Impacts of huanglongbing on fruit yield and quality and on flushing dynamics of Sicilian lemon trees.

INTRODUCTION: The severe Asian form of huanglongbing (HLB), a vascular disease associated with the phloem-limited bacterium 'Candidatus Liberibacter asiaticus', is transmitted by the Asian citrus psyllid (ACP) Diaphorina citri. Disease impacts are known for sweet oranges and acid limes but not lemons. METHODS: In a five-year study (2017-2021) we compared yield and fruit quality between naturally-infected and healthy 5-yr-old trees of Sicilian lemon 'Femminello', and shoot phenology on both lemon and 'Valencia' orange, both grafted onto 'Swingle' citrumelo, grown in southeastern São Paulo Stat

Nano mercury selenide as a source of mercury for rice.

Mercury (Hg) is a persistent and toxic metal while mercury selenide (HgSe) is generally considered as the environmental sink of Hg in its biogeochemical cycle. Recent studies found nano-sized HgSe (nano-HgSe) could be transformed by certain bacteria. This raises safety concerns about the application of selenium (Se) to curb Hg contamination in farmlands. Therefore, hydroponic experiments were performed in which rice plants were cultured with different concentrations of nano-HgSe and micro-sized HgSe (micro-HgSe) to explore their bioavailability and toxicity. It was found that both nano-HgSe an

Improved node culture methods for rapid vegetative propagation of switchgrass (Panicum virgatum L.).

BACKGROUND: Switchgrass (Panicum virgatum L.) is an important bioenergy and forage crop. The outcrossing nature of switchgrass makes it infeasible to maintain a genotype through sexual propagation. Current asexual propagation protocols in switchgrass have various limitations. An easy and highly-efficient vegetative propagation method is needed to propagate large natural collections of switchgrass genotypes for genome-wide association studies (GWAS). RESULTS: Micropropagation by node culture was found to be a rapid method for vegetative propagation of switchgrass. Bacterial and fungal contamin

Insights Into Manganese Solubilizing Bacillus spp. for Improving Plant Growth and Manganese Uptake in Maize.

Manganese (Mn) is an essential micronutrient for plant growth that is involved in the structure of photosynthetic proteins and enzymes. Mn deficiency is widespread mainly in dry, calcareous, and sandy soil, which leads to a significant decrease in crop yield. Mn-reducing bacteria promote the solubilization of Mn minerals, thus increasing Mn availability in soil. The present study aimed to assess the Mn solubilizing ability and plant growth-promoting potential of Bacillus spp. strains for maize plants with insoluble Mn compounds. Several Mn-solubilizing bacterial (MSB) strains were isolated fro

Methylmercury production in a paddy soil and its uptake by rice plants as affected by different geochemical mercury pools.

The formation of neurotoxic methylmercury (MeHg) in paddy fields and its accumulation by rice plants is of high environmental concern. The contribution of different geochemical mercury (Hg) pools in paddy soils to MeHg production and its accumulation by rice seedlings is not well-studied up to now. Therefore, we investigated the impact of different inorganic Hg forms, including HgCl2, nano-particulated HgS (nano-HgS), Hg bound with dissolved organic matter (Hg-DOM), β-HgS, and α-HgS, at levels of 5 mg Hg/kg soil and 50 mg Hg/kg soil, on the production of MeHg in the soil during rice growing se

COP9 signalosome subunit 5A affects phenylpropanoid metabolism, trichome formation and transcription of key genes of a regulatory tri-protein complex in Arabidopsis.

BACKGROUND: Trichomes and phenylpropanoid-derived phenolics are structural and chemical protection against many adverse conditions. Their production is regulated by a network that includes a TTG1/bHLH/MYB tri-protein complex in Arabidopsis. CSN5a, encoding COP9 signalosome subunit 5a, has also been implicated in trichome and anthocyanin production; however, the regulatory roles of CSN5a in the processes through interaction with the tri-protein complex has yet to be investigated. RESULTS: In this study, a new csn5a mutant, sk372, was recovered based on its altered morphological and chemical ph

A comparative transcriptomic analysis reveals the core genetic components of salt and osmotic stress responses in Braya humilis.

