PURPOSE: The differential gene expression of neural precursor cell expressed developmentally downregulated 9 (NEDD9) in human trabecular meshwork (HTM) cells after dexamethasone (Dex) treatment was confirmed through gene expression profiling. However, the regulatory mechanism of NEDD9 expression in HTM cells remains unknown. In this study, we investigated NEDD9 expression in HTM cells and gained a better understanding of glucocorticoid-induced glaucoma (GIG) pathophysiology.
METHODS: The Gene Expression Omnibus database and GEO2R tool were used to identify differentially expressed genes in th

Study of rare genetic disorder often provides fundamental insights into the pathology of common diseases. Cherubism is a rare craniofacial disorder in children characterized by the destruction of maxillary and mandibular bones due to expansile fibrous inflammatory lesions. Genetic study of cherubism families discovered that gain-of-function mutations in the signaling adaptor protein SH3BP2 are responsible for cherubism. Analysis of the mouse model revealed that cherubism is an autoinflammatory disorder that is caused by dysregulated signaling pathway mediated by toll-like receptors and spleen

DIAPH1 is a member of the family of Diaphanous Related Formins (DRFs) implicated in cell migration and cytokinesis. DRFs are maintained in an autoinhibited state by the intramolecular association between diaphanous inhibitory (DID) and diaphanous autoregulatory (DAD) domains. Actin polymerization requires the binding of activated RhoA to the GTPase binding domain (GBD) of DIAPH1 and the dissociation of DAD. In the presence of excess RhoA, actin polymerization is only partially activated. Using monomeric domain constructs of DIAPH1, the sequential binding affinities of RhoA and DAD to GBD-DID w

Cilia are essential for the development and function of many different tissues. Although cilia machinery is crucial in the eye for photoreceptor development and function, a role for cilia in early eye development and morphogenesis is still somewhat unclear: many zebrafish cilia mutants retain cilia at early stages due to maternal deposition of cilia components. An eye phenotype has been described in the mouse Arl13 mutant, however, zebrafish arl13b is maternally deposited, and an early role for cilia proteins has not been tested in zebrafish eye development. Here we use the zebrafish dzip1 mut

Podocyte dysfunction contributes to proteinuric chronic kidney disease. A number of key proteins are essential for podocyte function, including nephrin, podocin, CD2-associated protein (CD2AP), synaptopodin, and α-actinin-4 (ACTN4). Although most of these proteins were first identified through genetic studies associated with human kidney disease, subsequent studies have identified phosphorylation of these proteins as an important posttranslational event that regulates their function. In this review, a brief overview of the function of these key podocyte proteins is provided. Second, the role o

The actin cytoskeleton is a dynamic structure recognized for many years as integral to the coupling of external stimuli to cell activation and ensuing changes in morphology and movement. It is only recently, however, that a molecular understanding of actin involvement in these activities has emerged coincident with the identification of cytosolic signaling effectors that couple extracellular stimuli to induction of actin nucleation. Notable among these actin regulatory effectors are members of the Wiskott-Aldrich syndrome protein (WASp) family, a group of cytoskeletal adaptors imbued with the

Cellular plasticity allows organisms to adapt dynamically to both physiological and pathological contexts. Epithelial-mesenchymal transition (EMT) is a well-known example of this plasticity and is now widely recognized as a reversible and highly dynamic spectrum of cellular states rather than a simple binary switch. In this review, we summarize current knowledge on the Hippo pathway transcriptional co-activators YAP and TAZ, focusing on their role as a central hub that integrates mechanical, biochemical and metabolic signals from the microenvironment to control cell fate reprogramming. We disc

Osteoporosis is a systemic skeletal disorder fundamentally characterized by an imbalance between bone formation and bone resorption, ultimately leading to reduced bone mass, microarchitectural deterioration, and increased fracture susceptibility. As bone is a highly mechanoresponsive tissue, mechanical stimulation is indispensable for the maintenance of skeletal homeostasis, and insufficient loading is a recognized driver of bone loss. In recent years, Piezo1, integrins, and YAP/TAZ have emerged as key nodes in bone cell mechanotransduction. Piezo1 functions primarily as a mechanosensitive ion

The Hippo-YAP signaling pathway is crucial for tissue homeostasis, yet its specific contribution to recurrent implantation failure (RIF) and recurrent pregnancy loss (RPL) remains poorly understood. This study investigated Hippo-YAP signaling in physiological and pathological human endometrium, and its functional impact on endometrial receptivity and decidualization. We analyzed endometrial tissues from fertile controls, RIF patients, and RPL patients, complemented by mechanistic studies in Ishikawa cells and telomerase-immortalized human endometrial stromal cells. In fertile controls, YAP was

Spinal cord injury (SCI) triggers a cascade of secondary events, among which neuroinflammation plays a central role in driving tissue damage and functional deficits. Inflammasomes are multiprotein complexes that sense cellular stress and mediate caspase-1 activation, interleukin (IL)-1β and IL-18 maturation, and pyroptotic cell death. While the NLRP3 inflammasome has been extensively studied in SCI, emerging evidence suggests that other inflammasomes, including NLRP1, AIM2, and NLRC4, may also contribute to the neuroinflammatory process. This review summarizes current knowledge on non-NLRP3 in

