Effects of azadirachtin on the biology of Lutzomyia longipalpis (Diptera: Psychodidae: Phlebotominae) adult female, the main vector of American visceral leishmaniasis.

The effects of azadirachtin A added to the sucrose diet of the adult females on the mortality, oviposition, and hatching of the sand fly vector of American visceral leishmaniasis Lutzomyia longipalpis (Lutz & Neiva, 1912) were investigated. Concentrations of 0.1, 1.0, and 10.0 microg/mg of azadirachtin significantly increased insect mortality in comparison with control insects. The same dose also significantly reduced oviposition but not hatching. After a long development period, significantly fewer adult insects were obtained from eggs hatching by azadirachtin-treated females in a dose-respon

Targeting the malaria parasite inside the mosquito host.

Control of malaria uses a variety of tools including drugs, vaccines, and insecticidal strategies to reduce parasite transmission by mosquito vectors. A new tool that eliminates parasites in mosquitoes by delivering antimalarials to the insects rather than infected people is emerging as a simple, relatively inexpensive malaria control strategy. In this Chapter, we review antimalarials that show efficacy in mosquitoes, effective methods to deliver compounds into the insects, and strategies to optimize this new tool as a useful component of the arsenal of strategies to combat malaria.

Leishmania spp. DNA in Sand Flies in a Transition Area of the Cerrado and Atlantic Forest biomes, Mato Grosso do Sul, Brazil.

Leishmaniases are infectious diseases whose vectors are female sand flies that depend on the blood of vertebrates for egg maturation. In this study, we investigated the sand fly species and DNA from Leishmania spp. This entomological/epidemiological study was conducted in a rural area in Nova Andradina, Mato Grosso do Sul, Brazil. From March 2019 to February 2020, five modified Falcão light traps were installed at night once a month. The collected insects were separated by sex into pools of 10 specimens. A sample was used for species identification via microscopic analysis, and another sample

Residual survival and local dispersal drive reinfestation by Triatoma dimidiata following insecticide application in Guatemala.

Chagas disease is caused by the protozoan parasite Trypanosoma cruzi and transmitted by triatomine insect vectors. In Guatemala, insecticide spraying is an integral part of management of the main vector, Triatoma dimidiata. Spraying typically has low efficacy, which may be due to incomplete elimination from infested houses, within-village dispersal, or influx from other villages or sylvan environments. To evaluate how these mechanisms contribute to reinfestation, we conducted a time-course analysis of T. dimidiata infestation, abundance and household genetic structure in two nearby villages in

History of domestication and spread of Aedes aegypti--a review.

The adaptation of insect vectors of human diseases to breed in human habitats (domestication) is one of the most important phenomena in medical entomology. Considerable data are available on the vector mosquito Aedes aegypti in this regard and here we integrate the available information including genetics, behaviour, morphology, ecology and biogeography of the mosquito, with human history. We emphasise the tremendous amount of variation possessed by Ae. aegypti for virtually all traits considered. Typological thinking needs to be abandoned to reach a realistic and comprehensive understanding o

Understanding the role of insects in the acquisition and transmission of antibiotic resistance.

Antibiotic resistance (AR) is a global healthcare threat that requires a comprehensive assessment. Poorly regulated antibiotic stewardship in clinical and non-clinical settings has led to a horizontal dissemination of AR. A variety of often neglected elements facilitate the circulation of AR from antibiotic sinks like concentrated animal feeding operations and healthcare settings to other environments that include healthy human communities. Insects are one of those elements that have received underwhelming attention as vectors of AR, despite their well-known role in transmitting clinically rel

The microbiome of wild and mass-reared new world screwworm, Cochliomyia hominivorax.

Insect population control through continual releases of large numbers of sterile insects, called sterile insect technique (SIT), is only possible if one can mass-rear large quantities of healthy insects. Adaptation of insect stocks to rearing conditions and artificial feeding systems can have a multitude of negative effects such as inbreeding depression, reduced compatibility with wild strains, unintentional selection for traits that lower fitness after release, and an altered microbiome. Changes to insect microbiomes can have many effects on insects ranging from a reduction in sex pheromones

Screening for Viruses and Lemur-Associated Filara in Wild-Caught Mosquitoes From Madagascar.

