Upon formation of a biofilm, bacteria undergo several changes that prevent eradication with antimicrobials alone. Due to this resistance, the standard of care for infected medical implants is explantation of the infected implant and surrounding tissue, followed by eventual reimplantation of a replacement device. Recent studies have demonstrated the efficacy of heat shock for biofilm eradication. To minimize the heat required for in situ biofilm eradication, this study investigated the hypothesis that antibiotics, while ineffective by themselves, may substantially increase heat shock efficacy.

BACKGROUND: Foot infections are a major cause of morbidity in people with diabetes and the most common cause of diabetes-related hospitalization and lower extremity amputation. Staphylococcus aureus is by far the most frequent species isolated from these infections. In particular, methicillin-resistant S. aureus (MRSA) has emerged as a major clinical and epidemiological problem in hospitals. MRSA strains have the ability to be resistant to most β-lactam antibiotics, but also to a wide range of other antimicrobials, making infections difficult to manage and very costly to treat. To date, there

Candida species are common human commensals and cause either superficial or invasive opportunistic infections. The biofilm form of candida as opposed to its suspended, planktonic form, is predominantly associated with these infections. Alternative or adjunctive therapies are urgently needed to manage Candida infections as the currently available short arsenal of antifungal drugs has been compromised due to their systemic toxicity, cross-reactivity with other drugs, and above all, by the emergence of drug-resistant Candida species due to irrational drug use. Areas covered: Combination anti-Cand

Biofilms are polymicrobial communities that grow on surfaces in nature. Oral bacteria can spontaneously form biofilms on the surface of teeth, which may compromise the health of the teeth, or their surrounding (periodontal) tissues. While the oral bacteria exhibit high tropism for their specialized ecological niche, it is not clear if bacteria that are not part of the normal oral microbiota can efficiently colonize and grow within oral biofilms. By using an in vitro "supragingival" biofilm model of 6 oral species, this study aimed to investigate if 3 individual bacterial species that are not p

PURPOSE OF REVIEW: Biofilms have been observed on the sinus mucosa of patients with chronic rhinosinusitis and are associated with poor clinical outcomes after surgery. This article summarizes the recent studies which describe the efficacy of treating biofilms in chronic rhinosinusitis.
RECENT FINDINGS: Biofilms are polymicrobial communities comprised of bacteria that tend to be more antibiotic-resistant than when in planktonic forms. Antibiotic therapy against biofilms is usually associated with relapse following cessation of treatment and may also have an adverse effect on normal commensal

The objective of the present work was to study the contribution of biofilms to the development of chronic bacterial rhinosinusitis. A total of 50 patients with this pathology were available for the examination. Mucosal swabs were taken from the middle nasal passages of all the patients to be used for the detection of biofilms by luminescence microscopy. The lactoferrine content in mucosal secretion from the nasal cavity was determined by an immunoenzymatic assay. Two groups of the patients presenting with bacterial rhinosinusitis were distinguished in the study of impression smears from nasal

Escherichia coli-associated urinary tract infections (UTIs) are among the most common bacterial infections in humans. UTIs are usually managed with antibiotic therapy, but over the years, antibiotic-resistant strains of uropathogenic E. coli (UPEC) have emerged. The formation of biofilms further complicates the treatment of these infections by making them resistant to killing by the host immune system as well as by antibiotics. This has encouraged research into therapy using bacteriophages (phages) as a supplement or substitute for antibiotics. In this study we characterized 253 UPEC in terms

BACKGROUND: Bacterial biofilms have been identified on the sinonasal mucosa of patients with chronic rhinosinusitis (CRS) but also on control samples. Their role in the disease pathogenesis is unproven. The objective of this study was to further evaluate the role of biofilms in CRS by assessing whether they are associated with an inflammatory response.
METHODS: Mucosal samples were collected from 18 patients with CRS and 7 normal subjects. Bacteria on the mucosal surface were identified by Gram stain. Immune cells were identified by Giemsa stain and immunohistochemistry (IHC). The number of l

Pseudomonas aeruginosa is an opportunistic pathogen that produces sessile communities known as biofilms that are highly resistant to antibiotic treatment. Limited information is available on the exact role of various components of the matrix in biofilm-associated antibiotic resistance. Here we show that the presence of extracellular polysaccharide reduced the extent of biofilm-associated antibiotic resistance for one class of antibiotics. Minimal bactericidal concentration (MBC) for planktonic and biofilm cells of P. aeruginosa PA14 was measured using a 96 well microtiter plate assay. The MBC

