Plasma sterol profiling in autism spectrum disorder: insights from cerebrotendinous xanthomatosis screening and beyond.

Cerebrotendinous xanthomatosis (CTX) is a rare, treatable bile acid synthesis disorder characterized by increased levels of cholestanol. Studies indicate that autism spectrum disorder (ASD) may be an early manifestation of CTX. Independent of CTX, disturbances in sterol and bile acid metabolism are observed in ASD. Therefore, this study aimed to estimate the prevalence of CTX in a pediatric ASD cohort using cholestanol-based screening with reflex CYP27A1 sequencing and to compare plasma sterol profiles among children with ASD. We conducted a single-center, cross-sectional study including 103 p

Evaluation of plasma phytosterols in sitosterolemia, their kindreds and hyperlipidemia subjects.

BACKGROUND: Patients suffering from sitosterolemia with ABCG5/8 mutation typically present with early-onset or rapidly progressive atherosclerosis. Their kindreds with partial genetic deficiencies of ABCG5/8 are often considered healthy. However, discerning sitosterolemia from its familial kindreds and hyperlipidemia subjects has remained challenging. METHODS: Here we retrospectively recruited 7 families including 8 individuals diagnosed with sitosterolemia subjects, and 14 kindreds carrying single gene mutations. Additionally, 17 individuals with hyperlipidemia and 130 healthy controls serve

Chemical and Biochemical Features of Spinasterol and Schottenol.

Phytosterols, which are produced in plants, are structurally similar to cholesterol. Their basic structures consist of a cyclo pentano-perhydrophenanthrene nucleus composed of 3 hexane rings and of a pentane ring with an alkyl side chain. There are around more than 250 phytosterols and related compounds that have been identified in natural resources. Among them, spinasterol and schottenol, its dihydro analog, are often found in seeds, and consequently in seed oils, and in other botanical parts of some plant families such as Sapotaceae, Cactaceae, and Cucurbitaceae. Spinasterol and/or schotteno

Metabolic discrimination of Thymus, Zataria, and Ziziphora genera by NMR-based metabolomics.

Thyme is a commercial spice widely used in food, cosmetics, and pharmaceutical industries. Three popular genera in food and traditional medicine, including Zataria Boiss., Thymus L., and Ziziphora L., are considered as thyme-like plants in Iran. Thyme has been standardised based on phenolic monoterpenes, which are abundant in the essential oils of these three genera. Apart from monoterpenes, the flavonoid naringenin, the triterpene oleanolic acid, and phytosterols such as β-sitosterol were abundant in Zataria, Thymus, and Ziziphora samples, respectively. Therefore, employing current analytical

Rapid hydrogenation of unsaturated sterols and bile alcohols using microwaves.

This paper describes an operationally simple, rapid hydrogenation of unsaturated sterols and bile alcohols in a domestic microwave oven. This has been achieved by the addition of catalytic amounts of Pd/C in methylene chloride/propylene glycol solvents in the presence of ammonium formate followed by microwave irradiation. It is suggested that this methodology will be helpful in the identification of saturated and unsaturated sterols with different side-chain structures in rare diseases: sitosterolemia, cerebrotendinous xanthomatosis (CTX), as well as atherosclerosis and diabetes mellitus.
