Investigation of the role of ZurR in the physiology and pathogenesis of Listeria monocytogenes.

Listeria monocytogenes is a Gram positive pathogen that is ubiquitous in the environment. It is a facultative anaerobic rod that causes listeriosis, a disease with potentially lethal consequences for susceptible individuals. During infection, the pathogen is capable of sequestering metal ions to act as vital biocatalysts in cellular processes. The zinc uptake regulator (ZurR) is predicted to coordinate uptake of zinc from the external environment. An in-frame deletion of the zurR gene resulted in a mutant exhibiting a small colony phenotype and a smaller cell size. The zurR mutant was unaffect

Modulation of histone H3 lysine 56 acetylation as an antifungal therapeutic strategy.

Candida albicans is a major fungal pathogen that causes serious systemic and mucosal infections in immunocompromised individuals. In yeast, histone H3 Lys56 acetylation (H3K56ac) is an abundant modification regulated by enzymes that have fungal-specific properties, making them appealing targets for antifungal therapy. Here we demonstrate that H3K56ac in C. albicans is regulated by the RTT109 and HST3 genes, which respectively encode the H3K56 acetyltransferase (Rtt109p) and deacetylase (Hst3p). We show that reduced levels of H3K56ac sensitize C. albicans to genotoxic and antifungal agents. Inh

Heat-responsive long noncoding RNA and microRNA modules associated with thermotolerance in Moso bamboo.

Heat stress limits Moso bamboo (Phyllostachys edulis) growth and development, yet the roles of noncoding RNA networks in the heat response remain unclear. Here, by integrating mRNA, long noncoding RNA, and small RNA transcriptomes from seedlings exposed to 42 °C, we identified 2,889 differentially expressed long noncoding RNAs, 1,216 mRNAs, and 21 microRNAs, respectively. Target prediction suggested that differentially expressed long noncoding RNAs potentially regulate 578 genes, whereas differentially expressed microRNAs target 300 long noncoding RNAs and 186 mRNAs. These predicted targets we

The miR167-ARF8 Module Integrates HSFA1-HSFA2 and RVE4/8-ERF53/54 Transcriptional Cascades to Enhance Plant Thermotolerance.

Heat stress (HS) is a major environmental factor limiting plant survival and crop productivity, prompting the evolution of adaptive responses driven by transcriptional reprogramming coordinated by Class A1 HS TRANSCRIPTION FACTORS (HSFA1s). Here, we demonstrate that the miR167-AUXIN RESPONSE FACTOR 8 (ARF8) module functions as a central regulatory hub, enhancing thermotolerance by integrating both HSFA1-dependent and HSFA1-independent signaling cascades. Under HS, the expression of ARF6 and ARF8 is repressed, while miR167 is induced. Disruption of ARF6/ARF8 or overexpression of miR167 signific

Inhibition of H3K14 lactylation promotes diabetic corneal nerve regeneration via Wnt1/β-catenin signaling.

AIMS: This study bridges diabetes-induced metabolic alterations and corneal neuropathy, proposing lactylation as a key mechanistic and therapeutic target in diabetic keratopathy (DK). METHODS: We used a streptozotocin-induced Type 1 diabetes mellitus (T1DM) mouse model to examine metabolic reprogramming in trigeminal ganglion (TG) neurons and its effects on corneal nerve regeneration. Glycolysis, lactate levels, and histone lactylation (H3K14la) were analyzed via Western blot, qPCR, immunofluorescence, and enzymatic assays. Genome-wide H3K14la profiling was conducted using CUT&Tag, followed b

Mild microwave hyperthermia promotes mitotic catastrophe, induces time-delayed cGAS-STING activation and restores sensitivity to anti-PDL1 therapy in Pan02 pancreatic cancer model.

Pancreatic ductal adenocarcinoma (PDAC) responds poorly to immune checkpoint inhibitors (ICIs), representingindicating an unmet clinical need. Hyperthermia therapy, an approved anti-cancer approach with limited adverse effects, has been suggested to be capable of modulating the tumor microenvironment (TiME). This study examined the potential of microwave hyperthermia (Ht) to restructure the TiME of Pan02 PDAC model and restore Pan02 tumor's sensitivity to ICIs, followed by mechanistic investigation. In vitro experiments were performed using a 41 °C water bath, and in vivo studies established t

Pirin, a redox-sensitive modulator of beta-oxidation, generates hydroxyl radicals and interacts with CatR, the transcriptional repressor of the major vegetative catalase gene in Streptomyces.

