Expression of an immunogenic F1-V fusion protein in lettuce as a plant-based vaccine against plague.

Yersinia pestis is a pathogenic agent that causes the bubonic and pneumonic plague. The development of an efficient and low-cost oral vaccine against these diseases is highly desirable. In this study, the immunogenic fusion protein F1-V from Y. pestis was introduced into lettuce via Agrobacterium-mediated transformation, and putative transgenic lines were developed. The presence of the transgene in these putative transgenic lines was determined using polymerase chain reaction (PCR), and transgene integration and transgene copy number were confirmed following Southern blot analysis. The presenc

[Field experiment of transgenic cry1Ab insect resistant rice].

Gene cry1Ab was transformed into Minghui 63 (Oryza sativa L.) via Agrobacterium and several homozygous transgenic lines were obtained. In this study, the copy numbers, Bt protein contents, insect resistance, and main agronomic traits of these lines were examined. All these lines had single-copy number of cry1Ab through Southern blotting analysis. The line T(1Ab)-10 and its hybrid expressed the higher Cry1Ab proteins than any other lines and their corresponding hybrids when assayed by ELISA. After artificial infestation of first-instar YSB (Yellow stem borer) and natural affection of stemborers

Autoantibodies to brain components and antibodies to Acinetobacter calcoaceticus are present in bovine spongiform encephalopathy.

Bovine spongiform encephalopathy (BSE) is a neurological disorder, predominantly of British cattle, which belongs to the group of transmissible spongiform encephalopathies together with Creutzfeldt-Jakob disease (CJD), kuru, and scrapie. Autoantibodies to brain neurofilaments have been previously described in patients with CJD and kuru and in sheep affected by scrapie. Spongiform-like changes have also been observed in chronic experimental allergic encephalomyelitis, at least in rabbits and guinea pigs, and in these conditions autoantibodies to myelin occur. We report here that animals with BS

ocs element promoter sequences are activated by auxin and salicylic acid in Arabidopsis.

ocs elements are a group of promoter elements that have been exploited by two distinct groups of plant pathogens, Agrobacterium and certain viruses, to express genes in plants. We examined the activity of single and multiple ocs elements linked to a minimal plant promoter and the uidA reporter gene in transgenic Arabidopsis. beta-Glucuronidase activity was detected only in root tips and in callus tissue after auxin treatment. A more sensitive assay revealed that auxin treatment also increased ocs element activity in aerial parts of the plant, although the absolute levels of ocs element activit

Composite Cucurbitaceae Plants as a Tool to Study Physiological and Molecular Mechanisms of Root Development.

Composite plants are a convenient model for obtaining a whole plant with undisturbed regulatory links between the shoot and root system, while the roots carry the properties specified by the researcher. This approach allows expressing transgenes only in roots, which in some cases may be preferable over the expression of transgenes in all organs of a non-chimeric transgenic plant. Composite plants have been widely used to study the nitrogen fixation process, mycorrhizal symbioses, and the interaction of plant roots with various pathogens. The current protocol for Agrobacterium rhizogenes-mediat

LcNAC72-LcDREB1 Regulatory Module Mediates Low-Temperature Stress Response by Enhancing ROS Scavenging Capacity in Lonicera caerulea.

Lonicera caerulea (blue honeysuckle) is a high-value berry crop native to cold temperate regions with exceptional intrinsic freezing tolerance. However, research on the underlying mechanisms of cold resistance remains relatively scarce in blue honeysuckle. Here, we demonstrate that the L. caerulea transcription factors LcNAC72 interacts with LcDREB1, as revealed by yeast two-hybrid co-immunoprecipitation, luciferase complementation Imaging, and bimolecular fluorescence complementation assays. Agrobacterium-mediated transient overexpression showed that LcNAC72-LcDREB1 cooperatively modulate rea

Green Mold of Citrus: Recent Insights into Penicillium digitatum Pathogenicity and Biological Control Strategies.

P. digitatum, the causal agent of citrus green mold, remains the most destructive postharvest pathogen of citrus worldwide. Recent advances have greatly expanded our understanding of the molecular dialogue between P. digitatum and citrus hosts, revealing coordinated virulence strategies involving cell wall-degrading enzymes, major facilitator superfamily transporters, transcription factors, and secondary metabolism, alongside host defenses mediated by phytohormones and specialized metabolites. This review integrates genomic, transcriptomic, metabolomic, and functional genetic discoveries, incl

Genetic Manipulation in Sporothrix Species: Molecular Tools, Challenges, and Applications.

