Stereoselective total synthesis of (-)-spirofungin A by utilising hydrogen-bond controlled spiroketalisation.

The stereoselective total synthesis of the spiroketal containing Streptomyces metabolite (-)-spirofungin A (1) is described. A key step involved a spiroketalisation controlled by an intramolecular H-bond which favoured the desired spiroketal 4 (13:1 ratio). The presence of the intramolecular H-bond in 4 is possibly due to a 1,5-alkyne-oxygen interaction. Other key steps include an efficient cross-metathesis to form the spiroketal precursor, a tin mediated syn-aldol reaction and a Stille cross-coupling reaction to create the C22C23 bond. A final Wittig extension followed by deprotection gave (

Hydrogen-bond dynamics for water confined in carbon tetrachloride-acetone mixtures.

In a variety of biological scenarios water is found trapped within hydrophobic environments (e.g., ion channels). Its behavior under such conditions is not well understood and therefore is attracting enormous scientific attention. It is of particular interest to understand how the confining environment affects both the structure and dynamics of water. Within this scenario, we report molecular dynamics simulation results for water trapped in a mixture of acetone and carbon tetrachloride whose composition mimics the one employed in recently reported experiments [Gilijamse, J. J.; et al. Proc. Na

A ligand-mediated hydrogen bond network required for the activation of the mineralocorticoid receptor.

Ligand binding is the first step in hormone regulation of mineralocorticoid receptor (MR) activity. Here, we report multiple crystal structures of MR (NR3C2) bound to both agonist and antagonists. These structures combined with mutagenesis studies reveal that maximal receptor activation involves an intricate ligand-mediated hydrogen bond network with Asn770 which serves dual roles: stabilization of the loop preceding the C-terminal activation function-2 helix and direct contact with the hormone ligand. In addition, most activating ligands hydrogen bond to Thr945 on helix 10. Structural charact

General Synthesis of MOF Nanotubes via Hydrogen-Bonded Organic Frameworks toward Efficient Hydrogen Evolution Electrocatalysts.

The application scope of metal-organic frameworks (MOFs) can be extended by rationally designing the architecture and components of MOFs, which can be achieved via a metal-containing solid templated strategy. However, this strategy suffers from low efficiency and provides only one specific MOF from one template. Herein, we present a versatile templated strategy in which organic ligands are weaved into hydrogen-bonded organic frameworks (HOFs) for the controllable and scalable synthesis of MOF nanotubes. HOF nanowires assembled from benzene-1,3,5-tricarboxylic acid and melamine via a simple son

Competition between the hydrogen bond and the halogen bond in a [CH3OH-CCl4] complex: a matrix isolation IR spectroscopy and computational study.

Methanol (CH3OH) is the simplest alcohol and carbon tetrachloride (CCl4) is widely used as a solvent in the chemical industry. CH3OH and CCl4 are both important volatile substances in the atmosphere and CCl4 is an important precursor for atmospheric ozone depletion. Moreover, mixtures of CH3OH and CCl4 are an important class of non-aqueous mixtures as they exhibit a large deviation from Raoult's law. The specific interaction between CH3OH and CCl4 is not yet investigated experimentally. The interaction between CH3OH and CCl4 at the molecular level can be twofold: hydrogen bond (O-HCl) and halo

Tuning Supramolecular Structure in Trimethylglycine Cocrystals: Competition Between Hydrogen and Halogen Bonding upon Cl/Br Substitution.

Two novel cocrystals of zwitterionic trimethylglycine (TMG) with 2,6-dichlorophenol [TMG•2,6-dichlorophenol] (1:1) and 2,6-dibromophenol [TMG•2,6-dibromophenol] (1:2) are synthesized and structurally characterized using single crystal X-ray diffraction. To estimate the energy of various intermolecular interactions, periodic DFT calculations were performed followed by Bader analysis of the crystalline electron density. TMG molecules form dimers in [TMG•2,6-dichlorophenol] (1:1). Its supramolecular structure is governed by the primary charge-assisted H-bonds (~60 kJ/mol) and supported by C-H∙∙∙O

Hydrogen-bonding interface engineering enables efficient perchlorate removal via dual-site polarization and suppressed competitive hydration.

