Insights from Streptococcus pneumoniae glucose kinase structural model.

Streptococcus pneumonia is the common cause of sepsis and meningitis. Emergence of multiple antibiotic resistant strains in the community-acquired bacterium is catastrophic. Glucose kinase (GLK) is a regulatory enzyme capable of adding phosphate group to glucose in the first step of streptomycin biosynthesis. The activity of glucose kinase was regulated by the Carbon Catabolite Repression (CCR) system. Therefore, it is important to establish the structure-function relation of GLK in S. pneumoniae. However, a solved structure for S. pneumoniae GLK is not available at the protein data bank (PDB)

Modeling the structure of mAb 14B7 bound to the anthrax protective antigen.

The anthrax protective antigen (PA) is a key component of the tripartite anthrax toxin. Monoclonal antibody (mAb) 14B7 and its engineered, affinity-matured variants have been shown to be effective in blocking PA binding to cellular receptors and mitigating anthrax toxicity. Here, we perform computational structural modeling of the mAb 14B7-PA interaction. Our objectives are to determine the structure of the 14B7-PA complex, to deduce a structural explanation for the affinity maturation from the docking models, and to study the effect of inaccuracies in the antibody homology model on docking. Aging and symptoms of anxiety and depression: structural invariance of the tripartite model.

Negative affect measures were evaluated in a cross-sectional community sample of adults aged 18-93 (N = 335) to examine the structure of neuroticism, anxiety, and depressive symptoms in young, middle, and older adult cohorts. Structural equation modeling was used to contrast 3 nested models: a 1-factor general distress model; a 2-factor high negative-low positive affect model; and a 3-factor "tripartite model" reflecting a higher order Negative Affect factor that is common to depression and anxiety problems and 2 lower order factors, Low Positive Affect (mostly specific to depression) and Arou

Solution structures of chromium(VI) complexes with glutathione and model thiols.

Chromium(VI) complexes of the most abundant biological reductant, glutathione (gamma-Glu-Cys-Gly, I), are among the likely initial reactive intermediates formed during the cellular metabolism of carcinogenic and genotoxic Cr(VI). Detailed structural characterization of such complexes in solutions has been performed by a combination of X-ray absorption fine structure (XAFS) and X-ray absorption near-edge structure (XANES) spectroscopies, electrospray mass spectrometry (ESMS), UV-vis spectroscopy, and kinetic studies. The Cr(VI) complexes of two model thiols, N-acetyl-2-mercaptoethylamine (II) a

Predicting the outcome of eating disorders using structural equation modeling.

OBJECTIVE: There is a need for models that predict accurately the course of mental disorders. METHOD: Eating-disordered female inpatients were assessed longitudinally at the beginning of treatment (t1), at the end of treatment (t2), at 2 or 3-year follow-up (t3), and at 6-year follow-up (t4). The sample consisted of 196 women with bulimia nervosa (BN) purging type, 103 women with anorexia nervosa (AN), and 68 women with binge eating disorder (BED; N=367). Confirmatory factor analysis and path analysis were used to predict the women's status at 6-year follow-up. RESULTS: The results for BN an

Sequencing and model structure of a Naja naja atra protein fragment.

We report the amino acid sequence of a basic protein isolated from the snake venom of Naja naja atra. An automated Edman sequencer was used to determine the 65-residue sequence, aided by electrospray ionization/mass spectrometry. Online reduction and pyridylethylation of the peptide was performed to identify the cysteine residues. Trypsin, chymotrypsin and aspartic digestions were carried out to derive peptide fragments for further sequencing. Fragmented peptides were overlapped to obtain the complete sequence. Molecular mass measurements of the whole protein and its fragments were used as a c

Analytical results on the stability of age-structured recurrent epidemic models.

This paper examines the equilibrium and stability properties of relatively simple mathematical models for the transmission of infectious diseases such as measles, rubella, and mumps. We consider endemic diseases which are recurrent over a long time period. We discuss some simple models which incorporate an age structure into the population amongst whom the disease is spreading, since recent work has shown this feature to be important. The results are relevant in three ways: for predicting the long-term overall level of incidence of disease; for describing the oscillations of the incidence of d

A common structural model for central nervous system drugs and their receptors.

On the basis of the hypothesis that there is a common structural basis for central nervous system (CNS) drug action consisting primarily of an aromatic group and a nitrogen atom, a four-point model for a common pharmacophore is defined with use of five semirigid CNS-active drug molecules: morphine, strychnine, LSD, apomorphine, and mianserin. Two of the points of the model represent possible hydrophobic interactions between the aromatic group and the receptor, while the other two represent hydrogen bonding between the nitrogen atom and the receptor. The model is then extended by the inclusion

From Crisis to Meaning: A Structural Equation Model of Adjustment to Bereavement.

Experiencing the death of a loved one often leads to a crisis of meaning, prompting efforts to restore coherence through meaning-making. Although several models have addressed this process, few studies have empirically examined how distinct forms of meaning (situational and global) emerge and interrelate. The present study used structural equation modelling to investigate meaning-making in bereavement. A model was developed incorporating variables from existing frameworks (crisis of meaning, distress, deliberate and intrusive rumination, emotional expression) and existential constructs (uncert

Conflict Escalation in Intimate Relationships: An Event-Based Structural Model.

