t-[35S]butylbicyclophosphorothionate binding sites in invertebrate tissues.

Specific high affinity binding of the cage convulsant t-[35S]butylbicyclophosphorothionate (TBPS) was observed in membrane homogenates of housefly heads and crayfish abdominal muscles. [35S]TBPS binding in these two invertebrate tissues was inhibited by biologically active cage convulsants, picrotoxin analogs, and barbiturates. The housefly binding sites were inhibited most potently by several insecticides. Approximately 50% of total binding was displaceable by excess (0.1 mM) nonradioactive TBPS, picrotoxinin, ethyl bicyclophosphate, or dieldrin. Optimal binding assay conditions for housefly

Whole-Genome Resequencing Analysis Reveals Insights into Sex Determination and Gene Loci Associated with Sex Differences in Procambarus clarkii.

Since the molecular mechanisms underlying sex determination in Procambarus clarkii are still unclear, it is important to investigate the genetic basis of sex determination in crustaceans. Currently, the molecular mechanisms of sex determination and the gender-specific markers in this species remain poorly understood. In this study, a total of 14,046,984 SNPs and 2,160,652 InDels were identified through genome-wide resequencing of 89 individuals (45 females and 44 males). Further analysis confirmed that the candidate chromosome was Chr38, the sex determination system was identified as XY, and t

RNA-seq and 16S rRNA analysis revealed the effects of Agaricus bisporus polysaccharides on Aeromonas salmonicida-induced enteritis in crayfish.

To investigate the effect of Agaricus bisporus polysaccharides (ABPs) on Aeromonas salmonicida (A. salmonicida)-induced enteritis in crayfish, this study first established an enteritis model in crayfish via A. salmonicida infection. Subsequently, ABPs at a concentration of 0.5 mg/mL were administered to the enteritis-affected crayfish via anal perfusion at a dosage of 100 μL per individual. The intestinal tissues of the treated crayfish were collected to perform transcriptome analysis and intestinal microbiota composition analysis.Transcriptomic analysis of the crayfish intestines revealed tha

Paragonimus kellicotti flukes in Missouri, USA.

Paragonimiasis is an infection caused by lung flukes of the genus Paragonimus. In Asia, P. westermani infections are relatively common because of dietary practices. However, in North America, cases of paragonimiasis, which are caused by P. kellicotti flukes, are rare. Only 7 autochthonous cases of paragonimiasis were reported during 1968-2008. In 2009, we reported 3 new case-patients with paragonimiasis who had been seen at our medical center over an 18-month period. Six additional case-patients were identified in St. Louis, Missouri, USA, and treated at Washington University-affiliated health

Vibrio mimicus infection associated with crayfish consumption, Spokane, Washington, 2010.

We report a cluster of severe diarrheal disease caused by Vibrio mimicus infection among four persons who had consumed leftover crayfish the day after a private crayfish boil. Gastrointestinal illness caused by Vibrio mimicus has not been reported previously in Washington State. Three cases were laboratory confirmed by stool culture; using PCR, isolates were found to have ctx genes that encode cholera toxin (CT). Two of the cases were hospitalized under intensive care with a cholera-like illness. The illnesses were most likely caused by cross-contamination of cooked crayfish with uncooked cray

Methylmercury levels and bioaccumulation in the aquatic food web of a highly mercury-contaminated reservoir.

The low Ebro River basin (NE Spain) represents a particular case of chronic and long-term mercury pollution due to the presence of an industrial waste (up to 436 μg/g of Hg) coming from a chlor-alkali plant Albeit high total mercury (THg) levels have been previously described in several aquatic species from the surveyed area, methylmercury (MeHg) values in fish individuals have never been reported. Accordingly, in order to investigate bioaccumulation patterns at different levels of the aquatic food web of such polluted area, crayfish and various fish species, were analysed for THg and MeHg con

Impact of a toxic and a non-toxic strain of Microcystis aeruginosa on the crayfish Procambarus clarkii.

