Automated Identification of Ventricular Tachycardia Ablation Targets: Multicenter Validation and Workflow Characterization.

Decrement evoked potentials (EPs) (DeEPs) constitute an accepted method to identify physiological ventricular tachycardia (VT) ablation targets without inducing VT. The feasibility of automated software (SW) in the detection of arrhythmogenic VT substrate has been documented. However, multicenter validation of automated SW and workflow has yet to be characterized. The objective of this study was to describe the functionality of a novel DeEP SW (Biosense Webster, Diamond Bar, CA, USA) and evaluate the independent performance of the automated algorithm using multicenter data. VT ablation cases w

Role of imaging in current acute ischemic stroke workflow for endovascular therapy.

Ischemic stroke is caused by a thrombus that blocks an intracranial artery. Brain tissue beyond the blocked artery survives for a variable period of time because of blood and nutrients received through tiny vessels called collaterals. Imaging the brain and the vasculature that supplies it is therefore a vital first step in treating patients with acute ischemic stroke. In this review, we focus on current evidence for imaging selection of patients for endovascular therapy in the context of the recently positive clinical trials, such as Multicenter Randomized Clinical Trial of Endovascular Treatm

An evidence-based workflow for integrating molecular testing with histopathology in the workup of meningiomas.

BACKGROUND: Meningiomas are graded using histologic and molecular criteria according to the World Health Organization (WHO) classification. However, there is scant evidence considering the specific value of each individual atypical histological feature that contributes to WHO grading. There is also little evidence-based guidance as to which cases should receive molecular testing. METHODS: Detailed histological information was collected for 1058 resected meningiomas, including cases with genomic DNA methylation and copy number variant (CNV) data, next-generation sequencing, and RNA risk score.

New definition, diagnostic criteria, and diagnostic workflows for patients with suspected COMISA.

BACKGROUND: The co-occurrence of insomnia and sleep apnea was first reported in 1973; however, precise definition, diagnostic criteria, and diagnostic workflows have not been provided until now. As more studies have been conducted, this has led to heterogeneous patient classifications and increased variability in results obtained from these studies. METHODS: Through a narrative review including randomized controlled trials, as well as retrospective and prospective studies-we aimed to address the variations in diagnostic criteria. Additionally, we examined reviews, editorials, and other publis

An automated workflow for quantifying the formation of synuclein aggregates in human dopaminergic neurons.

Parkinson's disease (PD) is a neurodegenerative disorder characterized by alpha-synuclein (α-syn) aggregates termed Lewy bodies. To model PD pathology in vitro, preformed fibrils of α-syn (PFFs), which can be taken up by cells, provide a seed that drives misfolding and aggregation of endogenous α-syn, with new aggregates amplifying this process. External application of PFFs to dopaminergic neurons (DNs) increases aggregate formation, marked by α-syn phosphorylation at serine 129 (pS129-syn), a pathological PD marker. Building on this, we developed an automated synuclein seeding assay to quanti

Electronic Health Record Workflows in Acute Care Surgery: Ethnographic Study.

BACKGROUND: Systematic strategies to harness electronic health record (EHR) workflows, reduce redundancy, and support decision-making remain limited in acute care surgery (ACS). Understanding how EHR systems and workflows intersect with time-sensitive settings is critical to improving decision-making and outcomes in ACS. OBJECTIVE: This study aimed to evaluate how ACS clinicians leverage the EHR for decision-making and to identify opportunities and challenges for EHR-enabled decision support. METHODS: We conducted a qualitative ethnographic study over a 6-month period, combining in-depth int

tgLang: A Domain-Specific Language for Geometry Processing and Computational Imaging Workflows.

Geometry-processing and computational-imaging workflows combine heterogeneous data structures, topology-changing edits, dense numerical fields, visualization, and repeated experimental variation. These workflows are often clear as algorithms but obscured in software by traversal boilerplate, representation conversions, build-system boundaries, and ad hoc scripting conventions. This paper presents tgLang, a domain-specific language with explicit, runtime-enforced representation types that makes meshes, point clouds, curve networks, grids, two-dimensional images, and image stacks first-class exe

A simulation framework for evaluating electronic order workflows in integrated health records.

Electronic health record (EHR) systems are critical to modern healthcare delivery, yet the dynamic workflows that govern electronic order processing remain underexplored. Inefficiencies in these digital pathways can cause delays in care, repetitive workloads, and even patient harm. This study presents a discrete-event simulation framework used to reconstruct and evaluate EHR-based order workflows in a large integrated healthcare system. Using real-world data extracted from the Veterans Health Administration's Corporate Data Warehouse, the authors mapped order events to standardized state trans

RheoKit: A Workflow for Green-Kubo Viscosity Analysis in Tinker and Tinker-HP.

