Buffer-dependent variations in alginate oligosaccharide production and their biofunctional properties in colonic fermentation.

The inclusion of a buffer in the enzymatic reaction medium can significantly affect the production and biofunctional properties of alginate oligosaccharides (AOS). This comparative study evaluated the impact of buffer addition (AOSWB) versus no buffer (AOSWoB) on the bioactivity and technofunctional properties of AOS. AOSWB showed reduced chelating activity (193.83 ± 6.29 μmol TE/g dw) and lower reducing sugars (76.92 ± 0.50 mg GE/g dw), despite treatment selection optimized via principal component analysis (PCA). AOSWoB exhibited a bidentate bridge chelation mechanism with barium, affecting b

Continuous production and extraction of monodisperse alginate microgels via deterministic lateral displacement.

Over the past two decades, microfluidic approaches have been extensively explored for the production of monodisperse alginate hydrogel microparticles, particularly for applications such as 3D in vitro cell culture in tissue engineering. However, conventional methods often suffer from satellite droplet contamination and inefficient oil-to-water phase transfer, limiting purity and scalability. Here, we present an integrated microfluidic platform for the continuous, on-chip production, gelation, extraction, and purification of monodisperse alginate hydrogel microparticles. The device combines a f

Methacrylated alginate hydrogels for water absorption and metal ion removal in environmental applications.

Water contamination by heavy metals poses significant environmental risks, requiring efficient remediation strategies. In this study, methacrylated alginate hydrogels were synthesized and photocrosslinked under UV radiation to develop an effective adsorbent material. A design of experiments (DOE) approach was used to evaluate the effects of alginate concentration (20-30 g L-1) and UV exposure time (1-3 h) on swelling capacity and metal adsorption. The hydrogels exhibited tunable swelling behavior (135 ± 0.4% to 442.9 ± 26.2%) that was influenced by polymer network density. Adsorption tests rev

Microfluidic alginate encapsulation for stabilization of carotenoid-rich microalgal biomass of Chromochloris zofingiensis.

Carotenoids have considerable value because of their strong antioxidant activity; however, their practical applications are limited by their low stability under light exposure. In this study, a microfluidic flow-focusing technique was employed to encapsulate intact Chromochloris zofingiensis cells within alginate microcapsules to enhance the photostability of intracellular carotenoids. Microcapsules were produced using aqueous sodium alginate solutions containing microalgae, which were dispersed into mineral oil containing Span 80, followed by ionic gelation in calcium chloride solution. Spectral Trajectory Analysis of Drug Release from Bioprinted Alginate-Based Hydrogels.

The release of molecular probes from soft hydrogel matrices involves coupled transport and structural relaxation processes that are difficult to capture using predefined kinetic models. Here we investigate the release dynamics of indocyanine green from bioprinted alginate-based hydrogels, including alginate-gelatin and calcium-cross-linked alginate-poly-(vinyl alcohol) matrices, using a data-driven spectroscopic approach. Time-resolved UV-vis absorption spectra collected in the surrounding aqueous phase are analyzed by singular value decomposition to construct reduced-dimensional trajectories

One-Pot Strategy to Prepare Structurally Colored Alginate Dispersions for Functional Applications.

Structural colored hydrogels have emerged as a class of flexible optical materials with broad application prospects in smart color displays, wearable devices, sensors, and beyond. However, conventional preparation methods are often limited by time-consuming procedures and limited production scales. In this work, a structural colored sodium alginate (SA) dispersion was prepared by simply mixing an SA solution with an ultralow concentration of poly(styrene-methyl methacrylate-acrylic acid) colloidal emulsion and a small amount of cuttlefish ink. Depletion-induced interactions between the polymer

Disease-modifying efficacy of alginate-microencapsulated probiotics in an animal model of ulcerative colitis.

Ulcerative colitis (UC) can be challenging to control with current therapies. Modulating gut dysbiosis with probiotics is a promising approach, but the benefits depend on keeping the cell viability as they pass through gastrointestinal transit (GIT). This study developed an alginate microencapsulation system and evaluated its overall disease-modifying efficacy in an acetic acid-induced colitis model. Lactiplantibacillus plantarum and Levilactobacillus brevis were microencapsulated in calcium alginate beads via extrusion. Encapsulation efficiency, morphology, and viability under simulated GIT c

Sodium alginate-modified copper catalytic membrane for carbene N-H insertion.

