[Liver monooxygenase system inducers in the treatment of respiratory distress syndrome in newborn].

Physical and biochemical parameters of pulmonary surfactant (PS) were studied in 6-day-old rabbits with the respiratory distress syndrome (RDS) treated by benzonal and zixorin inductors. Surface-active characteristics of PS were impaired under conditions of RDS at the expense of deficiency of total phospholipids, specifically phosphatidylcholine (PC) and phosphatidylethanolamine (PEA). Treatment with benzonal and zixorin improved the surface-active characteristics of PS and increased the content of total phospholipids mainly at the expense of PC and PEA.

An investigation into the cytotoxic effects of microbubbles and their constituents on osteosarcoma and bone marrow stromal cells.

BACKGROUND: Ultrasound-responsive microbubbles offer a means of achieving minimally invasive, localised drug delivery in applications including regenerative medicine. To facilitate their use, however, it is important to determine any cytotoxic effects they or their constituents may have. The aim of this study was to test the hypothesis that phospholipid-shelled microbubbles are non-toxic to human bone-derived cells at biologically-relevant concentrations. METHODS: Microbubbles were fabricated using combinations of 1,2-distearoyl-sn-glycero-3-phosphocholine (DSPC), 1,2-dibehenoyl-sn-glycero-3-

Alterations of phospholipid metabolites in postmortem brain from patients with Alzheimer's disease.

To test the hypothesis that brain cell membranes degenerate in Alzheimer's disease (AD), we measured the levels of phospholipids, their water-soluble metabolites, and glycerophosphocholine (GPC) cholinephosphodiesterase activity in postmortem brain tissue from patients with AD and age-matched controls. We found significantly higher levels of the phospholipid catabolite GPC in AD brain. In contrast, choline and ethanolamine levels were significantly lower in AD, and phospholipid levels were slightly decreased. Furthermore, in AD the activity of the GPC-degrading enzyme GPC cholinephosphodiester

[Measurement of phospholipids in erythrocytes, plasma and platelets in a patient with hereditary spherocytosis].

The amount and distribution of phospholipids (PLs) were studied in erythrocytes, plasma and platelets of one patient with hereditary spherocytosis (HS). Phosphatidylethanolamine (PE) was always decreased. Total PLs amount was remarkably reduced in erythrocytes but normal in platelets. Since platelets do not contain those proteins which have been implicated in the red cell abnormalities of patients with HS, our findings reinforce the theory that a defective protein-lipid interaction causes the abnormal lipid loss in the erythrocytes of these patients.

Common and varied molecular responses of Escherichia coli to five different inhibitors of the lipopolysaccharide biosynthetic enzyme LpxC.

A promising yet clinically unexploited antibiotic target in difficult-to-treat Gram-negative bacteria is LpxC, the key enzyme in the biosynthesis of lipopolysaccharides, which are the major constituents of the outer membrane. Despite the development of dozens of chemically diverse LpxC inhibitor molecules, it is essentially unknown how bacteria counteract LpxC inhibition. Our study provides comprehensive insights into the response against five different LpxC inhibitors. All compounds bound to purified LpxC from Escherichia coli. Treatment of E. coli with these compounds changed the cell shape

Lipidomics reveals the reshaping of the mitochondrial phospholipid profile in cells lacking OPA1 and mitofusins.

Depletion or mutations of key proteins for mitochondrial fusion, like optic atrophy 1 (OPA1) and mitofusins 1 and 2 (Mfn 1 and 2), are known to significantly impact the mitochondrial ultrastructure, suggesting alterations of their membranes' lipid profiles. In order to make an insight into this issue, we used hydrophilic interaction liquid chromatography coupled with electrospray ionization-high resolution MS to investigate the mitochondrial phospholipid (PL) profile of mouse embryonic fibroblasts knocked out for OPA1 and Mfn1/2 genes. One hundred sixty-seven different sum compositions were re

In vitro and in vivo models of Huntington's disease show alterations in the endocannabinoid system.

In this study, we analyzed the components of the endocannabinoid system (ECS) in R6/2 mice, a widely used model of Huntington's disease (HD). We measured the endogenous content of N-arachidonoylethanolamine and 2-arachidonoylglycerol and the activity of their biosynthetic enzymes (N-acyl-phosphatidylethanolamine-hydrolyzing phospholipase D and diacylglycerol lipase, respectively) and hydrolytic enzymes [fatty acid amide hydrolase (FAAH) and monoacylglycerol lipase, respectively] and of their target receptors (type 1 cannabinoid receptor, type 2 cannabinoid receptor, and transient receptor pote

Tissue-specific differences in brain phosphodiesters in late-life major depression.

OBJECTIVE: Late-life depression has been hypothesized to have a neurodegenerative component that leads to impaired executive function and increases in subcortical white matter hyperintensities. Phosphorus magnetic resonance spectroscopy (MRS) can quantify several important phosphorus metabolites in the brain, particularly the anabolic precursors and catabolic metabolites of the constituents of cell membranes, which could be altered by neurodegenerative activity. METHODS: Ten patients with late-life major depression who were medication free at time of study and 11 aged normal comparison subjec

Rapid chromatographic method to decipher distinct alterations in lipid classes in NAFLD/NASH.