The occurrence of cyanobacteria in many water bodies where crayfish such as Procambarus clarkii are abundant leads to the possibility of toxin accumulation and food chain transfer. This paper describes the accumulation and depuration of microcystins from a microcystin and a non-microcystin producing strain of Microcystis aeruginosa, on the survivorship, growth and nutritional status of P. clarkii. Crayfish larvae were resistant to cyanobacteria and their toxins, surviving cyanobacteria densities during acute exposures. Juvenile crayfish tolerated toxic cyanobacteria better than non-toxic ones.

Effects of the anesthetics heptanol, halothane and isoflurane on gap junction conductance in crayfish septate axons: a calcium- and hydrogen-independent phenomenon potentiated by caffeine and theophylline, and inhibited by 4-aminopyridine.

This study has monitored junctional and nonjunctional resistance, [Ca2+]i and [H+]i, and the effects of various drugs in crayfish septate axons exposed to neutral anesthetics. The uncoupling efficiency of heptanol and halothane is significantly potentiated by caffeine and theophylline. The modest uncoupling effects of isoflurane, described here for the first time, are also enhanced by caffeine. Heptanol causes a decrease in [Ca2+]i and [H+]i both in the presence and absence of either caffeine or theophylline. A similar but transient effect on [Ca2+]i is observed with halothane. 4-Aminopyridine

Studies of axon-glial cell interactions and periaxonal K+ homeostasis--III. The effect of anisosmotic media and potassium on the relationship between the resistance in series with the axon membrane and glial cell volume.

The effect of anisosmotic physiological solutions and [K+]o on the resistance in series with the axon membrane were studied in medial giant axons of the crayfish, Procambarus clarkii, to determine if changes in series resistance are correlated with changes in glial cell volume and volume regulatory responses. Series resistance was estimated from computer analysed voltage waveforms generated by constant current and space clamp techniques using piggy-back axial wire current passing and glass pipette recording electrodes. Axons subjected to anisosmotic physiological solution in the range of 23 to

Ionic mechanism of the salicylate block of nerve conduction.

Benzoic acid, salicylic acid and 5-bromosalicylic acid have been studied for their mechanism of action on crayfish and squid giant axons. External application of 0.1 to 0.5 mM 5-bromosalicylic acid prolonged the falling phase of the action potential, and at concentrations higher than 1.5 mM, nervous conduction was blocked. Boltage clamp analyses showed that 5-bromosalicylic acid reduced both peak transient sodium conductance and steady-state potassium conductance. The curves relating the peak and steady-state conductances to membrane potential were shifted in the direction of depolarization. T

Tannic acid-stained microtubules with 12, 13, and 15 protofilaments.

Subunit structure in the walls of sectioned microtubules was first noted by Ledbetter and Porter (6), who clearly showed that certain microtubules of plant meristematic cells have 13 wall protofilaments when seen in cross section. Earlier, protofilaments of microtubular elements had been described in negatively stained material, although exact counts of their number were difficult to obtain. In microtubular elements of axonemes, some success has been achieved in visualizing protofilaments in conventionally fixed and sectioned material (8, 10); much less success has been achieved in identifying

WSSV promotes its replication by inhibiting Dorsal activation via CypA in crayfish.

As a key therapeutic target for various diseases, Cyclophilin A (CypA) exhibits a dual nature in its biological functions. While the molecular mechanisms underlying its role in antiviral immunity have been well elucidated in vertebrates, research in invertebrates has merely been limited to expression responses induced by pathogens, and the underlying mechanisms through which CypA mediates immune regulation remain to be urgently deciphered. In this study, CypA expression was up-regulated in red swamp crayfish (Procambarus clarkii) following infection with white spot syndrome virus (WSSV). Funct

Detection of genomic DNA of the crayfish plague fungus Aphanomyces astaci (Oomycete) in clinical samples by PCR.

A diagnostic procedure, based on a polymerase chain reaction method (PCR) was developed to detect infection of crayfish with the Oomycete Aphanomyces astaci. A set of oligonucleotide primers was designed to specifically amplify A. astaci DNA in the ITS region surrounding the 5.8S rDNA gene. The PCR amplifies a 115bp amplicon. The specificity of the primers was demonstrated by testing on 27 A. astaci strains and against 20 non-A. astaci Oomycetes and 5 fungal species. Most of the non-A. astaci Oomycete or fungal species included in the study are either known parasites of freshwater crayfish cut

Characterization and classification of five cysteine proteinases expressed by Paragonimus westermani adult worm.

