Resveratrol induces molecular changes in cholesterol homeostasis in SAMP8 mice cerebellum.

Resveratrol (Rsv) is a natural polyphenol with neuroprotective properties that modulates several pathways implicated in Alzheimer's disease (AD). Cholesterol homeostasis is disrupted in AD patients, and this imbalance plays a key role in amyloid precursor protein (APP) processing, β-amyloid aggregation and membrane stability. The effect of Rsv on the cerebellum, an emerging structure in cognitive networks and AD pathology due to its high connectivity with other brain regions, remains largely unexplored. This study aims to characterize the effects of Rsv on the cerebellum of SAMP8 mice, an anim

Regulation of Intestinal Butyrate Transporters by Oxidative and Inflammatory Status.

Beneficial effects of the microbiota-derived metabolite butyrate at the colonic level are well established, particularly through its relevance in colorectal cancer (CRC) and inflammatory bowel disease (IBD), two major intestinal pathologies. Therefore, the mechanisms involved in butyrate transport across colonic epithelial cell membranes (uptake transporters: monocarboxylate transporter 1 (MCT1) and sodium-coupled monocarboxylate transporter 1 (SMCT1); efflux transporters: breast cancer resistance protein (BCRP) and MCT1/monocarboxylate transporter 4 (MCT4)), which are determinant for its intr

SHADE AVOIDANCE 4 Regulates Magnesium Deficiency-Induced Root Hair Development by Regulating Auxin Transport in Arabidopsis.

Root hairs enlarge root surface and facilitate plant exploration for edaphic resources, particularly under nutrient-limiting conditions. While magnesium (Mg) deficiency is known to markedly stimulate root hair development, the underlying molecular mechanisms remain poorly characterised. Here, we elucidate the critical role of SHADE AVOIDANCE 4 (SAV4) in regulating Mg deficiency-induced root hair elongation through modulating epidermal auxin level in Arabidopsis. Root hair elongation under Mg deficiency was significantly suppressed in sav4 mutant, and SAV4 epidermal-specific expression partiall

Thioxanthone-Mediated Cytoprotection Against Cisplatin Toxicity: Exploring the Potential Involvement of P-Glycoprotein Through Computational and Experimental Approaches.

P-glycoprotein (P-gp), an efflux transporter highly expressed in renal tubules, plays a crucial role in the detoxification and protection of barrier/excretory tissues from harmful xenobiotics. Xanthones and thioxanthones (TXs) are known for their antimicrobial and antitumor activities and for their ability to modulate membrane transporters such as P-gp. Previous studies have reported that (thio)xanthonic derivatives enhance P-gp expression and/or activity in intestinal cells, reducing the intracellular accumulation of toxic substrates; however, their capacity to modulate P-gp in renal cells re

GPR81 Regulates MCT1 Membrane Translocation Through a PKA-Dependent Signaling Pathway in Rat Podocytes.

Podocytes and their foot processes form a functional layer of the glomerular filtration barrier. Due to their unique morphology and function, podocytes employ distinct nutrient pathways to maintain the bioenergetic balance, with lactate being one of several available energy substrates. Enhanced lactate intake modulates the redox state of the cell by increasing mitochondrial respiration and reactive oxygen species production. Monocarboxylate transporter 1 (MCT1) is the primary lactate transporter, and alterations in its surface expression may contribute to the regulation of lactate uptake in po

The mist of ferroptosis: The Orpheus journey of mitochondria - Exploring the symphony of cell fate.

Ferroptosis is a regulated cell death pathway characterized by iron-dependent accumulation of lipid peroxides and distinct mitochondrial morphological alterations, including reduced volume, increased membrane density, elevated membrane potential, cristae loss, and outer membrane rupture. These features starkly contrast with those observed in apoptosis, autophagy, and necrosis. As the cellular energy hub and metabolic nexus, mitochondria play multifaceted regulatory roles in ferroptosis through their integration of metabolic networks and redox homeostasis. This review systematically examines mi

K activity of CCD principal cells from normal and DOCA-treated rabbits.

We used liquid ion exchanger and conventional microelectrodes to evaluate the effects of mineralocorticoids on the intracellular K activity (aiK) and K transport properties of principal cells (PC) of isolated cortical collecting ducts (CCDs). Hoffman modulation optics and electrophysiological methods were used to identify PC. K activity was measured with two single-barreled electrodes. We found that aiK of PC from deoxycorticosterone acetate (DOCA)-treated rabbits (97.6 mM) was not different from controls (94.8 mM). The driving forces for K transport across the basolateral membrane favored cel

Neurochemical, pharmacokinetic, and behavioral effects of the novel selective serotonin reuptake inhibitor BMS-505130.

BMS-505130 is a potent and selective serotonin transport inhibitor; K(i) for binding to the serotonin transporter = 0.18 nM (K(i) values for binding to the norepinephrine and dopamine transporters = 4.6 and 2.1 microM, respectively). In platelet serotonin uptake studies BMS-505130 (5 mg/kg, p.o.) produced a robust inhibition of serotonin uptake. In microdialysis studies oral dosing with BMS-505130 produced a dose-dependent increase in cortical serotonin levels that reached a maximal effect of 200% above baseline at a dose of 1 mg/kg, p.o.; the peak serotonin response was transient in nature. Dopamine transporter-mediated cytotoxicity of beta-carbolinium derivatives related to Parkinson's disease: relationship to transporter-dependent uptake.