AIM: To establish a simple method to quantify lipid classes in liver diseases and to decipher the lipid profile in p62/IMP2-2/IGF2BP2-2 transgenic mice. METHODS: Liver-specific overexpression of the insulin-like growth factor 2 mRNA binding protein p62/IMP2-2/IGF2BP2-2 was used as a model for steatosis. Steatohepatitis was induced by feeding a methionine-choline deficient diet. Steatosis was assessed histologically. For thin layer chromatographic analysis, lipids were extracted from freeze-dried tissues by hexane/2-propanol, dried, redissolved, and chromatographically separated by a two-solve

Phospholipid antibodies in alcoholic liver disease.

Alcoholic liver injury has been reported to be directed preferentially against the proteins of the cell membrane, sparing the phospholipids. However, antiphospholipid antibodies against certain cell membrane phospholipids are known to be associated with a variety of diseases. We undertook this investigation to determine whether antiphospholipid antibodies were present in the serum of patients with alcoholic liver disease. We investigated seventy long-term alcoholic patients (> 80 gm ethanol/day for > 1 yr) and 8 normal nonalcoholic controls by means of enzyme-linked immunosorbent assay to dete

Role of Lipid Signaling by Platelet-Activating Factor Receptor in Tubular Epithelial Cells in Acute Kidney Injury-to-CKD Transition.

KEY POINTS: Platelet-activating factor receptor contributed to renal tubular epithelial cells G2/M arrest through suppressing mouse double minute 2-mediated p53 ubiquitin degradation in AKI-to-CKD transition. Phosphatidylethanolamine (18:0/18:1) served as a novel endogenous ligand of platelet-activating factor receptor, inducing the downstream signaling. WAY-639497, a small-molecule platelet-activating factor receptor antagonist identified by virtual screening, mitigated renal tubular epithelial cells G2/M arrest and AKI-to-CKD transition. BACKGROUND: AKI represents a critical clinical compli

Concentration-Dependent Modes of Melittin-Induced Membrane Destabilization: An Integration of Experimental and Computational Tools.

Melittin is a well-known antimicrobial peptide whose mechanism of action is still not entirely understood. In this study, we combine complementary experimental techniques with molecular dynamics simulations to determine melittin-membrane interactions at different scales. Using 1,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE): 1,2-dipalmitoyl-sn-glycero-3-phospho-(1'-rac-glycerol) (DPPG) membranes that mimic the Escherichia coli inner membrane, this integrative approach reveals a clear transition in melittin's mode of action with increasing peptide loading. At low concentrations, melittin p

Analysis of Interfacial Membrane Polarity of Brain-Related Lipids via Flattened Tetrahedron Phase Diagram.

Ternary phase diagrams are an important tool in analyzing the interactions between different lipid components in the membrane; however, the maximum analytical capacity of 3 lipids is their primary limitation. Phosphatidylethanolamine (PE) is one of the 4 most abundant lipids in the brain that has its phase characteristics and interactions with the other abundant brain lipids [sphingolipids, phosphatidylcholine (PC), and cholesterol] yet to be extensively analyzed. This study had two primary objectives: first, we examined DOPE (an analog for PE) as an important fourth lipid component that may h

Engineering yeast to induce the synthesis of GPI-APs with a permanent phosphoethanolamine on mannose 2 of the glycan moiety.

GPI-APs are a family of proteins attached to the plasma membrane by a glycoplipid that undergoes remodeling of the glycan and lipid structure during transport to the cell surface. We describe a protocol to induce the synthesis of a GPI-anchored protein whereby EtNP is added to Man2 but not removed. By temporally manipulating the expression of Gpi7p, the enzyme that adds EtNP to Man2, in ted1Δ dcr2Δ cells prior to the expression of a canonical GPI-AP (mNeon-Gas1p), EtNP is attached to Man2 of de novo synthesized mNeon-Gas1p and cannot be removed. This strategy provides a means to temporally and

Immunocytochemical localization of choline-phospholipids in postnatal mouse molars.

This localization was studied using anti-choline phospholipid monoclonal IgM antibody (MC22-33F). By immunofluorescence, the apical portions of enamel-secreting ameloblasts and dentine-secreting odontoblasts were positively stained with MC22-33F. By immunoelectron microscopy, lysosomes in preodontoblasts, odontoblasts, preameloblasts and ameloblasts as well as matrix vesicles in dentine were strongly stained with MC22-33F. On the other hand, plasma membrane of these cells did not stain with MC22-33F. The frequency of positively reacting lysosomes in ameloblasts and odontoblasts and of matrix v

Osteosarcoma and other tumors of bone.

Steady progress has been made in the identification of genetic alterations and prognostic factors in osteosarcoma. Imaging studies continue to be employed not only for preoperative staging, but to quantify factors such as tumor bulk and tumor response to neoadjuvant chemotherapy. Although there have been no major refinements of local or systemic therapy, continuing research into novel agents (eg, liposomal muramyl, tripeptide phosphatidylethanolamine, and antisense oligonucleotides to insulin-like growth factor receptors) hold promise for new therapies for this disease in the future.

Abnormal excretion of urinary phospholipids and sulfatide in patients with mitochondrial encephalomyopathies.

We found that patients with mitochondrial encephalomyopathies excreted urinary phosphatidylethanolamine, cardiolipin, and phosphatidylserine most likely derived from mitochondria and sulfatide which is specific to myelin or the kidney. It is of interest that four patients with myoclonus epilepsy with ragged-red fibers and one patient with chronic progressive external ophthalmoplegia all showed qualitatively similar abnormal excretion of such urinary lipids. It is conceivable that the urinary acidic phospholipids reflect abnormalities in the mitochondrial phospholipids, which are very important
