Plant Phenolics Inhibit Focal Adhesion Kinase and Suppress Host Cell Invasion by Uropathogenic Escherichia coli.

Traditional folk treatments for the prevention and management of urinary tract infections (UTIs) and other infectious diseases often include plants and plant extracts that are rich in phenolic and polyphenolic compounds. These have been ascribed a variety of activities, including inhibition of bacterial interactions with host cells. Here we tested a panel of four well-studied phenolic compounds - caffeic acid phenethyl ester (CAPE), resveratrol, catechin, and epigallocatechin gallate - for effects on host cell adherence and invasion by uropathogenic Escherichia coli (UPEC). These bacteria, whi

Benzoindoxazine derivatives containing carbazole for detection of CN- and its application in plant seed extracts and cell imaging.

Cyanide (CN-) is a highly toxic compound that exists in many substances and is harmful to the environment and human health. Therefore, it is of great significance to develop excellent CN- ion probes, especially solvent-induced on-off fluorescent probes. Based on the condensation reaction of indolo[2,1-b][1,3]oxazine molecules with aldehydes, probes (E)-13a-(2-(9-ethyl-9H-carbazol-3-yl)vinyl)-14,14-dimethyl-10-nitro-13a,14-dihydro-8H-benzo[e]benzo[5,6][1,3]oxazino[3,2-a]indole (NCO) and (E)-13a-(2-(9-benzyl-9H-carbazol-3-yl)vinyl)-14,14-dimethyl-10-nitro-13a,14-dihydro-8H-benzo[e]benzo[5,6][1,3

Evaluating the Utility of Permethylated Polysaccharide Solution NMR Data for Characterization of Insoluble Plant Cell Wall Polysaccharides.

Plant cell wall polysaccharide analysis encompasses the utilization of a variety of analytical tools, including gas and liquid chromatography, mass spectrometry (MS), and nuclear magnetic resonance (NMR) spectroscopy. These methods provide complementary data, which enable confident structural proposals of the many complex polysaccharide structures that exist in the complex matrices of plant cell walls. However, cell walls contain fractions of varying solubilities, and a few techniques are available that can analyze all fractions simultaneously. We have discovered that permethylation affords th

Effects of plant-derived anti-leukemic drugs on individualized leukemic cell population profiles in Egyptian patients.

Leukemias are a group of cancer types that originate from blood-forming tissues. In this disease, an abnormally large number of immature white blood cells is produced by the bone marrow. The relationship between treatments with plant-derived drugs and leukemia-associated immunophenotypes (LAIPs) of clinically isolated leukemia cells has yet to be established. The aim of the present study was to develop a preliminary clinical prognostic map for commonly expressed LAIPs in patients clinically diagnosed with leukemia, as well as to assess the potential involvement of LAIPs in the response rate to

Antioxidant activity of different extracts of Argentinian medicinal plants against arsenic-induced toxicity in renal cells.

Chronic toxicity of arsenic resulting from drinking water is a health problem encountered in humans, especially in South America and Asia, where a correlation between oxidative stress, tumor promotion, and arsenic exposure has been observed. Differential solvent extraction (petroleum ether (PE); dichloromethane (DCM); methanol (OL) and water (W)) was performed to compare the protective (antioxidant) activity of five Argentinian medicinal plants on arsenite-induced oxidative stress in Vero cells, assayed by hydroperoxide measurement. The results were analyzed using ANOVA followed by the LSD Fis

Metabolic profile of caecal micro-organisms from rats fed indigestible plant cell-wall components.

A fibre-free diet, or the same diet supplemented with 100 g cabbage or carrot cell-wall preparation/kg, was fed to rats for 28 days and the activities of a number of caecal microbial enzymes (azoreductase, aryl nitroreductase, beta-glucosidase, beta-glucuronidase, imidazole nitroreductase and nitrite reductase) were determined in vitro. The plant cell-wall preparations diluted the gut contents and decreased the number of bacteria per gram of caecal contents. Enzyme activities per gram of caecal contents were also decreased, with the exception of beta-glucosidase activity which was significantl

Ectopic NGAL expression can alter sensitivity of breast cancer cells to EGFR, Bcl-2, CaM-K inhibitors and the plant natural product berberine.

