Colchicine overdose-induced acute renal failure and electrolyte imbalance.

Colchicine has been used to control gouty arthritis for long time; colchicine overdose, however, causes multiple organ dysfunction. To date, no investigation has revealed the site of kidney lesion or dysfunction. This investigation describes the case of a male with a history of gout who ingested a large amount of colchicine and developed renal, hematopoietic, gastrointestinal, muscular, electrolytic, and hepatic disorder. Glucosuria was noted during hospital days. Colchicine intoxication is shown to induce proximal tubule damage. Severe electrolytes imbalance was noted, including hypomagnesemi

Toxic epidermal necrolysis-like reaction secondary to colchicine overdose.

Colchicine is a microtubule-inhibiting drug used to treat gout, familial Mediterranean fever and many other skin diseases. Intoxication with colchicine affects multiple organs, often fatally. Cutaneous sequelae of colchicine toxicity are rare. We describe the clinical and histological features of a toxic epidermal necrolysis-like exanthem in a patient who lethally overdosed on colchicine.

[Colchicine in toxic liver cirrhosis and primary biliary cirrhosis].

In animals, colchicine has been shown hepatoprotective, which is related to its anti-fibrotic and anti-inflammatory effect. In man, colchicine has been used for the treatment of alcoholic liver disease and primary biliary cirrhosis. Although controlled trials observed an improvement of liver function tests under colchicine, the survival rate in most studies was only slightly improved without reaching statistical significance. Therefore, at present, no final evaluation of colchicine for the treatment of alcoholic liver diseases and primary biliary cirrhosis can be given.

Arachidonic acid metabolites and colchicine in Behçet's disease (BD).

Vasculitis is accepted to be the basis of Behçet's disease (BD) which is a multisystem disease, and the arachidonic acid(AA) metabolites acting as balancing mediators in the organism are accepted to be responsible for the vasculitis. In this study, we examined the prostaglandin E2 (PGE2) and leukotriene C4 (LTC4) levels of the patients with BD before and after colchicine therapy. We found a statistical decrease in the PGE2 and LTC4 levels after colchicine therapy compared to the previous levels, concluding that colchicine inhibits the inflammation and the polymorphonuclear leukocyte (PML) chem

Colchicine intoxication: clinical pharmacology, risk factors, features, and management.

The use of colchicine for acute gouty arthritis dates to ancient times. In recent years, colchicine also has been used successfully for various other rheumatic and nonrheumatic conditions. Colchicine is a safe drug when used according to established therapeutic guidelines. However, toxicity can be considerable if ingested intentionally or if the recommended doses are exceeded. Colchicine intoxication is characterized by multi-organ involvement and by the poor prognosis associated with administration of large amounts of the drug. Therapy is basically supportive and symptomatic because of the ra

Hyper-IGD syndrome: a new case treated with colchicine.

We report a new case of hyper-IgD syndrome, a recently described disease characterized by recurrent episodes of fever with headache, bilateral cervical lymphadenopathy and, more rarely, abdominal pain and diarrhoea. Polyclonal increase of serum IgD is the most important laboratory finding. Etiopathogenesis and differences with familial Mediterranean fever are discussed. Moreover, good results obtained with colchicine treatment are also reported.

[Acute respiratory distress syndrome in adults in colchicine poisoning].

Two cases of adult respiratory distress syndrome were treated in a series of 26 patients suffering from colchicine overdose. The syndrome appeared between the 24th and 72nd hours. It was characterized by the presence of interstitial as well as alveolar oedema seen on chest roentgenograms. Haemodynamic investigation showed a hyperkinetic state with moderate precapillary pulmonary arterial hypertension. In addition, multivisceral phenomena were observed in all cases. Post-mortem examination revealed interstitial and alveolar pulmonary oedema with haemorrhagic or macrophagic alveolitis often acco

[Effects of colchicine treatment on rat intestinal disaccharidase activities].

