Characteristics of cell-membrane and extracellular proteoglycans in monkey submandibular gland.

We investigated the characteristics of cell-membrane and extracellular proteoglycans in the monkey submandibular gland using enzymatic digestion and Western blot analysis. Extracellular proteoglycans and cell-membrane proteoglycans were extracted from the phosphate-buffered saline (PBS) soluble fraction and microsomal fraction of submandibular glands, respectively, and purified partially by ion exchange chromatography. Cellulose acetate membrane electrophoresis and immunoblotting with 3G10 monoclonal antibody or 6B6 monoclonal antibody showed that the extracellular proteoglycans contain severa

Functional associations of CD38 with CD3 on the T-cell membrane.

CD38 is a multifunctional membrane surface glycoprotein expressed by different cells and tissues, including T cells at certain stages of their development. Besides its involvement in transmembrane signaling, CD38 play a role in cell adhesion processes. Structurally, membrane CD38 was reported as presenting lateral associations with molecules involved in recognition and signaling, namely with the TCR/CD3 complex in T cells. Here we report that ligation of CD38 by agonistic and non-agonistic monoclonal antibodies exerts different effects on T cells, the former inducing down-modulation of the ass

Modulation of glomus cell membrane currents of intact rat carotid body.

1. In order to understand better the relationship between sinus nerve chemoreceptor activity and changes in glomus cell membrane current, both were measured simultaneously in rat carotid bodies in vitro. Mean membrane resistance of intact glomus cells was 1327 +/- 140 M omega (n = 104, mean +/- S.E.M.) and membrane capacitance was 7.9 +/- 0.8 pF (n = 28). 2. Over the course of 15 min following the start of whole-cell recording, outward current increased by 169 +/- 48% (n = 19), but there was no significant change in holding current or membrane resistance. Reversal potential of the tail current

Anionic mechanisms of zinc uptake across the human red cell membrane.

1. Zinc is taken up into human red cells by two mechanisms that depend upon the presence of anions. One of these requires bicarbonate ions, is inhibited by 4,4'-diisothiocyanatostilbene-2,2'-disulphonic acid (DIDS) and appears to be catalysed by the anion exchanger. The second occurs in the presence of thiocyanate or salicylate ions and may represent transport of a neutral complex with Zn2+. 2. The initial rate of Zn2+ uptake via the anion exchanger is 64 +/- 13 mumol (10(13) cells x h)-1 microM-1 external Zn2+, in the presence of 5 mM-bicarbonate at pH 7.4 and 37 degrees C (+/- S.D.). This is

Destruction of basement membrane and cell infiltrates in oral lichen planus.

Immunohistochemical staining analysis of oral lichen planus (OLP) using monoclonal antibodies (MoAbs) against the components of basement membrane (BM) and infiltrated cells was performed in 27 cases. Integrity of lining epithelial BM was more strictly elucidated by staining for laminin and type IV collagen than by fibronectin. Staining patterns of laminin were similar to type IV collagen. In the cases examined, continuity of BM was proven by laminin staining in 10 cases, partially destructive BM was seen in 9 cases and complete destruction of BM in 8 cases. Neural cell recognition molecule F11: membrane interaction by covalently attached phosphatidylinositol.

The mode of membrane insertion of F11 130 kDa protein, a neural chick cell surface glycoprotein involved in neurite fasciculation, has been investigated. Up to 41% of total F11 130 kDa is released from adult chick brain plasma membranes by phosphatidylinositol specific phospholipase C (PI-PLC), whereas no release is mediated by lecithin/cephalin specific phospholipase C (PLC). PI-PLC dependent release of F11 is also observed from embryonal chick brain plasma membranes and from the surface of intact retinal cells. Biosynthetic labelling experiments demonstrate that F11 contains ethanolamine. Ta

Movement of acid equivalents across the mammalian smooth muscle cell membrane.

Factors affecting intracellular pH (pHi) in the smooth muscle of guinea pig ureter have been investigated using pH-sensitive microelectrodes. Associated acids and bases appear to have free passage across the cell membrane but results suggest very low permeability to charged acid equivalents, thus implicating carrier-mediated movements in many of the observed pHi transients. Recovery from acidosis in the nominal absence of CO2 was inhibited by removal of Na+ and by the presence of amiloride, indicating that Na+/H+ exchange was responsible. The presence of CO2 resulted in a faster recovery from

[Immunodiagnosis of kidney tubular cell injuries using specific anti-membrane antibodies].

In contrast to healthy persons, microvillous antigens of the proximal tubule were excreted at an increased rate in patients with kidney diseases as could be shown using specific antisera against brush border (BB) fragments (tissue-proteinuria, histuria). These urinary membrane components were immunologically completely identical with those antigens prepared from isolated kidney cell membranes. A glycoprotein of 240 000 dalton, containing mannose and N-acetylglucosamine was identified as a major immunoreactive constituent of the brush border surface and found to be part of a multienzyme complex

Spectrin oligomers of the red cell membrane extracts in hereditary spherocytosis.

Spectrin oligomers were extracted from the erythrocyte membrane at 37 degrees C with the low ionic strength buffer. The spectrin extractability of HS membranes was enhanced compared with controls in some cases. The major spectrin component in the extract was dimer which was, apart from a tetramer, purified by gel filtration on the Sepharose 4B column. No changes of dimer-tetramer interconversion in the extract from HS membranes were observed. Similarly, the protein composition of the membrane extracts from two types of red cells remained unchanged.

Changes in membrane lipids of the beta-cell during insulin secretion.

