Dysregulated B cells are central to many hematologic malignancies and autoimmune diseases, but current B cell-targeted therapies often fail to translate binding into effective signaling. Here, we present CD20 EryPatch, a living dynamic cellular patch that co-opts natural erythrocyte processes of morphological and biological changes, enabling a dual mechanism of micrometer-scale mechanotransduction and phagocytic "tag-and-clear" for enhanced B cell depletion. CD20 EryPatches are designed to adhere to B cells and anchor on CD20 receptors across extensive cell surface areas. Their progressive dis

Red cell membrane disorders are the most common type of inherited hemolytic disorders in the Japanese population. In hereditary spherocytosis (HS), the primary presentation is a loss of membrane surface area, leading to reduced deformability because of defects in the membrane proteins ankyrin, band 3, β-spectrin, α spectrin, or protein 4.2 (P4.2). Complete P4.2 deficiencies, which are inherited in an autosomal recessive manner, comprise a unique HS subgroup and are common in Japanese, but rare in other populations. In contrast, the principle presentation in hereditary elliptocytosis (HE) is me

Hemorheological abnormalities such as elevated whole blood viscosity, plasma viscosity, erythrocyte deformability and platelet aggregation, hematocrit and fibrinogen levels, are frequently examined as diagnostic tool and prognostic relevance in socially important hemorheological disorders. Distinct biological - morphological and functional platelet alterations, have been described in different addictions (heroin-, cocaine-, nicotine-, alcohol-, etc.). Chronic addictions could cause biochemical and conformational changes in platelets and their membranes, thus modulating platelet receptor expres

Hereditary spherocytosis is a common inherited disorder that is characterised by anaemia, jaundice, and splenomegaly. It is reported worldwide and is the most common inherited anaemia in individuals of northern European ancestry. Clinical severity is variable with most patients having a well-compensated haemolytic anaemia. Some individuals are asymptomatic, whereas others have severe haemolytic anaemia requiring erythrocyte transfusion. The primary lesion in hereditary spherocytosis is loss of membrane surface area, leading to reduced deformability due to defects in the membrane proteins ankyr

A unifying hypothesis for the genesis of cerebral malaria proposes that parasite antigens (released by replication in blood, surface molecules on parasitized erythrocytes, or merozoites) activate platelets that, in turn, contribute to the activation of the inflammatory response and increased levels of endothelial cell adhesion molecules (eCAMs). Increased levels of eCAMs result in further parasitized-erythrocyte sequestration and marked local inflammation that might disrupt the brain microvasculature, which cannot be repaired by the hemostasis system because of its procoagulant state. Disrupti

A clinical investigation was undertaken of the haemorheological effects of short-term administration of synthetic sex hormones. In a randomised controlled investigative trial, groups of 20 women taking ethynyloestradiol, norethisterone, combined norgestrel and ethynyloestradiol or no therapy had their blood viscosity and its major determinants measured before and after 3 months of treatment. Oestrogens and progestogens, singly or in combination, were found to cause a rise in blood viscosity. Oestrogens did so by raising haematocrit and plasma fibrinogen, parameters that are similarly raised in

INTRODUCTION: Hemorheology is the study of the flow properties of the blood and its elements, which, together with natural anticoagulants, are important determinants of cardiovascular events. This study aimed to assess hemorheological and natural anticoagulant profiles of patients with celiac disease (CeD) comprehensively.
METHODS: Our study is a case-control study (registered under ISRCTN49677481) comparing patients with CeD with age- and sex-matched control subjects (1:1). We measured erythrocyte deformability (ED) at high (3-30 Pa) and low shears (0.3-3 Pa), erythrocyte aggregation, whole

Certain rheological disorders such as enhanced aggregation and reduced deformability of erythrocytes are closely associated with microcirculatory disturbances, decreased tissue oxygen supply and represent a major problem in severely burnt patients [1-4]. Vitamin E (alpha-tocopherol) and vitamin A (retinol) exert an important plasma antioxidant properties against free radical-induced damage [11,12]. They possess chain-breaking properties and act as synergists [13]. Plasma antioxidant capacity may be decreased in severely burnt patients as a result of the enhanced consumption and inadequate rest

The membrane elastic shear modulus mu determined by a micropipette technique was found to be elevated by 25% to 200% for red blood cells (RBC) from 7 patients with glucose-6-phosphate dehydrogenase (G-6-PD) and 9 patients with pyruvate-kinase (PK) enzymopathies above the mean value for normal controls. All patients exhibit chronic nonspherocytic hemolytic anemias. A negative linear correlation (r = -0.72) between mu and the number of reticulocytes was obtained in G-6-PD deficiencies. In contrast, a positive correlation (r = 0.88) was found for red blood cells of patients with PK deficiency. A

Plasmodium knowlesi merozoites invade erythrocytes by a sequence of events: attachment, widespread deformation of the erythrocyte membrane, endocytosis, and resealing of the membrane and vesicle. The possible mechanisms by which these events occur are discussed. In particular, there is a detailed discussion of the role of the Duffy blood group system in invasion.

Specialists may gauge the severity of sickle cell disease crisis by quantifying the number of abnormal-looking and sickle-shaped erythrocytes in blood smears. State-of-the-art integral geometry-based descriptors for automatic classification of erythrocytes as normal cells, sickle cells or cells with other deformations have achieved excellent results. Unfortunately, they are computationally expensive, requiring powerful desktop computers and a great deal of memory to run. We propose two new integral geometry-based descriptors for the shape of erythrocytes. Like state-of-the-art techniques, the

Erythrocytes (red blood cells) are critical for the transport of oxygen and carbon dioxide around the body, reaching all tissues ultimately through narrow capillaries which requires significant deformations of the cells. As a result, they must withstand continuous mechanical stress and are specifically adapted with a lipid bilayer enriched in cholesterol and an underlying spectrin-based cytoskeleton for stability and flexibility. Thus, erythrocyte membranes have unique mechanical, biophysical and electrostatic properties suited to their function, which have long been of physiological and biome

BACKGROUND: In sickle cell disease (SCD), polymerization of hemoglobin S (HbS) leads to the formation of rigid, non-deformable sickled RBCs. Loss of RBC deformability, sickling and irreversible membrane damage causes abnormal blood rheology, and increases viscosity which contributes to vasoocclusion and other SCD pathophysiology. GBT440 (generic name voxelotor) is a novel anti-polymerization and anti-sickling agent currently undergoing clinical evaluation for the treatment of SCD.
OBJECTIVE: The purpose of this study was to determine the effects of GBT440 on deformability of sickle RBCs (SS R

A prominent characteristic of sickle cells is the presence of a subpopulation of severely dehydrated cells. Apart from its effect on the rate and extent of Hb S polymerization at low oxygen tension, dehydration has adverse effects on deformability of oxygenated sickle cells. In general, the degree of deformability defect induced by cellular dehydration measured in vitro depends on the method used to measure deformability. Methods in which the extent or rate of cellular deformation are measured are especially sensitive to intracellular Hb concentration, whereas filtration methods are more sensi

Two ideas explain the mechanism of shape regulation: the lipid bilayer coupled theory and the protein network scaffold theory. Recently, several important articles have been published on the former theory. However, many phenomena argue against the theory, including behavior of ghosts and triton shells, various types of manipulation of proteins, and fixation of the shape by the addition of large reagents outside the cell. Moreover, hereditary spherocytosis shows normal, uneven distribution of phospholipids, and hereditary and artificial defects of a membrane protein show spherocytes or elliptoc
