Fusobacterium infection facilitates the development of endometriosis through the phenotypic transition of endometrial fibroblasts.

Retrograde menstruation is a widely accepted cause of endometriosis. However, not all women who experience retrograde menstruation develop endometriosis, and the mechanisms underlying these observations are not yet understood. Here, we demonstrated a pathogenic role of Fusobacterium in the formation of ovarian endometriosis. In a cohort of women, 64% of patients with endometriosis but <10% of controls were found to have Fusobacterium infiltration in the endometrium. Immunohistochemical and biochemical analyses revealed that activated transforming growth factor-β (TGF-β) signaling resulting fro

Unraveling a Unique Encounter of Fusobacterium nucleatum With Empyema: A Case Report and Review of Literature.

We report a case of Fusobacterium nucleatum (F. nucleatum) empyema in a 34-year-old male with no significant past medical history or obvious risk factors who presented with shortness of breath and chest pain. His imaging showed complicated parapneumonic effusion which grew F. nucleatum. He was started on piperacillin-tazobactam. The patient's clinical condition deteriorated despite initial therapeutic efforts, leading to escalated antibiotic therapy and further investigations. The patient's subsequent clinical course included pigtail catheter placement with drainage of fluid requiring tpa and

A unique case of Fusobacterium nucleatum spondylodiscitis communicating with a pleural empyema through a fistula.

Species (spp.) belonging to the genus Fusobacterium are anaerobic commensals colonizing the upper respiratory tract, the gastrointestinal tract, and the genitals. Infections with Fusobacterium spp. have been reported at many anatomical sites, including pneumonias and pleural empyemas; however, there are very few published cases of Fusobacterium spp. causing spondylodiscitis or fistulas. Bone infections with Fusobacterium can spread directly to surrounding muscular tissue or by hematogenous transmission to any other tissue including pleurae and lungs. Similarly, pleural infections can spread Fu

Specific increase of Fusobacterium in the faecal microbiota of neonatal calves infected with Cryptosporidium parvum.

The faecal microbiota plays a critical role in host health, with alterations in the human faecal microbial composition associated with various conditions, particularly diarrhoeal diseases. However, little is known about microbial changes during cryptosporidiosis, one of the most important diarrhoeal diseases caused by protozoa in cattle. In this study, alterations in the faecal microbiota of neonatal calves as a result of Cryptosporidium parvum infection were investigated on a C. parvum-positive farm. Comparisons were made among groups of C. parvum-infected, rotavirus-infected, and the pathoge

Subacute osteomyelitis of the femur due to Fusobacterium nucleatum in a 7-year-old boy.

Subacute hematogenous osteomyelitis is an insidious infection, which commonly has a delayed diagnosis. We describe the case of a 7-year-old boy with subacute osteomyelitis, which was initially considered to be a bone tumor. Infection should be considered in all cases of bone pain, especially in children, even in the absence of typical systemic features of inflammation.

A case of adolescent pelvic inflammatory disease caused by a rare bacterium: Fusobacterium nucleatum.

BACKGROUND: Pelvic inflammatory disease (PID) is a polymicrobial infection that typically occurs in sexually active females. PID is rare in premenarchal and/or noncoital young women; however, there are case reports of PID occurring in virginal females. We present the first reported case of PID associated with F. nucleatum. In this unusual case of PID in an adolescent, the responsible organism may have originated from the patient's oral cavity. CASE: A 13-year-old noncoital Caucasian female presented to a tertiary children's hospital with fever and acute abdominal/pelvic pain. She had experien

Fusobacterium nucleatum: a keystone bridging pathogen in polymicrobial biofilms and host immune dysregulation.

Oral infectious diseases such as periodontitis stem from disrupted homeostasis between host immunity and polymicrobial biofilm communities. Fusobacterium nucleatum, a ubiquitous oral commensal, is a key intermediate colonizer with well-documented interspecies coaggregation activity. It has been proposed as a bridging pathobiont that contributes to microbial dysbiosis and immune dysregulation at the host-microbe interface, though its in vivo structural bridging function remains debated. This mini-review synthesizes recent advances in understanding the dual functions of F. nucleatum in interspec

Targeting Fusobacterium nucleatum in cancer therapy: A new frontier in solid tumor treatment.