Madagascar is a hotspot of biodiversity, but poverty and population growth provoke a high risk of conflict between food security and biodiversity conservation in this tropical country. Numerous vector-borne diseases, including viral infections, affect public health in Madagascar and a continuous expansion of anthropogenically used areas intensifies contact on the human-wildlife interface. However, data on human and animal pathogens in potential insect vectors is limited. Therefore, we conducted a parasitological and virological survey of 785 adult female mosquitoes between March and May 2016 a

Controlling rate of release of tsetse fly repellent blend by encapsulating in β-cyclodextrin nanoparticles.

Tsetse flies are major vectors of African trypanosomiasis, with devastating medical and veterinary consequences in sub-Saharan region of Africa. Insect repellents are promising tool for control of tsetse flies in the region. A four-component tsetse-repellent blend (δ-nonalactone, heptanoic acid, 4-methylguaiacol and geranyl acetone) previously formulated and optimized was encapsulated in β-cyclodextrin for a slow controlled release. Here, we explored various methods of microencapsulating (kneading, co-precipitation, heating or freeze-drying) tsetse fly repellent blend in β-cyclodextrin nanopar

Uncovering the Worldwide Diversity and Evolution of the Virome of the Mosquitoes Aedes aegypti and Aedes albopictus.

Aedes aegypti, the yellow fever mosquito, and Aedes albopictus, the Asian tiger mosquito, are the most significant vectors of dengue, Zika, and Chikungunya viruses globally. Studies examining host factors that control arbovirus transmission demonstrate that insect-specific viruses (ISVs) can modulate mosquitoes' susceptibility to arbovirus infection in both in vivo and in vitro co-infection models. While research is ongoing to implicate individual ISVs as proviral or antiviral factors, we have a limited understanding of the composition and diversity of the Aedes virome. To address this gap, we

Structural features of the salivary gland hypertrophy virus of the tsetse fly revealed by cryo-electron microscopy and tomography.

Glossina palipides salivary gland hypertrophy virus (GpSGHV) infects tsetse flies, which are vectors for African trypanosomosis. This virus represents a major challenge in insect mass rearing and has hampered the implementation of the sterile insect technique programs in the Member States of the International Atomic Energy Agency. GpSGHV virions consist of long rod-shaped particles over 9000Å in length, but little is known about their detailed structural organization. We show by cryo electron microscopy and cryo electron tomography that the GpSGHV virion has a unique, non-icosahedral helical s

Antiparasitic peptides.

: The most important parasitic diseases, malaria, leishmaniasis, trypanosomiasis, and schistosomiasis, are a great burden to mankind, threatening the life of millions of people worldwide and mostly affecting the poorest. Because drug resistance is increasing and vaccines are rarely available, novel chemotherapeutic compounds are necessary in order to treat these devastating diseases. Insects serve as vectors of many human parasitic diseases and have been shown to express a huge variety of antimicrobial peptides (AMPs). Therefore, research activity on insect-derived AMPs has been increasing in

Empirical Bayesian estimation of the disease transmission probability in multiple-vector-transfer designs.

Plant disease is responsible for major losses in agriculture throughout the world. Diseases are often spread by insect organisms that transmit a bacterium, virus, or other pathogen. To assess disease epidemics, plant pathologists often use multiple-vector-transfers. In such contexts, groups of insect vectors are moved from an infected source to each of n test plants that will then be observed for developing symptoms of infection. The purpose of this paper is to present new estimators for p, the probability of pathogen transmission for an individual vector, motivated from an empirical Bayesian

The insecticidal potential of scorpion beta-toxins.

Voltage-gated sodium channels are a major target for toxins and insecticides due to their central role in excitability, but due to the conservation of these channels in Animalia most insecticides do not distinguish between those of insects and mammals, thereby imposing risks to humans and livestock. Evidently, as long as modern agriculture depends heavily on the use of insecticides there is a great need for new substances capable of differentiating between sodium channel subtypes. Such substances exist in venomous animals, but ways for their exploitation have not yet been developed due to prob

Host range of Triatoma virus does not extend to Aedes aegypti and Apis mellifera.

Vector control is the most effective method to prevent transmission of Chagas disease. Control is mostly made through chemical insecticides although they have negative impact on wild pollinators, such as bees. Reducing pesticide use through biological alternatives could minimize the damage to these beneficial insects. Triatoma virus (TrV) is a pathogen able to kill triatomines and thus a valid candidate to be used as biological control agent. In this study we evaluate the capacity of TrV to infect an important beneficial insect (Apis mellifera) as well as a plague insect (Aedes aegypti). Resul