Braya humilis is a member of the Euclidieae tribe within the family Brassicaceae. This species exhibits a broad range of adaptations to different climatic zones and latitudes as it has a distribution that ranges from northern Asia to the arctic-alpine regions of northern North America. In China, B. humilis is mainly found on the Qinghai-Tibetan Plateau (QTP) and in adjacent arid regions. In this study, we sequenced a sample from an arid region adjacent to the QTP using the Illumina platform generating a total of 46,485 highly accurate unigenes, of which 78.41% were annotated by BLASTing versus

Proteomics analysis reveals novel host molecular mechanisms associated with thermotherapy of 'Ca. Liberibacter asiaticus'-infected citrus plants.

BACKGROUND: Citrus Huanglongbing (HLB), which is linked to the bacterial pathogen 'Ca. Liberibacter asiaticus' (Las), is the most devastating disease of citrus plants, and longer-term control measures via breeding or genetic engineering have been unwieldy because all cultivated citrus species are susceptible to the disease. However, the degree of susceptibility varies among citrus species, which has prompted efforts to identify potential Las resistance/tolerance-related genes in citrus plants for application in breeding or genetic engineering programs. Plant exposure to one form of stress has

Photoreceptor Specificity in the Light-Induced and COP1-Mediated Rapid Degradation of the Repressor of Photomorphogenesis SPA2 in Arabidopsis.

The Arabidopsis COP1/SPA E3 ubiquitin ligase is a key negative regulator that represses light signaling in darkness by targeting transcription factors involved in the light response for degradation. The COP1/SPA complex consists of COP1 and members of the four-member SPA protein family (SPA1-SPA4). Genetic analysis indicated that COP1/SPA2 function is particularly strongly repressed by light when compared to complexes carrying the other three SPAs, thereby promoting a light response after exposure of plants to extremely low light. Here, we show that the SPA2 protein is degraded within 5-15 min

Genetic and physiological determinants of Streptomyces scabies pathogenicity.

UNLABELLED: SUMMARY Common scab is a severe disease worldwide affecting tap root crops and potato tubers. It is caused by soil-borne filamentous bacteria belonging to the genus Streptomyces. Streptomycetes usually are saprophytic microorganisms, but a few species have acquired the ability to infect underground plant tissues. The predominant causal agent of potato scab worldwide is Streptomyces scabies. The production of phytotoxins called thaxtomins is essential for the virulence of common scab-causing agents. The genes involved in the biosynthetic pathway of thaxtomins and other virulence gen

In situ modification of herbivore-induced plant odors: a novel approach to study the attractiveness of volatile organic compounds to parasitic wasps.

Many parasitic wasps (parasitoids) exploit volatile organic compounds (VOCs) emitted by herbivore-infested plants in order to locate their hosts, but it remains largely unknown which specific compounds within the volatile blends elicit the attractiveness to parasitoids. One way of studying the importance of specific VOCs is to test the attractiveness of odor blends from which certain compounds have been emitted. We used this approach by testing the attraction of naive and experienced females of the two parasitoids Cotesia marginiventris and Microplitis rufiventris to partially altered volatile

Strategies for development of functionally equivalent promoters with minimum sequence homology for transgene expression in plants: cis-elements in a novel DNA context versus domain swapping.

The cauliflower mosaic virus 35S (35S) promoter has been extensively used for the constitutive expression of transgenes in dicotyledonous plants. The repetitive use of the same promoter is known to induce transgene inactivation due to promoter homology. As a way to circumvent this problem, we tested two different strategies for the development of synthetic promoters that are functionally equivalent but have a minimum sequence homology. Such promoters can be generated by (a) introducing known cis-elements in a novel or synthetic stretch of DNA or (b) "domain swapping," wherein domains of one pr

Enhancing chlorpyrifos degradation and plant growth through biochar-assisted microbial remediation.

The extensive use of chlorpyrifos (CPF) in agriculture has led to persistent soil contamination, posing serious risks to environmental and human health. Developing sustainable, in situ remediation strategies is therefore a priority. This study evaluated an integrated plant-microbe-biochar system for the remediation of CPF-contaminated soils. Indigenous CPF-degrading bacteria were isolated from agricultural soils and screened for degradation efficiency in mineral salts medium (MSM) and soil microcosms. The most efficient isolate, designated C78, was identified by 16S rRNA gene sequencing as Kle

The ADF4-RCAR12 module regulates ABA-mediated drought tolerance in Arabidopsis.

