Acetate but not propionate induces oxidative stress in bakers' yeast Saccharomyces cerevisiae.

The influence of acetic and propionic acids on baker's yeast was investigated in order to expand our understanding of the effect of weak organic acid food preservatives on eukaryotic cells. Both acids decreased yeast survival in a concentration-dependent manner, but with different efficiencies. The acids inhibited the fluorescein efflux from yeast cells. The inhibition constant of fluorescein extrusion from cells treated with acetate was significantly lower in parental strain than in either PDR12 (ABC-transporter Pdr12p) or WAR1 (transcriptional factor of Pdr12p) defective mutants. The constan

COQ9, a new gene required for the biosynthesis of coenzyme Q in Saccharomyces cerevisiae.

Currently, eight genes are known to be involved in coenzyme Q6 biosynthesis in Saccharomyces cerevisiae. Here, we report a new gene designated COQ9 that is also required for the biosynthesis of this lipoid quinone. The respiratory-deficient pet mutant C92 was found to be deficient in coenzyme Q and to have low mitochondrial NADH-cytochrome c reductase activity, which could be restored by addition of coenzyme Q2. The mutant was used to clone COQ9, corresponding to reading frame YLR201c on chromosome XII. The respiratory defect of C92 is complemented by COQ9 and suppressed by COQ8/ABC1. The latt

Novel proteins linking the actin cytoskeleton to the endocytic machinery in Saccharomyces cerevisiae.

The importance of coupling the process of endocytosis to factors regulating actin dynamics has been clearly demonstrated in yeast, and many proteins involved in these mechanisms have been identified and characterized. Here we demonstrate the importance of two additional cortical components, Ysc84p and Lsb5p, which together are essential for the organization of the actin cytoskeleton and for fluid phase endocytosis. Both Ysc84p and Lsb5p were identified through two-hybrid screens with different domains of the adaptor protein Sla1p. Ysc84p colocalizes with cortical actin and requires the presenc

The effect of azalomycin F on Ca2+ homeostasis in Trichoderma viride and Saccharomyces cerevisiae.

Azalomycin F (AMF), a macrocyclic lactone antibiotic, in concentrations of 10(-5) g/ml (10(-6) - 10(-5) mol/l) was found to stimulate both the 45Ca2+ influx and efflux in intact Trichoderma viride submerged mycelium and in cells of Saccharomyces cerevisiae without having Ca2+ ionophoric properties. AMF also inhibited ATP-dependent Ca2+ uptake in membrane fractions prepared from T. viride submerged mycelium. 45Ca2+ which had been accumulated in membrane fractions in an ATP-dependent manner was released upon addition of AMF. This release was observed in light organellar fractions (LOF) of S. cer

Telomere sequences at the novel joints of four independent amplifications in Saccharomyces cerevisiae.

Primary gene amplification, the mutation from one copy of a gene per genome to two or more genes per genome is a major mechanism of oncogene overexpression. We previously developed a system in the yeast Saccharomyces cerevisiae to phenotypically detect primary amplifications of a reporter cassette, ADH4:CUP1. We present here the sequence analysis of novel joints from four independent, spontaneous circular amplifications identified by the ADH4:CUP1 system. All four novel joints consist of C(1-3) A telomeric repeats joined to short (14- to 16-bp) CA-rich tracts between ADH4 and the telomere of c

Identification of Cryptosporidium parvum dihydrofolate reductase inhibitors by complementation in Saccharomyces cerevisiae.

There is a pressing need for drugs effective against the opportunistic protozoan pathogen Cryptosporidium parvum. Folate metabolic enzymes and enzymes of the thymidylate cycle, particularly dihydrofolate reductase (DHFR), have been widely exploited as chemotherapeutic targets. Although many DHFR inhibitors have been synthesized, only a few have been tested against C. parvum. To expedite and facilitate the discovery of effective anti-Cryptosporidium antifolates, we have developed a rapid and facile method to screen potential inhibitors of C. parvum DHFR using the model eukaryote, Saccharomyces

[Optimization of hydrolysis of protein substrates by yeast proteases from Saccharomyces carlsbergenis].

