The responses of sulfonamides, sulfonamide-resistance genes (sul) and soil bacterial communities to different fertilization regimes were investigated by performing a field experiment using paddy soil with no fertilizer applied, chemical fertilizer applied, organic fertilizer applied, and combination of chemical and organic fertilizer applied. Applying organic fertilizer increased the bacterial community diversity and affected the bacterial community composition. Eutrophic bacteria (Bacteroidetes, Gemmatimonadetes, and Proteobacteria) were significantly enriched by applying organic fertilizer.

To date, the determination of sulfonamide metabolites in animal-derived food has universal disadvantages of low throughput and no integrated metabolites involved. In this study, a powerful and reliable strategy for high-throughput screening of sulfonamide metabolites in goat meat was proposed based on an aqueous two-phase separation procedure (ATPS) combined with ultrahigh-performance liquid chromatography quadrupole-Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap). Noncovalent interactions including van der Waals force, hydrogen bonding, and hydrophobic effect were determined to

A method for the acylation of sulfonamides and transacylation of N-acylsulfonamides has been developed. The acylation proceeds smoothly in the presence of Cu(NTf2)2 (0.1 mol%) using carboxylic anhydrides, acyl chlorides, or isopropenyl acetate as acylating agents. Alternative catalysts active in this transformation include Zn(NTf2)2, Mg(NTf2)2, La(NTf2)3, and Ce(NTf2)3. The transacylation of N-acylsulfonamides takes place smoothly at room temperature in excess acyl chlorides in the presence of Cu(NTf2)2 (20 mol%) and furnishes analogous N-acylsulfonamides through exchange of the N-acyl group.

Background/Objectives: Microtubule-targeting agents represent a pillar of cancer chemotherapy; however, their clinical utility is constrained by significant toxicity, pharmacokinetic instability, and susceptibility to multidrug resistance transporters. This study aimed to explore the impact of replacing substituted phenyl rings with a naphthalene moiety in sulfonamide-based colchicine-site ligands, with the goal of identifying new antiproliferative candidates with improved profiles. Methods: We designed, synthesized, and evaluated a library of 35 naphthalene sulfonamides bearing varied aryl gr

Non-nucleoside reverse transcriptase inhibitors are key compounds of current antiretroviral therapy for people living with human immunodeficiency virus (HIV). Indole-3-sulfonamides have demonstrated good biological profile against HIV. Here we describe a high-throughput experimentation (HTE)-compatible route to access indole-3-sulfonamides through a selective activation of the S-N bond of a primary indole-3-sulfonamide. This selective activation strategy enables efficient installation of diverse amine substituents with broad functional-group tolerance. Cheminformatic tools were integrated into

Currently, the synthesis of bioactive sulfonamides using amino acid as a starting reagent has become an area of research interest in organic chemistry. Over the years, an amine-sulfonyl chloride reaction has been adopted as a common step in traditional sulfonamide synthetic methods. However, recent developments have shown amino acids to be better precursors than amines in the synthesis of sulfonamides. Although amines and amino acids have some structural similarities, using amino acids rather than amines in the synthesis of sulfonamides minimizes several drawbacks. Comparatively, amino acids a

Vascular endothelial growth factor receptor-2 (VEGFR-2), a tyrosine kinase receptor (TKR), plays a crucial role in angiogenesis and is overexpressed in most cancers. It is important for tumor angiogenesis, facilitating essential angiogenic cellular processes, such as promoting endothelial cell survival, proliferation, migration, and vascular permeability. Consequently, VEGFR-2 has become one of the main targets for anti-angiogenic therapy, with its inhibition serving as a crucial strategy for developing new drugs to mitigate angiogenesis-dependent cancers. Small-molecule drugs targeting VEGFR-

We report the first highly enantioselective carbene insertion into the N-H bonds of sulfonamides, affording α-amino esters in moderate to good yields with excellent enantioselectivity (up to 98% ee). The success of this transformation relies on the synergistic effect between a ruthenium carbene intermediate and a chiral phosphoric acid. Slight modification of the reaction conditions further enables the use of carbamates as N-nucleophiles, achieving exceptional enantiocontrol (up to 99% ee). Additionally, this catalytic system facilitates an asymmetric three-component reaction involving sulfona

