An Entropy-Driven Multipedal DNA Walker Microsensor for In Situ Electrochemical Detection of ATP.

Microelectrode- and nanoelectrode-based electrochemistry has become a powerful tool for the in situ monitoring of various biomolecules in vivo. However, two challenges limit the application of micro- and nanoelectrodes: the difficulty of highly sensitive detection of nonelectroactive molecules and the specific detection of target molecules in complex biological environments. Herein, we propose an electrochemical microsensor based on an entropy-driven multipedal DNA walker for the highly sensitive and selective detection of ATP. An ATP aptamer was immobilized on the microelectrode surface to fo

Real time measurements of water flow in amphibian gastric glands: modulation via the extracellular Ca2+-sensing receptor.

The mechanisms for the formation of the osmotic gradient driving water movements in the gastric gland and its modulation via the extracellular Ca(2+)-sensing receptor (CaR) were investigated. Real time measurements of net water flux in the lumen of single gastric glands of the intact amphibian stomach were performed using ion-selective double-barreled microelectrodes. Water movement was measured by recording changes in the concentration of impermeant TEA(+) ions ([TEA(+)](gl)) with TEA(+)-sensitive microelectrodes inserted in the lumen of individual gastric glands. Glandular K(+) (K(+)(gl)) an

Thyroid follicular cells: the resting membrane potential and the communication network.

Intracellular recordings of membrane potentials, input resistance and time constant have been made in vitro from the follicular cells of the rat, rabbit and guinea-pig thyroid glands using glass microelectrodes. The passive permeability properties of these cells have been investigated by altering the concentration of one or more ions in the superfusing fluid. Investigations into the intercellular coupling characteristics of the thyroid gland were made by inserting two microelectrodes into neighbouring communicating cells. The mean transmembrane potentials were between - 60 and - 70 mV in all t

Characteristics of sympathetic nerve activity in the rat sciatic nerve in response to microstimulation in a sympathetic fascicle in the contralateral side.

Microneurography is used for the monitor of various peripheral nerve activities. We recently reported that the electrical stimulation of peripheral sympathetic nerve fascicle via the microelectrode, i.e., microstimulation, temporarily reduced the blood glucose level in rats in case that the stimulation intensity was set high enough to induce small muscle contraction. However, the nature of microstimulation has little been clarified yet. Therefore, in the present study, we first detected sympathetic nerve signal microneurographically in the bilateral sciatic nerves of rats, and one of the micro

Analysis of hemorrhagic risk factors during deep brain stimulation surgery for movement disorders: comparison of the circumferential paired and multiple electrode insertion methods.

BACKGROUND: The most serious complication of deep brain stimulation (DBS) surgery is intracranial hemorrhage. The authors have assessed risk factors for hemorrhage in DBS surgery and compared two types of microelectrode insertion technique on hemorrhagic risk. METHODS: A total of 171 DBS procedures were performed on 110 patients (58 females, 52 males) by the same neurosurgeon at a single center between May 2005 and May 2010. We used two microelectrode insertion methods: multiple microelectrode insertion (MMI) and circumferential paired microelectrode insertion (CPMI). We analyzed the correlat

Colloidal deposition on remotely controlled charged micropatterned surfaces in a parallel-plate flow chamber.

This article describes a method to influence colloid deposition by varying the zeta potential at microelectrodes with remotely applied electric potentials. Deposition experiments were conducted in a parallel-plate flow chamber for bulk substrates of glass, indium tin oxide (ITO), and ITO-coated glass microelectrodes in 10 and 60 mM potassium chloride solutions. Colloid deposition was found to be a function of solution chemistry and the small locally delivered electric surface potentials. Electric fields and physical surface heterogeneity can be ruled out as cause of the observed deposition. Re

Nicotine-evoked nitric oxide release in the rat hippocampal slice.

The effects of cholinergic agonists on nitric oxide (NO) release in hippocampal slices from male Sprague-Dawley rats were investigated using electrochemical recording procedures using Nafion and O-phenylenediamine-treated carbon fiber microelectrodes. These microelectrodes are highly selective for NO versus other interferents. Acetylcholine (Ach) with neostigmine, or nicotine was delivered by pressure ejection from pipettes placed within 300 microm of the NO sensors. Both Ach and nicotine produced NO signals ranging from 0.04 to 2.14 microM in the CA1, CA3, and dentate gyrus of the rat hippoca

In vivo recordings from the human vagus nerve using ultrasound-guided microneurography.

