Survival after pathological complete response following neoadjuvant chemotherapy versus chemoradiotherapy for oesophageal squamous cell carcinoma.

BACKGROUND: Although several oesophageal squamous cell carcinoma (OSCC) studies have reported no definitive overall survival (OS) differences between neoadjuvant chemoradiotherapy (NACRT) and neoadjuvant chemotherapy (NAC), the higher pCR rate with NACRT has been viewed as a potential advantage. Beyond ongoing concerns about the validity of pCR as a surrogate endpoint, it remains uncertain whether survival differs between these modalities among patients with OSCC who achieve pCR. METHODS: An integrated analysis of individual patient data (IPD) from phase III trials evaluating perioperative th

Clinicopathological predictors of distant recurrence in breast cancer patients achieving pathological complete response after neoadjuvant chemotherapy.

BACKGROUND: Pathological complete response (pCR) after neoadjuvant chemotherapy (NAC) is associated with favorable prognosis in breast cancer; however, a subset of patients still experience distant recurrence. Data on long-term outcomes and predictors of recurrence after pCR remain limited. METHOD: This retrospective observational study was conducted as an extended follow-up analysis of a previously reported multicenter cohort. We included 365 patients with primary breast cancer who achieved pCR after NAC at four institutions between 2007 and 2016. Distant metastasis-free survival (DMFS) was

Ultrasound-based shrinkage patterns at mid-neoadjuvant chemotherapy for predicting pathological complete response in invasive breast cancer.

OBJECTIVE: To develop an ultrasound-based tumor shrinkage pattern and subsequently construct a predictive model for pathological complete response by analyzing changes in ultrasonic features and measurements at mid-treatment of neoadjuvant chemotherapy (NAC) in invasive breast cancer, ultimately evaluating its clinical applicability. METHODS: This study included all breast cancer patients who underwent NAC and subsequent surgery between January 2017 and December 2024. All patients received breast ultrasound examinations before and during NAC. All breast tumor images were reviewed by two exper

Pathological complete response and postmastectomy radiation therapy after primary systemic therapy: results from the ReSTORE study.

BACKGROUND: The ReSTORE study aimed to evaluate oncological outcomes and explore the association between pathological complete response (pCR), postmastectomy radiotherapy and nodal irradiation (PMRT), and recurrence. METHODS: This multicentre retrospective study included patients with breast cancer treated with chemotherapy-based primary systemic therapy (PST) followed by skin-or nipple-sparing mastectomy and immediate reconstruction. Locoregional recurrence-free survival (LRRFS) and disease-free survival (DFS) were analysed. RESULTS: A total of 256 patients were analysed, with a median foll

Use of sintilimab in primary adenosquamous carcinoma of the liver results in pathological complete response: a case report and literature review.

Adenosquamous cell carcinoma (ASC) is a rare and aggressive malignant tumor which consists of both adenocarcinoma (AC) and squamous cell carcinoma (SCC) component types. Although ASC can sometimes develop in the stomach, pancreas, gallbladder and thyroid, it rarely occurs in the liver. As such, primary ASC of the liver remains a poorly understood malignancy due to both the paucity of reported cases and scarcity of available published data. As such, while the use of immune checkpoint inhibitors (ICIs), including PD-1 and PD-L1 antagonists, has profoundly changed the treatment paradigm and outco

Pathological complete response to pembrolizumab in recurrent retroperitoneal dedifferentiated liposarcoma with high tumor mutational burden: a case report.

BACKGROUND: Dedifferentiated liposarcoma (DDLPS), a common subtype of retroperitoneal soft tissue sarcoma, generally shows a limited response to conventional chemotherapy. Recently, immune checkpoint inhibitors (ICIs) have demonstrated promising activity against DDLPS. However, predictive biomarkers for response remain unclear. We present a case of recurrent retroperitoneal DDLPS with a high tumor mutational burden (TMB), demonstrating a pathological complete response (pCR) to pembrolizumab. We also explore the association between the response to ICIs and the tumor microenvironment, including

Predictive factors for complete pathological response in hormone receptor-negative breast cancer patients undergoing neoadjuvant chemotherapy.