We present RheoKit, a workflow that automates viscosity calculations via the Green-Kubo formalism in Tinker 25 and Tinker-HP. Additionally, we introduce new user-facing enhancements to Tinker 25 and Tinker-HP that expose stress tensor data upon request. We validate the implementation and workflow by calculating the viscosity of water, ethanol, isopropanol, and toluene using the AMOEBA polarizable force field. Our results confirm the accuracy of our implementation and suggest that AMOEBA may benefit from parameter tuning for improved experimental agreement.

A critical review of analytical methods in the electroplating industry: Workflow, methods and limits.

Process control in electroplating industry requires rapid analytical response and methodological robustness of highly concentrated, multi-component chemical matrices. This critical review provides a comprehensive evaluation of the analytical workflows governing the electrodeposition sector, categorizing methodologies into preventative monitoring of the electrolytic solution and the characterization of the resulting metal coatings. We critically evaluate the figures of merit, operational limitations, and matrix tolerances of both state-of-the-art instrumental techniques and classical wet chemic

Preparing for Shotgun Sequencing in Forensic Genetics-A Workflow for Analysis of Monozygotic Twins.

BACKGROUND/OBJECTIVES: Identifying the true perpetrator among monozygotic (MZ) twins has long posed a challenge in forensic genetics and for police investigations because MZ twins are likely identical in their short tandem repeat (STR) profile. In this study, we propose a workflow to address this issue, anchored in a case study involving MZ twins. METHODS: The workflow includes shotgun sequencing of reference samples from one twin and trace samples, followed by a targeted amplicon-based approach (AmpliSeq™ Custom Panel) to validate the identified variants. RESULTS: Biological traces from the

Statistical Considerations for Research Reproducibility: Strategies for Building Transparent Analytic Workflows.

BACKGROUND: Reproducibility, the ability for independent investigators to obtain consistent results using the same data and analytic procedures, is foundational to scientific integrity, yet remains a challenge across research settings. Common practices such as manual data cleaning, point-and-click analyses, and creating tables by manually copying and pasting values from output to table shells increase the likelihood of error, reduce transparency, and impede efficient collaboration. As data become more complex and expectations for reproducibility from journals, funders, and the scientific commu

An institutional experience of a scripting-driven automation of the emergent palliative radiotherapy workflow.

BACKGROUND: Emergency palliative radiotherapy is often delivered under time-constrained and high-stress conditions, frequently bypassing conventional computed tomography (CT) simulation and relying on nonstandard workflows. These factors increase variability and the potential for error. PURPOSE: This work describes the development, implementation, and early clinical evaluation of a simulation-free, cone-beam CT (CBCT) based emergency radiotherapy workflow that leverages treatment planning system (TPS) scripting to standardize and automate deterministic steps while preserving clinical oversigh

Application of a Multiomics Imaging Workflow to Explore Asparlas Treatment in Solid Tumors.

In acute lymphoblastic leukemia (ALL), hypermethylation of the asparagine synthetase (ASNS) gene promoter, leading to low levels of ASNS in tumor cells, is recognized as a prognostic biomarker, and l-asparaginase-based treatments (e.g., Asparlas) are frequently administered to these patients. In these cancers, tumor cells rely on external asparagine, and its depletion in the bloodstream results in tumor cell apoptosis. A multiomics (imaging) workflow is required to evaluate key molecular changes and characterize solid tumors to explore the potential efficacy of Asparlas in solid tumors. This s

Using the Simcyp R Package for PBPK Simulation Workflows With the Simcyp Simulator.

Physiologically based pharmacokinetic and pharmacodynamic (PBPK/PD) modeling aims to understand how a drug is absorbed, distributed, metabolized, excreted, and acts in a human or animal body. The Simcyp Simulator is a well-known commercial PBPK/PD simulation software offering many options. It can, for example, use multiple compounds and population specification files to study the behavior of specific drug formulations in particular population groups. Such features can greatly speed up and optimize clinical research and drug development studies. On the other hand, the statistical coding languag

An Intelligent and Efficient Workflow for Path-Oriented 3D Bioprinting of Tubular Scaffolds.

Modern 3D printing is a valuable tool for tissue engineering (TE), and the fabrication of complex geometries such as tubular scaffolds with adaptable structure, for example, as replacements for intestines, bronchi, esophagus, or vessels, could contribute to standardized procedures in the future of regenerative medicine. However, high-precision bioprinting of scaffolds for tubular TE applications remain a major challenge and is an arduous endeavor with currently available three-axis bioprinters, which are limited to planar, layer-by-layer printing processes. In this work, a novel, straightforwa

Primary Cutaneous Cryptococcosis in an Immunocompetent Patient: Diagnostic Workflow and Choice of Treatment.

Cryptococcosis is an opportunistic infection in immunocompromised patients, involving mainly the lungs and central nervous system; however, the skin, eyes and genitourinary tract could also be involved as secondary sites of infection. Primary cutaneous cryptococcosis (PCC) is a distinct clinical entity that can occur in both immunocompetent and -compromised patients, usually trough skin injury. In immunocompetent patients, it is a very rare infection, presenting with non-specific clinical pictures and being challenging to diagnose. Herein, we present the case of an immunocompetent man with PCC