Optimum conditions for enzymatic hydrolysis of bovine blood in the presence of brewer's yeast Saccharomyces carlsbergensis were studied. In addition to the amount of blood, yeast, sodium citrate, and ethanol in the mixture, the optimum process duration and temperature were determined. The yield of the major end product (blood hydrolysate) increased from 11 to 55%, and the amino acid composition of the end product was brought to a balanced level as a result of hydrolysis optimization and addition of pancreatin to the reaction mixture.

Antibodies to Saccharomyces cerevisiae in patients with Crohn's disease and their possible pathogenic importance.

Saccharomyces cerevisiae (baker's yeast) may play an important part in the pathogenesis of Crohn's disease. Because of this the levels of IgG and IgA antibodies against three S cerevisiae strains (NCYC 77, NCYC 79, and NCYC 1108) were assayed in 49 patients with Crohn's disease, 43 with ulcerative colitis, 14 with coeliac disease, and 21 healthy controls. Coded serum samples were tested by ELISA. Similar antibody patterns to all three strains were found. IgG and IgA antibody levels were significantly raised in patients with Crohn's disease compared with healthy controls (p < 0.001 and p < 0.00

Specific Saccharomyces cerevisiae genes are expressed in response to DNA-damaging agents.

When exposed to DNA-damaging agents, the yeast Saccharomyces cerevisiae induces the expression of at least six specific genes. We have previously identified one damage inducible (DIN) gene as a gene fusion (din-lacZ fusion) whose expression increases in response to DNA-damaging treatments. We describe here the identification of five additional DIN genes as din-lacZ fusions and the responses of all six DIN genes to DNA-damaging agents. Northern blot analyses of the transcripts of two of the DIN genes show that their levels increase after exposure to DNA-damaging agents. Five of the din-lacZ fus

Bacterio-pharmacological activity of Saccharomyces boulardii in clindamycin-induced colitis in the hamster.

The effect of S. boulardii on experimental colitis induced by the single oral administration of 1 mg/kg of clindamycin in the Syrian hamster was studied. Oral administration of S. boulardii for 13 days, starting 3 days prior to the administration of clindamycin, significantly reduced the mortality rate and inhibited the growth of C. difficile in the caecum and colon. There was a marked improvement in the sub-mucosa and mucosa which showed major inflammatory lesions when clindamycin was administered alone.

Kinetics and activation energetics of death in Saccharomyces cerevisiae induced by sulfur dioxide.

Death of Saccharomyces cerevisiae induced by sulfur dioxide (K(2)S(2)O(2) was used as the SO(2) source) followed saturation kinetics. The enthalpy of activation of death was not affected by concentration over the range tested (5-150) mg/L of (K(2)S(2)O(2) at pH 3.4) and averaged 3.6 x 10(4) cal/mol as compared with 8.5 x 10(4) cal/mol for DeltaH of thermal death. The entropy of activation of death was hyperbolic function of the sulfur dioxide concentration, extrapolated at zero concentration to DeltaS(0) = 36.8 cal mol(-1) K(-1) and tended to DeltaDeltaS(max) = 13.2 cal mol(-1) K(-1) at satura

The impact of tetrabutylammonium lactate (TBAL) semiclathrate former on the viability of Saccharomyces cerevisiae.

Cell suspensions are stored long term via cryopreservation in mechanical ultra freezers or in liquid nitrogen with the addition of cryoprotective agents (CPAs). Cell viability is often compromised by ice formation, which causes osmotic stress from concentrated solutes and mechanical damage from ice recrystallization. There is a need for novel CPA formulations and alternative approaches. Some ionic polyalkyl-onium salts, miscible in water, form ice-like semiclathrate crystals. Semiclathrates can dissociate at temperatures greater than 0 °C and, unlike ice, incorporate semiclathrate forming solu

A Microfluidics-Based Assay for the Discovery of Gametogenic Rejuvenation Factors in Saccharomyces cerevisiae.

