Keratinocytes: A key player in skin immunity and their emerging roles in secondary lymphedema.

Keratinocytes, once viewed as inert epidermal scaffolds, are now recognized as active participants in the cutaneous immunity. They orchestrate local immune responses by releasing cytokines, chemokines, and alarmins that shape innate and adaptive inflammation. Dysregulated keratinocyte-immune interactions underline chronic inflammatory skin disorders, such as atopic dermatitis, psoriasis, and cutaneous lupus erythematosus. More recently, keratinocytes have been identified as key contributors to secondary lymphedema, a chronic inflammatory and fibrotic condition historically attributed to lympha

When the Matrix Warns: Unraveling Brain Extracellular Matrix Components as Danger Signals in Glioma Progression.

The brain constitutes a biologically unique environment tasked with executing the most sophisticated functions of information processing and storage. This singularity arises from several factors, including its status as an immunoprivileged organ. Moreover, the brain extracellular matrix (ECM) provides a structural scaffold for cells and simultaneously acts as an active mediator for information storage and transmission. Unsurprisingly, the ECM has become firmly embedded as a key component of the tetrapartite synapse concept, which has gathered considerable traction in recent decades. In the pre

HMGB1 functional plasticity in neurodegenerative diseases and its translational implications.

High mobility group box 1 (HMGB1) is a chromatin-associated protein that can be redistributed and released during cellular stress, when it acts as an extracellular alarmin within the broader family of damage-associated molecular patterns (DAMPs). Its functions depend on subcellular localization, redox and post-translational modification state, cellular source, mode of release, and molecular partners. This critical narrative review examines how these variables shape HMGB1 biology in Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS), Huntington's disease (HD

What Is-and What Is Not-Immunogenic Cell Death? Functional Definitions, Experimental Standards, and Common Pitfalls.

Immunogenic cell death (ICD) links tumor cell demise to the activation of anti-tumor immunity, but its adoption has also generated inconsistent definitions and frequent overinterpretation of surrogate biomarkers. Here, we synthesize mechanistic and methodological evidence showing that danger-associated molecular patterns (DAMPs), cytokine release, and endoplasmic reticulum stress report immunogenic potential rather than ICD itself. We propose that ICD should be defined by its functional immunological endpoint, namely efficient antigen presentation and antigen-specific adaptive immunity, ideall

Damage-Associated Molecular Patterns (DAMPs) in Retinal Disorders.

Damage-associated molecular patterns (DAMPs) are endogenous danger molecules released from the extracellular and intracellular space of damaged tissue or dead cells. Recent evidence indicates that DAMPs are associated with the sterile inflammation caused by aging, increased ocular pressure, high glucose, oxidative stress, ischemia, mechanical trauma, stress, or environmental conditions, in retinal diseases. DAMPs activate the innate immune system, suggesting their role to be protective, but may promote pathological inflammation and angiogenesis in response to the chronic insult or injury. DAMP

Key Events Participating in the Pathogenesis of  Alcoholic Liver Disease.

Alcoholic liver disease (ALD) is a leading cause of morbidity and mortality worldwide. It ranges from fatty liver to steatohepatitis, fibrosis, cirrhosis and hepatocellular carcinoma. The most prevalent forms of ALD are alcoholic fatty liver, alcoholic hepatitis (AH) and alcoholic cirrhosis, which frequently progress as people continue drinking. ALD refers to a number of symptoms/deficits that contribute to liver injury. These include steatosis, inflammation, fibrosis and cirrhosis, which, when taken together, sequentially or simultaneously lead to significant disease progression. The pathogene

Oxidative stress, DAMPs, and immune cells in acute pancreatitis: molecular mechanisms and therapeutic prospects.

Acute pancreatitis (AP) is a gastrointestinal disease characterized by inflammation of the pancreas and is associated with high rates of morbidity and mortality. The pathogenesis of AP involves a complex interplay of cellular and molecular mechanisms, including oxidative stress, damage-associated molecular patterns (DAMPs), and the infiltration of various immune cells. This review aims to provide a comprehensive overview of the molecular mechanisms underlying AP, the role of different immune cells in its progression and potential therapeutic perspectives. Oxidative stress, characterized by an

Mitochondrial DAMPs: Key mediators in neuroinflammation and neurodegenerative disease pathogenesis.

Neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), and amyotrophic lateral sclerosis (ALS) are increasingly linked to mitochondrial dysfunction and neuroinflammation. Central to this link are mitochondrial damage-associated molecular patterns (mtDAMPs), including mitochondrial DNA, ATP, and reactive oxygen species, released during mitochondrial stress or damage. These mtDAMPs activate inflammatory pathways, such as the NLRP3 inflammasome and cGAS-STING, contributing to the progression of neurodegenerative diseases. This review delv

[Molecular mechanisms underlying the pathogenesis of septic multiple organ failure].

Sepsis is defined as the body's overwhelming and life-threatening response to infection that can lead to tissue damage, organ failure, and death. Since bacterial infection is one of the main causes of sepsis, appropriate antimicrobial therapy remains the cornerstone of sepsis and septic shock management. However, since sepsis is a multifaceted chaos involving inflammation and anti-inflammation disbalance leading to the unregulated widespread release of inflammatory mediators, cytokines, and pathogen-related molecules leading to system-wide organ dysfunction, the whole body control to prevent t

Serum concentration of extracellular cold-inducible RNA-binding protein is associated with respiratory failure in COVID-19.