Complete pathological response (pCR) is a pivotal predictor of enhanced disease-free and overall survival rates in breast cancer patients. Accurate prediction of pCR is therefore of paramount clinical significance. This retrospective study aimed to delineate the factors associated with pCR through a comprehensive analysis encompassing clinical, pathological, and immunohistochemical profiling of patients diagnosed with hormone receptor-negative invasive ductal carcinomas. The study cohort was composed of 73 female patients. The cases were reviewed retrospectively using data from University Hosp

Pathological Complete Response to a Single Dose of Pembrolizumab-Based Chemoimmunotherapy for Squamous Cell Carcinoma of the Lung: A Case Report.

We herein describe a patient with non-small-cell lung cancer who achieved pCR with a single dose of pembrolizumab-based chemoimmunotherapy followed by surgery. A 61-year-old man was referred to our hospital with wheezing and an abnormal chest shadow. Squamous cell carcinoma of the left lower lobe, cT2aN1M0 stage IIB, was diagnosed and pembrolizumab-based chemoimmunotherapy was initiated at the patient's request. One month later, chest CT revealed new ground-glass opacities of the lungs, which were judged to be a CTCAE grade 2 pneumonitis due to an immune-related adverse event (irAE). Therefore

Neoadjuvant Chemotherapy with Low-Dose Nivolumab Leading to Complete Pathological Response in Non-small Cell Lung Carcinoma.

AIMS: The milk acidification rate of Streptococcus thermophilus strains can be affected by several factors, one of which is the hydrolysis of urea by the urease complex. To evaluate the technological suitability of S. thermophilus strains deprived of urease activity in milk fermentation, the genetic cluster related to urease enzymatic activity has been characterized in the type strain DSM 20167T. METHODS AND RESULTS: Amplification of the urease genes of S. thermophilus DSM 20167T was developed on the basis of the urease gene cluster of the phylogenetically related S. salivarius. Complete pathological response with diabetic ketoacidosis to the combination of sintilimab and anlotinib in an unresectable hepatocellular carcinoma patient: a case report.

Most hepatocellular carcinoma (HCC) patients have dismal prognoses because they are already in the advanced stage at the time of initial diagnosis and are unable to undergo upfront surgery. Recent studies of immune checkpoint inhibitors (ICIs) and antiangiogenic agents (AAAs) have shown encouraging results for unresectable HCC (uHCC). Here, we report a patient with uHCC who was treated with a combination of anlotinib and sintilimab (sintilimab 200 mg, intravenous glucose tolerance test, q21d and anlotinib 12 mg, orally, d1-14, q21d), an analog of the combination of lenvatinib and pembrolizumab

Development of a nomogram for the prediction of pathological complete response after neoadjuvant chemoradiotherapy in patients with esophageal squamous cell carcinoma.

Nomograms incorporating multiple prognostic factors are useful for individualized estimation of survival in cancer patients. However, nomograms for the prediction of pathological complete response (pCR) after neoadjuvant chemoradiotherapy (nCRT) in patients with esophageal cancer are scarce. Here, we describe the development of a nomogram for predicting pCR after nCRT in patients with esophageal squamous cell carcinoma (ESCC). We retrospectively reviewed the records of 392 ESCC patients who underwent nCRT followed by esophagectomy. Seventy percent of the participants (n = 274) were randomly as

The prevalence and clinical significance of residual occult breast cancer after neoadjuvant chemotherapy: reassessing surgical pathology in cases initially described as pathological complete response.

AIMS: Evaluation of pathological complete response (pCR) [no residual invasive carcinoma in the breast (RIC) or lymph node metastases (LNM) in surgical specimens following therapy] is typically based on evaluation of one level of haematoxylin and eosin (H&E) section. Not achieving pCR is associated with worse outcomes, and additional therapy may ensue. This study of patients with triple-negative (TNBC) or human epidermal growth factor receptor 2 (HER2)-positive (HER2+) breast cancer who underwent neoadjuvant therapy aims to assess whether occult residual disease (ORD) can be identified in deep

Prediction of pathological complete response after neoadjuvant therapy in breast cancer using pretreatment core biopsy pathology reports: a pathology-report-driven model with exploratory clinical-laboratory fusion.

Pathological complete response (pCR) after neoadjuvant therapy is an important response endpoint in breast cancer. We investigated whether routinely generated pretreatment core biopsy pathology reports could support pCR prediction and whether clinical, preoperative laboratory, and treatment information provided incremental predictive value. This single-center retrospective model-development study included 171 patients with breast cancer who underwent core biopsy, neoadjuvant therapy, and surgery at Xinxiang Central Hospital between March 1, 2016, and January 1, 2026; 64 achieved pCR and 107 di
