Parkinson's disease (PD) is characterized by progressive loss of dopaminergic neurons in the substantia nigra, with ferroptosis emerging as critical pathogenic mechanisms. Recent evidence suggests that STING activation can induce neuronal ferroptosis through autophagic degradation of GPX4. Taurochenodeoxycholic acid (TCDCA), a naturally occurring bile acid, has demonstrated neuroprotective properties through activation of Takeda G protein-coupled receptor 5 (TGR5). However, whether TCDCA can improve PD by modulating the cGAS-STING-ferroptosis axis remains unexplored. We investigated the effect

The cGAS-STING (cyclic GMP-AMP synthase (cGAS)-stimulator of interferon genes (STING)) axis integrates DNA damage and cellular stress with type I interferon (IFN) signalling to facilitate transcriptional changes underlying inflammatory stress responses. The cGAS-STING pathway responds to cytosolic DNA in the form of double-stranded DNA, micronuclei, and long interspersed nuclear element 1 (L1) retroelements. L1 retroelements are a class of self-propagating non-long terminal repeat transposons that have remained highly active in mammalian genomes. L1 retroelements are emerging as important indu

Abnormal glial activation promotes neurodegeneration in Alzheimer's disease (AD), the most common cause of dementia. Stimulation of the cGAS-STING pathway induces microglial dysfunction and sterile inflammation, which exacerbates AD. We showed that inhibiting STING activation can control microglia and ameliorate a wide spectrum of AD symptoms. The cGAS-STING pathway is required for the detection of ectopic DNA and the subsequent immune response. Amyloid-β (Aβ) and tau induce mitochondrial stress, which causes DNA to be released into the cytoplasm of microglia. cGAS and STING are highly express

Titanium dioxide nanoparticles (TiO2 NPs) pose potential reproductive toxicity risks, notably compromising the blood-testis barrier (BTB), for which Sertoli cells are pivotal in maintaining integrity. However, certain research gaps remain regarding the molecular mechanisms by which TiO2 NPs induce apoptosis in Sertoli cells. This study aims to investigate whether TiO2 NPs trigger apoptosis through cGAS-STING-mediated down-regulation of MCUb in mouse Sertoli (TM4) cells. TM4 cells were exposed to various concentrations of TiO2 NPs (0, 25, 50, 75 and 100 μg/mL) for different periods (0, 3, 6, 12

BACKGROUND: Sepsis-induced acute lung injury (ALI) is characterized by excessive inflammation and pyroptosis of lung epithelial cells. AU-rich element binding factor 1 (AUF1) is a key RNA-binding protein involved in mRNA decay and regulation of inflammatory responses. Understanding AUF1's role in ALI could reveal novel therapeutic targets.
METHODS: We employed both in vivo (cecal ligation and puncture, CLP) and in vitro (lipopolysaccharide, LPS-treated cells) models of sepsis-induced ALI. Survival rates, lung histopathology, and expression levels of ZDHHC21, STING palmitoylation, and pyroptos

Natural immunity, the first line for the body to defense against the invasion of pathogen, serves as the body's perception of the presence of pathogens depends on nucleic acid recognition mechanisms. The cyclic GMP-AMP synthase-stimulator of the interferon gene (cGAS-STING) signaling pathway is considered an essential pattern recognition and effector pathway in the natural immune system and is mainly responsible for recognizing DNA molecules present in the cytoplasm and activating downstream signaling pathways to generate type I interferons and some other inflammatory factors. STING, a crucial

Stimulator of interferon genes (STING) is an innate immune receptor activated by natural or synthetic agonists to elicit antitumoral immune response via type I IFNs and other inflammatory cytokines. Bacillus Calmette-Guerin (BCG) is the standard of care as intravesical therapy for patients with high-risk non-muscle invasive bladder cancer (NMIBC). There are limited options available for patients with NMIBC who developed BCG unresponsiveness. In this study, we characterized in vitro and in vivo antitumor effects of E7766, a macrocyle-bridged STING agonist, via intravesical instillation in two s

BACKGROUND: Cabozantinib, a tyrosine kinase inhibitor (TKI) approved for advanced hepatocellular carcinoma (HCC), has established clinical benefit although the underlying immunomodulatory mechanisms, particularly those involving mitochondrial stress and the cGAS/STING pathway, remain poorly defined. METHODS: We assessed cabozantinib’s effects on mitochondrial integrity and innate immune signaling in hepatoma cells and macrophage cell lines, analyzing mitochondrial depolarization, reactive oxygen species production, cytosolic release of mitochondrial DNA (mtDNA), activation of the cGAS/STING pa

Non‑alcoholic steatohepatitis (NASH) is characterized by hepatic steatosis, inflammation, and fibrosis, yet therapeutic options remain limited. Aberrant activation of stimulator of interferon genes (STING) signaling in macrophages drives hepatic inflammation. Saikosaponin D (SSd), a bioactive triterpenoid saponin from Bupleurum falcatum, exhibits hepatoprotective properties, but its role in modulating STING-mediated macrophage polarization in NASH remains unknown. A NASH mouse model was established in C57BL/6J mice by feeding a methionine-choline-deficient (MCD) diet for 4 weeks, followed by 8

