BACKGROUND: Papillon-Lefèvre syndrome (PLS) is a disorder that involves destruction of the periodontium and abnormal hyperkeratosis of the palms of the hands and soles of the feet. Mutations of the lysosomal protease cathepsin C gene (CTSC) have been associated with PLS. However, genotypic and phenotypic correlation has not been established. In the present study we investigated the CTSC gene in a Brazilian cohort affected by PLS.
METHODS: Eight consanguineous members of a kindred with PLS were studied. DNA was extracted and all exons of the gene amplified by the polymerase chain reaction (PCR

AIMS AND BACKGROUND: Laminin is a major glycoprotein component of basement membrane which is an important barrier to tumor cells which must be breached before metastatic spread can occur. Proteolytic enzymes play an important role in mediating the passage of cancer cells through the basement membrane (BM) and extracellular matrix. We have compared the patterns of laminin and cathepsin D (CD) expressions in a range of benign and malignant breast lesions to better understand the process of tumor progression.
METHODS: One hundred and sixty-two cases of breast samples comprising 18 fibroadenomas,

Alzheimer's disease (AD) is a complex disease with the possible involvement of several genes. The APOE*4 allele has been documented to be a major risk factor for sporadic late-onset AD, but it is neither necessary nor sufficient to cause the disease. Cathepsin G, a serine protease found commonly in the azurophillic granules of neutrophils, has been reported to possess some beta-secretase like properties, and thus may be involved in the processing of amyloid precursor protein (APP). Recently, an A-->G polymorphism has been reported in exon 4 of the cathepsin G gene, which changes the codon AAC

We found a novel G-->A change at nucleotide -25 within the promoter of the CTSS gene encoding the elastase cathepsin S. The CTSS -25G/A polymorphism could be detected by digestion with endonuclease BfmI. The frequency of the CTSS -25A allele was 0.457 in Caucasians and 0.431 in Canadian Inuit. Because of the importance of the CTSS gene product in vascular matrix remodeling, this polymorphism may be useful in the study of associations with atherosclerosis and related phenotypes.

The formation of A beta and A beta-containing fragments is likely a key event in the process of neural degeneration in Alzheimer's disease. The N-terminal residue (Asp-1) of A beta and its C-terminally extended sequences is liberated from the beta-amyloid precursor protein (beta APP) by beta-secretase(s). This activity appears highly increased by the presence (N-terminally to Asp-1) of a double-mutation (KM-->NL) found in several Swedish families affected by early onset Alzheimer's disease. By means of synthetic peptides encompassing the "normal' (N peptide) and mutated (delta NL peptide) sequ

The presence, localization and activities of cathepsin G in gingival tissue specimens and crevicular fluid (GCF) from 9 adult periodontitis patients and 6 controls with clinically healthy periodontium were studied by use of avidinbiotin-peroxidase complex method, Western and dot blotting, and spectrophotometric activity assay. In contrast to healthy gingival tissue specimens, gingival tissue specimens collected from adult periodontitis patients contained inflammatory cells in lamina propria, beneath the oral sulcular epithelium, 10-50% of which were cathepsin G positive polymorphonuclear neutr

Numerous studies have demonstrated that cathepsin D as a major lysosomal acid protease plays an important role in the degradation of protein in several tissues. An important function of the retinal pigment epithelium is to interact with the photoreceptor cells in the renewal process. During the renewal process, the RPE cell phagocytosis discarded photoreceptor discs which are then degraded in the RPE phagolysosomes. It is believed that cathepsin D plays a main role in the degradation of rod outer segments and rhodopsin into glycopeptides. The cellular localization of cathepsin D immunoreactivi

The interaction of a series of sulfonate and phosphate esters derived from N-hydroxysuccinimide with human leukocyte cathepsin G was investigated. The synthesized compounds were found to be time-dependent inhibitors of the enzyme. The composite interplay of steric and electronic effects leads to the formation of acyl enzymes of variable stability, ultimately resulting in partial or full recovery of enzymatic activity. Compounds acting via phosphorylation of the active site serine inactivated the enzyme rapidly and irreversibly.

Cyst fluids from 55 premenopausal women with gross cystic breast disease were classified by K+/Na+ ratio: 19 with ratio over 1 (type I) and 36 with ratio less than 1 (type II). Immunoradiometric assay of cathepsin D in both types of cyst fluids revealed the presence of large amounts of this proteinase. The average concentration of cathepsin D in type I cyst fluids was 63.3 nmol/l, which was significantly higher than that corresponding to type II cyst fluids (35.1 nmol/l). Immunoprecipitation analysis of intracystic cathepsin D demonstrated that this protein was present as the 52 kD non-process

An investigation is described of the expression of the cysteine proteinase cathepsin L during placental development. In addition, whether cathepsin L expression is linked to c-rasHa expression in development, as it is in metastatic cells, is examined. Large amounts of cathepsin L and its transcript are present in the mouse placenta, more than six times more than in adult kidney and liver. Throughout gestation, cathepsin L and its transcript are located in the giant cells and spongiotrophoblasts of the placenta. Several forms of different mobility on denaturing gels are found in the placenta. T