Thermosets are indispensable for their lightweight and excellent thermomechanical properties but suffer from nonrecyclability, exacerbating plastic pollution. Covalent Adaptable Networks (CANs), leveraging dynamic covalent bonds, address this challenge. This study explores β-amino sulfonamides (BASA) as novel dynamic and thermoreversible linkers for CANs. Compared to previously investigated β-amino amides (BAAs), small molecule BASA studies show a faster amine exchange reaction via (retro)aza-Michael reactions in a temperature window from 140 to 180 °C. More importantly, in contrast to the acr

We report the mechanism of the iron-catalyzed oxidative α-amination of ketones with sulfonamides. Using linear free energy relationships, competition experiments, and identification of reaction intermediates, we have found that the mechanism of this reaction proceeds through rate-limiting electron transfer to 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ) from an iron enolate in the process of forming an α-DDQ adduct. The adduct then serves as the electrophile for substitution with sulfonamide nucleophiles, accelerated by iron and additional DDQ. This mechanistic study rules out formation of

A general method for the light-driven intermolecular anti-Markovnikov hydroamination of alkenes with primary sulfonamides, sulfamides, and sulfamates is presented. The reaction is mediated by a ternary catalyst system composed of an iridium(III) chromophore, a fluorinated alkoxide base, and a thiol H-atom donor. We hypothesize that the reactions proceed through a proton coupled electron-transfer (PCET) mechanism wherein implementation of the alkoxide base imparts additional thermochemical driving force for the homolytic activation of strong N-H bonds that were previously inaccessible using thi

Antibiotics play a key role in the fight against bacterial diseases. However, bacteria quickly learn how to minimize the effects of antibiotics and strengthen their resistance. Thus, the fight against them becomes more and more difficult and there is a constant search for new bactericidal compounds. It is important in this type of search to determine the basic properties of compounds such as pKa, hydrogen bond formation, or hydrophobicity. Here, we present the results of our in silico study of five sulfonamide derivatives differing in alkylamine substituent length. Based on our results, we pro

Both co-cultivation and co-substrate addition strategies have exhibited massive potential in microalgae-based antibiotic bioremediation. In this study, glucose and sodium acetate were employed as co-substrate in the cultivation of microalgae-bacteria consortium for enhanced sulfadiazine (SDZ) and sulfamethoxazole (SMX) removal. Glucose demonstrated a two-fold increase in biomass production with a maximum specific growth rate of 0.63 ± 0.01 d-1 compared with sodium acetate. The supplementation of co-substrate enhanced the degradation of SDZ significantly up to 703 ± 18% for sodium acetate and 2

We describe the synthesis and biological evaluation of a series of novel aryl sulfonamides that exhibit potent inhibition of NaV1.5. Unlike local anesthetics that are currently used for treatment of Long QT Syndrome 3 (LQT-3), the most potent compound (-)-6 in this series shows high selectivity over hERG and other cardiac ion channels and has a low brain to plasma ratio to minimize CNS side effects. Compound (-)-6 is also effective inshortening prolonged action potential durations (APDs) in a pharmacological model of LQT-3 syndrome in pluripotent stem cell-derived cardiomyocytes (iPSC-CMs). Un

Sulfonamides (SAs) are a kind of antibiotics widely used in medical treatment and livestock breeding. However, they have poor degradability in human and animal intestines, and will enter the sewage treatment system through the discharge of feces and urine. The aerobic activated sludge (AAS) in wastewater treatment plant was found to be able to effectively transform SAs. This article summarizes the advances in biodegradation of SAs in aerobic activated sludge system, which includes the biodegradation mechanisms, the main biodegradation pathways, and the environmental factors affecting the degra

Liquid manure is frequently used as soil fertilizer due to its high nutrient content. It can also contain residues of pharmaceuticals, such as antibiotics, if farm animals are medicated. The anaerobic fermentation process in biogas plants is discussed as one way to reduce the input of antibiotics into the environment.
