Preanalytical Factors Influencing the Stability of Ethanol in Antemortem Blood and Urine Samples.

The quantitative analysis of ethanol in blood and other biological specimens is a commonly requested service from forensic science and toxicology laboratories worldwide. The measured blood alcohol concentration (BAC) constitutes important evidence when alcohol-related crimes are investigated, such as drunken driving or drug-related sexual assault. This review article considers the importance of various preanalytical factors that might influence changes in the ethanol concentration in blood after collection and before analysis or reanalysis after various periods of storage. When blood samples w

Uropharmacology: X. Central nervous system stimulants and depressants.

Several drugs that are utilized primarily for their effects on the central nervous system also affect lower urinary tract function. Most of these effects are produced by the action of these drugs on adrenergic and cholinergic receptors or by direct action of lower urinary tract musculature. Central nervous system stimulants and depressants which are known to affect the storage or evacuation role of the lower urinary tract are discussed.

Mephenytoin as a probe for CYP2C19 phenotyping:effect of sample storage, intra-individual reproducibility and occurrence of adverse events.

AIMS: To further evaluate mephenytoin as a probe for CYP2C19 phenotyping. METHODS: Healthy subjects (n = 2638) were phenotyped using the urinary (S)-mephenytoin to (R)-mephenytoin ratio. This method was evaluated for (a) the stability of the S/R-ratio following sample storage, (b) the intraindividual reproducibility of the ratio, and (c) the occurrence of adverse events. RESULTS: After prolonged storage, the S/R-ratio of samples from extensive metabolisers (EM) increased up to 85%. In 1.5% of the cases (1 out 66), this led to incorrect classification of phenotype. In EMs, but not in poor met

Hypoxic storage of red blood cells improves metabolism and post-transfusion recovery.

BACKGROUND: Blood transfusion is a lifesaving intervention for millions of recipients worldwide every year. Storing blood makes this possible but also promotes a series of alterations to the metabolism of the stored erythrocyte. It is unclear whether the metabolic storage lesion is correlated with clinically relevant outcomes and whether strategies aimed at improving the metabolic quality of stored units, such as hypoxic storage, ultimately improve performance in the transfused recipient. STUDY DESIGN AND METHODS: Twelve healthy donor volunteers were recruited in a two-arm cross-sectional stu

Efficacy of multipurpose solutions for rigid gas permeable lenses.

The use of multipurpose solutions for cleaning and disinfecting rigid gas permeable lenses has replaced single purpose solutions, but there are no reports of the efficacy of these multipurpose solutions, or of the effects of storage conditions on their disinfecting capacities. This study investigated activity against four bacterial and two fungal species, and the effects of storage in a refrigerator, at room temperature, at elevated temperature in both dry and humid conditions and with exposure to sunlight. The disinfecting solutions were challenged with the micro-organisms initially upon open

Structurally optimized photo-crosslinkable casein-PEG hydrogels for enhanced encapsulation and functional stability of resveratrol.

Casein-based gel delivery systems often exhibit limited loading efficiency and functional performance under conventional formulation approaches. We present a novel UV-induced photo-crosslinking strategy for generating porous casein-polyethylene glycol gels, enhancing resveratrol stabilization and enabling controlled delivery during processing and storage. Structural characterization by FTIR and 13C NMR supported the formation of an enhanced covalent network via ester and amide linkages. Formulation optimization showed that 10% casein achieved a crosslinking degree of 93.32 ± 0.43% (p < 0.05),

Cannabinoid Stability in Antemortem and Postmortem Blood.

In toxicological testing, drug stability is important when providing quantitative results and interpretation of findings, as well as when collecting correlation data. The goal of this study was to expand on previous stability studies and to evaluate other cannabinoids encountered in forensic toxicology. In this 6-month study, the stability of Δ-9-tetrahydrocannabinol (THC), 11-hydroxy-THC, 11-nor-9-carboxy-THC, Cannabinol and Cannabidiol in antemortem and postmortem blood was evaluated in refrigerated (4°C) and frozen (-4°C) storage conditions. A dose-optimization trial of laronidase (Aldurazyme) in patients with mucopolysaccharidosis I.

Recombinant human alpha-l-iduronidase (Aldurazyme), laronidase) is approved as an enzyme replacement therapy to treat the lysosomal storage disorder, mucopolysaccharidosis type I (MPS I) at a dose of 0.58 mg/kg by once-weekly intravenous infusion. To assess whether alternate dosing regimens might provide a better reduction in lysosomal storage, a 26-week, randomized, open-label, multinational dose-optimization trial was conducted. The pharmacodynamic effect and safety of the approved laronidase dose was compared to three alternative regimens (1.2mg/kg every 2 weeks; 1.2mg/kg every week; 1.8 mg

Harnessing dicarboxylate-based salts for enhancing the performance of amorphous solid dispersions: The case of olanzapine.

