Single-nucleus transcriptomics reveals cell fate transitions and transcriptional programs driving bulblet formation in Lilium.

Bulblet formation is a critical type of direct vegetative regeneration in plants, where modified leaf bases undergo a developmental transition to generate new bulb structures. However, the cellular trajectories driving this scale-to-bulblet transition remain poorly understood. Here, we applied single-nucleus sequencing to construct a high-resolution cellular atlas of lily bulblet formation. We identified a population of meristem-like cells located adjacent to vascular bundle sheath cells that exhibits progenitor-like features and transcriptional associations with vascular and parenchyma cell s

Electron microscopic study on the development of Babesia ovis (Piroplasmia) in the salivary glands of the vector tick Rhipicephalus bursa.

The formation of Babesia ovis sporozoites in salivary gland cells of the vector tick Rhipicephalus bursa was studied by electron microscopy. The kinetics of B. ovis were found lying intracellularly on the second day after infestation (a.i.) of the ticks. The parasites enlarged rapidly losing all features of the motile form. Invaginations of the cell membrane initiated a fragmentation of this developmental stage. On the third day a.i the parasite (measuring 40 x 25 microns) was divided into numerous single membrane-bounded cytomeres, each provided with at least one lobed nucleus. On the fourth

A pyrene-based two-photon excitable fluorescent probe to visualize nuclei in live cells.

The two-photon absorption properties of a pyrene-pyridinium dye (1) were studied for potential application in two-photon spectroscopy. When probe 1 was used in cellular two-photon fluorescence microscopy imaging, it allowed the visualization of nuclei in live cells with a relatively low probe concentration (such as 1 μM). Spectroscopic evidence further revealed that probe 1 interacted with DNA as an intercalator. The proposed DNA intercalation properties of probe 1 were consistent with the experimental findings that suggested that the observed nucleus staining ability is dependent on the subst

[Neuroendocrine carcinoma with significantly vacuolar nucleus at the esophagogastric junction: A case report].

Neoplasms characterized by the expression of markers of neuroendocrine differentiation in neoplastic cells are defined as neuroendocrine neoplasms. A case of neuroendocrine carcinomas (NECs) with a small amount of papillary adenocarcinoma and significantly vacuolar nucleus at the esophagogastric junction was reported in this article. A 77-year-old male had dysphagia for one week. Endoscopy revealed early-stage esophagogastric junction carcinoma, and biopsy was diagnosed as poorly differentiated carcinoma. Endoscopic submucosal dissection was performed. Histologically, the papillary adenocarcin

Splanchnic and Pelvic Spinal Afferent Pathways Relay Sensory Information From the Mouse Colorectum Into Distinct Brainstem Circuits.

The distal colon and rectum (colorectum) are innervated by two distinct spinal (splanchnic and pelvic) afferent nerve pathways. This study aimed to identify where the sensory information relayed by splanchnic and pelvic afferents integrates within the brainstem. Microinjection of transneuronal viral tracer (herpes simplex virus-1 H129 strain expressing EGFP, H129-EGFP) into the distal colon was used to assess the brainstem structures receiving ascending input from the colorectum. H129-EGFP+ cells were distributed in structures involved in ascending sensory relay, descending pain modulation, an

The sorting of blood group active proteins during enucleation.

Enucleation represents the critical stage during red blood cell development when the nucleus is extruded from an orthochromatic erythroblast in order to generate a nascent immature reticulocyte. Extrusion of the nucleus results in loss of a proportion of the erythroblast plasma membrane, which surrounds the nucleus, the bulk of the endoplasmic reticulum and a small region of cytoplasm. For this reason enucleation provides an important point in erythroblast differentiation at which proteins not required for the function of the erythrocyte can be lost, whilst those that are important for the str

Human Cytomegalovirus Enters the Primary CD34+ Hematopoietic Progenitor Cells Where It Establishes Latency by Macropinocytosis.