BACKGROUND: Although the existence of biofilms on the sinus mucosa of patients with chronic rhinosinusitis (CRS) is now well established, the role that these structures play remains unclear. It is thought that biofilms may contribute to the recalcitrant and persistent nature that characterizes CRS, but little research exists documenting the effect that they have on postoperative mucosal outcomes. This article presents a retrospective analysis of sinus surgical patients and correlates the presence of biofilms with mucosal outcomes. This study was performed to evaluate the role that bacterial bi

BACKGROUND: Candidozyma auris (formerly Candida auris) is an emerging fungal pathogen which is highly transmissible in clinical settings, likely due to its ability to persist on skin and hard surfaces.
METHODS: We investigated the biofilm-forming ability of three C. auris isolates on non-porous plastic surfaces and developed a relevant and repeatable laboratory model for evaluating decontamination interventions.
FINDINGS: The isolate from clade I (South Asian) developed a robust biofilm under realistic environmental conditions, driving persistence of viable cells on dry surfaces up to 28 day

Staphylococcus aureus presents significant challenges in clinical settings due to its ability to form resilient biofilms, which drive persistent infections on medical implants and at wound sites. These bacteria are the leading cause of biomaterial-associated infections, often leading to implant failure. Moreover, S. aureus can form biofilms on cardiovascular implants, further complicating device-related infections. Cavitation microstreaming, generated by ultrasound-activated microbubbles through their oscillations, has emerged as a promising method for disrupting biofilms. However, the underly

Pseudomonas aeruginosa is a ubiquitous gram-negative bacterium that causes diverse opportunistic infections in vulnerable populations. P. aeruginosa is innately resistant to many therapeutic agents, and biofilm-mediated infections demonstrate enhanced recalcitrance to antimicrobials. It has been known for over 25 years that iron plays a critical role in P. aeruginosa biofilm physiology, and numerous studies have revealed mechanistic insights into how this metallonutrient affects the regulatory, structural, and nutritional requirements of biofilm communities. This mini-review presents the curre

UNLABELLED: Staphylococcus aureus is a leading cause of biofilm-associated infections, in which communities of bacterial cells are encased in an extracellular matrix composed of polysaccharides, proteins, and extracellular DNA (eDNA) that protect bacteria from host immune defense and antibiotics. Despite their importance, the mechanisms by which matrix components are released from bacterial cells and incorporated into the biofilm matrix remain poorly understood. Using a drip-flow biofilm system, we showed that membrane vesicles (MVs) were associated with the biofilm matrix formed by S. aureus

We investigated the oscillatory expansion of Bacillus subtilis biofilms on minimal salts glycerol glutamate (MSgg) agar by modulating the concentration of the nutrient glutamate and the rigidity of the surface via agar concentration. The concentration of glutamate and agar influenced the parameters of biofilm oscillation. Specifically, elevated glutamate levels boosted the initial growth rate (within the first 15 h) and prolonged the oscillation period. Conversely, increasing the agar concentration physically restrained colony expansion and dampened the amplitude of growth rate variations, lea

Dental caries is initiated by the adherence of Streptococcus mutans to tooth surfaces, leading to biofilm (plaque) formation and the production of enamel-eroding acids. Sortase A (SrtA) plays a critical role in this process because it anchors many protein adhesins to the bacterial cell wall, making it a promising target for plaque prevention. Our previous research demonstrated that edible polyphenols from maple extracts-nortrachelogenin-8'-O-β-D-glucopyranoside, lariciresinol, isoscopoletin, and (-)-epicatechin gallate (ECG)-inhibit SrtA in Listeria monocytogenes and act as antibiofilm agents.

BACKGROUND: Klebsiella pneumoniae is a significant nosocomial pathogen that has developed resistance to multiple antibiotics, often forming biofilms that enhance its virulence. This study investigated the efficacy of a novel nanoformulation, AgNPs@chitosan-NaF, in combating K. pneumoniae biofilms.
METHODS: Antimicrobial susceptibility testing was performed to assess the antibiotic resistance profile of K. pneumoniae isolates. The antibiofilm activity of AgNPs@chitosan-NaF was evaluated using crystal violet staining and scanning electron microscopy. The underlying mechanisms of action were inv

Polymicrobial infections, involving the simultaneous presence of two or more microbial species at a single infection site, are an emerging clinical challenge with increasing incidence across a wide range of healthcare settings. These infections often result in more severe disease outcomes compared to monomicrobial infections due to complex interspecies interactions that enhance microbial survival, virulence, and resistance. One of the most concerning aspects of polymicrobial infections is their strong association with multidrug resistance (MDR). The Synergistic interactions among microbial spe
