In plant immunity, pathogen-activated intracellular nucleotide binding/leucine rich repeat (NLR) receptors mobilize disease resistance pathways, but the downstream signaling mechanisms remain obscure. Enhanced disease susceptibility 1 (EDS1) controls transcriptional reprogramming in resistance triggered by Toll-Interleukin1-Receptor domain (TIR)-family NLRs (TNLs). Transcriptional induction of the salicylic acid (SA) hormone defense sector provides one crucial barrier against biotrophic pathogens. Here, we present genetic and molecular evidence that in Arabidopsis an EDS1 complex with its part

BACKGROUND: Addressing global food security under rapid population growth and climate change requires sustainable strategies to protect crops from pathogens while reducing reliance on agrochemicals. Salicylic acid (SA), a key phytohormone in plant defense, is a promising natural elicitor for crop protection. However, its rapid metabolism and phytotoxicity restrict practical use. To overcome these limitations, we developed SA-amino acid conjugates designed to improve protective efficacy of SA while minimizing its toxicity.
RESULTS: Six SA-amino acid conjugates were synthesized through optimize

BACKGROUND: The plant immune response to DNA is highly self/nonself-specific. Self-DNA triggered stronger responses by early immune signals such as H2O2 formation than nonself-DNA from closely related plant species. Plants lack known DNA receptors. Therefore, we aimed to investigate whether a differential sensing of self-versus nonself DNA fragments as damage- versus pathogen-associated molecular patterns (DAMPs/PAMPs) or an activation of the DNA-damage response (DDR) represents the more promising framework to understand this phenomenon.
RESULTS: We treated Arabidopsis thaliana Col-0 plants w

BACKGROUND AND AIMS: We have recently shown that an Arabidopsis thaliana double mutant of type III phosphatidylinositol-4-kinases (PI4Ks), pi4kβ1β2, constitutively accumulated a high level of salicylic acid (SA). By crossing this pi4kβ1β2 double mutant with mutants impaired in SA synthesis (such as sid2 impaired in isochorismate synthase) or transduction, we demonstrated that the high SA level was responsible for the dwarfism phenotype of the double mutant. Here we aimed to distinguish between the SA-dependent and SA-independent effects triggered by the deficiency in PI4Kβ1 and PI4Kβ2.

UNLABELLED: Bdellovibrio and like organisms (BALOs) predate on Gram-negative bacteria selectively, which holds promise as an alternate to combat against multidrug-resistant bacterial pathogens. In this study, two Bdellovibrio strains designated MPR17 and MPR18 were isolated from water samples by double-layer agar technique using P. fluorescens as prey and investigated for their predatory potentials against six pathogenic Gram-negative, MDR and XDR bacteria. Both the BALOs preyed on all the tested bacteria including P. syringae, B. glumae, V. parahaemolyticus, P. mirabilis, M. bovis, and A. sch

C(2)H(4) is associated with plant defense, but its role during the hypersensitive response (HR) remains largely uncharacterized. C(2)H(4) production in tobacco (Nicotiana tabacum) following inoculation with HR-eliciting Pseudomonas syringae pathovars measured by laser photoacoustic detection was biphasic. A first transient rise (C(2)H(4)-I) occurred 1 to 4 h following inoculation with HR-eliciting, disease-forming, and nonpathogenic strains and also with flagellin (flg22). A second (avirulence-dependent) rise, at approximately 6 h (C(2)H(4)-II), was only seen with HR-eliciting strains. Tobacco

Upon localized attack by necrotizing pathogens, plants gradually develop increased resistance against subsequent infections at the whole-plant level, a phenomenon known as systemic acquired resistance (SAR). To identify genes involved in the establishment of SAR, we pursued a strategy that combined gene expression information from microarray data with pathological characterization of selected Arabidopsis (Arabidopsis thaliana) T-DNA insertion lines. A gene that is up-regulated in Arabidopsis leaves inoculated with avirulent or virulent strains of the bacterial pathogen Pseudomonas syringae pv

Histidine kinases, used for environmental sensing by bacterial two-component systems, are involved in regulation of bacterial gene expression, chemotaxis, phototaxis, and virulence. Flavin-containing domains function as light-sensory modules in plant and algal phototropins and in fungal blue-light receptors. We have discovered that the prokaryotes Brucella melitensis, Brucella abortus, Erythrobacter litoralis, and Pseudomonas syringae contain light-activated histidine kinases that bind a flavin chromophore and undergo photochemistry indicative of cysteinyl-flavin adduct formation. Infection of

An Arabidopsis thaliana mutant, esa1, that shows enhanced susceptibility to the necrotrophic pathogens Alternaria brassicicola, Botrytis cinerea and Plectosphaerella cucumerina, but has wild-type levels of resistance to the biotrophic pathogens Pseudomonas syringae pv. tomato and Peronospora parasitica. The enhanced susceptibility towards necrotrophic pathogens correlated with a delayed induction of phytoalexin accumulation and delayed induction of the plant defensin gene PDF1.2 upon inoculation with pathogens. Antibody activity to cotton dust bacteria in the nasal secretions of cotton-mill cardroom workers was studied using the ELISA technique. Secretion samples were collected by inserting a roll of filter paper into the nasal cavities of the subjects. The amount of secretion was significantly less among cotton-mill workers than workers in a wood factory. No significant differences were seen between byssinotic and nonbyssinotic cotton-mill workers. No significant differences were observed between cotton and wood workers regarding antibody activity against Pseudomonas syringae and Enterobacter agglom

