3,5-Dicaffeoylquinic acid attenuates poststroke muscle atrophy through GPX4 dependent ferroptosis inhibition.

BACKGROUND: Ischemic stroke-induced sarcopenia (ISS) is a major contributor to poststroke motor dysfunction and severely impairs quality of life. Ferroptosis, an iron- and lipid peroxidation- dependent form of programmed cell death, has not been fully explored in ISS. PURPOSE: To evaluate the natural compound 3,5-dicaffeoylquinic acid (3,5-DCQA) as a novel anti-ferroptosis agent that markedly attenuates ISS via glutathione peroxidase 4 (GPX4)-mediated ferroptosis inhibition. METHODS: A permanent middle cerebral artery occlusion (pMCAO) rat model was established. Cerebral injury was assessed

Synergistic Effects of Pyrrosia lingua Caffeoylquinic Acid Compounds with Levofloxacin Against Uropathogenic Escherichia coli: Insights from Molecular Dynamics Simulations, Antibiofilm, and Antimicrobial Assessments.

Urinary tract infections (UTIs), primarily caused by uropathogenic Escherichia coli (UPEC), have high morbidity and recurrence rates. Resistance to levofloxacin hydrochloride (LEV), a commonly used treatment for UTIs, is increasingly problematic, exacerbated by biofilm formation mediated by interactions between cyclic di-GMP (c-di-GMP or CDG) and YcgR. In this study, we identified three caffeoylquinic acid compounds from Pyrrosia lingua-chlorogenic acid (CGA), sibiricose A5 (Si-A5), and 3-O-caffeoylquinic acid methyl ester (CAM)-that target YcgR through molecular docking. Biological assays rev

Shotgun chemome characterization of Artemisia rupestris L. Using direct infusion-MS/MSALL.

In comparison of liquid chromatography, direct infusion is a superior choice to achieve high-throughput measurements. The specificity and selectivity of tandem mass spectrometry (MS/MS) actually result in a so-called MS separation potential when chemical characterization of herbal medicines. Here, a MS/MSALL program was introduced to promote DI-MS/MS to be an eligible tool for shotgun chemome characterization of Artemisia rupestris L. that is currently drawing worldwide interests because of the promising antiviral activity. After MS1 spectral acquisition for the crude extract, the gas phase fr

Nanomolar Inhibitors of Trypanosoma brucei RNA Triphosphatase.

UNLABELLED: Eukaryal taxa differ with respect to the structure and mechanism of the RNA triphosphatase (RTPase) component of the mRNA capping apparatus. Protozoa, fungi, and certain DNA viruses have a metal-dependent RTPase that belongs to the triphosphate tunnel metalloenzyme (TTM) superfamily. Because the structures, active sites, and chemical mechanisms of the TTM-type RTPases differ from those of mammalian RTPases, the TTM RTPases are potential targets for antiprotozoal, antifungal, and antiviral drug discovery. Here, we employed RNA interference (RNAi) knockdown methods to show that Trypa
