Alkylphospholipids (APLs) represent a novel class of anticancer drugs that disrupt lipid homeostasis by inhibiting lipid transport to the endoplasmic reticulum and altering cellular lipid metabolism. In this study, we use protein-free membrane models to investigate how the APLs miltefosine, edelfosine and perifosine influence membrane dynamics through lipid-APL interactions, resulting in the stabilization of planar membranes. This stabilization impedes critical remodelling processes such as membrane budding and fusion, essential for cellular processes. Our findings demonstrate that APLs modula

Gram-positive organisms are responsible for some of the most serious of human infections. Resistance to front-line antimicrobial agents can complicate otherwise curative therapy. These organisms possess multiple drug resistance mechanisms, with drug efflux being a significant contributing factor. Efflux proteins belonging to all five transporter families are involved, and frequently can transport multiple structurally unrelated compounds resulting in a multidrug resistance (MDR) phenotype. In addition to clinically relevant antimicrobial agents, MDR efflux proteins can transport environmental

The cell membrane transport capacity and surface targets of multiple myeloma (MM) cells heavily influence chemotherapy and immunotherapy. Here, it is found that caveolin-1 (CAV1), a primary component of membrane lipid rafts and caveolae, is highly expressed in MM cells and is associated with MM progression and drug resistance. CAV1 knockdown decreases MM cell adhesion to stromal cells and attenuates cell adhesion-mediated drug resistance to bortezomib. CAV1 inhibition in MM cells enhances natural killer cell-mediated cytotoxicity through increasing CXCL10, SLAMF7, and CD112. CAV1 suppression r

Despite significant advances in early detection and therapeutic opportunities, cancer remains a critical global health challenge. The greatest barrier to reducing cancer-associated mortality is the persistent problem of therapeutic resistance and metastasis. Standard treatment approaches, including surgery combined with chemotherapy and radiation, are effective for many patients with localised disease. However, a substantial proportion of patients will experience recurrence and therapy resistance, leading to treatment failure and poor outcomes. This adverse prognosis for patients highlights th

Efficient and selective ion transport under nanoscale confinement underlies many emerging technologies, yet current nanofluidic membranes remain predominantly optimized for cation transport. The development of high-performance anion management systems is still in its early stages. Chitin-a sustainable polysaccharide featuring a Bouligand‑type hierarchical structure and readily modifiable functional groups-offers a uniquely advantageous platform for creating positively charged nanofluidic materials that address this gap. In this perspective, we highlight how directional disassembly can unlock a

Carnitine is a vital molecule in human metabolism, prominently involved in fatty acid β-oxidation within mitochondria. Predominantly sourced from dietary intake, carnitine also derives from endogenous synthesis. This review delves into the complex network of carnitine transport and distribution, emphasizing its pivotal role in human fertility. Together with its role in fatty acid oxidation, carnitine modulates the acety-CoA/CoA ratio, influencing carbohydrate metabolism, lipid biosynthesis, and gene expression. The intricate regulation of carnitine homeostasis involves a network of membrane tr

A new glycerol-based trifunctional block copolymer (TBC) of propylene oxide and ethylene oxide and its conjugate with succinic acid (TBC-SA) were studied as a drug delivery system and compared with Pluronic L61. TBCs have multiple effects on the plasma membrane of human cells, e.g. increasing its fluidity and ion permeability, inhibiting ATPase activity of efflux transporter P-glycoprotein through reversible membrane destabilization. Such membrane-modulating properties attributed to the unimer form of copolymers increase in the order Pluronic L61≪TBC<TBC-SA and correlate with an ability of TBC

The ATP-binding cassette transporter A1 (ABCA1) functional variant R230C (rs9282541) is associated with low plasma HDL-C levels, yet its atomic-scale mechanism remains unclear. We evaluated the structural and dynamic impact of R230C compared to wild-type (WT) ABCA1 using 200 ns molecular dynamics simulations in a lipid raft membrane. While the tertiary fold was preserved, R230C exhibited reduced overall flexibility (lower RMSD and localized RMSF rigidification) alongside a slightly expanded conformation (increased Rg and SASA). Localized fluctuations near residue 230 in extracellular domain 1

INTRODUCTION: Phalaenopsis-type Dendrobium is a commercially important tropical orchid that is highly sensitive to chilling, which can cause premature leaf senescence and abscission.
METHODS: We evaluated the protective effect of exogenous 24-epibrassinolide (EBR) and integrated time-resolved RNA-seq with physiological traits previously characterized in the same experimental framework. A concentration of 2.0 mg/L was selected for transcriptomic analysis based on the refined dose-response experiment. WGCNA was performed using 18 EBR-treated libraries. Global GO and KEGG outputs were retained a

TRPM4 is a Ca(2+)-activated nonselective cation channel. The channel is activated by an increase of intracellular Ca(2+) and is regulated by several factors including temperature and Pi(4,5)P2. TRPM4 allows Na(+) entry into the cell upon activation, but is completely impermeable to Ca(2+). Unlike TRPM5, its closest relative in the transient receptor potential family, TRPM4 proteins are widely expressed in the body. Currents with properties that are reminiscent of TRPM4 have been described in a variety of tissues since the advent of the patch clamp technology, but their physiological role is on