A novel copper-based catalytic membrane (Cu@SA-CM) was fabricated via surface cross-linking using sodium alginate as a green modifier. Characterization via FTIR, FESEM, EDS, and XPS confirms that copper is uniformly dispersed on the membrane and predominantly exists in the +2 oxidation state (Cu2+). This membrane efficiently catalyzes N-H insertion of carbenes, generated from diazo compounds, into N-heterocycles under mild conditions with high selectivity. In a continuous-flow system, Cu@SA-CM maintained consistent performance over 20 cycles, attributed to sodium alginate's dual role in coordi

Organocatalytically-Tuned In Situ Formed Alginate Hydrogel Enables Sustained Therapeutic Release.

In situ hydrogelation of alginate represents a versatile strategy for biomaterial design, enabling gel formation directly under physiological conditions without external triggers. In this study, oxidized alginate (Alg-O) was covalently crosslinked with guanidine hydrazide (GH) through acid-catalysed hydrazone linkages to yield stable Alg-Gl hydrogels at ambient conditions. Systematic evaluation of gelation parameters demonstrated that both pH and organocatalyst selection modulate gelation kinetics and minimum gelation concentration, with mildly acidic environments and anthranilic acid catalysi

Fabrication and controlled release behavior of metal-polyphenol-decorated alginate hydrogel beads.

Hydrogel-based controlled release systems are widely used in pharmaceutical and agricultural fields. To achieve controlled release of hydrophilic drugs, which otherwise release rapidly through hydrated polymer networks, we develop a facile ferric ion co-crosslinking strategy to fabricate composite hydrogel beads. The highly porous composite hydrogel beads were prepared by blending recycled household formaldehyde adsorbent (RFA) with sodium alginate (Alg) at a 1:1 (w/w) ratio, followed by Fe3+-induced crosslinking and subsequent surface functionalization with tannic acid (TA). Through altering

Alginate-Amphotericin B bifunctional conjugates have improved solubility and reduced toxicity.

Alginates are a family of linear anionic polysaccharides produced by brown algae and some bacteria and consist of 1 → 4 linked β-d-mannuronic acid (M), and its C5-epimer α-l-guluronic acid (G). Research has shown that oligoguluronates possess biofilm disruption properties, which combined with their high water-solubility and low toxicity, represent ideal candidates for creating novel bifunctional antifungal compounds. The progressive increase in antifungal resistance is a major global human health threat. This intrinsic resistance illustrates the need to develop improved antifungal drugs with g

Alginate-Encapsulated HDES as a Green Sorbent for Elemental Preconcentration Prior to Determination by FAAS.

This study presents the development, characterization, and application of a new solid-phase extraction (SPE) sorbent for the preconcentration of Cd, Co, Cu, Fe, Mn, Ni, and Zn in water samples prior to analysis by flame atomic absorption spectrometry (FAAS). The sorbent was prepared by encapsulating hydrophobic deep eutectic solvents (HDES) in calcium alginate, a strategy that prevents HDES accumulation in FAAS and avoids nebulization and transport interferences. The developed method allows the analyte to be eluted into a clean aqueous phase (HNO3 3.5% v/v), containing negligible amounts of HD

Injectable biochar-reinforced alginate hydrogels for hemostasis under wet and deformable conditions.

Uncontrolled bleeding under wet and deformable conditions remains a major challenge for conventional hemostatic materials, which often suffer from limited tissue adhesion and poor mechanical adaptability. Here, we developed an injectable sodium alginate hydrogel ionically cross-linked with Ca2⁺ and reinforced with hazelnut branch-derived biochar (HB) to improve structural stability and local hemostatic performance. The HB-containing hydrogel exhibited pronounced shear-thinning behavior, high injectability, enhanced wet tissue adhesion, and self-healing efficiency exceeding 80%, while achieving

Preparation and functional evaluation of an alginate-targeted monoclonal antibody against Pseudomonas aeruginosa.

