Coumarin-Tagged Dinuclear Trithiolato-Bridged Ruthenium(II)⋅Arene Complexes: Photophysical Properties and Antiparasitic Activity.

The synthesis, characterization, photophysical and biological properties of 13 new conjugate coumarin-diruthenium(II)⋅arene complexes against Toxoplasma gondii are presented. For all conjugate organometallic unit/coumarins, an almost complete loss of fluorescence efficacy was observed. However, the nature of the fluorophore, the type of bonding, the presence and length of a linker between the coumarin dye and the ruthenium(II) moiety, and the number of dye units influenced their biological properties. The in vitro activity against a transgenic T. gondii strain grown in human foreskin fibroblas

Revealing Silylation of C(sp2 )/C(sp3 )-H Bonds in Arylphosphines by Ruthenium Catalysis.

The first aromatic C-H silylation between arylphosphines and hydrosilanes enabled by a ruthenium complex has been developed. The excellent ortho-selectivity results from a four-membered metallacyclic intermediate involving phosphorus chelation. The developed system can be extended to the benzylic C-H silylation of arylphosphines. Diverse silylated arylphosphines are produced, exhibiting broad functional group compatibility. Further functionalization of the products under mild conditions renders the formed compounds useful building blocks.

Neoadjuvant proton beam irradiation vs. adjuvant ruthenium brachytherapy in transscleral resection of uveal melanoma.

BACKGROUND: Uveal melanoma is the most common primary ocular malignancy in adults in the USA and Europe. The optimal treatment of large uveal melanoma is still under debate. Radiation therapy has its limitation due its eye-threatening secondary complications and is therefore often combined with surgical excision of the tumor. METHODS: In a retrospective interventional review, we evaluated in total 242 patients with uveal melanoma that underwent transscleral tumor resection with a predefined protocol, either with adjuvant ruthenium brachytherapy (Ru-106 group, n 136,), or with neoadjuvant prot

Characterization of the Activities of Dinuclear Thiolato-Bridged Arene Ruthenium Complexes against Toxoplasma gondii.

The in vitro effects of 18 dinuclear thiolato-bridged arene ruthenium complexes (1 monohiolato compound, 4 dithiolato compounds, and 13 trithiolato compounds), originally designed as anticancer agents, on the apicomplexan parasite Toxoplasma gondii grown in human foreskin fibroblast (HFF) host cells were studied. Some trithiolato compounds exhibited antiparasitic efficacy at concentrations of 250 nM and below. Among those, complex 1 and complex 2 inhibited T. gondii proliferation with 50% inhibitory concentrations (IC50s) of 34 and 62 nM, respectively, and they did not affect HFFs at dosages o

Unraveling the photoluminescence response of light-switching ruthenium(II) complexes bound to amyloid-β.

Photoluminescent molecules are widely used for real-time monitoring of peptide aggregation. In this Article, we detail both experimental and computational modeling to elucidate the interaction between [Ru(bpy)2dppz](2+) and amyloid-β (Aβ(1-40)) aggregates. The transition from monomeric to fibrillar Aβ is of interest in the study of Alzheimer's disease. Concentration-dependent experiments allowed the determination of a dissociation constant of 2.1 μM, while Job plots provided a binding stoichiometry of 2.6 Aβ monomers per [Ru(bpy)2dppz](2+). Our computational approach that combines molecular do

Ru-OSO coordination photogenerated at 100 K in tetraammineaqua(sulfur dioxide)ruthenium(II) (±)-camphorsulfonate.

The photoinduced O-bound coordination mode in RuSO(2) complexes, previously observed only at 13 K, has been generated at 100 K in tetraammineaqua(sulfur dioxide)ruthenium(II) (±)-camphorsulfonate. This coordination state, often denoted MS1, decays to the η(2)-bound MS2 state, with an estimated half-life of 3.4(8) h and a long-lived population of 2.9(4)% at 120 K.

Photoconversion bonding mechanism in ruthenium sulfur dioxide linkage photoisomers revealed by in situ diffraction.

Three new ruthenium-sulfur dioxide linkage photoisomeric complexes in the [Ru(NH(3))(4)(SO(2))X]Cl(2)·H(2)O family (X = pyridine (1); 3-chloropyridine (2); 4-chloropyridine (3)) have been developed in order to examine the effects of the trans-ligand on the nature of the photo-induced SO(2) coordination to the ruthenium ion. Solid-state metastable η(1)-O-bound (MS1) and η(2)-side S,O-bound (MS2) photoisomers are crystallographically resolved by probing a light-induced crystal with in situ diffraction. This so-called photocrystallography reveals the highest known photoconversion fraction of 58(3

Synthesis of glycoconjugate carbonic anhydrase inhibitors by ruthenium-catalysed azide-alkyne 1,3-dipolar cycloaddition.

