Physical activity and inflammation: effects on gray-matter volume and cognitive decline in aging.

Physical activity has been positively associated with gray-matter integrity. In contrast, pro-inflammatory cytokines seem to have negative effects on the aging brain and have been related to dementia. It was investigated whether an inactive lifestyle and high levels of inflammation resulted in smaller gray-matter volumes and predicted cognitive decline across 6 years in a population-based study of older adults (n = 414). Self-reported physical activity (fitness-enhancing, health-enhancing, inadequate) was linked to gray-matter volume, such that individuals with inadequate physical activity had

Influence of population versus convenience sampling on sample characteristics in studies of cognitive aging.

PURPOSE: We examined whether differences in findings of studies examining mild cognitive impairment (MCI) were associated with recruitment methods by comparing sample characteristics in two contemporaneous Australian studies, using population-based and convenience sampling. METHOD: The Sydney Memory and Aging Study invited participants randomly from the electoral roll in defined geographic areas in Sydney. The Australian Imaging, Biomarkers and Lifestyle Study of Ageing recruited cognitively normal (CN) individuals via media appeals and MCI participants via referrals from clinicians in Melbou

Transplanted bone marrow stromal cells improve cognitive dysfunction due to aging hypoperfusion in rats.

BACKGROUND: Aging is an important risk factor for vascular dementia, and D-galactose (D-gal) injection can simulate the pathology of aging. Two-vessel occlusion of common carotid arteries (2VO) is the most popular model for vascular dementia. This study was aimed to investigate the possibility of D-gal injection plus 2VO simulating cognitive impairment of aging vascular dementia; and whether transplanted bone marrow stromal cells (BMSCs) can improve the cognitive function induced by D-gal injection plus 2VO. METHODS: Totally 30 male Sprague-Dawley rats were divided into 5 groups equivalently:

Cognitive Reserve and Psychological Well-Being in Aging Population of Community-Dwellers in Spain.

INTRODUCTION: Greater brain resilience-the capacity of the brain to maintain normal functioning and cognition with aging-is associated with a slower cognitive decline. Cognitive reserve (CR) is one component of brain resilience and may be supported by educational, occupational, and leisure experiences. PURPOSE: This exploratory, cross-sectional study examined associations among psychological well-being, engagement in leisure activities associated with cognitive reserve, and cognitive performance in older community dwellers (older adults living independently) in Spain. General brain resilience

Loneliness as a Pathway Linking Hearing Decline to Cognitive Aging: Longitudinal and Genetic Evidence.

Age-related hearing loss is linked to loneliness and poorer cognitive health, but it remains unclear whether loneliness helps explain associations between hearing difficulties and cognitive performance or dementia, and whether these patterns reflect causal pathways or shared underlying liability. In this preregistered study, we triangulated analyses across multiple data sources spanning approximately 18 years of observational data with 8 sources of molecular genetic information to examine whether loneliness helps explain the association between hearing difficulty and cognitive performance, Alz

The type 2 deiodinase Thr92Ala polymorphism and cognitive aging: current evidence and biological context.

Type 2 iodothyronine deiodinase (DIO2) plays a central role in regulating thyroid hormone (TH) metabolism in the brain by catalyzing the conversion of thyroxine (T4) to the active triiodothyronine (T3), thereby ensuring adequate local TH signaling in neural tissues. Among DIO2 genetic variants, the Thr92Ala (rs225014) polymorphism has been extensively investigated in relation to TH homeostasis and diverse clinical phenotypes. Accumulating evidence suggests a potential association between the Thr92Ala variant and cognitive aging; however, findings across studies remain inconsistent. Proposed me

AI-Driven Digital Twin Architecture for Multimodal Prediction and Adaptive Intervention in Cognitive Aging.

Age-related cognitive dysfunction, including mild cognitive impairment and dementia, underscores the need for scalable and personalized predictive models. We present a conceptual artificial intelligence-driven digital twin framework to support early detection, real-time monitoring, and adaptive intervention. The system is structured around 4 core processes: perception, analytics, decision-making, and adaptive feedback, and is organized across 5 functional layers: data acquisition, integration, modeling, reasoning, and application. Multimodal behavioral, physiological, and clinical data are har

Aging on the road: neural and behavioral correlates of cognitive control during distracted driving.

OBJECTIVE: Aging increases crash risk through failures to prioritize safety-critical actions amid competing information. The neural basis of adaptive cognitive control under these demands remains insufficiently understood. METHODS: This study examined age effects on braking prioritization in 44 drivers (23 younger, 21 older) using a car-following simulation with lane keeping, in which participants withheld sign responses whenever brake signals took priority. Participants completed Sign (button press to classify road signs), Brake (pedal press to the brake lights of the car ahead), and dual-de

Food-Derived Omega-3 Fatty Acids and Cognitive Aging: Integrating Nutritional Neuroscience and Geroscience.

