An Advanced Detection System for In Situ Hybridization Using a Fluorescence Resonance Energy Transfer-based Molecular Beacon Probe.

In situ hybridization (ISH) is a powerful method for detecting specific RNAs at the cellular level. Although conventional ISH using hapten-labeled probes are useful for detecting multiple RNAs, the detection procedures are still complex and required longer time. Therefore, we introduced a new application of fluorescence resonance energy transfer (FRET)-based molecular beacon (MB) probes for ISH. MCF-7 cells and C57BL/6J mouse uterus were used for ISH. MB probes for ERα mRNA and 28S rRNA were labeled with Cy3/BHQ-2 and 6-FAM/DABCYL, and conventional probes were labeled with digoxigenin. Fluores

Förster Resonance Energy Transfer between Fluorescent Organic Semiconductors: Poly(9,9-dioctylfluorene-alt-benzothiadiazole) and 6,13-Bis(triisopropylsilylethynyl)pentacene.

In the present study, an investigation of the electronic excitation energy transfer between two p-type fluorescent semiconductors, F8BT [poly(9,9-dioctylfluorene-alcohol-benzothiadiazole] and TIPS-P [6,13-bis(triisopropylsilylethynyl)pentacene], has been carried out in a chloroform solution using steady-state and time-domain fluorescence techniques. The spectral overlap integral between donor (F8BT) emission and acceptor (TIPS-P) absorption is 2.04 × 1015 nm4/(M cm), and the corresponding critical transfer distance is 53.12 Å. In donor decay dynamics, at the lower acceptor concentrations, the

Dynamically probing ATP-dependent RNA helicase A-assisted RNA structure conversion using single molecule fluorescence resonance energy transfer.

RNA helicase A (RHA) as a member of DExH-box subgroup of helicase superfamily II, participates in diverse biological processes involved in RNA metabolism in organisms, and these RNA-mediated biological processes rely on RNA structure conversion. However, how RHA regulate the RNA structure conversion was still unknown. In order to unveil the mechanism of RNA structure conversion mediated by RHA, single molecule fluorescence resonance energy transfer was adopted to in our assay, and substrates RNA were from internal ribosome entry site of foot-and-mouth disease virus genome. We first found that

Thermal evaluation of a hermetic transcutaneous energy transfer system to power mechanical circulatory support devices in destination therapy.

Current generation left ventricular assist devices (LVADs) are powered by a percutaneous driveline. The high prevalence of driveline infections has motivated the development of transcutaneous energy transfer (TET) systems which eliminate driveline associated complications by wirelessly delivering power across the skin. Destination therapy (DT) requires long-term reliable operation of the TET electronics suggesting the use of hermetic packaging techniques as used in all other chronically implanted devices. TET coils dissipate heat during operation and in order for the technology to be suitable

Visualizing Endogenous Sulfur Dioxide Derivatives in Febrile-Seizure-Induced Hippocampal Damage by a Two-Photon Energy Transfer Cassette.

Febrile seizure (FS), a frequently encountered seizure disorder in pediatric populations, can cause hippocampus damage. It has been elucidated that sulfur dioxide (SO2) content is overproduced during the development of FS and related brain injury. Thus, monitoring in situ the level of endogenous SO2 in FS-related models is helpful to estimate the pathogenesis of FS-induced brain injury, but the effect detection method remains to be explored. Herein, we developed a two-photon energy transfer cassette based on an acedan-anthocyanidin scaffold, TP-Ratio-SO2, allowing us to achieve this purpose. T

Electrochemiluminescence energy transfer-promoted ultrasensitive immunoassay using near-infrared-emitting CdSeTe/CdS/ZnS quantum dots and gold nanorods.

The marriage of energy transfer with electrochemiluminescence has produced a new technology named electrochemiluminescence energy transfer (ECL-ET), which can realize effective and sensitive detection of biomolecules. To obtain optimal ECL-ET efficiency, perfect energy overlapped donor/acceptor pair is of great importance. Herein, we present a sensitive ECL-ET based immunosensor for the detection of tumor markers, using energy tunable CdSeTe/CdS/ZnS double shell quantum dots (QDs) and gold nanorods (GNRs) as the donor and acceptor, respectively. Firstly a facile microwave-assisted strategy for

Compact and versatile nickel-nitrilotriacetate-modified quantum dots for protein imaging and Förster resonance energy transfer based assay.