Cathepsin B and L activities were determined with 7-amino-4-methyl-coumarin conjugates as substrates using small tissue specimens of gastric cancer and benign gastric ulcer. Both cathepsin B and L activities were higher in homogenates of gastric cancer tissue than in regenerative mucosa of benign gastric ulcer. Cathepsin B activity was much higher in cancer tissue than in tissue surrounding the tumor. In contrast, the activity in regenerative mucosa of benign gastric ulcer was not elevated as compared with the tissue surrounding the ulcer. Similar results were obtained with cathepsin L activit

The overtime study of changes in the activity of cathepsin D, a lysosomal enzyme, in the splenocytes of CBA mice after their infection with virulent and avirulent Shigella strains of the same origin and with the same antigenic structure has been made. As the result of two months of observations, changes in the activity of this enzyme in the cytoplasmic and lysosomal cell fractions have been found to occur in phases. The activity of cathepsin D has been shown to depend on the virulence of Shigella strains used for inoculation. Virulent Shigella strains induce the pronounced and prolonged activa

Cathepsin B in the bovine ciliary body and iris was studied biochemically using alpha-N-benzoyl-D,L-arginine-2-naphthylamide as substrate. The enzyme was purified to 210-fold from the autolyzed extract. The partially purified enzyme had a pH optimum at 6.0 and a molecular weight of 27,000. The apparent Km value for the substrate was 1.6 mM. The enzyme was activated by disodium ethylenediamine tetraacetate, cysteine, and dithiothreitol. The enzyme activity was inhibited strongly by leupeptin and partially by hyaluronate and chondroitin sulfate A.

Cathepsin D has been purified from rabbit thyroids, and its action on thyroglobulin has been examined. The enzyme was obtained in an electrophoretically homogenous form by gel filtration, followed by ion exchange chromatography and affinity chromatography with immobilized pepstatin. In some preparations, the enzyme occurred in a high molecular weight form. The ability of cathepsin D to hydrolyze [125I]thyroglobulin to fragments with a molecular weight of less than 100K was determined by polyacrylamide gel electrophoresis in sodium dodecyl sulfate. This activity showed a pH optimum of 3.5, was

The diagnosis of lymphangioleiomyomatosis (LAM) can be straightforward when LAM cells are abundant, but may be difficult with subtle lesions mimicking emphysema or with lung architecture obscured by parenchymal collapse. A variety of markers have been proposed for immunohistochemical confirmation of LAM. The is increasing evidence from sequencing studies that tuberous sclerosis 2 (TSC2) is frequently mutated in sporadic LAM, and TSC2 expression/loss can now be demonstrated by immunohistochemistry but TSC2 immunohistochemistry has not been reported in LAM. Here we examined the immunohistochemic

This study investigates the expression level of cathepsin D (CTSD) in the serum of patients with acute myeloid leukemia (AML) and its clinical significance and evaluates its potential value as a prognostic biomarker for AML. Bioinformatics analysis was performed to examine the expression profile and prognostic correlation of CTSD in AML. A total of 63 newly diagnosed AML patients and 50 healthy controls were enrolled in validation. Serum CTSD levels were measured using enzyme-linked immunosorbent assay, and their associations with clinical characteristics, treatment response, and prognosis wer

Myeloid azurophil granules provide a rich source of intracellular leukemia antigens. Cathepsin G (CG) is a serine protease that has higher expression in acute myeloid leukemia (AML) blasts in comparison to normal myeloid progenitors. Based on the unique biology of HLA-A*0201 (HLA-A2), in which presentation of leader sequence (LS)-derived peptides is favored, we focused on the LS-CG-derived peptide CG1 (FLLPTGAEA). We previously detected CG1/HLA-A2 complexes on the surface of primary HLA-A2+ AML blasts and cell lines, and immunity targeting CG1/HLA-A2 in leukemia patients. T cell receptor (TCR)

Cathepsin C (CTSC) mediates neutrophil serine protease (NSP) maturation, contributing to inflammatory cascades, making it a key therapeutic target. In this study, through large-scale screening of a natural product library, marrubiin, a diterpenoid lactone compound, was identified as a potent CTSC inhibitor, which holds potential value in the treatment of inflammatory diseases. It inhibited human recombinant CTSC (IC50 = 57.5 nM) and intracellular CTSC (IC50 = 51.6 nM) with acceptable cytotoxicity, and reduced the activity and protein levels of downstream NSPs in vitro. Functionally, marrubiin

Tuberculosis (TB) outcomes vary widely, from asymptomatic infection to mortality, yet most animal models do not recapitulate human phenotypic and genotypic variation. The genetically diverse Collaborative Cross mouse panel models distinct facets of TB disease that occur in humans and allows identification of genomic loci underlying clinical outcomes. We previously mapped a TB susceptibility locus on mouse chromosome 2. Here, we identify cathepsin Z (Ctsz) as a lead candidate underlying this TB susceptibility and show that Ctsz ablation leads to increased bacterial burden, pulmonary inflammatio
