Cell-penetrating peptides (CPPs) represent a class of short peptides capable of efficiently traversing biological membrane barriers, demonstrating substantial potential in tumor-targeted drug delivery. Drugs modified with CPPs can enter cells through direct membrane penetration or endocytic pathways, and CPPs have been extensively investigated in various antitumor drug delivery systems. This review initially categorizes CPPs into cationic peptides, amphipathic peptides, and hydrophobic peptides according to their charge characteristics, and subsequently elucidates the molecular mechanisms unde

Antimicrobial resistance has become a major global health challenge, creating an urgent need for novel anti-infective molecules. Antimicrobial peptides (AMPs) constitute a promising alternative to conventional antibiotics because of their broad-spectrum antimicrobial activity and immunomodulatory properties. Despite the remarkable diversity of protozoa, very few protozoan AMPs have been experimentally identified. Here, we employed the deep learning algorithm AMPlify to systematically screen the proteomes of Blastocystis hominis, Trypanosoma cruzi, Leishmania major, Toxoplasma gondii, and Entam

Constrained macrocyclic peptides can engage protein surfaces that resist both small molecules and biologics but producing them at library scale still depends on chemical synthesis and macrocyclisation workflows that are slow, specialised and hard to parallelise. We describe a recombinant pipeline for disulphide-stapled cyclic peptides, compatible with 96 well plates, that lowers this barrier. Peptide-encoding sequences are introduced by extension PCR and in vivo assembly (IVA) cloning at about $2 per variant, expressed as small ubiquitin-like modifier (SUMO) fusions secreted to the oxidising p

We report a new method to synthesize N-formylkynurenine (Nfk)-containing peptides. Nfk is a known product of Trp metabolism; yet access to peptides containing this residue remains challenging. Tryptophan (Trp) within peptides can be oxidized to Nfk using sodium periodate (NaIO4) at room temperature in 24 h. This reaction is chemoselective for Trp, and we have demonstrated its compatibility on thirteen peptides with all natural amino acids, except Cys and Met. Nfk residues within peptides could be hydrolysed to kynurenine (Kyn) with 1% TFA in water. Overall, this report presents a mild and sele

Fecal coprolites preserve ancient microbiomes and are a potential source of extinct but highly efficacious antimicrobial peptides (AMPs). Here, we develop AMPLiT (AMP Lightweight Identification Tool), an efficient tool deployable to portable hardware for AMP screening in metagenomic datasets. AMPLiT demonstrates AUPRC performances of 0.9486 ± 0.0003 and reasonable overall training time of 3200 ± 53 s. By computationally utilizing AMPLiT, we analyze seven ancient human coprolite metagenomes, identifying 160 AMP candidates. Of 40 representative peptides synthesized, 36 (90%) peptides demonstrate

Therapeutic angiogenesis is constrained by the inability to localize, sustain, and finely tune vascular signaling. Here, we detail a supramolecular peptide system that enables angiogenic programming by decoupling mechanical integrity from receptor-level bioactivity. We integrate a vascular endothelial growth factor (VEGF)-mimetic amphiphile (SLan) with a mechanically robust β sheet peptide (K1) to yield injectable nanofiber hydrogels with tunable stiffness (100 to 1000 pascals) and preserved vascular endothelial growth factor receptor 2 (VEGFR2) affinity. Constrained bicyclic peptides (Bicycle molecules) with high affinity for biological targets have emerged as potentially powerful therapeutic agents, particularly for the in vivo targeting of cancer receptors. However, their antibody-mimetic properties have yet to be explored for use in diagnostic immunoassays. These synthetically derived compounds serve as biorecognition scaffolds that allow for facile site-selective modification and large-scale production. A phage display screen against various constructs of the SARS-CoV-2 nucleocapsid (N) protein identified several Bicycle molecules with bin