Outbreaks of plant diseases pose a severe threat to crop production and global food security. The extensive and prolonged use of conventional chemical bactericides has exacerbated pathogen resistance, creating an urgent demand for novel antibacterial agents with high biosafety profiles. This study screened a series of cyclic terpenoids against three phytopathogenic bacteria: Xanthomonas oryzae pv. oryzae (Xoo), Xanthomonas axonopodis pv. citri (Xac), and Pseudomonas syringae pv. actinidiae (Psa), with Nopol showing the strongest inhibitory activity against Xoo (EC50 = 8.86 mg·L-1). Tabtoxin resistance protein (TTR) is an enzyme that renders tabtoxin-producing pathogens, such as Pseudomonas syringae, tolerant to their own phytotoxins. Here, we report the crystal structure of TTR complexed with its natural cofactor, acetyl coenzyme A (AcCoA), to 1.55A resolution. The binary complex forms a characteristic "V" shape for substrate binding and contains the four motifs conserved in the GCN5-related N-acetyltransferase (GNAT) superfamily, which also includes the histone acetyltransferases (HATs). A single-step mechanism is proposed to explain the function of three conserved resi

BACKGROUND: Plant bacterial diseases have severely affected the yield and quality of agricultural products, among which kiwifruit canker and bacterial wilt disease, caused by Pseudomonas syringae pv. actinidiae (Psa) and Ralstonia solanacearum (Rs), respectively, seriously restrict the development of the related industry. However, there are few effective green agents available to target these diseases.
RESULTS: Herein, we synthesized 41 novel derivatives of the natural product tryptanthrin, among which partial compounds showed noticeable antibacterial activity against Psa and Rs. Compounds T7

Host susceptibility to pathogens can be shaped by genetic, ecological, and evolutionary factors. The ability to predict the spread of disease therefore requires an integrated understanding of these factors, including effects of pests on pathogen growth and competition between pathogens and commensal microbiota for host resources. We examined interactions between the leaf-mining moth Cameraria ohridella, the bacterial causal agent of bleeding canker disease Pseudomonas syringae pv aesculi, and the bark-associated microbiota of horse chestnut (Aesculus hippocastanum) trees. BACKGROUND: Autoimmune hepatitis (AIH) is a chronic, progressive inflammatory liver disease mediated by autoimmune mechanisms. 3-O-Acetyloleanolic acid (OAA), a triterpenoid isolated from the stems of syringa oblata Lindl. has demonstrated therapeutic potential for liver and immune-related disorders. The farnesoid X receptor (FXR) plays a critical role in regulating bile acid metabolism within the enterohepatic circulation and modulates metabolic dysregulation and immune homeostasis across multiple tissues.
PURPOSE: This study investigated the protective effect of OAA against concanavalin A (

Lycotoxin I, from the wolf spider (Lycosa carolinensis), is an amphipathic pore-forming peptide that has antimicrobial and anti-insect activity. Constitutive expression of a lycotoxin I modified for oral toxicity to insects in tobacco (Nicotiana tabacum) conferred significantly enhanced resistance to larvae of the corn earworm (Helicoverpa zea) and cigarette beetle (Lasioderma serricorne). Gene expression levels of modified lycotoxin I were negatively correlated to the survival of corn earworm larvae. In addition, pathogenic symptoms caused by Pseudomonas syringae pathovar tabaci and Alternari

Plant WRKY transcription factors act as either positive or negative regulators of plant basal disease resistance. To comprehensively characterise the complicated functional network, we isolated OsWRKY77 from rice seedlings treated with salicylic acid. OsWRKY77 is a typical WRKY transcription factor, based on in its protein structure analysis, nuclear localisation of the fused OsWRKY77-GFP protein and gel electrophoretic mobility shift assay binding, which demonstrated that OsWRKY77 was able to bind a W-box. Transgenic Arabidopsis lines overexpressing OsWRKY77 repressed growth of Pseudomonas sy

Plants possess inducible systemic defense responses when locally infected by pathogens. Bacterial infection results in the increased accumulation of the mobile metabolite azelaic acid, a nine-carbon dicarboxylic acid, in the vascular sap of Arabidopsis that confers local and systemic resistance against the pathogen Pseudomonas syringae. Azelaic acid primes plants to accumulate salicylic acid (SA), a known defense signal, upon infection. Mutation of the AZELAIC ACID INDUCED 1 (AZI1) gene, which is induced by azelaic acid, results in the specific loss of systemic immunity triggered by pathogen o

Plants often respond to attack by insect herbivores and necrotrophic pathogens with induction of jasmonate-dependent resistance traits, but respond to attack by biotrophic pathogens with induction of salicylate-dependent resistance traits. To assess the degree to which the jasmonate- and salicylate-dependent pathways interact, we compared pathogenesis-related protein activity and bacterial performance in four mutant Arabidopsis thaliana lines relative to their wild-type backgrounds. We found that two salicylate-dependent pathway mutants (cep1, nim1-1) exhibited strong effects on the growth of