Perchlorate (ClO4⁻) contamination poses serious risks to ecosystems and human health due to its high mobility, persistence, and thyroid-disrupting toxicity. Current technologies struggle with the rapid, selective removal of ClO4⁻, particularly in complex water matrices. In this study, we addressed this challenge by developing a hydrogen-bond interface-engineered adsorbent, Mn/N-coordinated porous carbon modified with formic acid (Mn-NC···HCOOH), to achieve efficient and rapid ClO4⁻ removal. The novelty of this work lies in the engineering of dual active sites (Mn and N-coordinated C), coupled

Structural flexibility and hydrogen bonding of aqueous vitamin C: Raman and UV spectroscopic and computational insights.

Raman and UV spectroscopic experiments, supported by quantum mechanical modeling of the conformations, vibrational and electronic absorption spectra, and optical rotation of aqueous vitamin C, reveal qualitative differences in the conformational behavior and hydrogen bonding of its neutral form, ascorbic acid AH2, and the deprotonated form, the ascorbate anion AH-. AH2, which predominates in solutions with concentrations c ≥ 10-3 M, can adopt about eighty conformations with similar free energies. The three most populated conformers, which together account for about forty percent of the ensembl

Thermodynamic Stability in Transition Metal-Hydrogen Dications: Potential Energy Curves, Spectroscopic Parameters, and Bonding for VH2.

Seventeen electronic states of the dication VH2+ were characterized by the SA-CASSCF/icMRCI methodology using very extended basis sets; 11 were described for the first time. Potential energy curves were constructed and the associated spectroscopic parameters evaluated. Triplet and quintet states correlating with the V2+ + H channel are thermodynamic stable. For states dissociating into the channel V+ + H+, avoided crossings at large distances give rise to thermodynamic metastability but do not affect the characterization of the bound region. Configuration state functions with the 3σ orbital /d

Rapidly Screening the Correlation between the Rotational Mobility and the Hydrogen Bonding Strength of Confined Water.

Automated Deuterium Relaxation-Ordered SpectroscopY in solution (ADROSYS), an automated two-dimensional deuterium NMR methodology, discriminates between D2O populations (as well as deuterium-labeled alcohol groups) whose properties differ as a result of being confined inside nanoscale volumes. In this contribution, a proof-of-principle demonstration on reverse micelles (RMs) yields the insight that as the length scale of the confinement decreases from several nanometers down to less than a nanometer, the position of the signal peak migrates through the two-dimensional (2D) spectrum, tracing ou

Solvent Effects on Optical Rotation: On the Balance between Hydrogen Bonding and Shifts in Dihedral Angles.

Optical rotations of several conformers of four fluorinated molecules containing the 1-naphthalene or 4-(benzyloxy)phenyl group at the stereocenter have been calculated both in the gas phase and in an aqueous environment. For the compounds containing the 4-(benzyloxy)phenyl group, solvent effects on the optical rotations have also been investigated in chloroform as solvent. Optical rotations have been obtained by time-dependent density functional theory (TDDFT) with the CAM-B3LYP functional and the aug-cc-pVDZ basis set at λ = 589 nm. Implicit and explicit solvent effects were investigated thr

Highly enantioselective phosphination and hydrophosphonylation of azomethine imines: using chiral squaramide as a hydrogen bonding organocatalyst.

Enantioselective phosphination and hydrophosphonylation reactions between azomethine imines and diarylphosphine oxides or dialkyl phosphites were respectively developed by the use of a chiral squaramide as the hydrogen bonding organocatalyst, which afforded two types of phosphorus containing product in high yields with good to excellent enantioselectivities.

New insights into the dual fluorescence of methyl salicylate: effects of intermolecular hydrogen bonding and solvation.