Canonical signal peptides (SPs) are short N-terminal sequences that direct nascent proteins into the secretory pathway, but their role extends far beyond protein targeting. Advances in sequencing and computational tools have enabled their systematic identification across proteomes, highlighting SPs as critical regulators of protein biogenesis, including endoplasmic reticulum (ER) targeting, translocation, folding, and proteostasis. Clinically, mutations affecting SP function underlie a distinct group of human disorders, while SP-derived fragments are emerging as diagnostic biomarkers and thera

In this review, we provide a comprehensive analysis of the role of natural peptides-particularly those derived from amphibian skin secretions-as multitarget-directed ligands (MTDLs) in the context of Alzheimer's disease (AD). Given the multifactorial nature of AD, where cholinergic dysfunction intersects with amyloid-β aggregation, tau hyperphosphorylation, oxidative stress, metal ion imbalance, and monoamine oxidase dysregulation, therapeutic strategies capable of modulating several pathological pathways simultaneously are urgently needed. We begin by revisiting the cholinergic hypothesis and

Neurodegenerative and degenerative disorders are in part, driven by imbalances in cellular inflammatory and oxidation-reduction (redox) states. This vulnerability triggers the activation of highly selective and tightly regulated cellular defense networks against oxidative stress (OS), primarily involving the glutathione (GSH) and the thioredoxin (Trx) enzymatic complexes. These systems operate through reversible oxidation/reduction reactions involving the thiol groups of cysteine (Cys) residues, maintaining redox homeostasis and protecting cells from oxidative damage. To reinforce this defense

Melanin produced by melanocytes, primarily determines human skin color and protects against ultraviolet radiation. However, excessive melanin deposition can lead to skin conditions such as freckles, age spots, and moles, potentially causing aesthetic concerns and psychological distress. Consequently, there is significant research interest in developing safe and effective whitening products that inhibit melanin synthesis. Bioactive peptides represent a promising compound category that effectively reduces melanin synthesis with minimal side effects. This review explores melanin pigmentation, ide

Dental caries is an avoidable and complex condition impacting billions of individuals worldwide, posing a specific concern among younger generations, despite the progress of oral hygiene products. This deterioration occurs due to the acid demineralization of tooth enamel, leading to the loss of minerals from the enamel subsurface. The remineralisation of early enamel carious lesions could prevent the cavitation of teeth. The enamel protein amelogenin constitutes 90% of the total enamel matrix protein and plays a key role in the bio mineralisation process. The aim of this study is to investigat

The underdevelopment of adjuvant discovery and diversity, compared to core vaccine technology, is evident. On the other hand, antibiotic resistance is on the list of the top ten threats to global health. Immunomodulatory peptides that target a pathogen and modulate the immune system simultaneously are promising for the development of preventive and therapeutic molecules. Since investigating innate immunity in insects has led to prominent achievements in human immunology, such as toll-like receptor (TLR) discovery, we used the capacity of the immunomodulatory peptides of arthropods with concomi

Antimicrobial resistance is a major public health concern. Antimicrobial peptides (AMPs) are one of the host defense mechanisms responding efficiently against multidrug-resistant microbes. Since the process of screening AMPs from a large number of peptides is still high-priced and time-consuming, the development of a precise and rapid computer-aided tool is essential for preliminary AMPs selection ahead of laboratory experiments. In this study, we proposed AMPs recognition models using a new peptide encoding method called amino acid index weight (AAIW). Four AMPs recognition models including a

Mitochondrial-derived peptides (MDPs) are a novel class of bioactive microproteins encoded by short open-reading frames (sORF) in mitochondrial DNA (mtDNA). Currently, three types of MDPs have been identified: Humanin (HN), MOTS-c (Mitochondrial ORF within Twelve S rRNA type-c), and SHLP1-6 (small Humanin-like peptide, 1 to 6). The 12 S ribosomal RNA (MT-RNR1) gene harbors the sequence for MOTS-c, whereas HN and SHLP1-6 are encoded by the 16 S ribosomal RNA (MT-RNR2) gene. Special genetic codes are used in mtDNA as compared to nuclear DNA: (i) ATA and ATT are used as start codons in addition t