Within the emerging field of nutritional neuroscience, omega-3 fatty acids are among the most extensively investigated dietary bioactive compounds with potential relevance to cognitive aging. As populations continue to age worldwide, identifying modifiable nutritional factors that support cognitive health has become an important public health priority. Docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), obtained primarily from marine sources, are integral components of neuronal membranes and serve as precursors of specialized pro-resolving lipid mediators. Decoupling MCI-specific signatures from shared neurobiological substrates of cognitive aging via deep learning.

The specific neuroanatomy of mild cognitive impairment (MCI) is obscured by its clinical heterogeneity and confounding effects from normative variation. This problem is compounded by the inability of conventional neuroimaging methods to disentangle these overlapping influences. Leveraging data from the Beijing Aging Brain Rejuvenation Initiative (BABRI, n = 918) and the Alzheimer's Disease Neuroimaging Initiative (ADNI, n = 1293), this study employed a conditional variational autoencoder (CVAE) to: (1) systematically distinguish between aging-related cognitive decline and MCI-specific cognitiv

AMPK activation by Aldometanib from Bigujing improves metabolic and cognitive dysfunction in aging mice.

Aging-related cognitive decline is closely associated with disrupted energy metabolism and impaired autophagic clearance in the brain. Aldometanib, an active ingredient from the traditional formulation Bigujing, exerts systemic metabolic benefits, but its effects on brain aging and the underlying mechanisms remain unclear. This study aims to evaluate the efficacy of Aldometanib in mitigating cognitive decline in D-galactose-induced aging mice, and to elucidate its potential mechanisms involving AMPK-mediated energy metabolism and autophagy regulation. Male C57BL/6 J mice were treated with D-ga

Central Sensitization and Cognitive Vulnerability: A Neurobiological Framework for CNS Resilience and Brain Aging.

OBJECTIVE: Preservation of central nervous system (CNS) resilience during midlife is increasingly recognized as an important determinant of healthy cognitive aging, as biological processes contributing to Alzheimer's disease and related dementias (ADRD) may begin decades before clinically apparent impairment. Identifying measurable physiological states associated with diminished CNS resilience during this period is therefore an important objective of brain-aging research. Central sensitization is a neurophysiological process characterized by enhanced CNS responsiveness, impaired inhibitory mod

Platelet factor 4 in cognitive, immune and hematopoietic aging: emerging evidence and translational challenges.

Platelet Factor 4 (PF4) is a CXC chemokine abundantly sequestered within platelet α-granules. Its modulatory roles in age-related dysfunction across cognitive, immune, and hematopoietic systems have garnered increasing research attention in recent years. PF4 modulates an array of biological pathways associated with the attenuation of aging-related cellular defects; these pathways are activated by physical activity, young plasma exposure, and Klotho-related signaling cascades. Mechanistically, PF4 engages CXCR3 and LDLR to activate downstream PI3K/Akt and MAPK signaling axes, exerting neuroprot

Palmitoylethanolamide and Luteolin in Brain Aging and Cognitive Decline: Biological Rationale and Current Evidence.

Cognitive decline is closely associated with biological aging and with interconnected processes, including chronic low-grade inflammation, oxidative stress, mitochondrial dysfunction, and cellular senescence. Nutraceutical strategies capable of modulating several of these pathways may represent a complementary approach to supporting cognitive health. This narrative review examines the biological rationale and available evidence regarding palmitoylethanolamide (PEA), luteolin, and their co-ultramicronized formulation (PEA-Lut) in cognitive impairment and dementia. A cognitive-motor framework for spatial navigation in aging and early-stage Alzheimer's disease.

Spatial navigation is essential for wellbeing and independence and shows significant declines as part of age-related neurodegenerative disorders, such as Alzheimer's disease. Navigation is also one of the earliest behaviors impacted by this devastating disease. Neurobiological models of aging and spatial navigation have focused primarily on the cognitive factors that account for impaired navigation abilities during the course of healthy aging and early stages of preclinical and prodromal Alzheimer's disease. The contributions of physical factors that are essential to planning and executing mov

Aging aggravates cognitive dysfunction in spontaneously hypertensive rats by inducing cerebral microvascular endothelial dysfunction.

Hypertension in the elderly can seriously lead to cerebral microvascular damage and promote the development of vascular cognitive impairment. While endothelial function is crucial in cerebral microvascular protection, it is unclear whether aging exacerbates hypertension-induced cognitive dysfunction through endothelial dysfunction. In this study, we injected D-galactose (D-gal) into 24 spontaneous hypertension rats (SHR) and 24 Wistar-Kyoto rats (WKY) for 12 weeks to induce aging. Firstly, the results of behavioral experiments showed that compared with WKY and SHRs injected with D-gal for 0 we