INTRODUCTION: Drought is one of the most severe abiotic stresses limiting crop productivity, and abscisic acid (ABA)-induced stomatal closure is a key mechanism underlying plant drought resistance. Although ABA signaling has been extensively characterized in Arabidopsis thaliana, the role and regulatory mechanisms of microfilaments (F-actin) in this process remain poorly understood. Previously, we identified the actin-binding protein CAP1 as an interactor of the ABA receptor RCAR12 and a phosphorylation target of OPEN STOMATA 1 (OST1), suggesting a potential link between actin dynamics and ABA

Assessing phenotypic diversity and trait relationships within (Syzygium cumini (L.) Skeels) using morpho-biochemical traits.

BACKGROUND: Jamun (Syzygium cumini (L.) Skeels) is a multipurpose fruit tree with notable nutritional, medicinal, and ecological potential. Uttar Pradesh, a major jamun growing state in India, harbours considerable variability in jamun, which remains underexplored. METHODS: To assess phenotypic variability and identify superior types, 123 genotypes of jamun were evaluated using morphological descriptors. Biochemical attributes were also assessed for a subset of 15 genotypes from the full panel. RESULTS: Significant variation was observed for most qualitative descriptors, with clear differenc

NtFATB silencing impairs lipid homeostasis and enhances cold sensitivity associated with reduced the ICE-CBF-COR expression in Nicotiana tabacum.

Fatty acyl-acyl carrier protein thioesterases B (FATB) plays a critical role in regulating fatty acid chain length and saturation in plants. However, the function of FATB genes in tobacco remains poorly understood. In this study, we identified and characterized NtFATB, a FATB homolog in tobacco. Phylogenetic analysis revealed that NtFATB is closely related to FATB proteins from Nicotiana tomentosiformis and several Solanaceae species. Sequence alignment and domain prediction identified conserved catalytic residues and two characteristic acyl-ACP thioesterase domains. Expression analysis showed

Coumarin application modulates antioxidant defense to enhance resistance against Fusarium in mung bean.

Plant-derived bioactive compounds contribute a major role in plant defense against fungal pathogens. Among these, coumarin proved to be a major controller in root-pathogen interactions, exhibiting diverse protective mechanisms. This study investigates the effects of coumarin (50 mg.L- 1), applied through seed priming and foliar spray, on the biochemical responses of Mung bean and its impact on root colonization by Fusarium solani. Mung bean seeds pre-treated with coumarin were sown under both infected and non-infected conditions. Once the seedling established up to two-leaf stage, weekly folia

Conservation and divergence of abiotic stress-responsive gene co-expression networks in diploid and tetraploid cotton.

Cotton (Gossypium spp.) is highly susceptible to abiotic stresses, including temperature extremes, drought, and salinity. The diploid species Gossypium arboreum retains stress adaptive traits that are poorly represented in modern tetraploid cultivars, yet the evolutionary and regulatory bases of these ploidy associated differences remain insufficiently understood. Here, we performed a comparative transcriptomic analysis of 54 seedling leaf RNA-seq libraries from Gossypium arboreum, Gossypium hirsutum, and Gossypium barbadense subjected to four abiotic stress treatments. By integrating standard

A multifunctional sodium alginate/chitosan composite hydrogel for saline-alkali soil remediation: Synthesis, characterization, and proof-of-concept for promoting crop growth.

Soil salinization threatens agriculture by restricting water uptake, inducing ionic stress, and destabilizing soil structure. We developed a vapor-phase acidification method to prepare a sodium alginate/chitosan (SA-CTS) dual-network hydrogel incorporating biochar (BC) and palygorskite (Pal) as functional fillers. The BC/Pal/SA-CTS composite absorbed 66.08 g/g in deionized water and 36.37 g/g in 0.9% NaCl, retaining approximately 55% of its DI-water swelling capacity under saline conditions. Component-resolved assays conducted at an equal total amendment rate suggested that the SA-CTS hydrogel