Recently, the tumor-infiltrating microbiome (TIM) has gained increasing attention due to its pivotal role in carcinogenesis, tumor progression, resistance to chemotherapy, and immune evasion. Although the composition of these microbial communities is not yet fully elucidated, emerging studies have demonstrated that certain bacteria exert pro-tumoral effects, either through enzymatic activities or by modulating the host immune response. Among them, Fusobacterium nucleatum (Fn) represents one of the most pathogenic residents of several solid tumors, making it a novel and promising target in the

Fusobacterium nucleatum enhances enterotoxigenic Bacteroides fragilis-mediated neuron loss and intestinal motility dysfunction.

Gut dysmotility is a common long-term effect of enterotoxigenic Bacteroides fragilis (ETBF) colonization but the underlying mechanisms are not clear. Here, using mouse colonization models, we found that ETBF-secreted B. fragilis toxin (BFT) causes loss of colonic myenteric neurons, which are crucial regulators of intestinal motility. BFT induced apoptosis in these neurons through the NOD1-RIPK2-CASP9-CASP3 pathway, leading to gut dysmotility that persisted even after the clearance of ETBF. Analyses of human inflammatory bowel disease and colorectal cancer patient cohorts showed a positive corr

Prediction of non-coding RNAs in Fusobacterium nucleatum-infected mice using machine learning.

BACKGROUND: The anaerobic commensal Fusobacterium nucleatum is scarce in healthy subgingival dental biofilms but is highly prevalent in periodontal pockets. Numerous genome-wide association studies and gene expression studies using microarrays or RNA sequencing (RNA-Seq) have been performed to better understand the genetic architecture of periodontal disease. However, these investigations have limited predictive capacity for identifying RNAs, particularly non-coding RNAs (ncRNAs). The mechanism of regulation of ncRNAs by F. nucleatum to alter disease progression in mice has not been thoroughly

Tumor-Associated Fusobacterium nucleatum and Aggressive Architectural Features in Stage II Colorectal Cancer.

Tumor budding (TB) and poorly differentiated clusters (PDCs) are histopathological indicators of invasive potential in colorectal cancer (CRC). Concurrently, Fusobacterium nucleatum (F. nucleatum) has emerged as a modulator of CRC progression. The relationship between intratumoral F. nucleatum and aggressive tumor architecture remains unknown. We evaluated 257 stage II primary CRCs. TB and PDC were assessed at the invasive front. Intratumoral F. nucleatum DNA was quantified from FFPE samples using droplet digital PCR. Associations between F. nucleatum load and molecular and clinicopathological

ST3GAL1 mediates sialylation to promote adhesion of Fusobacterium nucleatum to colorectal cancer cells.

Fusobacterium nucleatum is a commensal bacterium in the oral cavity and is closely related to several oral infectious diseases. In addition to oral colonization, F. nucleatum is enriched in colorectal cancer (CRC) and is closely associated with CRC progression, while the mechanism of F. nucleatum adhesion to cancer cells has not been fully explained. Sialylation is of high levels in CRC and host cell surface sialoglycans are considered as potential targets for microorganism adhesion. However, the effect of excessive sialylation in CRC on the adhesion of gut microorganisms remains unknown. Here

Detection of Fusobacterium nucleatum in Colorectal Adenomas Reveals Associations with Immune Molecular Signatures.

Fusobacterium nucleatum has been implicated in colorectal cancer, but its role in adenomas remains unclear. We applied an RNA-based detection of F. nucleatum in formalin-fixed paraffin-embedded adenoma tissue and explored the mutational landscape and transcriptomic profile of F. nucleatum+ patients in comparison to F. nucleatum- patients. Bespoke F. nucleatum probes successfully detected F. nucleatum in 11% of adenomas. F. nucleatum+ patients exhibited a positively enriched anti-bacterial defence response and immune-related transcriptomic signatures, as well as a proliferative profile. These f

Influence of Fusobacterium nucleatum on Tumor Necrosis Factor Alpha Therapy in Ulcerative Colitis.

Inflammatory bowel disease (IBD), including ulcerative colitis (UC), involves chronic gastrointestinal inflammation, with tumor necrosis factor alpha (TNF-α) playing a key role. Anti-TNF therapy is widely used, but not all UC patients respond, suggesting additional contributing factors. Gut microbiota alterations, particularly dysbiosis, may influence treatment outcomes. This study examines the relationship between Fusobacterium nucleatum (F. nucleatum) density and TNF-α expression in UC patients receiving anti-TNF therapy. Biopsy samples from responders (n = 10), nonresponders (n = 10), and h