The generation of compact quantum dots (QDs) probes is of critical importance for visualizing molecular interaction occurring in biological context, particularly when using the Förster resonance energy transfer (FRET) approach. This Article reports novel water-soluble compact CdSe/ZnS QDs prepared by ligand exchange reaction using thiolated nitrilotriacetate (NTA). The resulting NTA-QDs revealed higher stability and remarkable conjugation efficiency compared to the other QDs prepared with different ligands by using the ligand exchange method. The Ni-NTA group is a well-known binding moiety for

Enhancing Cr(VI) bioreduction via Na+-stimulated intracellular energy metabolism and electron transfer using methane as electron donor.

Anaerobic oxidation of methane (AOM) coupled with Cr(VI) bioreduction process offers a cost-effective pathway to simultaneously mitigate methane emissions and remediate Cr(VI) contamination. However, its practical application is limited by insufficient intracellular energy generation and low electron transfer efficiency. This study elucidated the regulatory role and mechanism of Na+, a transmembrane electrochemical gradient driver, on the metabolic activity and electron transfer of this coupled system. Batch experiments demonstrated that NaCl addition significantly improved Cr(VI) reduction ef

Intramolecular Vibrational Energy Transfer in the Precatalyst [Mn(ppy)(CO)4] Tracked by Dual-Frequency 2D Infrared Spectroscopy.

The thermal dissociation of carbon monoxide is a fundamental entry step to many catalytic cycles involving metal carbonyl (MCO) complexes as it reveals a vacant coordination site at the metal center, enabling substrate coordination. Overcoming the dissociation barrier requires sufficient accumulation of energy in vibrational modes with displacement vectors along the reaction coordinate (M-CO distance). Hence, understanding the energy transfer to and from these vibrations is essential in developing a detailed understanding of a precatalyst's activation pathway. Here, the intramolecular vibratio

Energy migration, charge transfer, and charge dissociation in self-assembling nonfullerene acceptor aggregates with zincporphyrin-nonfullerene acceptor dyads.

The synergy between self-assembling donor-acceptor-donor type nonfullerene acceptors (TACIC-Br) and zincporphyrin-nonfullerene acceptor linked molecules (ZnP-TACIC) provides a compelling model for examining key multi-step processes, including energy migration, charge transfer (CT), and charge dissociation (CD) in photosynthesis and organic photovoltaics (OPVs). Remarkably, TACIC-Br molecules exhibited a strong tendency to aggregate, even in the good solvent CHCl3. However, when the proportion of the poor solvent (MeOH) exceeded 40% in a CHCl3/MeOH mixture (v/v), these aggregates displayed an u

A Fluorescence Resonance Energy Transfer-Based Assay Targeting Tumor-Derived Extracellular Vesicles for Highly Specific Pancreatic Cancer Detection.

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most challenging cancers to detect, with significant limitations compared to screening methods for lung, breast, colon, and cervical cancers. Current methods are often plagued by high false-positive rates, necessitating costly, complex, and invasive confirmatory procedures. These challenges arise from the low incidence of PDAC and the need for highly specific and sensitive screening methods. To address these limitations, we developed EV-FRET, a fluorescence resonance energy transfer (FRET)-based assay designed as a rapid, specific, non

Low-potential pyrene-coordinated MOFs and CoSOH nanosheets: An electrochemiluminescence energy resonance transfer system for aflatoxin B1 detection.

BACKGROUND: Of the mycotoxins, aflatoxin is the most significant. The detection of aflatoxin B1 (AFB1) is crucial for ensuring food safety, as this highly carcinogenic toxin readily contaminates crops such as grains and nuts, and timely detection can effectively prevent associated health risks. The selection of luminophores is of paramount importance in the detection of ECL (electrochemiluminescence). Tetraphenylethylene (TPE) and its derivatives are the most representative molecules in aggregation-induced electrochemiluminescence. The energy resonance transfer phenomenon in ECL-RET (electroch

Charge transfer dynamics and tuning of reorganization energy in graphene-encapsulated co-based alloy catalysts for fuel cells.