Viral entry is targeted by immunological and pharmacological measures to inhibit viral infection. Human cytomegalovirus (HCMV) entry into cells where it initiates productive infection has been well studied, but its entry into cell types where it establishes latency has not. Therefore, we examined the entry of HCMV into CD34+ hematopoietic progenitor cells where the virus establishes latency. We determined that HCMV enters into the primary CD34+ hematopoietic progenitor cells in which it establishes latency by macropinocytosis. The capsid-associated tegument protein pp150 is released from matur

Intra-nucleus mosaic pattern (InMop) and whole-cell Haralick combined-descriptor for identifying and characterizing acute leukemia blasts on single cell peripheral blood images.

Acute leukemia is usually diagnosed when a test of peripheral blood shows at least 20% of abnormal immature cells (blasts), a figure even lower in case of recurrent cytogenetic abnormalities. Blast identification is crucial for white blood cell (WBC) counting, which depends on both identifying the cell type and characterizing the cellular morphology, processes susceptible of inter- and intraobserver variability. The present work introduces an image combined-descriptor to detect blasts and determine their probable lineage. This strategy uses an intra-nucleus mosaic pattern (InMop) descriptor th

Effects of metals on chromosomes of higher organisms.

An analysis of the available data on the clastogenic effects of metals and their compounds on higher organisms indicates some general trends. Following chronic exposure to subtoxic doses, a decrease in mitotic frequency and an increase in the number of chromosomal abnormalities are observed. These effects are usually directly proportional to the dose applied and the duration of treatment within the threshold limits. Recovery after acute treatment is inversely related to the dosage. The ultimate expression of the effects depends on certain factors, including the mode and vehicle of administrati

Effect of Fas and Bcl-2 DNA Damages Response Expression in Stem Cells on Apoptosis of Nucleus Pulposus of Intervertebral Disc.

Источник пометил публикацию как отозванную

The nucleus pulposus is an elastic jelly composed of crisscross fibrous reticular structures, namely, chondrocytes and proteoglycan mucoid matrix. Embryo and adult SC can resist the accumulation of genetic damage and repair them through various DNA repair mechanisms, thus preventing them from spreading to daughter cells. Fresh medullary tissue was fixed with 10% formaldehyde solution, embedded in paraffin, and sectioned at 4 m. The nucleus pulposus was stained with HE, and its degeneration was observed under light microscope. The average apoptotic index (AI) of 20 denatured nuclei was 50.230,

Injectable ECM-mimetic dynamic hydrogels abolish ferroptosis-induced post-discectomy herniation through delivering nucleus pulposus progenitor cell-derived exosomes.

Discectomy-induced ferroptosis of nucleus pulposus cells (NPCs) contributes to postoperative lumbar disc herniation (LDH) recurrence and intervertebral disc degeneration (IDD). We discover that nucleus pulposus progenitor cells (NPPCs) could imprint ferroptosis resistance into NPCs through exosome-dependent intercellular transmission of miR-221-3p. Based on these findings, we first develop synthetically-tailored NPPC-derived exosomes with enhanced miR-221-3p expression and NPC uptake capacity, which are integrated into an injectable hydrogel based on extracellular matrix (ECM) analogues. Somatosensory context alters auditory responses in the cochlear nucleus.

The cochlear nucleus, the first central auditory structure, performs initial stimulus processing and segregation of information into parallel ascending pathways. It also receives nonauditory inputs. Here we show in vivo that responses of dorsal cochlear nucleus (DCN) principal neurons to sounds can change significantly depending on the presence or absence of inputs from the somatosensory dorsal column nucleus occurring before the onset of auditory stimuli. The effects range from short-term suppression of spikes lasting a few milliseconds at the onset of the stimulus to long-term increases or d

Associated roles of hemolysin and p60 protein for the intracellular growth of Bacillus subtilis.

