A syndrome of insulin resistance resembling leprechaunism in five sibs of consanguineous parents.

Leprechaunism is a rare autosomal recessive disorder associated with extreme insulin resistance with paradoxical hypo-glycaemia. It is characterised by prenatal and postnatal growth retardation, reduced subcutaneous tissue, coarse features, acanthosis nigricans, enlarged genitalia, and death in the first year of life. Defects in both the insulin receptor and postreceptor steps of the insulin action pathway have been reported. At the molecular level, several mutations have been described. The patients reported here are from a Yemeni family with a syndrome of insulin resistance similar to leprec

Progressive supranuclear palsy-like syndrome in two siblings of a consanguineous marriage.

Two siblings of a consanguineous marriage showed identical clinical features consisting of supranuclear vertical ophthalmoplegia, bradykinesia, rigidity, stooped posture, pseudobulbar palsy, and dementia, all beginning in the sixth decade. There was no history of encephalitis or of exposure to known chemicals. L-Dopa therapy was only partially and temporarily effective. Autopsy of one patient revealed extensive neurofibrillary degeneration with prominent involvement of the limbic system. There were no senile plaques or Lewy bodies. Under electron microscopy, there were paired helical filaments

Maroteaux-Lamy syndrome in a large consanguineous kindred: biochemical and immunological studies.

We describe a large consanguineous German-Acadian ("Cajun") family from a rural area in Louisiana in which 11 persons in two generations had the Maroteaux-Lamy syndrome. The mutant arylsulfatase B enzyme in this family was similar to the mutant enzyme in previously studied families in its cross-reactivity with specific antibodies to the enzyme, but it differed in both its electrophoretic mobility and its residual enzymatic activity. These findings indicate that a different mutational event leading to Maroteaux-Lamy syndrome occurred in this family.

Congenital alopecia, psychomotor retardation, convulsions in two sibs of a consanguineous marriage.

A family is described in which two sibs of a consanguineous marriage have alopecia, convulsions, EEG anomalies and mental retardation. Although the children have significant features resembling those described by Moynahan, this syndrome appears to be different in the mode of inheritance and in other aspects (sensorineural hearing loss in the male, syndactyly in the female).

Bilateral renal agenesis in 2 male sibs born to consanguineous parents.

Two boys with bilateral agenesis of kidneys and ureters were the product of a consanguineous marriage. This family and previous reports of familial bilateral renal agenesis support the supposition that a minor proportion of cases of BRA is caused by the homozygous state of an autosomal recessive gene.

Neonatal Glanzmann's Thrombasthenia Presenting as Refractory Post-circumcision Hemorrhage in a Region of High Consanguinity: A Case Report.

Glanzmann's thrombasthenia (GT) is a rare autosomal recessive disorder characterized by deficient or dysfunctional platelet glycoprotein IIb/IIIa (αIIbβ3 integrin), the principal fibrinogen receptor. In neonates, GT is often clinically silent until unmasked by a hemostatic challenge such as surgery or trauma. Standard coagulation screens are normal, making timely diagnosis difficult and potentially leading to life-threatening complications. We report the case of a seven-day-old Saudi boy from Najran who developed severe, refractory hemorrhage following circumcision. Initial surgical hemostasis

RNU4-2 monoallelic variants as a leading cause of syndromic neurodevelopmental disorder, including in patients with parental consanguinity.

We analysed rare variants in the non-coding RNU4-2 gene as a potential cause of neurodevelopmental disorder (NDD) and intellectual disability (ID) in a large cohort of individuals enriched for parental consanguinity.Genome sequencing (GS) data from 22 928 individuals in our Biodatabank were queried for rare, monoallelic variants in RNU4-2 From these, 4918 patients presented with NDD/ID. Human Phenotype Ontology (HPO)-encoded clinical information was extracted and analysed using the ontologyX R package.Nearly 50% of the 4918 patients with NDD/ID reported parental consanguinity. Consanguinity and adverse fetal outcomes- a population-based cohort study from a multiethnic population in the Middle East.

BACKGROUND: Consanguinity occurs widely in the Middle East and is associated with reproductive loss, congenital malformations, metabolic disorders, and autosomal recessive disorders. Qatar has a high consanguinity rate; however, large-scale population-based studies evaluating perinatal outcomes are lacking. METHODS: A retrospective cohort study of pregnancies ≥ 20 weeks of gestational age in Qatar from a maternity registry was conducted. Women were divided into consanguineous unions (CU) and non-consanguineous unions (NCU) based on their relationship with their spouses. The CUs were further di

Parental Consanguinity and Family History in Relation to Psoriasis and the Role of Sex: A Case-Control Study.

INTRODUCTION: Psoriasis is caused by an interplay between intrinsic and extrinsic factors. Parental consanguinity increases homozygosity in the genome of the offspring, which in turn increases disease risk. The association between parental consanguinity and psoriasis in the offspring remains unexplored. Therefore, this study sought to evaluate the association of parental consanguinity and family history with psoriasis in the offspring and to determine whether sex modulates the aforementioned associations. METHODS: A case-control study enrolled adults aged 21 years and more. Psoriasis cases (n

ABO-incompatible Pediatric Liver Transplantation With Antibody and B-cell Depletion-free Immunosuppressive Protocol in High Consanguinity Communities.