Saccharomyces cerevisiae progressively ages through asymmetrical cell divisions, retaining age-associated damage in the mother cell while producing young daughter cells. Gametogenesis completely resets the replicative lifespan of aged cells, thus rewinding the aging clock for the next generation. Although gametogenic rejuvenation in yeast has been described before, techniques to quantify the resetting of replicative lifespan at high throughput have been lacking. Previous methods for quantifying rejuvenation following gametogenesis were limited to low-throughput, labor-intensive approaches that

Simultaneous synthesis of resveratrol by dual-compartment metabolic engineering in Saccharomyces cerevisiae.

Resveratrol, a plant-derived polyphenolic compound, is widely used in medicine, health care, and nutrition. Synthetic biologyenables the microbial synthesis of resveratrol as apromising alternative to traditional plant extraction. However, an insufficient precursor supply remains a critical bottleneck limiting the overall yield. Here, for the first time, a dual-compartment engineering strategy was developed to simultaneously synthesize resveratrol in the peroxisomes and cytoplasm of Saccharomyces cerevisiae and to increase resveratrol synthesis in the presence of a sufficient supply of precurs

A haploid wild yeast resource for exploring the natural ecology of Saccharomyces cerevisiae.

Saccharomyces cerevisiae occurs predominantly in the diploid state in nature, limiting genetic analyses of wild populations. Here, we establish a haploid collection from 32 Taiwanese S. cerevisiae isolates through targeted HO disruption. This resource spans predomesticated Asian wild lineages and enables the investigation of reproductive isolation and ecological trait variation. Although all pairwise hybridizations formed zygotes, many yielded reduced spore viability, revealing strong postzygotic barriers. Genome analyses associated reduced hybrid fertility with lineage-specific structural var

Control of alcoholic fermentation through modulation of nitrogen metabolism in Saccharomyces cerevisiae.

Saccharomyces cerevisiae sake strains exhibit high alcoholic fermentation performance. Comparative transcriptomic analysis revealed that the expression of genes required for nitrogen sensing and metabolism, including amino acid biosynthesis and uptake, was markedly lower in the sake strain than in the laboratory strain. Thus, we hypothesized that changes in nitrogen metabolism affect the fermentation capability of S. cerevisiae. To evaluate the impact of altered nitrogen metabolism on alcoholic fermentation, we focused on the transcription activators Gcn4p, Gln3p, and Gat1p, and the protein ki

Regulating protein glycosylation modification enhances the synthesis of taxadiene in Saccharomyces cerevisiae.

Utilizing synthetic biology techniques to construct recombinant engineered cells for the synthesis of paclitaxel and its key precursors has emerged as an effective method to address the supply-demand imbalance and protect rare plant resources. Taxadiene, a critical precursor of paclitaxel, exhibits limited yield in eukaryotic systems, constraining its biosynthetic potential. Previous research has demonstrated that glycosylation modifications in Saccharomyces cerevisiae substantially impact the regulation of exogenous protein expression. In this study, we found that knocking out endogenous prot

The advances in creating Crabtree-negative Saccharomyces cerevisiae and the application for chemicals biosynthesis.

Saccharomyces cerevisiae is a promising microbial cell factory. However, the overflow metabolism, known as the Crabtree effect, directs the majority of the carbon source toward ethanol production, in many cases, resulting in low yields of other target chemicals and byproducts accumulation. To construct Crabtree-negative S. cerevisiae, the deletion of pyruvate decarboxylases and/or ethanol dehydrogenases is required. However, these modifications compromises the growth of the strains on glucose. This review discusses the metabolic engineering approaches used to eliminate ethanol production, the