Neutrophil gelatinase-associated lipocalin (NGAL, a.k.a Lnc2) is a member of the lipocalin family and has diverse roles. NGAL can stabilize matrix metalloproteinase-9 from autodegradation. NGAL is considered as a siderocalin that is important in the transport of iron. NGAL expression has also been associated with certain neoplasias and is implicated in the metastasis of breast cancer. In a previous study, we examined whether ectopic NGAL expression would alter the sensitivity of breast epithelial, breast and colorectal cancer cells to the effects of the chemotherapeutic drug doxorubicin. While

Effects of nemorosone, isolated from the plant Clusia rosea, on the cell cycle and gene expression in MCF-7 BUS breast cancer cell lines.

BACKGROUND: Breast cancer is the cause of considerable morbidity and mortality in women. While estrogen receptor antagonists have been widely used in breast cancer treatment, patients have increasingly shown resistance to these agents and the identification of novel targeted therapies is therefore required. Nemorosone is the major constituent of the floral resin from Clusia rosea and belongs to the class of polycyclic polyisoprenylated benzophenones of the acylphloroglucinol group. The cytotoxicity of nemorosone in human cancer cell lines has been reported in recent years and has been related

A plant extract of Ribes nigrum folium possesses anti-influenza virus activity in vitro and in vivo by preventing virus entry to host cells.

Infections with influenza A viruses (IAV) are still amongst the major causes of highly contagious severe respiratory diseases not only bearing a devastating effect to human health, but also significantly impact the economy. Besides vaccination that represents the best option to protect from IAV infections, only two classes of anti-influenza drugs, inhibitors of the M2 ion channel and the neuraminidase, often causing resistant IAV variants have been approved. That is why the need for effective and amply available antivirals against IAV is of high priority. Here we introduce LADANIA067 from the

Effect of plant derived antimicrobials on Salmonella enteritidis adhesion to and invasion of primary chicken oviduct epithelial cells in vitro and virulence gene expression.

Salmonella Enteritidis (SE) is a major foodborne pathogen in the United States and one of the most frequently reported Salmonella serotypes globally. Eggs are the most common food product associated with SE infections in humans. The pathogen colonizes the intestinal tract in layers, and migrates to reproductive organs systemically. Since adhesion to and invasion of chicken oviduct epithelial cells (COEC) is critical for SE colonization in reproductive tract, reducing these virulence factors could potentially decrease egg yolk contamination. This study investigated the efficacy of sub-inhibitor

A Potential Difference-Driven Cascade Effect of Cathode Biofilm-Plant Physiology Governs the Efficiency-Energy-Risk Trade-Off in Constructed Wetland-Microbial Electrolysis Cells.

Conventional potential difference optimization in microbial electrolysis cell-integrated constructed wetlands (ECWs) treating antibiotic-laden wastewater frequently prioritizes single-dimensional removal efficiency, critically overlooking trade-offs with energy consumption and ecological risks. By evaluating four ECWs operated under varying potential differences treating chloramphenicol (CAP) wastewater via physicochemical metrics, multiomics, structural equation modeling, and comprehensive risk assessment, we identified a potential difference-driven "cathode biofilm-plant physiology" cascade

Mechanistic Exploration of the anticancer activity of Justicia adhatoda Plant Leaf Ethanolic Extract against Colon Cancer Cells: An in silico and in vitro Approach.

OBJECTIVE: Being the 3rd most prevalent cancer, colorectal cancer (CRC) has a high rate of mortality, a poor prognosis, and a clinical outcome. Treatment with commercial synthetic anti-cancer drugs also imparts a number of co-morbidities, further complicating the condition. Hence, there is a need to identify a potential anticancer agent to decrease the mortality rate and provide better clinical outcome in CRC patients. Pharmacologically active agents from natural sources are the currently recognized alternatives to synthetic drugs due to their minimal toxicity. In this study, a widely recogniz

Isorhynchophylline, a Potent Plant Alkaloid, Induces Apoptotic and Anti-Metastatic Effects in Human Hepatocellular Carcinoma Cells through the Modulation of Diverse Cell Signaling Cascades.

