Drinking amid abundant illicit drugs. The Vietnam case.

We studied drinking patterns and problems of 451 US Army enlisted men after their return from Vietnam. Before Vietnam, nearly half were regular drinkers and one quarter had drinking problems. Problem drinking declined in Vietnam as opiate use rose sharply; half had tried opiates and 20% were opiate dependent. After Vietnam, opiate use decreased (now less than 2% opiate dependent) and problem drinking again became ascendant. At time of interview, 17% had drunk in an alcoholic pattern at some period in their lives and another 41% had had problems from drinking. The younger men were more likely t

Hydroxycinnamic Acid Amides Emerge as Multifunctional Molecules Involved in Regeneration and Volatile Signalling During Wound Responses.

Hydroxycinnamic acid amides (HCAAs) are phenylpropanoid-derived metabolites traditionally associated with antimicrobial defence and cell wall reinforcement during plant-pathogen interactions. However, their contribution to plant responses to mechanical injury, particularly in wound healing and damage-induced signalling, remains poorly understood. In this study, we investigated the wound-induced responses in tomato (Solanum lycopersicum) using a 35S::THT system in which tyramine hydroxycinnamoyl transferase (THT), a key enzyme controlling HCAAs biosynthesis, is overexpressed. By integrating met

Rhodanine and 2,4-dioxothiazolidine-based amides: synthesis and evaluation as dual-action cholinesterase inhibitors and antioxidants.

This study presents the synthesis and biological evaluation of substituted 2-(2,4-dioxothiazolidin-3-yl)-N-phenylacetamides and their thioxo analogues as potential multitarget agents for neurodegenerative diseases. A diverse library of amides was designed to explore structure-activity relationships and synthesized using two complementary approaches: a carbodiimide-mediated coupling and a microwave-assisted procedure. The compounds were screened for acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibition and antioxidant activity. Several derivatives displayed micromolar dual inh

Direct Asymmetric Synthesis of Chiral β-Hydroxy-α-Amino Amides via PLP-Inspired Biomimetic Aldol Reaction.

Chiral β-hydroxy-α-amino amides represent important structural motifs ubiquitous in pharmaceuticals and bioactive molecules. Current synthetic approaches typically rely on multistep conversion from the corresponding β-hydroxy-α-amino acids, resulting in poor step-economy and potential α-racemization during amidation. However, direct asymmetric synthesis of these compounds via aldol reactions remains highly challenging because of the low reactivity of glycinamides and the instability of the corresponding aldol products. Herein, we report a chiral pyridoxal-catalyzed biomimetic aldol reaction of

Isolation and Characterization of Glycosylated Fatty Acid Amides from the Norwegian Deep-Sea Sponge Phakellia sp.

Marine organisms from deep-sea environments have attracted considerable attention in drug discovery because they produce structurally diverse natural products. However, Norwegian deep-sea ecosystems remain largely unexplored in terms of natural product chemistry and bioactivity. In this study, a deep-sea sponge belonging to the genus Phakellia was selectively collected using a low-impact, remotely operated vehicle approach from the Mohn's Treasure area in the Norwegian Sea at a depth of 2858 m, representing one of the deepest sponge samples investigated in Norwegian waters to date. Development of Aryl Cyano Amides for Fluorescence Discrimination of Tau and Amyloid Beta Deposits in Tissue.

The accumulation of amyloid aggregates such as amyloid beta (Aβ) and tau deposits in the brain is a hallmark of many neurodegenerative diseases including Alzheimer's disease. Recent advances in optical imaging have shown that fluorescent probes can detect amyloids in living patients, potentially aiding in diagnosis. Here, we investigate structural modifications of amyloid-targeting Aryl Cyano Amide-based fluorophores aimed at enhancing their ability to discriminate between different amyloid aggregates, such as Aβ and tau. We identify two structural parameters that synergistically enable increa

Zirconium-Catalyzed Reductive Sulfonamidation of Amides and Its Application to Site-Selective N‑Alkylation of Pharmaceuticals.

The direct catalytic reductive amination of amides remains a challenging transformation, particularly when using weakly nucleophilic amines. However, this strategy offers several synthetic advantages, particularly due to the wide availability of amides, as well as N-sulfonyl and N-sulfinyl amines. We present a mild catalytic approach for the monoalkylation of sulfonamides, sulfamates, sulfamides, and sulfinamides using amides. This protocol exploits the oxophilic character of zirconium, an early, earth-abundant metal, to drive dual reductive cycles and enable the critical amination step. This

Additive-Controlled Divergent Synthesis of Amides and Ketones via Ni/Photoredox-Catalyzed Deoxygenative Functionalization of Alcohols.

