Divergent Synthesis of Chromenoindoles and Spiroindolines via Domino Reaction of Indolyl-Substituted Isocyanides with Quinone Esters.

A dearomative spirocyclization of tryptamine-derived isocyanides with quinone esters is developed for the divergent synthesis of structurally complex chromeno[2,3-b]indole and polycyclic spiroindoline scaffolds. This domino reaction features the formation and conversion of the six-membered dihydropyran ring with an amendable N,O-aminal moiety.

Pyrroloquinoline Quinone Modifies Lipid Profile, but Not Insulin Sensitivity, of Palmitic Acid-Treated L6 Myotubes.

Pyrroloquinoline quinone (PQQ) is a novel stimulator of mitochondrial biogenesis and cellular energy metabolism. This is the first study investigating regulatory mechanisms and metabolic responses underlying PQQ's action in palmitate-exposed L6 myotubes. Particularly, we assessed alterations in lipid content and composition, expression of metabolic enzymes, and changes in glucose transport. The experiments were conducted using muscle cells subjected to short (2 h) and prolonged (24 h) incubation with PQQ in a sequence of pre- and post-palmitic acid (PA) exposure. We demonstrated the opposite e

Chemoproteomics of an Indole-Based Quinone Epoxide Identifies Druggable Vulnerabilities in Vancomycin-Resistant Staphylococcus aureus.

The alarming global rise in fatalities from multidrug-resistant Staphylococcus aureus (S. aureus) infections has underscored a need to develop new therapies to address this epidemic. Chemoproteomics is valuable in identifying targets for new drugs in different human diseases including bacterial infections. Targeting functional cysteines is particularly attractive, as they serve critical catalytic functions that enable bacterial survival. Here, we report an indole-based quinone epoxide scaffold with a unique boat-like conformation that allows steric control in modulating thiol reactivity. We ex

Straightforward Synthesis of Bifunctional Phosphorus Phenols via Phosphination of In Situ Generated o-Quinone Methides.

An efficient and practical approach towards bifunctional phosphorus phenols has been developed through a reaction of diphenylphosphine oxide and the o-quinone methides in situ generated from 2-tosylalkyl phenols under basic conditions. This protocol features simple experimental procedures under mild conditions and is easily scaled up. With this method, a variety of diarylmethyl phosphine oxides can be produced with up to 92% yield.

New ortho-quinone methide formation: application to three-component coupling of isocyanides, aldehydes and phenols.

Herein, we wish to report a new three-component formation of heterocyclic scaffolds based on a one-pot process from simple phenols. The key step of this procedure involves an ortho-quinone methide formation from Mannich adducts under alkylative conditions. The transient o-quinone methide has been trapped in situ with indole and diketone using lithium perchlorate as catalyst. The interest of this procedure has been furthermore demonstrated by a new three-component aminobenzofuran formation from phenols, aldehydes and isocyanides.

Functional implications of antiestrogen induction of quinone reductase: inhibition of estrogen-induced deoxyribonucleic acid damage.

Recent studies have shown that the antiestrogens tamoxifen and raloxifene may protect against breast cancer, presumably because of a blockade of estrogen receptor (ER)-mediated transcription. Another possible explanation is that antiestrogen-liganded ER transcriptionally induces genes that are protective against cancer. We previously reported that antiestrogen-liganded ERbeta transcriptionally activates the major detoxifying enzyme quinone reductase (QR) [NAD(P)H:quinone oxidoreductase]. It has been established that metabolites of estrogen, termed catecholestrogens, can form DNA adducts and ca

Dose-specific production of chlorinated quinone and semiquinone adducts in rodent livers following administration of pentachlorophenol.

Production of chlorinated quinoid metabolites was investigated in the livers of Sprague-Dawley rats and B6C3F1 mice following single oral administration of pentachlorophenol (PCP) (0-40 mg/kg body weight) and in male Fischer 344 rats, following chronic ingestion of PCP at 1,000 ppm in the diet for 6 months (equivalent to 60 mg PCP/kg body weight/day). Analyses of the rates of adduction in the livers of Sprague-Dawley rats and B6C3F1 mice suggested that the production of tetrachloro-1,2-benzosemiquinone (Cl4-1,2-SQ) adducts was proportionally greater at low doses of PCP (less than 4-10 mg/kg bo

Synthesis and anti-inflammatory structure-activity relationships of thiazine-quinoline-quinones: inhibitors of the neutrophil respiratory burst in a model of acute gouty arthritis.

