Nutrient availability and sodium pyruvate regulate the resuscitation of viable but nonculturable Staphylococcus aureus.

Staphylococcus aureus (S. aureus) can enter a viable but nonculturable (VBNC) state when induced by citric acid at 4 °C, posing hidden risks to food safety upon resuscitation. However, the effects of nutritional conditions and sodium pyruvate (SP) on this process remain unclear. This study demonstrates that 0.5 × TSB, TSB, 2 × TSB, and SP resuscitate VBNC cells following sigmoidal kinetics, with an efficacy ranking of SP > 2 × TSB > TSB > 0.5 × TSB. SP significantly accelerated resuscitation through enhanced early enzymatic activity, a transient increase in intracellular ROS levels, and rapid

Bacillus subtilis spores inactivated by short pulses of soft X-rays in the water window.

INTRODUCTION: Bacterial endospores are highly resistant to environmental stressors. Their resistance complicates sterilization efforts, leading to exploration of novel inactivation techniques. Radiation in the water window spectral range (284-543 eV), typically studied using synchrotron sources, offers high contrast for bio-imaging and potential for efficient spore inactivation due to its high absorption and shallow penetration. METHODS: A compact pulsed laser-plasma soft X-ray source was used to irradiate Bacillus subtilis spores in the water window spectral range. A combination of STEM-EDS

Reducing culture underestimation in urine: flow cytometry-guided validation of low-dose DTT pre-treatment for biofilm dispersal.

A substantial proportion of symptomatic patients with urinary tract infections (UTIs) have negative or low-count urine cultures despite clinically compatible presentations. Biofilm-associated bacterial aggregation and the presence of viable bacteria not recovered by standard culture may contribute to this diagnostic gap. Flow cytometry (FACS) can detect these viable cells, but culturability remains essential for species identification and antimicrobial susceptibility testing. We evaluated whether low-concentration dithiothreitol (DTT) can safely disperse urinary biofilm-associated aggregates a

Effects of Ultrasound-Mediated Treatments on Dental Biofilm Attachment and Viability.

OBJECTIVE: Dental biofilms are responsible for the majority of oral cavity diseases (e.g., caries, periodontitis and gingivitis). In this work, we evaluate the effects of therapeutic ultrasound on a Streptococcus mutans biofilm model. The research aim is to assess whether therapeutic ultrasound can be implemented in standard practice for the treatment and prevention of oral infections. METHODS: Streptococcus mutans biofilms were grown on a tooth-mimicking substrate and subsequently exposed to chlorhexidine (CHX) and therapeutic ultrasound treatments. We examined the effects of low duty cycle

Yeast's vacuole a privileged niche that protects intracellular bacteria against antibiotics.

Detection of Helicobacter pylori, Staphylococcus, Nocardia and Cyanobacteria inside the yeast Candida tropicalis raised the question whether treating yeast with antibiotics mix (ABM) eliminates intracellular bacteria. Live/Dead staining showed occurrence of viable bacteria inside the vacuole of C. tropicalis. Amplification of bacterial 16S rRNA genes from yeast DNA with the size of 521, 750, 606 and 450 bp were similar to those from control H. pylori, Staphylococcus, Nocardia and Cyanobacteria, respectively. To eliminate intracellular bacteria yeast cultures in yeast-glucose (YG) broth were tr

Nitrogen supplementation during wine fermentation, cooling rate and SO₂ addition timing influence yeast viability and acetaldehyde production in the post-fermentation process.

The post-fermentation phase of still wine plays a crucial role in determining wine quality. This study evaluates the impact of key winery practices (including nitrogen supplementation during fermentation, cooling methods, and SO₂ addition) on yeast lees viability and acetaldehyde production during the post-fermentation stage. Two commercial Saccharomyces cerevisiae strains with distinct technological characteristics were assessed under a real vinification protocol involving a decanting rest, two rackings, and bottling. Nitrogen was supplemented either as ammonium alone (NH₄+) or as a mix of am

Proteins associated with environmental survival of the pathogen Neisseria meningitidis.

Previously, we reported the persistence of the bacterial pathogen Neisseria meningitidis on fomites, indicating a potential route for environmental transmission. The current goal was to identify proteins that vary among strains of meningococci that have differing environmental survival. We carried out a proteomic analysis of two strains that differ in their potential for survival outside the host. The Group B epidemic strain NZ98/254 and Group W carriage strain H34 were cultured either at 36 °C, 5% CO2, and 95% relative humidity (RH) corresponding to host conditions in the nasopharynx, or at l

Studying the viability and growth kinetics of vancomycin-resistant Enterococcus faecalis V583 following femtosecond laser irradiation (420-465 nm).

Enterococcus faecalis is among the most resistant bacteria found in infected root canals. The demand for cutting-edge disinfection methods has rekindled research on photoinactivation with visible light. This study investigated the bactericidal activity of femtosecond laser irradiation against vancomycin-resistant Enterococcus faecalis V583 (VRE). The effect of parameters such as wavelength and energy density on the viability and growth kinetics of VRE was studied to design an optimized laser-based antimicrobial photoinactivation approach without any prior addition of exogenous photosensitizers

The synergistic effect of photodynamic and sonodynamic inactivation against Candida albicans biofilm.