Isorhynchophylline (Rhy) is an active pharmacological component of Uncaria rhynchophylla that has been reported previously to exert significant antihypertensive and neuroprotective effects. However, very little is known about its potential anti-cancer activities. This study was carried out to evaluate the anticancer effects of Rhy against various human carcinoma cell lines. We found that Rhy exhibited substantial cytotoxic effect against human hepatocellular carcinoma HepG2 cells when compared with other human carcinoma cell lines including those of lung, pancreas, prostate, head and neck, bre

Mobile virus-derived siRNAs drive plant antiviral silencing through reiterated 3-4 cell immunization.

The movement of virus-derived small interfering RNAs (vsiRNAs) between plant cells has prompted the widely held assumption that they help immunize as-yet-uninfected tissues. Ascertaining this role is challenging because it requires uncoupling vsiRNA mobility from viral movement while preserving viral virulence. Consequently, the contribution of non-cell-autonomous RNAi to plant antiviral immunity remains essentially unknown. Here, we investigate the naturally phloem-restricted infection of turnip yellows virus (TuYV) in the stereotypical Arabidopsis root system at cellular resolution. We uncov

Plant cell wall-plasma membrane attachments mediate stress resilience through cellulose synthase complexes and remorins.

The outer cell surface of an organism is the frontline for detecting and responding to environmental stimuli. In plants, this interface consists of the plasma membrane that lies beneath the cell wall and remains associated with it through attachment sites. These wall-membrane attachments become evident upon hyperosmotic shock, when severe water loss causes the membrane to retract from the wall. Despite their long-standing observation, the molecular identity and function of these attachments remain poorly understood. Here, we identified two mechanisms governing wall-membrane attachments: one de

An integrated automated pipeline for cloning, transient expression, and purification augmenting the plant cell pack platform for biopharmaceutical screening.

Biopharmaceuticals, particularly monoclonal antibodies (mAbs), are a rapidly expanding class of therapeutics and have benefitted from high-throughput screening (HTS) strategies for protein engineering. Obtaining such complex biopharmaceuticals from plants might be advantageous because transient expression can yield grams of product within a week. For HTS, this system was scaled down as plant cell pack (PCP) technology to microtiter plate format and automated on a liquid-handling station, but is limited by manual cloning and protein purification. Therefore, we first generated a modular vector l

Dehydrogenase WRD1 regulates plant growth by modulating cell wall formation, ROS homeostasis, and auxin signaling in rice.

Nucleotide sugars serve as the substrates for the biosynthesis of cell wall polysaccharides. Although the enzymes such as uridine diphosphate (UDP)-glucose dehydrogenase (UGD) involved in nucleotide sugar biosynthesis have been biochemically characterized, their biological roles in plants remain largely unexplored. In this study, we identify a rice mutant weakness and root hair defective 1 (wrd1), which exhibits pleiotropic phenotypes including dwarfism, reduced tiller number, and abnormal root architecture. WRD1 encodes a UGD that catalyzes the conversion of UDP-Glc to UDP-GlcA. The WRD1 muta

Rapid design and production of Alpha-1 antitrypsin fusion proteins in a plant-based cell-free expression system.

Alpha-1 antitrypsin deficiency (AATD) is a rare disease, causing significant lung damage, that is currently treated with FDA approved replacement therapy comprised of weekly infusions of costly plasma-derived AAT. Fusing AAT with a crystallizable fragment (Fc) region could potentially extend the plasma half-life, reducing dose frequency. However, computational structural simulations predict steric hindrance of the Fc domain with the linker that effects the binding of AAT-Fc to neutrophil elastase and require functional experimental evaluation. Using the Nicotiana tabacum BY-2 cell derived "Alm

Mussel and plant cell wall-inspired soy protein adhesives: High strength, flame retardancy, and mildew resistance.

Traditional soybean adhesives lack water resistance and are prone to microbial attack. Inspired by mussel adhesion and plant cell wall chemistry, we engineered a high-performance soybean-based adhesive with robust water and mildew resistance. The adhesive is equipped with a multiscale bonding network that is composed of tannic acid (TA) that has been depolymerised and grafted with epoxy compounds (E-DTA), borax (B), and soybean meal (SM). The adhesive's overall performance was improved by the construction of a highly cohesive cross-linked framework through the formation of imine bonds (Schiff