UNLABELLED: The changes in morphology and proliferation kinetics produced by colchicine in the intestinal mucosa are known. Contrarily, the functional changes have yet not been investigated systematically. The aim of this study is to investigate the effects of colchicine treatment on rat intestinal disaccharidase activities. MATERIAL AND METHODS: For the experiments male SPF-Wistar rats, body weight 180--200 g (supplied by Mus-Rattus AG, 8011 Brunnthal, FRG) were used. The animals were kept under constant conditions (6 rats per cage) and fed Altromin (Altromin Gmb H,Lage/Lippe), tap water ad

Comparative effectiveness of sodium-glucose cotransporter 2 inhibitors along with colchicine versus colchicine alone among coronary artery disease and type 2 diabetes patients.

BACKGROUND: Colchicine, an anti-inflammatory agent, has been demonstrated to reduce adverse cardiovascular events in coronary artery disease (CAD) patients. Sodium-Glucose Cotransporter 2 Inhibitors (SGLT2i) provide cardiovascular prevention beyond glycemic control including anti-inflammatory effect, and reduced heart failure risk. Given their complementary mechanisms, combining colchicine and SGLT2i may offer synergistic benefits in CAD patients with T2DM, yet the incremental effects of this combination remain underexplored. OBJECTIVE: This study aimed to assess the comparative effectiveness

Adsorption of colchicine, hydroxychloroquine, ibuprofen, and strychnine to activated charcoal.

INTRODUCTION: Activated charcoal is commonly used in the management of xenobiotics to reduce systemic absorption, but data supporting its efficacy for certain xenobiotics remain limited. Quantitative data describing adsorption of colchicine, hydroxychloroquine, ibuprofen, and strychnine help inform emergency treatment options for the management of a xenobiotic ingestion. OBJECTIVE: To evaluate the adsorption of colchicine, hydroxychloroquine, ibuprofen, and strychnine to activated charcoal by quantifying adsorption kinetics and fitting isotherm data to characterize binding behavior and xenobi

Colchicine and dialysis patients (CAD): protocol for a feasibility study.

INTRODUCTION: Colchicine has been shown to reduce cardiovascular events and may improve outcomes in arteriovenous fistulas used for haemodialysis due to antiproliferative effects. However, it is often avoided in patients receiving dialysis. Therefore, a large trial assessing the potential benefits of colchicine in dialysis patients cannot begin without further data on feasibility. The primary objective of this study is to assess the feasibility of carrying out future trials of colchicine in dialysis patients. METHODS AND ANALYSIS: This is an open-label, single centre, single arm study with 10

Colchicine and cardiovascular outcomes in MINOCA: A retrospective cohort study.

BACKGROUND: MINOCA accounts for 5 %-15 % of myocardial infarctions and is defined by <50 % coronary stenosis without an alternative diagnosis, with heterogeneous mechanisms that complicate care. Given the role of inflammation and microvascular dysfunction, we evaluated whether colchicine improves outcomes. METHODS: We conducted a retrospective cohort study using TriNetX, a federated health research network of 134 million patients from 102 healthcare organizations. Adults (≥18 years) with a primary diagnosis of acute myocardial infarction (AMI) and no revascularization after cardiac catheteriz

Drug-Induced Dysphagia and Diarrhea: A Case of Colchicine Toxicity.

We report a 70-year-old woman with 1 year of progressive diarrhea, dysphagia, and 100-pound weight loss, resulting in severe protein-calorie malnutrition. Endoscopy revealed mid-esophageal webs, erosions, erythema, gastric antrum and duodenum congestion, and mucosal granularity in the sigmoid colon. Biopsies from the esophagus, duodenum, and colon showed ring mitoses, prominent dyskeratosis/apoptosis, epithelial atypia, and regenerative changes consistent with colchicine toxicity. Her symptoms resolved within 3 days of discontinuing colchicine.

Efficacy of colchicine in addition to anakinra in patients with recurrent pericarditis.

AIM: Anakinra, an anti IL-1 agent targeting IL-1 alfa and beta, is available for the treatment of recurrent pericarditis in cases with corticosteroid dependence and colchicine resistance after failure of conventional therapies. However, it is unclear if the combination with colchicine, a non-specific inhibitor of the inflammasome targeting the same inflammatory pathway of IL-1, could provide additional benefit to prevent further recurrences. The aim of the present observational study is to assess whether the addition of colchicine on top of anakinra could prolong the time to first recurrence a