Amides and ketones are essential carbonyl compounds with widespread applications in pharmaceuticals, materials science, and synthetic chemistry. Herein, we present a Ni/photoredox dual-catalyzed strategy for the divergent synthesis of amides and ketones from isocyanates and alcohols. This transformation is facilitated by N-heterocyclic carbene (NHC)-mediated activation of alcohols and is selectively controlled by the choice of additive: NaOAc promotes amide formation, whereas nBu4NPO4H2 directs the reaction toward ketone synthesis. This approach offers a versatile and practical route to access

Nα-Aroyl-N-Aryl-Phenylalanine Amides: A Promising Class of Antimycobacterial Agents Targeting the RNA Polymerase.

Tuberculosis (TB), caused by Mycobacterium tuberculosis, remains the leading cause of death from a bacterium in the world. The global prevalence of clinically relevant infections with opportunistically pathogenic non-tuberculous mycobacteria (NTM) has also been on the rise. Pharmacological treatment of both TB and NTM infections usually requires prolonged regimens of drug combinations, and is often challenging because of developed or inherent resistance to common antibiotic drugs. Medicinal chemistry efforts are thus needed to improve treatment options and therapeutic outcomes. Nα-aroyl-N-aryl

Analysis of structure-activity relationship of indol-3-yl-N-phenylcarbamic amides as potent STING inhibitors.

A structure-activity relationship (SAR) study of stimulator of interferon gene (STING) inhibition was performed using a series of indol-3-yl-N-phenylcarbamic amides and indol-2-yl-N-phenylcarbamic amides. Among these analogs, compounds 10, 13, 15, 19, and 21 inhibited the phosphorylation of STING and interferon regulatory factor 3 (IRF3) to a greater extent than the reference compound, H-151. All five analogs showed stronger STING inhibition than H-151 on the 2',3'-cyclic GMP-AMP-induced expression of interferon regulatory factors (IRFs) in a STINGR232 knock-in THP-1 reporter cell line. Cobalt-Catalyzed Aerobic Aminocyclization of Unsaturated Amides for the Synthesis of Functionalized γ- and δ-Lactams.

We report the cobalt-catalyzed aminocyclization of unsaturated N-acyl sulfonamides in the presence of oxygen to provide γ- and δ-lactam aldehydes. Use of an optically active cobalt catalyst resulted in the formation of enantiomerically enriched γ-and δ-lactam alcohols. The γ-lactam aldehydes and alcohols obtained were elaborated into useful building blocks.

Photochemical Deracemization of Primary Allene Amides by Triplet Energy Transfer: A Combined Synthetic and Theoretical Study.

The photochemical deracemization of 2,4-disubstituted 2,3-butadienamides (allene amides) was investigated both experimentally and theoretically. The reaction was catalyzed by a thioxanthone which is covalently linked to a chiral 1,5,7-trimethyl-3-azabicyclo[3.3.1]nonan-2-one skeleton providing a U-shaped arrangement of the sensitizing unit relative to a potential hydrogen-bonding site. Upon irradiation at λ = 420 nm in the presence of the sensitizer (2.5 mol %), the amides reached at -10 °C a photostationary state in which one enantiomer prevailed. The enantioenriched allene amides (70-93% ee)

Radical-Mediated Activation of Esters with a Copper/Selectfluor System: Synthesis of Bulky Amides and Peptides.

Herein, we describe a new approach for the activation of esters via a radical-mediated process enabled by a copper/Selectfluor system. A variety of para-methoxybenzyl esters derived from bulky carboxylic acids and amino acids can be easily converted into the corresponding acyl fluorides, directly used in the one-pot synthesis of amides and peptides. As a proof of concept, this method was applied to the iterative formation of sterically hindered amide bonds.

Copper-catalyzed aerobic oxidative C-C bond cleavage of simple ketones for the synthesis of amides.

A Cu-catalyzed oxidative amidation of simple ketones with amines via carbon-carbon (C-C) bond cleavage has been developed. A number of aryl and alkyl ketones could be easily converted to amides using cheap copper salt as the catalyst and O2 as the oxidant with a wide range of amines, including primary and secondary amines. This method shows a notable advantage of the broad scope for the substrate, thus providing a practical approach to amides. A plausible mechanism is proposed based on the preliminary experiments.

Fluorine walk: The impact of fluorine in quinolone amides on their activity against African sleeping sickness.

Human African Trypanosomiasis, also known as African sleeping sickness, is caused by the parasitic protozoa of the genus Trypanosoma. If there is no pharmacological intervention, the parasites can cross the blood-brain barrier (BBB), inevitably leading to death of the patients. Previous investigation identified the quinolone amide GHQ168 as a promising lead compound having a nanomolar activity against T. b. brucei. Here, the role of a fluorine substitution at different positions was investigated in regard to toxicity, pharmacokinetics, and antitrypanosomal activity. This 'fluorine walk' led to