Pseudomonas aeruginosa is a critical-priority pathogen responsible for life-threatening infections in immunocompromised individuals, yet its extensive antigenic heterogeneity has impeded the development of immunotherapeutics. Alginate, a structurally conserved surface polysaccharide that forms the backbone of the biofilm matrix of mucoid P. aeruginosa strains in chronic infections, represents a compelling target. Here, we report the generation and functional evaluation of mAb 546B, a murine IgG1 monoclonal antibody directed against the synthetic mannuronic acid tetrasaccharide epitope 1. This

Enhanced phoxim biodegradation by immobilizing Novosphingobium sp. RL4 on attapulgite-sodium alginate.

BACKGROUND: Residual phoxim pollution presents a potential threat to natural ecosystems and human health. The immobilization of degrading strains on natural adsorbent materials is a common strategy to enhance the degradation of target compounds in the environment by the strains. METHODS: A phoxim-degrading bacterial strain was isolated from the rhizosphere soil of rhubarb (Rheum palmatum L.), which had been exposed to long-term phoxim contamination. To enhance its stability and practical applicability, sodium alginate (SA) was utilized as a carrier material, while biochar (BC) and attapulgite

Phage-loaded alginate films and coatings for biofilm inhibition and control in food packaging.

Food packaging plays a crucial role in ensuring food safety; however, microbial contamination, particularly from biofilm-forming bacteria, remains a challenge. This study explores the use of sodium alginate-based films and coatings loaded with a bacteriophage cocktail targeting Escherichia coli and Pseudomonas fluorescens to mitigate biofilm formation in food packaging, with a focus on cheese products. The coatings were applied to parchment paper, polystyrene, and films prepared with sodium alginate loaded with bacteriophages, and assessed for their antimicrobial efficacy. Biofilm inhibition a

Dual-Cross-Linked Alginate Hydrogels as a Strategy to Improve the Antifungal Properties of Posaconazole.

Background/Objectives: Despite the continuous development of medicine, the treatment of dermatological fungal infections is difficult due to their chronic nature, recurrence, and resistance of some pathogens to standard therapies. In order to improve the effectiveness of treatment, not only are new active substances with antifungal activity synthesized, but new, unconventional carriers are also developed for substances already used. Methods: Therefore, the focus of this research was to evaluate the possibility of using a combination of two cross-linking techniques for sodium alginate ionic cro

Alginate-Gelatin Hydrogel Scaffold Model for Hypoxia Induction in Glioblastoma Embedded Spheroids.

Glioblastoma (GBM) is a highly aggressive and malignant brain tumor, characterized by hypoxia in its microenvironment, which drives its growth and resistance to treatments. Hypoxia-inducible factor 1 (HIF-1) plays a central role in GBM progression by regulating cellular adaptation to low oxygen availability, promoting processes such as angiogenesis and cell invasion. However, studying and modeling GBM under hypoxic conditions is complex, especially due to the limitations of animal models. In this study, we developed a glioma spheroid model using an alginate-gelatin hydrogel scaffold, which ena

Alginate gels: Chemistry, gelation mechanisms, and therapeutic applications with a focus on GERD treatment.

Alginate, a natural polysaccharide derived primarily from marine algae, has become popular in biomedical research due to its versatile gelation properties and biocompatibility. This review explores the chemistry, gelation mechanisms, and therapeutic applications of alginate gels, with a particular focus on their role in gastroesophageal reflux disease (GERD) management. Alginate's structure, comprised of guluronic and mannuronic acid blocks, allows for gel formation by ionic cross-linking with divalent cations like calcium ions, generating a stable "egg-box" structure. Creep of Alginate-Gelatin-Hyaluronic acid strands and cell viability after bioprinting.

The success of 3D bioprinting in tissue engineering relies on i) precise bioink deposition for creating intricate tissue architectures and ii) good cell viability after printing. However, printed strands made of hydrogels are susceptible to time-dependent deformation -known as creep- which can compromise printing accuracy. Creep might be reduced by increasing the crosslink density, but this could be deleterious for cells in the bioink. Therefore, this study investigates the impact of creep on the printability of an Alginate-Gelatin-Hyaluronic acid bioink. Creep data were fitted with a linear r