Sporothrix species are thermally dimorphic fungi responsible for sporotrichosis, a globally prevalent subcutaneous mycosis and an emerging zoonotic threat, particularly in South America. The high virulence of Sporothrix brasiliensis and its efficient transmission from cats to humans have intensified recent outbreaks, underscoring the importance of understanding the pathogenic mechanisms. While several putative virulence factors have been identified, such as melanin production, cell wall remodeling, extracellular vesicles, and thermotolerance, functional studies remain hampered by limited molec

Pathogens without borders: a review on cross-kingdom transmission strategies and pathogenicity of plant and human pathogens.

Emerging evidence indicates that certain plant pathogens-once thought to be restricted to the plant kingdom-can opportunistically infect humans, particularly those who are immunocompromised. This paradigm-shifting recognition of cross-kingdom pathogenicity underscores complex microbial adaptability and highlights an underexplored public health concern. This review is a compilation of current knowledge on bacterial (Pantoea, Burkholderia, Agrobacterium), fungal (Fusarium, Aspergillus, Curvularia), and viral (e.g., TMV, PMMoV) plant pathogens implicated in human disease. Shared eukaryotic cellul

DipTRANS: an improved method for in planta transformation and genome engineering in Nicotiana benthamiana.

Plant transformation remains constrained by labor-intensive tissue culture. Our previous work showed that direct delivery of developmental regulators (DRs) can induce de novo meristems on plants, offering a promising transformation approach. In this resource article, we introduced DipTRANS (Direct in planta Transformation), an optimized, soil-based heritable transformation platform for Nicotiana benthamiana that bypasses sterile culture entirely. DipTRANS is built on DR-induced de novo meristem formation. After optimizing parameters, including regulator combinations, Agrobacterium strain, and

Forward and reverse genetic dissection of morphogenesis identifies filament-competent Candida auris strains.

Candida auris is an emerging healthcare-associated pathogen of global concern. Recent reports have identified C. auris isolates that grow in cellular aggregates or filaments, often without a clear genetic explanation. To investigate the regulation of C. auris morphogenesis, we applied an Agrobacterium-mediated transformation system to all four C. auris clades. We identified aggregating mutants associated with disruption of chitin regulation, while disruption of ELM1 produced a polarized, filamentous growth morphology. We developed a transiently expressed Cas9 and sgRNA system for C. auris that

Cell cycle transition from S-phase to G1 in Caulobacter is mediated by ancestral virulence regulators.

Zinc-finger domain transcriptional regulators regulate a myriad of functions in eukaryotes. Interestingly, ancestral versions (MucR) from Alpha-proteobacteria control bacterial virulence/symbiosis. Whether virulence regulators can also control cell cycle transcription is unknown. Here we report that MucR proteins implement a hitherto elusive primordial S→G1 transcriptional switch. After charting G1-specific promoters in the cell cycle model Caulobacter crescentus by comparative ChIP-seq, we use one such promoter as genetic proxy to unearth two MucR paralogs, MucR1/2, as constituents of a quadr

Expression of poplar chitinase in tomato leads to inhibition of development in colorado potato beetle.

The previously described poplar chitinase, WIN6, is induced during infestation by gypsy moth (Lymantria dispar L.) larvae, thus suggesting a role in defense against insect pests. To test this hypothesis, we produced tomato seedlings infected with a recombinant potato virus X (PVX), which produces WIN6, and tested its insecticidal properties on Colorado potato beetle [CPB; Leptinotarsa decemlineata (Say)], which is a serious pest of tomatoes and other crops. The advantage of PVX is that plant material is ready for insect bioassay within 3-4 weeks of constructing the recombinant virus. Consideri

Penicillin derivatives induce chemical structure-dependent root development, and application for plant transformation.