Red blood cells (RBCs) modulate nitric oxide (NO) bioavailability in the vasculature. Extracellular free hemoglobin (Hb) in the vascular lumen can cause NO bioavailability related complications seen in pathological conditions such as pancreatitis, sickle cell disease and malaria. In addition, the role of extracellular free Hb has been critical to estimate kinetic and transport properties of NO-RBCs interactions in 'competition experiments'. We recently reported a strong dependence of NO transport on RBC membrane permeability and hematocrit. NO donors combined with anti-inflammatory drugs are a

EAAC1-mediated glutamate transport concentrates glutamate across plasma membranes of brain neurons and epithelia. In brain, EAAC1 provides a presynaptic uptake mechanism to terminate the excitatory action of released glutamate and to keep its extracellular concentration below toxic levels. Here we report the effect of well known anxiolytic compounds, benzodiazepines, on glutamate transport in EAAC1-stably transfected Chinese hamster ovary (CHO) cells and in EAAC1-expressing Xenopus laevis oocytes. Functional properties of EAAC1 agreed well with already reported characteristics of the neuronal

Recent developments in the structure of erythrocyte band 3 and its role in hereditary spherocytosis and distal renal tubular acidosis are described. The crystal structure of the N-terminal cytoplasmic domain provides a basis for understanding the organization of ankyrin and other peripheral membrane proteins around band 3. Band 3 also binds integral membrane proteins, including the Rh protein complex and CD47. Band 4.2 is important in these associations, which link the Rh complex to the skeleton. It is suggested that band 3 forms the scaffold for a protein assembly that could transduce signals

PURPOSE: Quercetin is one of the most abundant dietary flavonoids and has attracted considerable interest because it can modulate xenobiotic metabolism by inhibiting and regulating drug-metabolizing enzymes and membrane transporters. However, the clinical relevance of these interactions remains uncertain because available evidence is fragmented and largely derived from preclinical studies. This systematic review critically evaluates quercetin's effects on human phase I cytochrome P450 enzymes, phase II conjugation pathways, drug transporters, and clinically relevant pharmacokinetic drug intera

Recent work in humans and mouse has confirmed the involvement of the host defence β-defensin peptides in male fertility. We discuss here the work that has implicated β-defensins in sperm function including the identification of the epididymis as the predominant site of expression of the peptides and the in vivo consequences of mutation and deletion. The potential dual role of these peptides in the regulation of infection and control of sperm maturation is compelling and may combine their antimicrobial activity with the ability of these molecules to interact with cell membrane receptors and mod

Drug resistance as a remarkable issue in cancer treatment is associated with inflammation which occurs through complex chemical reactions in the tumor microenvironment. Recent studies have implicated that glucocorticoids and NSAIDs are mainly useful combinations for inflammatory response modulation in chemotherapeutic protocols for cancer treatment. Immunosuppressive actions of glucocorticoids and NSAIDs are mainly mediated by the transrepression or activation regulation of inflammatory genes with different DNA-bound transcription factors including AP-1, NFAT, NF-κB, STAT and also, varying fun

Aiming to develop safe and efficient cognitive enhancers, this study reports the design, synthesis, and biological evaluation of novel α-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) receptor modulators derived from the lead compound CX717. Twenty-four derivatives were designed, synthesized, and characterized by 1H-NMR, 13C-NMR, and high resolution MS (HR-MS). Molecular docking studies indicated that, compared to CX717, the compounds exhibited lower binding energy to the AMPA receptor. Cognition-improved activity was evaluated by passive avoidance, active avoidance, novel object r

BACKGROUND & AIMS: Copper transport in hepatocytes is regulated by the interaction of multiple pumps, chaperones, and accessory proteins. Intracellular chloride channels are essential for copper metabolism in yeast but their role in Cu transport in hepatocytes is unknown. The aim of this study was to determine whether chloride channels are modulators of copper incorporation into ceruloplasmin (CP).
METHODS: The effects of chloride concentration and chloride channel expression on secretion of holoCp and apoCp was measured by gel electrophoresis and immunoblotting. ClC family chloride channel e

The aim of this study was to investigate the role of phosphorylation/dephosphorylation mechanisms at the blood-brain barrier (BBB) in the uptake of organic cations. The experiments were performed using RBE4 cells, an immortalized, rat brain microvessel endothelial cell line, an in vitro model of the BBB. The modulation of the uptake of 1-methyl-4-phenylpyridinium (MPP(+)), a model organic cation, at the apical membrane of RBE4 cells was studied. Agents that stimulate protein kinase A, but not protein kinase C, produced a moderate inhibition (approximately 18% reduction) of uptake of [(3)H]MPP(

BACKGROUND: Absorption of non-haeme iron occurs mainly in the duodenum. It involves the divalent metal transporter 1 (DMT1) in the uptake of ferrous Fe(II) iron and the basolateral transporter ferroportin/IREG-1/MTP-1/SLC40A1 in its release. Whether ferroportin functions in this process at other sites in the enterocyte is unknown. In this study the effect of a blocking antibody to ferroportin on the uptake and release of iron was evaluated in enterocyte-like cells (IEC-6 and Caco-2) and in freshly isolated duodenal enterocytes from rats.
METHODS: Uptake of 1 microM Fe(II) and its release by c