Three new members of the cysteine proteinase gene family of Paragonimus westermani have been isolated and classified. Comparisons of the predicted amino acid sequences of PwCP2 (U69121), PwCP4 (U56958), and PwCP5 (U33215) were performed with those of the previously reported PwCP1 (U69120) and PwCP3 (U56865) sequence. The amino acid alignment showed conservation of the cysteine, histidine, and asparagine residue that form the catalytic triad. With 57 cysteine proteinases including PwCP1-5, we conducted phylogenetic analysis using neighbor joining method (NJ). A resultant unrooted tree revealed

Impact of the crayfish Procambarus clarkii on Schistosoma haematobium transmission in Kenya.

The Louisiana red swamp crayfish, Procambarus clarkii, which was introduced into east Africa in the 1950s or 1960s, has since widely dispersed. Previous work by our group has shown that P. clarkii can reduce populations of the molluscan intermediate hosts of human schistosomes through predatory and competitive interactions. Here, we investigate whether crayfish can reduce populations of Bulinus africanus and consequently, Schistosoma haematobium prevalence in school children. Children from 6 primary schools in the Machakos and Kitui Districts of Kenya were selected for study. Schools were divi

Two types of identified ascending interneurons with distinct GABA receptors in the crayfish terminal abdominal ganglion.

More than half of the identified ascending interneurons originating in the terminal abdominal ganglion of the crayfish received inhibitory sensory inputs from hair afferents innervating the tailfan on the side contralateral to their main branches. Biochemical aspects of this transverse lateral inhibition of ascending interneurons were examined by the use of neurophysiological and pharmacological techniques. Local application of gamma-aminobutyric acid (GABA) and its agonist muscimol into the neuropil induced membrane hyperpolarization of identified ascending interneurons with an increase in me

Repetitive axonal discharges elicited by serotonin and intracellular adenosine 3',5'-cyclic monophosphate in the crayfish neuromuscular junction.

Experiments were carried out in the opener muscle of the claw of small crayfish. After pretreatment of the preparation with serotonin (5-HT), application of the membrane permeant analogue of adenosine 3',5'-cyclic monophosphate (cAMP), 8-bromoadenosine 3',5'-monophosphate was capable of evoking reversibly repetitive discharges in the inhibitory and excitatory axon. Reducing phosphodiesterase activity with application of either 3-isobutyl-1-methylxanthine or theophylline also elicited repetitive axonal discharges after 5-HT treatment. Moreover, application of forskolin dissolved in ethanol caus

Purification from black widow spider venom of a protein factor causing the depletion of synaptic vesicles at neuromuscular junctions.

The aqueous extract of the venom glands of black widow spiders was fractionated on a column of Sephadex G-200 and then on a column of DEAE-Sephadex A-50 pH 8.2. A protein fraction was obtained that caused a great increase in the frequency of occurrence of miniature end plate potentials at the frog neuromuscular junction, and caused swelling of the nerve terminals and depleted them of their vesicles. The fraction consists of a least four protein components that are similar in their molecular weights (about 130,000) and isoelectric points (ranging from pH 5.2 to 5.5) and are immunologically indi

The impact of ultra-high pressure processing on crayfish tropomyosin allergenicity and microstructure preservation: Insights into texture and flavors.

This study investigated the effectiveness of ultra-high pressure (UHP) processing in reducing the allergenicity of crayfish tropomyosin (TM). TM content significantly decreased after UHP treatment (from 151.72 to 94.15 μg/mL with increasing UHP strength; from 37.00 to 18.52 μg/mL with prolonged UHP duration). Additionally, conformational properties were altered. In a BALB/c mouse model, allergic sensitization was effectively reduced by UHP treatment, as indicated by decreased IgE (from 8.36 to 4.33 μg/mL), IgG1 (from 214.02 to 84.41 μg/mL), histamine (from 10.21 to 6.01 ng/mL), mMCP-1 (from 16