Graphene-encapsulated metal alloy nanocatalysts with a metal@graphene type architecture are unique catalysts with multifunctional properties. However, effectively integrating these physicochemical properties to enhance electrocatalytic performance still presents a significant challenge. Here, using density functional theory (DFT) calculations, we first reveal that tuning the composition of CoxPdy (x ≫ y) alloys optimizes structural reorganization energy and enhances charge transfer from the metal core to the graphene shell, thereby improving oxygen reduction reaction (ORR) activity. Interlayer-engineered MXene Nanosheets confining CoGa-LDH enable ultrafast charge transfer kinetics for high-energy potassium-ion supercapacitors.

Two-dimensional MXenes, with their accordion-like morphology and facile exfoliation into monolayers, offer an ideal platform for immobilizing electroactive materials. However, composites fabricated from MXenes through conventional methods exhibit inhomogeneous dispersion, sluggish charge redistribution, and poor interfacial coupling, limiting their potassium-ion storage performance. Herein, an interlayer-engineered CoGa-LDH/MXene heterostructure is fabricated via Peltier effect-driven rotational hydrothermal synthesis with tunable Co/Ga ratios. This approach ensures atomic-level confinement of

Monte carlo simulation study on the dose and dose-averaged linear energy transfer distributions in carbon ion radiotherapy.

Dose-averaged linear energy transfer (LETd) is conventionally evaluated from the relative biological effectiveness (RBE)-LETd fitted function used in the treatment planning system. In this study, we calculated the physical doses and their linear energy transfer (LET) distributions for patterns of typical CIRT beams using Monte Carlo (MC) simulation. The LETd was then deduced from the MC simulation and compared with that obtained from the conventional method. The two types of LETd agreed well with each other, except around the distal end of the spread-out Bragg peak. Furthermore, an MC simulati

Protein Labeling Facilitates the Understanding of Protein Corona Formation via Fluorescence Resonance Energy Transfer and Fluorescence Correlation Spectroscopy.

Once nanoparticles enter into the biological milieu, nanoparticle-biomacromolecule complexes, especially the protein corona, swiftly form, which cause obvious effects on the physicochemical properties of both nanoparticles and proteins. Here, the thermodynamic parameters of the interactions between water-soluble GSH-CdSe/ZnS core/shell quantum dots (GSH-QDs) and human serum albumin (HSA) were investigated with the aid of labeling fluorescence of HSA. It was proved that the labeling fluorescence originating from a fluorophore (BDP-CN for instance) could be used to investigate the interactions b

Free Functional Muscle Transfer and One Bone Forearm for Upper-Extremity Limb Salvage After High-Energy Ballistic Trauma.

Upper-extremity limb salvage following high-energy trauma poses unique challenges of massive soft tissue injury in the setting of large bone defects, traumatic segmental neurovascular injuries, and functional deficits. These complex injuries require multidisciplinary care to achieve requisite revascularization, bone stabilization, and preservation of remaining options for soft tissue coverage. This case presents a 45-year-old man who sustained a high-velocity gunshot resulting in a dysvascular limb. Through shared decision-making, upper-extremity limb salvage was pursued. An electrochemiluminescence sensor for 17β-estradiol detection based on resonance energy transfer in α-FeOOH@CdS/Ag NCs.

An electrochemiluminescence (ECL) resonance energy transfer system is constructed for 17β-estradiol (E2) detection using α-FeOOH@CdS nanospheres as the ECL-active substrates and Ag NCs as an efficient quencher. CdS QDs loaded onto three-dimensional (3D) urchin-like α-FeOOH nanospheres (α-FeOOH@CdS nanospheres) exhibited excellent ECL responses, which is attributed to dual-amplification of α-FeOOH frameworks. The 3D hierarchical structure of the α-FeOOH nanospheres provided abundant sites for loading ECL-active species, thus significantly improving the ECL performance of substrates; While Fe3+