In this paper, we propose a new and complete mechanism for dual fluorescence of methyl salicylate (MS) under different conditions using a combined experimental (i.e., steady-state absorption and emission spectra and time-resolved fluorescence spectra) and theoretical (i.e., time-dependent density function theory) study. First, our theoretical study indicates that the barrier height for excited state intramolecular proton transfer (ESIPT) reaction of ketoB depends on the solvent polarity. In nonpolar solvents, the ESIPT reaction of ketoB is barrierless; the barrier height will increase with inc

beta Turns, water cage formation and hydrogen bonding in the structures of L-valyl-L-phenylalanine.

L-Valyl-L-phenylalanine has been crystallized as an orthorhombic dihydrate (1) in the shape of needles and as a monoclinic trihydrate (2) with Z = 16 (P2(1), Z' = 8) in the shape of thin plates. Peptide molecules in these two structures occur in three basic conformations, termed c(1), c(2A) and c(2B). c(2B) has not been observed previously for dipeptides. Together with c(1) it forms a model pair for Type I and Type II beta-turns in protein structures. The crystal packing of (2) is remarkable in that some of the L-Val side chains are exposed to the solvent region of the crystal rather than bein

Intramolecular hydrogen bonding and conformational studies of bridged thebaine and oripavine opiate narcotic agonists and antagonists.

A conformational study of a series of oripavine derivatives is reported using the PCILO semiempirical quantum mechanical method. Low-energy conformers of carbinol substituents on C7-C19-R1R2OH are found with and without intramolecular hydrogen bonding to the C6-OCH3 group. The relative energies of these conformers depend on the R1 and R2 groups and the diastereoisomerism of the alcohol. The results are consistent with available NMR and IR studies of intramolecular hydrogen bonding and with crystallographic data. The importance of interaction between specific conformations of C19 carbinols and

Probing Hydrogen Atom Transfer at a Phosphorus(V) Oxide Bond Using a "Bulky Hydrogen Atom" Surrogate: Analogies to PCET.

Recent computational studies suggest that the phosphate support in the commercial vanadium phosphate oxide (VPO) catalyst may play a critical role in initiating butane C-H bond activation through a mechanism termed reduction-coupled oxo activation (ROA) similar to proton-coupled electron transfer (PCET); however, no experimental evidence exists to support this mechanism. Herein, we present molecular model compounds, (Ph2N)3V═N-P(O)Ar2 (Ar = C6F5 (2a), Ph (2b)), which are reactive to both weak H atom donors and a Me3Si• (a "bulky hydrogen atom" surrogate) donor, 1,4-bis(trimethylsilyl)pyrazine.

Dissecting Hydrophobic and Hydrogen-Bonding Contributions in the Rim Binding of Pyromellitic Diimides to β-Cyclodextrin.

The relative contributions of hydrophobic interactions and hydrogen bonding in cyclodextrin host-guest systems remain a fundamental challenge in supramolecular chemistry. In the present study, a series of pyromellitic diimide (PMDI) derivatives bearing alkyl substituents of different sizes was employed to investigate the origin of rim-binding interactions with β-cyclodextrin (β-CD). NMR, ROESY and ICD studies established that PMDIs interact with β-CD through the narrow rim, with the alkyl substituents penetrating the cavity. The PMDI chromophore remained outside the cavity of the β-CD. In cont

Multiple hydrogen bonding enables large-area doped phosphorescent glasses with robust stability and high-temperature afterglow.

Molecular room-temperature phosphorescence (RTP) materials with compact and ordered structures organized by intermolecular interactions have proven to be a newly-emerged strategy for high-performance luminescence. However, typical crystalline materials exhibit intrinsic brittleness and compromised optical transparency due to their highly ordered packing, thereby restricting their applicability in diverse functional systems. Herein, a universal non-conjugated molecule (1,2,3,4-butane tetracarboxylic acid) with abundant hydrogen-bonding positions is introduced as a host matrix for supramolecular