The effect of insulin secretion on the turnover of phospholipids in the pancreatic beta-cell was determined by following the fate, during secretion, of phospholipids prelabelled with 3H-glycerol. The secretion of insulin in response to glucose (20mM) plus isobutyl-methylxanthine (0.5mM) was associated with a calcium dependent increase in the turnover of the major classes of phospholipids and an increase production of 1:2 diacylglycerol and triacylglycerol. In addition, changes in the fatty acid composition of the beta-cell, during insulin secretion, were determined. There was an increase in th

Plasma membrane damage by environmental materials enhances cell-cell fusion and impairs defense functions of macrophages.

INTRODUCTION: Many environmental materials can activate macrophages and trigger inflammatory responses. However, whether and how these exposures alter macrophage function in host defense remains elusive. METHODS: Bone marrow-derived macrophages were treated with various environmental materials, including laundry detergent, and their membrane damage was measured by Annexin V staining. Alterations in macrophage function were assessed by cell-cell fusion, in vitro phagocytosis, and in vivo clearance of Candida albicans. RESULTS AND DISCUSSION: Many environmental hazards damage the plasma membra

Immune cell-derived membrane nanovesicles: A promethean fire for autoimmune disease therapy through immune cell mimicry.

Autoimmune diseases (AIDs) constitute a heterogeneous group of disorders characterized by immune dysregulation, loss of self-tolerance, and chronic inflammation, which leads to tissue damage and organ dysfunction. Current therapies for AIDs are often limited by their lack of specificity, systemic side effects, and insufficient restoration of immune tolerance. Recent advances in nanotechnology and bioengineering have introduced immune and associated cell-derived membrane vesicles (IACMVs) as a promising therapeutic platform. Derived from macrophages, dendritic cells, neutrophils, platelets, or

Plasma Membrane Damage by Environmental Materials Enhances Cell-Cell Fusion and Impairs Immune Functions of Macrophages.

Macrophages are the most abundant phagocytes and play an essential role in host defense. Previous studies have shown that many environmental materials can activate macrophages and trigger inflammatory responses. However, whether these exposures alter macrophage function in host defense remains unclear. This study found that many environmental materials (such as carbon nanotubes, tungsten carbide [WC], and detergents) can damage the plasma membranes of macrophages. This damage leads to decreased reactive oxygen species (ROS) production and phagocytosis but elevated cell-cell fusion. In vivo, ai

Long-Term Cell-Membrane-Coated Ultrabright Nanospheres for Targeted Cancer Cell Imaging and Hydrophobic Drug Delivery.

Nanoparticle-based imaging agents have gained massive attention for the targeted cancer cell imaging of early stage disease diagnosis. Among these, organic dye-entrapped and -assembled nanoparticles have been considered as potential imaging agents. However, they are limited by poor brightness, low stability, low reproducibility, and selective surface engineering, which limit their translational potential. The molecular assembly of amphiphilic precursors and the chosen organic fluorophore can augment the brightness and stability of the engineered nanoimaging agents. Herein, we describe cancer c

Cell-Impermeable Buffering Fluorogenic Probes for Live-Cell Super-Resolution Imaging of Plasma Membrane Morphology Dynamics.

Super-resolution fluorescence imaging has emerged as a potent tool for investigating the nanoscale structure and function of the plasma membrane (PM). Nevertheless, the challenge persists in achieving super-resolution imaging of PM dynamics due to limitations in probe photostability and issues with cell internalization staining. Herein, we report assembly-mediated buffering fluorogenic probes BMP-14 and BMP-16 exhibiting fast PM labeling and extended retention time (over 2 h) on PM. The incorporation of alkyl chains proves effective in promoting the aggregation of BMP-14 and BMP-16 into nonflu

Red Blood Cell Membrane Cholesterol May Be a Key Regulator of Sickle Cell Disease Microvascular Complications.

Cell membrane lipid composition, especially cholesterol, affects many functions of embedded enzymes, transporters and receptors in red blood cells (RBC). High membrane cholesterol content affects the RBCs' main vital function, O2 and CO2 transport and delivery, with consequences on peripheral tissue physiology and pathology. A high degree of deformability of RBCs is required to accommodate the size of micro-vessels with diameters significantly lower than RBCs. The potential therapeutic role of high-density lipoproteins (HDL) in the removal of cholesterol and its activity regarding maintenance

Antimicrobial Peptide AMP-17 Affects Candida albicans by Disrupting Its Cell Wall and Cell Membrane Integrity.

BACKGROUND: Candida albicans is associated with high mortality among immunocompromised patients. Resistance to and toxic side effects of antifungal drugs require the development of alternative antifungal agents. AMP-17 is a novel antimicrobial peptide derived from Musca domestica that exerts excellent antifungal effects against the Candida species. In this article, we discuss the potential mechanism of AMP-17 against C. albicans from the perspective of affecting the latter's cell external structure. METHODS: Recombinant AMP-17 was prepared by prokaryotic expression system, and its anti-C. alb

Sequestrocytes: a manifestation of transcellular cross-bonding of the red cell membrane in sickle cell anemia.

An unusual form of red cells (called sequestrocytes) that circulate in the peripheral blood of patients with sickle cell anemia has been identified. In wetmount light microscopy preparations, sequestrocytes appear as massively vacuolated erythrocytes. We have shown by transmission electron microscopy and scanning electron microscopy that the morphology of sequestrocytes can be accounted for on the basis of transcellular cross-bonding of the cell membrane. Initially, endocytotic vesicles span the cytoplasmic space, forming putative cytoskeletal fusion zones. Points of fusion of the membrane ske
