and analysed the inhibitory effects of this dispersion on Streptococcus mutans and non-mutans streptococci. The shikonin dispersion showed pronounced anti-S. mutans activity, and inhibited growth of and biofilm formation by this bacter

BACKGROUND: Combining antimicrobial photodynamic therapy (aPDT) with phytochemicals may enhance antibiofilm activity while reducing reliance on higher photosensitizer concentrations. This study evaluated toluidine blue O (TBO)-mediated aPDT and the adjunctive effect of naringenin (NAR) against Streptococcus mutans biofilms.
MATERIALS AND METHODS: Minimum inhibitory concentrations (MICs) of TBO and NAR against S. mutans were determined. Checkerboard assays assessed drug interaction using the fractional inhibitory concentration index (FICI). Mature S. mutans biofilms were allocated to eight gro

Streptococcus mutans strains expressing the collagen-binding protein Cnm are strongly associated with severe and recurrent dental caries, yet the mechanistic basis for their clinical enrichment alongside Candida albicans remains unclear. Here, we investigated whether Cnm contributes to early cross-kingdom interactions that promote dual-species biofilm development. Using purified proteins, we found that C. albicans binds robustly to Cnm at levels comparable to those of glucosyltransferase B (GtfB) and higher than those of other S. mutans adhesins tested. Preincubation of collagen with Cnm inhib