Carbonic anhydrase IX (CA IX) is a recently validated target for the development of new cancer therapies. In this Letter we describe the synthesis and CA inhibition of a novel series of carbohydrate-based 1,5-disubstituted-1,2,3-triazole benzenesulfonamides. The key step of our synthesis is the regioselective Huisgen's 1,3-dipolar cycloaddition reaction (1,3-DCR) from carbohydrate azide substrates and 4-ethynylbenzenesulfonamide using a ruthenium-catalysed azide-alkyne cycloaddition (RuAAC). Our findings identified a number of triazole inhibitors (compounds 18, 19, 21-23, and 26) that block CA

Highly productive CNN pincer ruthenium catalysts for the asymmetric reduction of alkyl aryl ketones.

Chiral pincer ruthenium complexes of formula [RuCl(CNN)(Josiphos)] (2-7; Josiphos = 1-[1-(dicyclohexylphosphano)ethyl]-2-(diarylphosphano)ferrocene) have been prepared by treating [RuCl(2)(PPh(3))(3)] with (S,R)-Josiphos diphosphanes and 1-substituted-1-(6-arylpyridin-2-yl)methanamines (HCNN; substituent = H (1 a), Me (1 b), and tBu (1 c)) with NEt(3). By using 1 b and 1 c as a racemic mixture, complexes 4-7 were obtained through a diastereoselective synthesis promoted by acetic acid. These pincer complexes, which display correctly matched chiral PP and CNN ligands, are remarkably active catal

trans-Di-chlorido-tetra-kis-(pyridine-4-carbo-nitrile-κN)ruthenium(II) toluene hemisolvate.

The title compound, [RuCl2(C6H4N2)4]·0.5C7H8, resulted from the reaction of four equivalents of pcp (pyridine-4-carbonitrile) with ruthenium(II) chloride to form a propeller-like arrangement, which is positioned along the twofold axis of the C2/c space group. The packing of the complex in the structure, resulting from the overlap at the carbonitrile groups of the complexes, results in a grid-like assembly with Ru⋯Ru diagonal distances of 17.0973 (9) and 20.0289 (5) Å, and a near 50% calculated void volume.

Emerging therapeutic strategies for triple-negative breast cancer using ruthenium, iridium, and rhenium complexes.

Triple-negative breast cancer (TNBC) is an aggressive disease with limited treatment options due to the absence of ER, PR, and HER2 receptors. This review summarises the recent progress in the use of ruthenium, iridium, and rhenium complexes as alternative therapeutic agents for TNBC. These metal complexes are designed to respond to tumor-specific biomarkers or the tumor microenvironment, enabling selective uptake and enhanced anticancer activity. Through redox reactions and light activation, they generate reactive oxygen species, disrupt mitochondrial function, and induce cancer cell death. O

Coreactant-Filled Hydrogel at Ruthenium-Labeled Brain Tissue Section for Electrochemiluminescence Imaging in Gel.

Here, an electrochemiluminescence (ECL) imaging technique in gel is established to visualize single ruthenium-labeled brain tissue sections using a coreactant (e.g., tripropylamine, TPA) filled agarose hydrogel, instead of the coreactant in solution. The confined space between the gel and the tissue section restricts the diffusion of intermediates during the ECL reaction, resulting in enhanced and localized ECL emission, thereby improving the microstructural resolution. Accordingly, spatial heterogeneity in the ECL signals on the mouse midbrain tissue sections was observed, which primarily res

Ruthenium Compounds Differentially Inhibit Group IA and IIA Snake Venom Phospholipase A2 Anticoagulant Activity.

Neurotoxicity caused by snake venom phospholipase A2 (PLA2) activity derived from coral snakes (e.g., Micrurus tener, Micrurus fulvius, group IA PLA2) and some rattlesnakes (e.g., Crotalus scutulatus, group IIA PLA2) is medically significant. Of interest, the catalytic site of PLA2 also binds to activated clotting factor X, causing anticoagulation. Given that ruthenium (Ru)-containing compounds have been demonstrated to inactivate hemotoxic venoms in a solvent-dependent manner (e.g., 0.9% NaCl, phosphate-buffered saline), we wished to determine if RuCl3 would cause solvent-dependent inhibition

Stability of ruthenium oxide-coated microelectrode arrays for chronic stimulation of rat somatosensory cortex.

Intracortical microstimulation (ICMS) of the primary somatosensory cortex offers the potential for restoring sensory function in neuroprosthetic applications; however, achieving stable long-term performance from implanted electrode materials remains a challenge. Recently, ruthenium oxide (RuOx) has emerged as a viable faradaic coating material suitable for ICMS application due to its high-charge injection capacity, low impedance, and stable electrochemical properties. However, its long-term in vivo performance remains an area of active investigation. In this study, we evaluated the viability o

Light-Activated Ruthenium Nanoclusters Reprogram the Metabolic-Quorum Sensing Axis for Precision Periodontitis Therapy.

Periodontitis is a polymicrobial disease driven by metabolic interdependencies, wherein commensal bacteria fuel the virulence of keystone pathogens like Porphyromonas gingivalis (P. gingivalis). In particular, Veillonella parvula (V. parvula) supplies essential nutrients to the keystone pathogen P. gingivalis, enabling its virulence even at low abundance. Therefore, targeting V. parvula nitrate metabolism to deprive P. gingivalis of nutrients and attenuate its virulence represents a promising therapeutic strategy for controlling periodontitis progression. We engineered L-cysteine-capped ruthen