Pirins are iron-containing proteins conserved throughout evolution, which have been implicated in diverse cellular processes, mostly associated with stress. In prokaryotes, Pirins are present in many taxonomic groups and can be present in multiple copies, and only a few of these proteins have been studied. In Streptomyces ambofaciens a Pirin-like protein, PirA, is a redox-sensitive negative modulator of AcdB, a very long-chain acyl-CoA dehydrogenase (vLCAD), which catalyzes the first committed step of the beta-oxidation pathway. In this study, we first classified the Pirins in different prokar

The nuclear condensates of ESE3/EHF induce cellular senescence without the associated inflammatory secretory phenotype in pancreatic ductal adenocarcinoma.

Senescent cells are in a stable state of cell cycle arrest, leading to a natural barrier to tumorigenesis. Senescent cells secrete a pool of molecules, including cytokines, chemokines, proteases, and growth factors, termed the senescence-associated secretory phenotype (SASP), paradoxically contributing to pro-tumorigenic processes. However, the mechanism for regulating senescence and SASP in tumor cells remains unclear. Here, SPiDER senescence probe-based CRISPR/Cas9 library screening has identified ETS homologous factor (EHF) could effectively induce cellular senescence but without SASP, whic

Effects of Exposure to the Endocrine-Disrupting Chemical Bisphenol A During Critical Windows of Murine Pituitary Development.

Critical windows of development are often more sensitive to endocrine disruption. The murine pituitary gland has two critical windows of development: embryonic gland establishment and neonatal hormone cell expansion. During embryonic development, one environmentally ubiquitous endocrine-disrupting chemical, bisphenol A (BPA), has been shown to alter pituitary development by increasing proliferation and gonadotrope number in females but not males. However, the effects of exposure during the neonatal period have not been examined. Therefore, we dosed pups from postnatal day (PND)0 to PND7 with 0

Peripheral myelin protein 22 is regulated post-transcriptionally by miRNA-29a.

Peripheral myelin protein 22 (PMP22) is a dose-sensitive, disease-associated protein primarily expressed in myelinating Schwann cells. Either reduction or overproduction of PMP22 can result in hereditary neuropathy, suggesting a requirement for correct protein expression for peripheral nerve biology. PMP22 is post-transcriptionally regulated and the 3'untranslated region (3'UTR) of the gene exerts a negative effect on translation. MicroRNAs (miRNAs) are small regulatory molecules that function at a post-transcriptional level by targeting the 3'UTR in a reverse complementary manner. We used cul

Interplay between two RND systems mediating antimicrobial resistance in Brucella suis.

The RND-type efflux pumps are responsible for the multidrug resistance phenotype observed in many clinically relevant species. Also, RND pumps have been implicated in physiological processes, with roles in the virulence mechanisms of several pathogenic bacteria. We have previously shown that the BepC outer membrane factor of Brucella suis is involved in the efflux of diverse drugs, probably as part of a tripartite complex with an inner membrane translocase. In the present work, we characterize two membrane fusion protein-RND translocases of B. suis encoded by the bepDE and bepFG loci. MIC assa

Cisplatin disrupts OCT1-DNMT1-piRNA epigenetic regulatory axis to suppress GAB2-mediated aggressiveness in OSCC.

Chemotherapy-induced ncRNA-mediated plasticity is an emerging concept in cancer research. To that end, we observed a cisplatin-responsive regulatory program centered on piRNA activation. OSCC cells exposed to cisplatin markedly promoted the piRNA expression, with piR-hsa-30937 showing the most prominent upregulation. Mechanistically, cisplatin disrupts the OCT1-DNMT1 repressive complex that mediates DNA methylation of transcription factor binding sites of piR-hsa-30937, to derepress its expression. Functionally, piR-hsa-30937 targets GAB2 and sensitizes OSCC cells to cisplatin by suppressing p

Intercalated Motifs Contribute to Transcription Regulation in an Acidosis Model.

The formation and function of intercalated motifs (iMs) in cytosine-rich regions of the human genome have long been debated. While these pH-sensitive structures are unlikely to govern gene expression in normal tissues, their contribution to acidosis-specific transcriptional reprogramming cannot be ruled out. Here, we focused on previously reported iM-forming sequences from the regulatory regions of early response genes. We tested their impact on transcription via luciferase-based assays under normal conditions, as well as under conditions favoring or disfavoring iMs (acidic media or iM-destabi

Transcriptional and epigenomic analysis of insulin sensitivity in human adipocytes.