KEY POINTS: The vagus nerve is the largest cranial nerve and innervates many structures in the neck, thorax and abdomen. Although single-unit recordings from the vagus nerve have been performed in experimental animals for several decades, no recordings have ever been made from the human vagus nerve. The vagus nerve is routinely stimulated clinically, yet we know little of its physiology in humans. We describe the methodology and provide preliminary results of the first intraneural single-unit recordings from the cervical vagus in awake humans, using tungsten microelectrodes inserted into the n

Intracellular ionized calcium concentration in muscles from humans with malignant hyperthermia.

Ca2+ selective microelectrodes have been used to determine the free myoplasmic [Ca2+] in human skeletal muscle obtained from patients who had developed early signs associated with malignant hyperthermia (MH) during anesthesia. Intercostal muscle biopsies were performed under local anesthesia in four MH patients 15 days to 4 months after developing the MH crisis and in three control subjects. We used only microelectrodes that showed a Nernstian response between pCa3 and pCa7 (30.5 mV per decade at 37 degrees C). Membrane resting potential (V(m)) and calcium potential (V(Ca)) were obtained from

Electrophysiologic mapping for deep brain stimulation for movement disorders.

Electrophysiologic mapping remains an integral component of deep brain stimulation (DBS) surgical procedures, particularly in movement disorder cases where functional maps are used to guide DBS lead placement in patients with Parkinson's disease, dystonia, or tremor. Overall, the goal of the surgical procedure is to implant the distal end of a chronic, multicontact depth electrode into a specific brain region for the purpose of delivering therapeutic electrical stimulation. Regions that are currently targeted for patients with movement disorders include the subthalamic nucleus, the ventral int

Novel Multiparametric Bioelectronic Measurement System for Monitoring Virus-Induced Alterations in Functional Neuronal Networks.

Development and optimisation of bioelectronic monitoring techniques like microelectrode array-based field potential measurement and impedance spectroscopy for the functional, label-free and non-invasive monitoring of in vitro neuronal networks is widely investigated in the field of biosensors. Thus, these techniques were individually used to demonstrate the capabilities of, e.g., detecting compound-induced toxicity in neuronal culture models. In contrast, extended application for investigating the effects of central nervous system infecting viruses are rarely described. In this context, we wan

Flexible graphene-based neurotechnology for high-precision deep brain mapping and neuromodulation in Parkinsonian rats.

Deep brain stimulation (DBS) is a neuroelectronic therapy for the treatment of a broad range of neurological disorders, including Parkinson's disease. Current DBS technologies face important limitations, such as large electrode size, invasiveness, and lack of adaptive therapy based on biomarker monitoring. In this study, we investigate the potential benefits of using nanoporous reduced graphene oxide (rGO) technology in DBS, by implanting a flexible high-density array of rGO microelectrodes (25 µm diameter) in the subthalamic nucleus (STN) of healthy and hemi-parkinsonian rats. We demonstrate

Direct-Print 3D Electrodes for Large-Scale, High-Density, and Customizable Neural Interfaces.

Silicon-based microelectronics can scalably record and modulate neural activity at high spatiotemporal resolution, but their planar form factor poses challenges in targeting 3D neural structures. A method for fabricating tissue-penetrating 3D microelectrodes directly onto planar microelectronics using high-resolution 3D printing via 2-photon polymerization and scalable microfabrication technologies are presented. This approach enables customizable electrode shape, height, and positioning for precise targeting of neuron populations distributed in 3D. The effectiveness of this approach is demons

Construction by dielectrophoresis of microbial aggregates for the study of bacterial cell dormancy.

A study of the effect of aggregate size on the resuscitation of dormant M. smegmatis was conducted by constructing cell aggregates with defined sizes and shapes using dielectrophoresis and monitoring the resuscitation process under controlled laboratorial conditions in a long-term cell feeding system. Differently sized cell aggregates were created on the surface of indium tin oxide coated microelectrodes, their heights and shapes controlled by the strength of the induced electric field and the shape of the microelectrodes. Both two-dimensional (ring-patterned) and three-dimensional cell aggreg

Aberrant accumulation of age- and disease-associated factors following neural probe implantation in a mouse model of Alzheimer's disease.

Objective. Electrical stimulation has had a profound impact on our current understanding of nervous system physiology and provided viable clinical options for addressing neurological dysfunction within the brain. Unfortunately, the brain's immune suppression of indwelling microelectrodes currently presents a major roadblock in the long-term application of neural recording and stimulating devices. In some ways, brain trauma induced by penetrating microelectrodes produces similar neuropathology as debilitating brain diseases, such as Alzheimer's disease (AD), while also suffering from end-stage