Sixteen new thiazine-quinoline-quinones have been synthesised, plus one bicyclic analogue. These compounds inhibited neutrophil superoxide production in vitro with IC(50)s as low 60 nM. Compounds with high in vitro anti-inflammatory activity were also tested in a mouse model of acute inflammation. The most active compounds inhibited both neutrophil infiltration and superoxide production at doses 2.5 micromol/kg, highlighting their potential for development as novel NSAIDs.

Dermal absorption and percutaneous penetration of p-phenylenediamines (PPDs) and p-phenylenediamine quinones (PPD-Qs): Mechanisms and implications for human dermal exposure risks.

Dermal absorption constitutes a major exposure route for p-phenylenediamine antioxidants (PPDs) and their quinone derivatives (PPD-Qs), however, limited understanding of their penetration patterns and mechanisms constrains accurate risk assessment. This study employs human biomonitoring, an in vivo rat skin exposure model, and molecular docking simulations to investigate the dermal exposure risks and percutaneous penetration mechanisms of five PPDs and their corresponding PPD-Qs. These compounds were frequently detected in hand (n = 80) and forehead wipe samples (n = 74) from automobile disman

Integrated in vitro multi-endpoint framework for assessing 6PPD-Quinone cytotoxicity in fish and mammalian cell lines.

The tire wear-derived quinone N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine quinone (6PPD-Q) has attracted global attention due to its pronounced toxicity across diverse biological systems. To provide a quantitative evaluation of 6PPD-Q toxicity, we performed integrated cellular and phenotypic profiling of 6PPD-Q exposure across five cell lines: three fish-derived (PCK, CIK, RTG-2) and two mammalian cell lines (CHO-K1, Neuro-2a). Beyond conventional MTT viability assays, LDH release measurements, morphological analysis, apoptosis quantification, and cell cycle profiling, quantitative anal

Human exposure to tire-derived p-phenylenediamines and 6PPD-quinone: Associations with thyroid nodules and mechanistic insights.

N-Substituted p-phenylenediamines (PPDs), widely used as antioxidants in rubber products, and their transformation product 6PPD-quinone (6PPD-Q) have recently emerged as contaminants of increasing environmental and health concern. However, human biomonitoring data and associated health implications remain limited. In this study, urinary concentrations of six PPDs and 6PPD-Q were quantified in 368 individuals from South China, including patients with thyroid nodules (TNs, n = 120) and controls (n = 248). PPDs were widely detected, with detection frequencies ranging from 40% to 89%, indicating u

Molecular insights into the anti-COVID-19 activity of reduced (quinol) and oxidized (quinone) forms of coenzyme Q10.

Coronavirus disease 2019 (COVID-19), caused by SARS-CoV-2, was declared a global pandemic by the World Health Organization in March 2020 and rapidly emerged as a systemic mitochondrial disorder rather than a pure respiratory illness. The disease disrupts mitochondrial bioenergetics, redox balance, and electron transport, with significant impairment of the quinone-quinol pool. Coenzyme Q10 (CoQ10), a central component of the mitochondrial respiratory chain, shuttles electrons between complexes I/II and III through dynamic interconversion between its oxidized quinone (CoQ; QN) and reduced quinol

Integrated machine learning and experimental validation reveal S6K2 as a key target of 6PPD-quinone in bladder cancer.

Tire and Road Wear Particles (TRWP) are pervasive environmental contaminants, yet the molecular mechanisms linking their toxic derivative, 6PPD-quinone (6PPD-Q), to bladder cancer (BLCA) progression remain obscure. This study integrates network toxicology with experimental validation to elucidate this complex pathogenicity. We screened six representative TRWP compounds and utilized a comprehensive machine learning framework involving 113 model combinations, identifying the Gradient Boosting Machine (GBM) as the optimal classifier. Crucially, SHAP interpretability analysis revealed RPS6KB2 (S6K

Respiratory toxicity mechanism of 6PPD and 6PPD-quinone: An integrated study based on network toxicology and molecular docking.

The widespread distribution of environmental contaminants poses a significant threat to public health. N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD) and its ozone-derivative, 6PPD-quinone (6PPD-Q), are emerging pollutants that propagate through particulate matter and aerosols, exerting detrimental effects on the respiratory system. However, their specific pathogenic mechanisms remain unclear. This study employs integrated network toxicology and molecular docking to elucidate the molecular basis of 6PPD/6PPD-Q-induced respiratory toxicity. Respiratory hazard potential was predicted