We investigated five penicillin derivatives that are popularly used for transformation experiments with Agrobacterium rhizogenes-penicillin G, carbenicillin, ampicillin, amoxicillin and cephalexin-for their effects on the growth and morphology of Beta vulgaris, Capsicum annuum and Glehnia littoralis roots. Attention was given to the relationship between their chemical structures and functions. Ampicillin was found to stimulate root elongation but inhibit root branching, whereas carbenicillin inhibited root elongation but promoted root branching. Root cultures were also exposed to hydrolyzed pr

Brucella outer membrane lipoproteins share antigenic determinants with bacteria of the family Rhizobiaceae.

Brucellae have been reported to be phylogenetically related to bacteria of the family Rhizobiaceae. In the present study, we used a panel of monoclonal antibodies (MAbs) to Brucella outer membrane proteins (OMPs) to determine the presence of common OMP epitopes in some representative bacteria of this family, i.e., Ochrobactrum anthropi, Phyllobacterium rubiacearum, Rhizobium leguminosarum, and Agrobacterium tumefaciens, and also in bacteria reported to serologically cross-react with brucella, i.e., Yersinia enterocolitica O:9, Escherichia coli O:157, and Salmonella urbana. In particular, most

Biodehalogenation of low concentrations of 1,3-dichloropropanol by mono- and mixed cultures of bacteria.

The degradation of low concentrations of 1,3-dichloro-2-propanol (1,3-DCP) and related halohydrins by whole cells and cell-free extracts of soil bacteria has been investigated. Three bacteria (strains A1, A2, A4), isolated from the same soil sample, were distinguished on the basis of cell morphology, growth kinetics and haloalcohol dehalogenase profiles. Strain A1, probably an Agrobacterium sp., dehalogenated 1,3-DCP with the highest specific activity (0.33 U mg protein-1) and also had the highest affinity for 1,3-DCP (Km, 0.1 mM). Non-growing cells of this bacterium dehalogenated low concentr

Identification and characterization of the Brucella abortus phosphoglucomutase gene: role of lipopolysaccharide in virulence and intracellular multiplication.

Smooth lipopolysaccharide (LPS) of Brucella abortus has been reported to be an important virulence factor, although its precise role in pathogenesis is not yet clear. While the protective properties of LPS against complement are well accepted, there is still some controversy about the capacity of rough mutants to replicate intracellularly. The B. abortus phosphoglucomutase gene (pgm) was cloned, sequenced, and disrupted. The gene has a high index of identity to Agrobacterium tumefaciens pgm but is not part of the glycogen operon. A B. abortus null mutant lacks LPS O antigen but has an LPS core

Establishment, Culture, and Scale-up of Brugmansia candida Hairy Roots for the Production of Tropane Alkaloids.

Brugmansia candida (syn. Datura candida) is a South American native plant that produces tropane alkaloids. Hyoscyamine, 6β-hydroxyhyoscyamine (anisodamine), and scopolamine are the most important ones due to their anticholinergic activity. These bioactive compounds have been historically and widely applied in medicine and their demand is continuous. Their chemical synthesis is costly and complex, and thereby, these alkaloids are industrially produced from natural producer plants. The production of these secondary metabolites by plant in vitro cultures such as hairy roots presents certain advan

Attenuated signature-tagged mutagenesis mutants of Brucella melitensis identified during the acute phase of infection in mice.

For this study, we screened 1,152 signature-tagged mutagenesis mutants of Brucella melitensis 16M in a mouse model of infection and found 36 of them to be attenuated in vivo. Molecular characterization of transposon insertion sites showed that for four mutants, the affected genes were only present in Rhizobiaceae. Another mutant contained a disruption in a gene homologous to mosA, which is involved in rhizopine biosynthesis in some strains of Rhizobium, suggesting that this sugar may be involved in Brucella pathogenicity. A mutant was disrupted in a gene homologous to fliF, a gene potentially

Regulation of Brucella virulence by the two-component system BvrR/BvrS.

The Brucella BvrR/BvrS two-component regulatory system is highly similar to the regulatory and sensory proteins of Sinorhizobium and Agrobacterium necessary for endosymbiosis and pathogenicity in plants, and very similar to a putative system present in the animal pathogen Bartonella. Mutations in the bvrR or bvrS genes hamper the penetration of B. abortus in non-phagocytic cells and impairs intracellular trafficking and virulence. In contrast to virulent Brucella, BvrR/BvrS mutants do not recruit small GTPases of the Rho subfamily required for actin polymerization and penetration to cells. Dys