Candida albicans biofilm can cause diseases that are resistant to conventional antifungal agents. Photodynamic (PDI), sonodynamic (SDI), and sonophotodynamic (SPDI) inactivation have arisen as promising antimicrobial strategies. This study evaluated these treatments mediated by curcumin against C. albicans biofilms. For this, C. albicans biofilms were submitted to PDI, SDI, or SPDI with different light and ultrasound doses, then, the viability assay was performed to measure the effectiveness. Finally, a mathematical model was suggested to fit acquired experimental data and understand the syner

Contribution of Complement System pathways to the killing of Leptospira spp.

The Complement System (CS) plays an important role in the immune response against leptospirosis and can be activated by the Alternative and Lectin Pathways (Innate Immunity) and by the Classical Pathway (Acquired Immunity). Here we analyzed a broad range of nonpathogenic and pathogenic Leptospira strains considering their interaction with each CS pathway. We determined bacterial survival rate and CS protein deposition in the presence of purified proteins, specific component depleted sera and NHS treated with the chelating agents EDTA (inhibits all three activation pathways) or EGTA (inhibits t

Staphylococcal Enterotoxin A Production and Inactivation in Bread during the Production Processes.

Staphylococcus aureus food poisoning is caused by the intoxication of staphylococcal enterotoxin (SE) produced in foods. Staphylococcal food poisoning is mostly due to staphylococcal enterotoxin type A (SEA) among SEs. There have been many studies on the growth and SEA production of S. aureus in various foods, but few studies in bread. Thus, the SEA production by S. aureus in dough during fermentation and the SEA inactivation in dough during baking were studied in the normal production processes of bread in this study. No growth of S. aureus or SEA production in dough, whose total weight was a

The cluster model of ultraviolet disinfection explains tailing kinetics.

AIMS: To develop a new mathematical model derived from first principles to define the kinetics of ultraviolet disinfection and to explain the phenomenon known as tailing. The theory presented interprets tailing as the result of photoprotection due to cumulative Mie scattering effects in clustered populations of micro-organisms. METHODS AND RESULTS: Mie scattering effects at ultraviolet wavelengths are used to compute a shielding constant for each micro-organism based on the average projected diameter. An intrinsic rate constant, hypothesized to be a characteristic property of the microbial ge

'Big things in small packages: the genetics of filamentous phage and effects on fitness of their host'.

This review synthesizes recent and past observations on filamentous phages and describes how these phages contribute to host phentoypes. For example, the CTXφ phage of Vibrio cholerae encodes the cholera toxin genes, responsible for causing the epidemic disease, cholera. The CTXφ phage can transduce non-toxigenic strains, converting them into toxigenic strains, contributing to the emergence of new pathogenic strains. Other effects of filamentous phage include horizontal gene transfer, biofilm development, motility, metal resistance and the formation of host morphotypic variants, important for

Survival of Pseudomonas aeruginosa exposed to sunlight resembles the phenom of persistence.

During exposure of Pseudomonas aeruginosa stationary phase cells to natural solar radiation, a reduction in the rate of loss of bacterial viability was observed when survival fractions were lower than 1/10,000. This reduction was independent of the growth medium used and of the initial bacterial concentration, and was also observed when irradiation was performed with artificial UVA radiation (365nm, 47Wm(-2)). These results indicate the presence of a small bacterial subpopulation with increased tolerance to radiation. Such a tolerance is non-heritable, since survival curves comparable to those

Potential of bacteriophage ΦAB2 as an environmental biocontrol agent for the control of multidrug-resistant Acinetobacter baumannii.

BACKGROUND: Multidrug-resistant Acinetobacter baumannii (MDRAB) is associated with nosocomial infections worldwide. To date, the use of a phage to prevent infections caused by MDRAB has not been demonstrated. RESULTS: The MDRAB-specific phage ϕAB2 was stable at 4°C and pH 7 in 0.5% chloroform solution, and showed a slight decrease in plaque-forming units (PFU)/ml of 0.3-0.9 log after 330 days of storage. The addition of ϕAB2 at a concentration of at least 10⁵ PFU/ml to an A. baumannii M3237 suspension killed >99.9% of A. baumannii M3237 after 5 min, regardless of A. baumannii M3237 concentrat

PPARγ-mediated increase in glucose availability sustains chronic Brucella abortus infection in alternatively activated macrophages.

Eradication of persistent intracellular bacterial pathogens with antibiotic therapy is often slow or incomplete. However, strategies to augment antibiotics are hampered by our poor understanding of the nutritional environment that sustains chronic infection. Here we show that the intracellular pathogen Brucella abortus survives and replicates preferentially in alternatively activated macrophages (AAMs), which are more abundant during chronic infection. A metabolic shift induced by peroxisome proliferator-activated receptor γ (PPARγ), which increases intracellular glucose availability, is ident

Contact-free cold atmospheric plasma treatment of Deinococcus radiodurans.

In this study we investigated the sensitivity of Deinococcus radiodurans to contact-free cold atmospheric plasma treatment as part of a project to establish new efficient procedures for disinfection of inanimate surfaces. The Gram-positive D. radiodurans is one of the most resistant microorganisms worldwide. Stationary phases of D. radiodurans were exposed to cold atmospheric plasma for different time intervals or to ultraviolet C (UVC) radiation at dose rates of 0.001-0.0656 J cm⁻², respectively. A methicillin-resistant Staphylococcus aureus strain (MRSA) served as control for Gram-positive b