Previous studies have shown that the use of dicarboxylic acid salts can improve the performance of drug amorphous solid dispersions (ASDs), such as in the case of olanzapine (OLN). However, these studies focused only on the use of limited ASD matrices/carriers, e.g. polyvinyl alcohol, overlooking thus the critical impact of their selection on the drug's physical stability and dissolution performance. This study evaluates the performance of ASDs containing fumarate (FUM) and succinate (SUC) drug salts in comparison to ASDs prepared with the drug base, by utilizing the same model drug (i.e., OLN

CRISPR/Cas9-Mediated Generation of Niemann-Pick C1 Knockout Cell Line.

Generating a cholesterol storage phenotype of Niemann-Pick Type C (NPC) disease is important for investigating the mechanisms of intracellular cholesterol trafficking, as well as screening drugs for potential treatment of NPC disease. The use of the CRISPR/Cas9 technology to knockout specific genes within the genome of mammals has become routine in the past few years. Here, we describe a protocol for producing a cellular NPC cholesterol storage phenotype in HeLa cells using the CRISPR-Cas9 system to disrupt the NPC1 gene. The protocol details the steps for single guide RNA oligo cloning, cell

Novel cationic cellulose beads for oral delivery of poorly water-soluble drugs.

Cellulose beads emerge as carriers for poorly water-soluble drugs due to their eco-friendly raw materials and favorable porous structure. However, drug dissolution may be limited by their poor swelling ability and the presence of closed pores caused by shrinkage of the pristine cellulose beads. In this study, novel cellulose beads that can swell in acidic environment were prepared by introducing ethylenediamine (EDA) on dialdehyde cellulose (DAC), thereby addressing the shrinkage and closed pore problem of cellulose beads. The effect of the ratio of EDA on the swelling behavior and amine conte

Transmission of hepatitis C virus by needle-stick injury in community settings.

BACKGROUND: Hepatitis C virus (HCV) is predominantly transmitted by blood-to-blood contact, typically by sharing of needles by injecting drug users. Discarded needles could act as a vector for transmission of this infection. METHODS: Two cases of HCV seroconversion following a needle-stick injury in a community setting were identified. The effects of specimen processing and storage conditions on detection of HCV RNA were assessed to provide information about the likelihood of discarded needles containing infectious HCV. RESULTS: Consistent with a role for discarded needles in viral transmiss

Local effects of antimuscarinics.

Overactive bladder (OAB) syndrome has been estimated to occur in nearly 17% of the population. The most common drug treatments for OAB are antimuscarinic agents that act to increase bladder capacity and decrease the urge to urinate during the storage phase. An increasing number of studies have focused on te role and mechanism of muscarinic acetylcholine receptors for the regulation of afferent activity during urine storage. Interactions between muscarinic receptors in the urothelium, afferent nerves, or myofibroblasts and locally released acetylcholine might be involved in the emergence of det

Application and comparison of lyophilisation protocols to enhance stable long-term storage of filovirus pseudotypes for use in antibody neutralisation tests.

AIMS: Filoviruses encompass highly pathogenic viruses placing significant public health burden on countries affected. Efforts for improved diagnostics and surveillance are needed. The requirement for high-containment can be circumvented by using pseudotype viruses (PV), which can be handled safely, in tropism, drug screening, vaccine evaluation, and serosurveillance studies. We assessed the stability and functionality after long-term storage of lyophilised filovirus pseudotypes for use in neutralisation assays. METHODS AND RESULTS: We generated a panel of filovirus lentiviral pseudotypes foll

Formulation Characteristics of Solid-Dispersible Self-Emulsifying Drug Delivery Systems for Dual Drug Delivery.

Background: Oral delivery of chemotherapeutic agents remains challenging due to gastrointestinal degradation, poor intestinal permeability, and extensive first-pass metabolism, which collectively limit bioavailability. Lipid-based drug delivery systems offer a promising strategy to overcome these barriers. This study aimed to develop a freeze-dried, solid-dispersible self-emulsifying drug delivery system (SEDDS) using a water-in-oil-in-water (w/o/w) double emulsion approach for the co-encapsulation of hydrophilic (doxorubicin) and lipophilic (ellipticine) agents to enhance oral delivery. Deaggregation of micronized insoluble drugs by incorporating mannitol form α.

Micronization is frequently employed to increase the dissolution of poorly soluble drugs, but it easily led to powder aggregation and difficult to mix well on the micro level with poor content uniformity and erratic dissolution behavior. Mannitol is the most commonly used pharmaceutical excipient, and its β form (β-mannitol) is commercially available and extensively investigated, whereas form α (α-mannitol) remain poorly understood. Here, this study demonstrated that α-mannitol could significantly eliminate aggregation phenomena of micronized drugs (i.e., lurasidone hydrochloride, indomethacin

Preservative Efficacy Testing of Refrigerated Pharmaceuticals: Choice of Challenging Isolate and Storage Temperature.

The antimicrobial effectiveness test assesses the performance of the preservatives and/or antimicrobial substances added to multiuse sterile or nonsterile dosage forms to protect these products from microbiological growth. In this study, insulin products underwent the European Pharmacopoeia and the United States Pharmacopeia antimicrobial preservative effectiveness tests at both room and refrigerated temperatures. Notably, a psychrotolerant strain, Serratia marcescens, originally isolated from a refrigerated pharmaceutical product, was included in the antimicrobial effectiveness testing. When