Endogenous or exogenous beta-carboline (betaC) derivatives structurally related to the selective dopaminergic neurotoxin 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and its active metabolite 1-methyl-4-phenylpyridinium (MPP(+)) may contribute to dopaminergic neurodegeneration in Parkinson's disease (PD). We addressed the importance of the dopamine transporter (DAT) for selective dopaminergic toxicity by testing the differential cytotoxicity and cellular uptake of 12 betaCs in human embryonic kidney HEK-293 cells ectopically expressing the DAT gene. Cell death was measured using [4,5-Di

Cocaine-induced renal disease.

Cocaine has anaesthetic, vasoconstrictive and CNS stimulatory effects. Presently, it is used clinically as a local anaesthetic and abused as a recreational drug. It has been implicated in both acute and chronic renal failure and has been reported to affect every aspect of the nephron. This article will review the spectrum of cocaine-induced kidney disease and attempt to give insight into the pathophysiological mechanisms involved.

Ca(2+)-CaM regulating viability of Candida guilliermondii under oxidative stress by acting on detergent resistant membrane proteins.

UNLABELLED: Reactive oxygen species (ROS) play a vital role in reducing viability of yeast cells. The Ca(2+)-CaM signaling pathways are involved in regulating the intracellular ROS level in yeast cells under stress. Detergent resistant membranes (DRMs), the sterol-rich microdomains, participate in a wide range of cellular processes including growth, trafficking and death in yeast cells. In the present study, we found that Trifluoperazine (TFP), an antagonist of CaM, could increase the viability of Candida guilliermondii cells under H2O2 stress. Based on comparative analysis of DRM sub proteomi

Multidrug transport by the ABC transporter Sav1866 from Staphylococcus aureus.

Sav1866 is an ATP-binding cassette (ABC) protein from the pathogen Staphylococcus aureus and is a homologue of bacterial and human multidrug ABC transporters. Recently, the three-dimensional crystal structure of Sav1866 was determined at 3.0 A resolution [Dawson, R. J., and Locher, K. P. (2006) Nature 443, 180-185]. Although this structure is frequently used to homology model human and microbial ABC multidrug transporters by computational methods, the ability of Sav1866 to transport multiple drugs has not been described. We obtained functional expression of Sav1866 in the drug-sensitive, Gram-

Cholesterol defect is marked across multiple rodent models of Huntington's disease and is manifest in astrocytes.

Brain cholesterol, which is synthesized locally, is a major component of myelin and cell membranes and participates in neuronal functions, such as membrane trafficking, signal transduction, neurotransmitter release, and synaptogenesis. Here we show that brain cholesterol biosynthesis is reduced in multiple transgenic and knock-in Huntington's disease (HD) rodent models, arguably dependent on deficits in mutant astrocytes. Mice carrying a progressively increased number of CAG repeats show a more evident reduction in cholesterol biosynthesis. In postnatal life, the cholesterol-dependent activiti

A failure in energy metabolism and antioxidant uptake precede symptoms of Huntington's disease in mice.

Huntington's disease has been associated with a failure in energy metabolism and oxidative damage. Ascorbic acid is a powerful antioxidant highly concentrated in the brain where it acts as a messenger, modulating neuronal metabolism. Using an electrophysiological approach in R6/2 HD slices, we observe an abnormal ascorbic acid flux from astrocytes to neurons, which is responsible for alterations in neuronal metabolic substrate preferences. Here using striatal neurons derived from knock-in mice expressing mutant huntingtin (STHdhQ cells), we study ascorbic acid transport. When extracellular asc

Carrier-Mediated Selective Transport of PFAS across Liquid Membranes.

Per- and polyfluoroalkyl substances (PFAS) are persistent pollutants whose environmental mobility and biological accumulation are governed by transport across phase boundaries. Although biological PFAS transport has been extensively investigated, synthetic systems capable of mediating selective PFAS transport remain underdeveloped. Here, we report a family of synthetic carriers (P1-7) that enable structure-dependent, directional PFAS transport across liquid membranes under mild pH gradients. Binding interactions were characterized by ultraviolet-visible (UV-Vis) and nuclear magnetic resonance

An activity-modulated transport route across myelin (TRAM) for motor-driven organelle transfer in oligodendrocytes.

Myelin sheaths comprise compacted layers of membrane wrapped around axons. Each sheath, if 'unwrapped', has a cytoplasm-filled space at its perimeter. Transmission electron microscopy reveals that this space contains microtubules and organelles; however, whether these are developmental remnants or serve a specific function remains unknown. Live imaging of myelinating oligodendrocytes in mice showed microtubule-dependent organelle transport within myelin sheath cytoplasmic spaces. Further, movement of myelin-located peroxisomes was modulated by neuronal electrical activity, in vitro and in vivo

PI3-kinase and mTOR inhibitors differently modulate the function of the ABCG2 multidrug transporter.

The ATP-binding cassette (ABC) transporter ABCG2 plays an important role in tissue detoxification and confers multidrug resistance to cancer cells. Identification of expressional and functional cellular regulators of this multidrug transporter is therefore intensively pursued. The PI3-kinase/Akt signaling axis has been implicated as a key element in regulating various cellular functions, including the expression and plasma membrane localization of ABCG2. Here we demonstrate that besides inhibiting their respective target kinases, the pharmacological PI3-kinase inhibitor LY294002 and the downst

Multidrug efflux pumps of Gram-positive bacteria.

Gram-positive organisms are responsible for some of the most serious of human infections. Resistance to front-line antimicrobial agents can complicate otherwise curative therapy. These organisms possess multiple drug resistance mechanisms, with drug efflux being a significant contributing factor. Efflux proteins belonging to all five transporter families are involved, and frequently can transport multiple structurally unrelated compounds resulting in a multidrug resistance (MDR) phenotype. In addition to clinically relevant antimicrobial agents, MDR efflux proteins can transport environmental
