BACKGROUND: Retinal ischemia/reperfusion (RIR) is implicated in various forms of optic neuropathies, yet effective treatments are lacking. RIR leads to the death of retinal ganglion cells (RGCs) and subsequent vision loss, posing detrimental effects on both physical and mental health. Apigenin (API), derived from a wide range of sources, has been reported to exert protective effects against ischemia/reperfusion injuries in various organs, such as the brain, kidney, myocardium, and liver. In this study, we investigated the protective effect of API and its underlying mechanisms on RGC degenerati

Noise exposure is one of the most common causes of sensorineural hearing loss. Although many studies considered inflammation to be a major contributor to noise-induced hearing loss, the process of cochlear inflammation is still unclear. Studies have found that activation of the NF-κB signaling pathway results in the accumulation of macrophages in the inner ear plays an important role in hair cell damage. In this study, tandem mass tag (TMT) technique was used to analyze the changes in basilar membrane proteome expression before and after acoustic injury. After noise exposure, the nicotinamide

Neuroinflammation plays a vital role in Alzheimer's disease (AD) pathogenesis and other neurodegenerative disorders (NDs). Presently, only symptomatic treatments are available and no disease-modifying drugs are available for AD and other NDs. Thus, targeting AD-associated neuroinflammation with anti-inflammatory compounds and antioxidants has recently been given much focus. Now, flavonoids are being increasingly investigated as therapeutic agents to treat inflammation; apigenin has a neuroprotective effect. Iron dyshomeostasis plays a key role in sustaining the neuroinflammatory phenotype, hig

Apigenin, a flavonoid found in many plants, has various biological properties. We aimed to investigate the anti-inflammatory and anti-oxidative activity of apigenin against carbon tetrachloride (CCl4)-induced acute liver injury in mice and hydrogen peroxide (H2O2)-induced oxidative stress in HepG2 cells and possible mechanism. In vivo, apigenin significantly reduced alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activity in serum of mice challenged by CCl4 and markedly alleviated the lipid peroxidation as indicated by the increased level of superoxide dismutase (SOD), redu

Tumor microenvironments, particularly hypoxia and inflammation, heavily influence colorectal cancer (CRC) pathogenesis by altering polyamine metabolism. Identifying natural compounds targeting these vulnerabilities remains critical. Integrating untargeted metabolomics, network pharmacology, and a human endogenous metabolite library screen, we identified apigenin (API) as a potent anti-CRC candidate. API significantly inhibited the proliferation, migration, and invasion of RKO and HCT116 cells in vitro and suppressed xenograft tumor growth in vivo. Crucially, high-throughput screening revealed

Sonodynamic therapy (SDT), characterized by its non-invasiveness and high tissue penetration depth, has emerged as a promising novel antitumor treatment modality. Nevertheless, the development of efficient sonosensitizers for SDT still poses a challenge. In this study, a covalent organic framework based on zinc phthalocyanines (ZnPc-COF) was synthesized. It was discovered that ZnPc-COF exhibited significantly enhanced sonodynamic activity compared to free ZnPc. This study focuses on the adverse impacts of highly expressed HIF-1α at tumor sites on tumor treatment. Therefore, by leveraging the p

Diabetic nephropathy (DN) is the most common microvascular complication of diabetes and leading cause of end-stage renal disease (ESRD) requiring dialysis. Mesangial cell proliferation induced by hyperglycemia is a hallmark of DN. Apigenin, a compound abundantly present in fruits and vegetables, exhibit anti-inflammatory and antioxidant properties and have shown potential in alleviating DN. In this study, for the first time, the effect of Apigenin on mesangial cell proliferation in an experimental DN model induced by Streptozotosin (STZ) injection was evaluated through Gata3, Pdgfr-β and Nf-κb

Apigenin, identified as 4', 5, 7-trihydroxyflavone, is a natural flavonoid compound that has many interesting pharmacological activities and nutraceutical potential including anti-inflammatory and antioxidant functions. Chronic, low-grade inflammation and oxidative stress are involved in both the initiation and progression of hypertension and hypertension-induced cardiac hypertrophy. However, whether or not apigenin improves hypertension and cardiac hypertrophy through modulating NADPH oxidase-dependent reactive oxygen species (ROS) generation and inflammation in hypothalamic paraventricular n

The coronavirus disease (COVID-19) endemic has urged an intensive search for effective treatments to fight the infections. The lack of effective medication against SARS-CoV-2 infections has led to alternative treatment approaches, with medicinal plants as promising candidates in the ongoing development of anti-SARS-CoV-2 therapeutics. In this present study, apigenin-6-C-β-glucoside-4'-O-α-apiofuranoside (AGA), a phytochemical compound found in Christia vespertilionis methanolic leaves extract (MeCV) was screened for anti-SARS-CoV-2 via in silico molecular docking approach on angiotensin-conver