Kelch-like protein 3 (KLHL3) is a substrate adaptor of Cullin3-RING ubiquitin ligase (CRL3), and KLHL3-CUL3 complex plays a vital role in the ubiquitination of specific substrates. Mutations and abnormal post-translational modifications of KLHL3-CUL3 affect substrate ubiquitination and may related to the pathogenesis of Gordon syndrome (GS), Primary Hyperparathyroidism (PHPT), Diabetes Mellitus (DM), Congenital Heart Disease (CHD), Pre-eclampsia (PE) and even cancers. Therefore, it is essential to understand the function and molecular mechanisms of KLHL3-CUL3 for the treatment of related disea

Targeted protein degradation via molecular glues represents a powerful modality for modulating "undruggable" proteins. Herein, through proteomic profiling of a CRBN-binding library and rigorous structure-activity relationship (SAR) refinement, we report the discovery of dWBP4-1: a first-in-class, highly selective, CRBN-dependent molecular glue degrader of the spliceosome-associated scaffold protein WBP4. dWBP4-1 induces rapid, nanomolar degradation of WBP4 via a canonical G-loop-mediated mechanism, exhibiting exceptional proteome-wide selectivity with negligible transcriptomic or alternative s

Bone-muscle diseases, such as osteoporosis, rheumatoid arthritis, sarcopenia and cachexia, represent a growing global health concern, particularly among aging populations and older adults. These multifactorial disorders are characterized by progressive decline in bone density and muscle mass, increasing the chances of immobility and eventually disability. Such manifestations are driven by a complex molecular crosstalk between bones and muscles. This review highlights the key role of the irisin-sclerostin-inflammation triad in the pathophysiology of musculoskeletal degeneration. Irisin is a myo

To date it is only partially understood how the brain is cleared of waste products resulting from its high metabolic activity, although this process has important implications for the development and progression of neurodegenerative diseases. Lymphatic vessels play a central role in maintaining fluid and tissue homeostasis, and the recent description of meningeal lymphatic vessels within the dura mater of mice, human and zebrafish has raised considerable interest in unraveling the function of these vessels. In zebrafish, brain lymphatic endothelial cells (BLECs) constitute an additional mening

Recently, the development of protein degraders (protein-degrading compounds) has prominently progressed. There are two remarkable classes of protein degraders: proteolysis-targeting chimeras (PROTACs) and molecular glue degraders (MGDs). Almost 70 years have passed since thalidomide was initially developed as a sedative-hypnotic drug, which is currently recognized as one of the most well-known MGDs. During the last two decades, a myriad of PROTACs and MGDs have been developed, and the molecular mechanism of action (MOA) of thalidomide was basically elucidated, including identifying its molecul

BACKGROUND: Mutations to the co-chaperone protein BAG3 (B-cell lymphoma-2-associated athanogene-3) are a leading cause of dilated cardiomyopathy (DCM). These mutations often impact the C-terminal BAG domain (residues 420-499), which regulates heat shock protein 70-dependent protein turnover via autophagy. While mutations in other regions are less common, previous studies in patients with DCM found that co-occurrence of 2 BAG3 variants (P63A, P380S) led to worse prognosis. However, the underlying mechanism for dysfunction is not fully understood.

During embryonic development, the forebrain roof plate undergoes invagination, leading to separation of the cerebral hemispheres. Any defects in this process, in humans, lead to middle interhemispheric holoprosencephaly (MIH-HPE). In this study, we have identified a previously unreported downstream mediator of retinoic acid (RA) signaling, CNKSR2, which is expressed in the forebrain roof plate in the chick embryo. Knockdown of CNKSR2 affects invagination, cell proliferation and patterning of the roof plate, similar to the phenotypes observed upon inhibition of RA signaling. We further demonstr

OBJECTIVE: Recurrent pregnancy loss (RPL) is a common infertility-related complication that affects approximately 1-3 % of women worldwide. Known causes of etiology are found in approximately half the cases but the other half remain unexplained. It is estimated that several thousands of genes contribute to reproductive success in mammals and the genetic causes of RPL cannot be fully addressed through targeted genetic tests. In recent years, massive parallel sequencing technologies has helped discovering many causal mutations in hereditary diseases such as RPL.

BACKGROUND: Maffucci's syndrome is characterized by the coexistence of multiple enchondromas and soft-tissue hemangiomas. It has been clear that somatic mosaic isocitrate dehydrogenase type 1 (IDH1) or isocitrate dehydrogenase type 2 (IDH2) mutations are associated with Maffucci's syndrome and Ollier disease, but the mechanisms underlying hemangiomas of the Maffucci's syndrome is still obscure. This study aimed to determine the mechanism of hemangiomas in Maffucci's syndrome.
METHODS: We received a 26-year-old female patient with typical Maffucci's syndrome, and exome sequencing was conducted