UNLABELLED: The success of orthotopic liver transplantation as a life-saving treatment has led to new indications and a greater competition for organ grafts. Pediatric patients with acute liver-related crises can benefit from orthotopic liver transplantation, but organ availability in the limited time can be a major obstacle. Crossing ABO blood group barriers could increase the organs available to such patients. METHODS: From November 2010 to June 2015, 176 children aged 0.2-to18 y were transplanted in the King Faisal Specialist Hospital and Research Center. Out of those, 19 children were tra

Consanguinity and founder effect for Gaucher disease mutation G377S in a population from Tabuleiro do Norte, Northeastern Brazil.

Gaucher's disease (GD) is caused by a β-glucocerebrosidase deficiency, leading to the accumulation of glucocerebroside in the reticuloendothelial system. The prevalence of GD in Tabuleiro do Norte (TN) (1:4000) is the highest in Brazil. The purpose of this study was to present evidence of consanguinity and founder effect for the G377S mutation (c.1246G>A) among GD patients in TN based on enzyme, molecular and genealogical studies. Between March 2009 and December 2010, 131 subjects at risk for GD (GC in dried blood ≤2.19 nmol/h/ml) and 5 confirmed GD patients from the same community were submit

Consanguinity-Associated Genetic Disorders in Saudi Arabia: A Hypothesis on the Future Role of CRISPR-Based Gene Editing.

Background Consanguineous marriage remains highly prevalent in Saudi Arabia and is strongly associated with increased rates of inherited genetic disorders, including sickle cell disease and β-thalassemia. The persistence of such marriages, driven by cultural and social factors, limits the effectiveness of existing preventive strategies such as premarital screening and genetic counselling. Methodology A cross-sectional, survey-based study was conducted between November and December 2025 among 139 participants across Saudi Arabia. The questionnaire was distributed via WhatsApp messaging groups a

Whole-Transcriptome Analysis by RNA Sequencing for Genetic Diagnosis of Mendelian Skin Disorders in the Context of Consanguinity.

BACKGROUND: Among the approximately 8000 Mendelian disorders, >1000 have cutaneous manifestations. In many of these conditions, the underlying mutated genes have been identified by DNA-based techniques which, however, can overlook certain types of mutations, such as exonic-synonymous and deep-intronic sequence variants. Whole-transcriptome sequencing by RNA sequencing (RNA-seq) can identify such mutations and provide information about their consequences. METHODS: We analyzed the whole transcriptome of 40 families with different types of Mendelian skin disorders with extensive genetic heteroge

A community-based study of mucopolysaccharidosis type VI in Brazil: the influence of founder effect, endogamy and consanguinity.

Mucopolysaccharidosis type VI (MPS VI - Maroteaux-Lamy syndrome) is a globally rare lysosomal storage disease caused by a deficiency of arylsulfatase B. However, in Monte Santo, a poor and isolated rural region in Northeast Brazil with large family sizes and high rates of community endogamy and parental consanguinity (α = 0.00483), 9 living and 4 now deceased individuals in 11 kindreds have been diagnosed with MPS VI, all with the same p.H178L missense founder mutation. A further 33 deceased persons have been identified by family members as exhibiting the disease phenotype. Detailed pedigrees

Analysis of Genomic Testing of Congenital Heart Disease in a Cohort From a Consanguineous Population.

Congenital heart disease (CHD) is the most common type of birth defect with tremendous genetic heterogeneity. The aim of our study was to unravel the molecular pathology and the magnitude of recessive genotypes (RGs) in a cohort of CHD from our consanguineous population. In probands with CHD who were consecutively recruited, genetic analysis was conducted based on the most likely etiology to either copy number variants (CNVs) analysis, multigene panel, whole exome sequencing (WES), or whole genome sequencing (WGS). A total of 201 families were recruited, of whom 59.7% were consanguineous. The

Geospatial mapping of consanguineous marriage in India: Evidence from a cross-sectional study.

Consanguineous marriage remains a culturally embedded practice in many parts of India. This study examines the spatial distribution and socio-demographic correlates of consanguineous marriage across districts in India. Data were derived from the fifth round of the National Family Health Survey (NFHS-5), conducted in 2019-2021. Moran's Index and Univariate Local Indicator of Spatial Association (LISA) were used for analysis. The prevalence of consanguineous marriage decreased from 14.4% in 1992-93 to 10.8% in 2019-21. Out of 707 districts, 97 districts have high consanguineous marriages above 2

Homozygous TNNI3 frameshift variant in a consanguineous family with lethal infantile dilated cardiomyopathy.

BACKGROUND: Dilated cardiomyopathy (DCM) is characterized by dilatation of the left ventricle, systolic dysfunction, and normal or reduced thickness of the left ventricular wall. It is a leading cause of heart failure and cardiac death at a young age. Cases with neonatal onset DCM were correlated with severe clinical presentation and poor prognosis. A monogenic molecular etiology accounts for nearly half of cases. FAMILY DESCRIPTION: Here, we report a family with three deceased offspring at the age of 1 year old. The autopsy of the first deceased infant revealed a DCM. The second infant prese