OBJECTIVES: DM is a rare type I IFN (IFN-I)-driven autoimmune disease, and anti-nuclear matrix protein 2 (NXP2) antibody is related to severe muscle disease and poor prognosis. Circulating cell-free DNA (ccf-DNA), including ccf-mitochondrial DNA and ccf-nuclear DNA, activates the cGAS/STING pathway to induce IFN-I production in autoimmune diseases. We investigated whether serum-derived ccf-DNA had a pathogenic effect on skeletal muscle in anti-NXP2 antibody-positive DM.
METHODS: Serum ccf-DNA levels were measured, and correlations between ccf-DNA and clinicopathological indicators were perfor

Immunotherapy targeting the immune functions of the tumor microenvironment (TME) is beneficial for colorectal cancer; however, the response rate is poor. Ciprofloxacin is a fluoroquinolone-class antibiotic that is used to treat bacterial infections. The purpose of this study is to assess the mechanism of ciprofloxacin that enhances anti-PD1 in colorectal cancer. We found that ciprofloxacin induced cytosolic DNA, including single-stranded and double-stranded DNA, formation in mouse CT26 colorectal adenocarcinoma cells. Molecules in DNA-sensing signaling such as cGAS, STING, and IFNβ mRNA and pr

BACKGROUND: Glaucoma is an optic neurodegenerative disease. Retinal ganglion cells (RGCs) are the fundamental neurons in the trabecular meshwork, and their loss is the main pathological reason for glaucoma. The present study was to investigate mechanisms that regulate RGCs survival.
METHODS: A mouse model of glaucoma was established by injecting hypertonic saline into the limbal veins. RGCs apoptosis was detected by using flow cytometry. Protein expressions in RGCs in response to DNA damage inducer cisplatin treatment were detected by immunofluorescence and western blot. The expressions of in

We previously generated STING N153S knock-in mice that have a human disease-associated gain-of-function mutation in STING. Patients with this mutation (STING N154S in humans) develop STING-associated vasculopathy with onset in infancy (SAVI), a severe pediatric autoinflammatory disease characterized by pulmonary fibrosis. Since this mutation promotes the upregulation of antiviral type I interferon-stimulated genes (ISGs), we hypothesized that STING N153S knock-in mice may develop more severe autoinflammatory disease in response to a virus challenge. To test this hypothesis, we infected heteroz

BACKGROUND: Hypoglycemia in diabetes markedly increases the risk of coronary artery events, but the mechanism by which it damages vascular endothelium through nitric oxide (NO) regulation of innate immune pathways remains unclear.
OBJECTIVE: This study aimed to investigate the mechanism by which NO may induce vascular endothelial injury under hypoglycemic conditions, potentially through activation of the cyclic GMP-AMP synthase/stimulator of interferon genes (cGAS/STING) pathway.
METHODS: The mouse model of diabetic hypoglycemia and a primary endothelial cell model of low-high glucose cyclin

Radiation and bacillus Calmette-Guérin (BCG) instillations are used clinically for treatment of urothelial carcinoma, but the precise mechanisms by which they activate an immune response remain elusive. The role of the cGAS-STING pathway has been implicated in both BCG and radiation-induced immune response; however, comparison of STING pathway molecules and the immune landscape following treatment in urothelial carcinoma has not been performed. We therefore comprehensively analyzed the local immune response in the bladder tumor microenvironment following radiotherapy and BCG instillations in a

Inflammatory signals from immunological cells may cause damage to intestinal epithelial cells (IECs), resulting in intestinal inflammation and tissue impairment. Interferon-γ-inducible protein 16 (IFI16) was reported to be involved in the pathogenesis of Behçet's syndrome (BS). This study aimed to investigate how inflammatory cytokines released by immunological cells and IFI16 participate in the pathogenesis of intestinal BS. RNA sequencing and real-time quantitative PCR (qPCR) showed that the positive regulation of tumor necrosis factor-α (TNF-α) production in peripheral blood mononuclear cel

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are neurodegenerative disorders that overlap in their clinical presentation, pathology and genetic origin. Autoimmune disorders are also overrepresented in both ALS and FTD, but this remains an unexplained epidemiologic observation1-3. Expansions of a hexanucleotide repeat (GGGGCC) in the C9orf72 gene are the most common cause of familial ALS and FTD (C9-ALS/FTD), and lead to both repeat-containing RNA and dipeptide accumulation, coupled with decreased C9orf72 protein expression in brain and peripheral blood cells4-6. Here w

AIM: Persistent activation of STING pathway is the basis for several autoimmune diseases. STING is activated by cGAMP, which is produced by cGAS in the presence of DNA. Results/methodology: HPLC-based medium throughput screening for inhibitors of cGAS identified suramin as a potent inhibitor. Unlike other reported cGAS inhibitors, which bind to the ATP/GTP binding site, suramin displaced the bound DNA from cGAS. Addition of suramin to THP1 cells reduced the levels of IFN-β mRNA and protein. Suramin did not inhibit lipopolysaccharide- or Pam3CSK4-induced IL-6 mRNA expression.
CONCLUSION: Suram

The cytosolic DNA sensor cyclic GMP-AMP synthase (cGAS) recognizes exogenous double-stranded DNA and produces 2'3'-cyclic GMP-AMP (2'3'-cGAMP), activating the stimulator of interferon genes (STING) and innate immunity. Bovine cGAS functions remain poorly understood. Herein, the coding sequence of the bo-cGAS gene was obtained and its recognition function was investigated. Bo-cGAS consists of 1542 nucleotides and the encoding acid sequence contained high sequence homology to that of other livestock. Bo-cGAS was localized in the endoplasmic reticulum and was abundant in the lung. Bo-cGAS and bo-