Polycystic Ovarian Syndrome (PCOS) is a complex endocrine and metabolic disorder affecting women of reproductive age, characterized by hyperandrogenism, insulin resistance, chronic inflammation, and ovulatory dysfunction. Conventional therapies, such as oral contraceptives, insulin sensitizers, and anti-androgens, primarily offer symptomatic relief and are often associated with chronic adverse effects, underscoring the need for safer and more holistic alternatives. Naturally occurring bioactives have emerged as promising adjunct or alternative therapeutic agents in this context. This review cr

Apigenin is a phytochemical obtained from Chamomilla recutita. Its role in interstitial cystitis is not yet known. The present study is aimed at understanding the uroprotective and spasmolytic effects of apigenin in cyclophosphamide-induced interstitial cystitis. The uroprotective role of apigenin was analyzed by qRT-PCR, macroscopic analysis, Evans blue dye leakage, histological evaluation, and molecular docking. The spasmolytic response was measured by adding cumulative concentrations of apigenin to isolated bladder tissue pre-contracted with KCl (80 mM) and carbachol (10-9-10-4) on non-incu

INTRODUCTION: Lupus patients need alternatives to steroids and cytotoxic drugs. We recently found that apigenin, a non-mutagenic dietary flavonoid, can sensitize recurrently activated, normal human T cells to apoptosis by inhibiting nuclear factor-kappa-B (NF-kappaB)-regulated Bcl-xL, cyclooxygenase 2 (COX-2), and cellular FLICE-like inhibitory protein (c-FLIP) expression. Because sustained immune activation and hyperexpression of COX-2 and c-FLIP contribute to lupus, we treated SNF1 mice that spontaneously develop human lupus-like disease with apigenin.
METHODS: SNF1 mice with established lu

Triple-negative breast cancer (TNBC) is aggressive with limited treatments. Although the natural flavonoid Apigenin (API) shows anti-tumor potential, its mechanism in TNBC remains unclear. This study investigated API's role in inducing ferroptosis and inhibiting glycolysis to suppress TNBC. Molecular docking predicted API's binding to ferroptosis- and glycolysis-related proteins. In vitro, 4T1 and MDA-MB-231 TNBC cells were used to assess API's effects on viability, migration, invasion, and key metabolic markers (Fe2+, MDA, ROS, GSH, lactic acid, glucose, ATP, OCR, ECAR), and protein expressio

Obesity constitutes a growing global health concern, affecting individuals during the aging process. Flavonoids that increase tissue NAD+ levels, such as apigenin (Api), are proposed for the treatment of obesity and age-related diseases. Thus, this study aimed to provide initial preclinical evidence of Api in this setting using middle-aged rats induced to obesity by a high-calorie diet (HCD). Forty-seven 15-month-old male Wistar rats were assigned to five groups: standard diet (SD) or HCD, each with (Api at 50 mg/kg) or without treatment (vehicle), plus vehicle control group. Obesity induction

BACKGROUND: In Sjögren's syndrome (SS)-an autoimmune disease characterized by dry mouth and eyes-salivary gland epithelial cells (SGECs) undergo ferroptosis, which disrupts their integrity and impairs saliva secretion. Apigenin, a phytoestrogen, is known to activate estrogen signalling and alleviate xerostomia in ovariectomized mice; however, its effect on SGEC survival and function in SS remains unclear. We hypothesized that apigenin alleviates SS symptoms and progression by inhibiting ferroptosis in SGECs and aimed to elucidate the underlying mechanism.
METHODS: Apigenin (50 mg/kg) was oral

Vision impairment in retinal degenerative diseases such as age-related macular degeneration is primarily associated with photoreceptor degeneration, in which oxidative stress and inflammatory responses are mechanistically involved as central players. Therapies with photoreceptor protective properties remain to be developed. Apigenin-7-diglucuronide (A7DG), a flavonoid glycoside, is present in an assortment of medicinal plants with anti-inflammatory or ant-oxidant activities. However, the pharmacological significance of A7DG remains unknown in vivo. The current study isolated A7DG from Glechoma

A tenet of contemporary obstetrics is that a significant proportion of preterm births involve bacterial infection. Bacterial endotoxin induces pro-inflammatory cytokines, prostaglandins and proteases via the pro-inflammatory pathway nuclear factor-κB (NF-κB), which plays a key role in initiating uterine contractions and rupture of foetal membranes. In non-gestational tissues, the phytophenols curcumin, naringenin and apigenin exert anti-inflammatory properties via inhibition of NF-κB. The aim of this study was to determine whether these treatments regulate pro-inflammatory and pro-labour media