Research on intimate partner violence has traditionally focused on discrete aggressive acts and their frequency, with less attention to how appraisal, emotion, and response readiness are organized within specific conflict episodes. This study advances an event-based structural model of conflict escalation in intimate relationships by examining how appraisal processes, negative emotional activation, escalation tendency, and behavioral responding cohere within standardized conflict events. Using a vignette-based survey of 2,702 adults in long-term intimate relationships, participants responded t

Measuring and modelling the structural dynamics of poricidal anthers: a mechatronic approach.

The mechanical dynamics of poricidal stamens play a crucial role in buzz pollination, but they have only been studied in a few species showing comparatively less curvature. This paper presents an integrated experimental-computational approach to characterize the dynamic behaviour of the morphologically more complex stamens of Medinilla magnifica. An experimental set-up employing a custom-built shaker and a laser triangulation sensor is developed to measure the stamen motion with high spatial and temporal resolution. The resulting displacement transmissibility reveals three distinct resonances,

Building the model: a review of input structures in extreme temperature-health.

BACKGROUND: The health impacts of extreme temperatures have been extensively studied through epidemiological models. However, limited attention has been paid to the specification of these models, particularly regarding input structure and model selection. Although exposure metrics and statistical techniques have evolved over time, a comprehensive synthesis of the variables included in these models, and the rationale behind their inclusion, is still lacking. This gap limits the comparability of studies and may compromise the robustness of temperature-health evidence. METHODS: We conducted a sy

Dynamical analysis of a spatially heterogeneous disease model with degenerate structure.

This study analyzes a reaction-diffusion model for cocoa black pod disease (CBPD) with space-dependent parameters and partial degeneracy, which assumes that susceptible and infected cocoa pods remain immobile within the region. Spores produced by infected pods, which drive both direct and indirect infections, are assumed to disperse at distinct rates depending on their mode of transmission. Firstly, we establish the model's well-posedness, including the boundedness and global existence of solutions. Furthermore, to address the non-compactness of solution semiflow, we demonstrate the asymptotic

Patient-Specific Mobilization: A Structured, Systemwide Model of Mobility Care.

Hospital-associated functional decline resulting from immobility and prolonged bed rest is a widespread yet underrecognized complication in acute care. Despite strong evidence linking immobility directly to severe in-hospital complications and significant postdischarge impairments - including reduced independence, diminished quality of life, and impaired ability to perform activities of daily living (ADLs) - many U.S. hospitals lack standardized mobility care programs. Mobility practices vary widely and are typically driven by individual clinician knowledge, institutional culture, and informal

A Dual-age Structured Epidemiological Model with Waning Immunity and Reinfection.

Waning immunity and reinfection are critical features of many infectious diseases, but epidemiological models often fail to capture the interaction between an individual's immunity history and their current infection status, or do so only simplistically. We develop a dual-age structured model tracking immunity age (time since last recovery) and infection age (time since infection) to analyze epidemic dynamics under waning immunity and reinfection. The model is formulated as a system of age-structured partial differential equations describing susceptible and infected populations stratified by b

Dynamising One Health Through a Structured Global Networking Model.

Global environmental change, biodiversity loss, emerging infectious diseases, antimicrobial resistance, and widening social inequalities increasingly interact to create complex risks for human, animal, and ecosystem health. Although international One Health frameworks and funding mechanisms are essential, implementation at national and especially community levels often remains uneven, fragmented, and weakly evaluated. This Perspective presents a structured, reproducible networking model designed to strengthen community-level One Health implementation through voluntary global collaboration amon

A structure-function neuronal network model of the rat nervous system.

A consensus description of global vertebrate nervous system organization-its basic plan or wiring diagram-has not emerged after centuries of research. Our approach to the problem is based on published experimental neuroanatomical connection data that describe the adult female and male rat nervous system's intrinsic neuronal network organization at the interregional level of scale that is inherited and species specific. This bilateral network has 924 region nodes with a projected 81,582 directed and weighted axonal connections (edges) between them, giving a network density of 9.6%. On average t

Modeling COVID-19 transmission: effects of age structure and vaccination.

A mathematical model for COVID-19 dynamics is developed, incorporating age structure, disease progression, and vaccination. Addressing gaps in existing literature, the model integrates heterogeneous intercohort mixing for realistic disease transmission, with a primary focus on Pakistan and global applicability. Well-posedness is established via the abstract Cauchy problem framework. Threshold parameters and stability analysis identify conditions for disease persistence or eradication. An age-free sub-model gives additional insights. Numerical simulations using the finite differences method con

A structural and mechanistic model for BSEP dysfunction in PFIC2 cholestatic disease.

BSEP (ABCB11) transports bile salts across the canalicular membrane of hepatocytes, where they are incorporated into bile. Biallelic mutations in BSEP can cause Progressive Familial Intrahepatic Cholestasis Type 2 (PFIC2), a rare pediatric disease characterized by hepatic bile acid accumulation leading to hepatotoxicity and, ultimately, liver failure. The most frequently occurring PFIC2 disease-causing mutations are missense mutations, which often display a phenotype with decreased protein expression and impaired maturation and trafficking to the canalicular membrane. To characterize the mutat