UNLABELLED: Uncontrolled release of damage-associated molecular patterns (DAMPs) is suggested to be a major trigger for the dysregulated host immune response that leads to severe COVID-19. Cold-inducible RNA-binding protein (CIRP), is a newly identified DAMP that aggravates inflammation and tissue injury, and induces respiratory failure in sepsis. Whether CIRP contributes to the pathogenesis of respiratory failure in COVID-19 has not yet been explored. AIM: To investigate if the concentration of extracellular CIRP (eCIRP) in serum associates with respiratory failure and lung involvement by ch

ADP-ribosylation in evasion, promotion and exacerbation of immune responses.

ADP-ribosylation is the addition of one or more (up to some hundreds) ADP-ribose moieties to acceptor proteins. This evolutionary ancient post-translational modification (PTM) is involved in fundamental processes including DNA repair, inflammation, cell death, differentiation and proliferation, among others. ADP-ribosylation is catalysed by two major families of enzymes: the cholera toxin-like ADP-ribosyltransferases (ARTCs) and the diphtheria toxin-like ADP-ribosyltransferases (ARTDs, also known as PARPs). ARTCs sense and use extracellular NAD, which may represent a danger signal, whereas ART

Adding insult to injury - Inflammation at the heart of cardiac fibrosis.

The fibrotic response has evolutionary worked in tandem with the inflammatory response to facilitate healing following injury or tissue destruction as a result of pathogen clearance. However, excessive inflammation and fibrosis are key pathological drivers of organ tissue damage. Moreover, fibrosis can occur in several conditions associated with chronic inflammation that are not directly caused by overt tissue injury or infection. In the heart, in particular, fibrotic adverse cardiac remodeling is a key pathological driver of cardiac dysfunction in heart failure. Cardiac fibroblast activation

COPD: the role of neutrophils in inflammation, pathophysiology, and as drug targets.

Neutrophils are innate immune effector cells that play a vital role in host defense against infection. They exert antimicrobial activities through degranulation, phagocytosis, intracellular killing, production of reactive oxygen species, and neutrophil extracellular traps. Dysregulation of neutrophil abundance, phenotype, and immune activity is a commonly observed inflammatory feature of chronic obstructive pulmonary disease (COPD), a chronic inflammatory disorder of the lungs with no effective cure. Here, we review the clinical association and involvement of neutrophils in COPD pathogenesis,

The regulatory effects of luteolin, calycosin, and formononetin on the NLRP3/IL-33/ILC2s axis in the treatment of allergic rhinitis: mechanistic analysis and therapeutic potential.

Allergic rhinitis (AR), a common IgE-mediated inflammatory condition of the nasal mucosa, presents with nasal itching, episodic sneezing, and runny nose. Emerging evidence indicates that type 2 innate lymphoid cells (ILC2s) are key players in AR development. Epithelial-derived alarmins (IL-33, IL-25, TSLP) activate ILC2s, leading to Th2 cytokine production (IL-4, IL-5, IL-13) that enhances inflammation. Recent research shows that NOD-like receptor protein 3 (NLRP3) can function as a transcriptional regulator of interleukin-33 (IL-33), offering new mechanistic insights into ILC2s dysregulation.

The role of hormones in ILC2-driven allergic airway inflammation.

Group 2 innate lymphoid cells (ILC2s) are a type of innate immune cells that produce a large amount of IL-5 and IL-13 and two cytokines that are crucial for various processes such as allergic airway inflammation, tissue repair and tissue homeostasis. It is known that damaged epithelial-derived alarmins, such as IL-33, IL-25 and thymic stromal lymphopoietin (TSLP), are the predominant ILC2 activators that mediate the production of type 2 cytokines. In recent years, abundant studies have found that many factors can regulate ILC2 development and function. Hormones synthesized by the body's endocr

Tau liquid-liquid phase separation is modulated by the Ca2+ -switched chaperone activity of the S100B protein.

Aggregation of the microtubule-associated protein tau is implicated in several neurodegenerative tauopathies including Alzheimer's disease (AD). Recent studies evidenced tau liquid-liquid phase separation (LLPS) into droplets as an early event in tau pathogenesis with the potential to enhance aggregation. Tauopathies like AD are accompanied by sustained neuroinflammation and the release of alarmins at early stages of inflammatory responses encompass protective functions. The Ca2+ -binding S100B protein is an alarmin augmented in AD that was recently implicated as a proteostasis regulator actin

Proteomic analysis of human kidney biopsies unveils emerging acute kidney injury very early after liver graft reperfusion.

BACKGROUND: Acute kidney injury (AKI) is a frequent complication after liver transplantation (LT) and is associated with morbidity, mortality, and late development of chronic kidney disease. Risk factors for AKI after LT include patient, perioperative and graft-related factors. The exact renal molecular mechanisms behind AKI in LT are unclear. METHODS: Alterations in the proteome were investigated in kidney biopsies from 21 patients undergoing LT using quantitative proteomics. The most upregulated protein was validated using immunohistochemistry. In addition, serum levels of interleukin (IL)-

[Early identification and protective strategies for pulmonary and extrapulmonary organ injury secondary to severe pneumonia].

Severe pneumonia is one of the critical illness with the highest incidences and mortality rates in the intensive care unit. Its core threat lies not in the pulmonary infection itself, but in the subsequent multiple organ dysfunction syndrome (MODS). This review systematically delineates how severe pneumonia collectively accelerates organ injury through systemic inflammatory responses, type 2 immunity mediated by airway epithelial alarmins, endothelial thrombo-inflammatory pathways, as well as inappropriate oxygen therapy and mechanical ventilation. It further reviews the key points for early i