Hemolysin expressing Bacillus subtilis strain (B. subtilis ble/hlA) was used as a carrier for listerial protein p60 to study the impact of this protein on bacterial virulence independent of other gene products of Listeria monocytogenes. Bacillus subtilis ble/hlyA exhibited longer cell chains than B. subtilis ble/hlyA/iap. Recombinant Bacillus strains are able to adhere to the mouse macrophage-like J774 and human epithelial-like Int407 cell lines. The bacterial number of B. subtilis ble/hlyA/iap strain that adhered to the Int407 cell lines was 2.52-fold higher, and its invasion level strain was

Effects of prenatal alcohol exposure on the sexually dimorphic nucleus of the preoptic area of the hypothalamus in male and female rats.

Prenatal alcohol exposure can produce a variety of behavioral and physiological alterations, including changes in sexually dimorphic behaviors. It has been proposed that alcohol alters these behaviors by altering prenatal androgen and/or other steroid levels. This study was designed to examine the effects of prenatal alcohol exposure on a sexually dimorphic neuroanatomical structure, namely, the sexually dimorphic nucleus of the preoptic area of the hypothalamus (SDN-POA). This nucleus is larger in males than females and is sensitive to perinatal sex steroid exposure. The SDN-POA was examined

Distinct Populations of Amygdala Somatostatin-Expressing Neurons Project to the Nucleus of the Solitary Tract and Parabrachial Nucleus.

Rostral forebrain structures, such as the central nucleus of the amygdala (CeA), send projections to the nucleus of the solitary tract (NST) and the parabrachial nucleus (PBN) that modulate taste-elicited responses. However, the proportion of forebrain-induced excitatory and inhibitory effects often differs when taste cell recording changes from the NST to the PBN. The present study investigated whether this descending influence might originate from a shared or distinct population of neurons marked by expression of somatostatin (Sst). In Sst-reporter mice, the retrograde tracers' cholera toxin

Efferent connections of the nucleus of the lateral olfactory tract in the rat.

The efferent connections of the nucleus of the lateral olfactory tract (LOT) were examined in the rat with the Phaseolus vulgaris leucoagglutinin (PHA-L) technique. Our observations reveal that layers II and III of LOT have largely segregated outputs. Layer II projects chiefly ipsilaterally to the olfactory bulb and anterior olfactory nucleus, bilaterally to the anterior piriform cortex, dwarf cell cap regions of the olfactory tubercle and lateral shell of the accumbens, and contralaterally to the lateral part of the interstitial nucleus of the posterior limb of the anterior commissure. ten-1, an essential gene for germ cell development, epidermal morphogenesis, gonad migration, and neuronal pathfinding in Caenorhabditis elegans.

ten-m (odz) is the only pair-rule gene discovered in Drosophila that encodes a transmembrane protein and not a transcription factor. The vertebrate Ten-m orthologues have been implicated in pattern formation and neuronal development. To investigate the role of this protein in development, we characterize here the structure and function of the Caenorhabditis elegans orthologue ten-1. We found that two promoters control the expression of two different ten-1 transcripts. This results in the expression of type II transmembrane protein variants differing in their intracellular domains. Both ten-1 t

Plasticity of basal cells during postnatal development in the rat epididymis.

Our previous study has shown that basal cells sense luminal factors by forming a narrow body projection that can cross epithelial tight junctions. As a first step toward characterizing the structural plasticity of basal cells, in this study, we followed their appearance and morphology in the rat epididymis and vas deferens (VD) during postnatal development and examined their modulation by androgens in adulthood. Immunofluorescence labeling for cytokeratin 5 showed that basal cells are absent at birth. They progressively appear in a retrograde manner from the VD and cauda epididymis to the init

Compaction of chromatin domains regulates target search times of proteins.

Protein molecules must efficiently locate specific DNA sequences within the densely packed chromatin of the cell nucleus. We investigate how the spatial organisation of chromatin, specifically its organisation into Topologically Associating Domains (TADs), fundamentally affects this search process. Using exact analytical theory and simulations of different models of chromatin, we show that target search within compact, highly connected chromatin domains can leverage intersegmental jumps to significantly decrease search times. Further, we establish that there exists an optimal degree of polymer