Insulin resistance is a key feature of type 2 diabetes (T2D) and is also associated with a wide variety of other disease states. We isolated adipocytes from subjects with and without insulin resistance and subjected them to transcriptional and epigenomic profiling, allowing us to identify genes, cis-regulatory elements, and pathways that associate with insulin resistance in this critical cell type. We focused on several differentially enriched regions near the IRS1 gene that were close to, but distinct from, single-nucleotide polymorphisms associated with T2D and insulin resistance. CRISPR int

BDNF regulates pituitary stem cell engagement toward precursor state.

Following their engagement toward differentiation, tissue stem cells often transit through a precursor state that is difficult to define because of its transient nature; similarly, the precise role of lineage precursors in the implementation of tissue architecture and function is unknown. In the present work, we used two mouse models of deficient feedback regulation to characterize precursors of the pituitary corticotrope lineage that regulates the stress response. Both the POMC knockout and adrenalectomized mouse models develop glucocorticoid deficiency and compensatory accumulation of cortic

Consumption of clinically relevant cannabidiol doses during pregnancy alters postnatal behavior in offspring.

Pregnant people use cannabidiol (CBD), a non-psychoactive cannabinoid of cannabis, due to its perceived safety and to treat side-effects such as nausea, insomnia, pain, and anxiety. However, CBD crosses the placenta and accumulates in the fetal brain, where it can activate or repress the function of several molecular targets that are important for brain development. While consumption of high doses of CBD during pregnancy have been shown to disrupt offspring neurodevelopment and postnatal behavior, lower, more clinically relevant doses have not been assessed. Here, we show that oral consumption

Metformin sensitises hepatocarcinoma cells to methotrexate by targeting dihydrofolate reductase.

Metformin, the first-line drug for type II diabetes, has recently been considered an anticancer agent. However, the molecular target and underlying mechanism of metformin's anti-cancer effects remain largely unclear. Herein, we report that metformin treatment increases the sensitivity of hepatocarcinoma cells to methotrexate (MTX) by suppressing the expression of the one-carbon metabolism enzyme DHFR. We show that the combination of metformin and MTX blocks nucleotide metabolism and thus effectively inhibits cell cycle progression and tumorigenesis. Mechanistically, metformin not only transcri

Expression of the DNA-Binding Factor TOX Promotes the Encephalitogenic Potential of Microbe-Induced Autoreactive CD8+ T Cells.

Infections are thought to trigger CD8+ cytotoxic T lymphocyte (CTL) responses during autoimmunity. However, the transcriptional programs governing the tissue-destructive potential of CTLs remain poorly defined. In a model of central nervous system (CNS) inflammation, we found that infection with lymphocytic choriomeningitis virus (LCMV), but not Listeria monocytogenes (Lm), drove autoimmunity. The DNA-binding factor TOX was induced in CTLs during LCMV infection and was essential for their encephalitogenic properties, and its expression was inhibited by interleukin-12 during Lm infection. TOX r

The methyltransferase Setdb2 mediates virus-induced susceptibility to bacterial superinfection.

Immune responses are tightly regulated to ensure efficient pathogen clearance while avoiding tissue damage. Here we report that Setdb2 was the only protein lysine methyltransferase induced during infection with influenza virus. Setdb2 expression depended on signaling via type I interferons, and Setdb2 repressed expression of the gene encoding the neutrophil attractant CXCL1 and other genes that are targets of the transcription factor NF-κB. This coincided with occupancy by Setdb2 at the Cxcl1 promoter, which in the absence of Setdb2 displayed diminished trimethylation of histone H3 Lys9 (H3K9m

HDAC inhibitors restore C-fibre sensitivity in experimental neuropathic pain model.

BACKGROUND AND PURPOSE: Hypoesthesia is a clinical feature of neuropathic pain. The feature is partly explained by the evidence of epigenetic repression of Nav 1.8 sodium channel in the dorsal root ganglion (DRG). EXPERIMENTAL APPROACH: We investigated the possibility of trichostatin A (TSA), valproic acid (VPA) and suberoylanilide hydroxamic acid (SAHA) to reverse the unique C-fibre sensitivity observed following partial ligation of sciatic nerve in mice. KEY RESULTS: Nerve injury-induced down-regulation of DRG Nav 1.8 sodium channel and C-fibre-related hypoesthesia were reversed by TSA, VP
